Abstract

Dehydration, the decrease of body fluid and electrolytes, is the most common disorder in the elderly. Total body water (TBW) is influenced by various factors including body composition, such as fat mass (FM) and fat-free mass (FFM). FM can be represented by body mass index (BMI), waist circumference (WC), and waist-to-hip ratio (WHR) while FFM can be represented by muscle mass (MM). This study aimed to determine the correlation of BMI, WC, WHR and MM with TBW in the elderly. This cross-sectional research was conducted on elderly people in four nursing homes in Jakarta between November and December 2020. The sample was 87 people who meet the inclusion and exclusion criteria with the total sampling method. Fat mass data collection were carried out by trained personnel by measuring BMI, WC, WHR. MM data were collected using the Maltron Bioscan 916. Correlation analysis was carried out using the independent t-test and linear regression test. The results showed that the average TBW was 28.5 ± 5.4 liters (men were significantly higher than women). Further analysis showed that BMI, WC, and MM were positively correlated with TBW, but not WHR. In linear regression analysis, MM had the greatest correlation with TBW (p<0.05). This study validated that elderly people with lower muscle mass tend to have lower total body fluids; therefore, elderly people are advised to maintain muscle mass through diet and exercise modification to maintain total body fluids and reduce the risk of dehydration.

Abstract

Introduction: Parkinson's disease (PD) is the second most common neurodegenerative disease. Identification of PD biomarkers is critical for early diagnosis and development of therapeutic targets. There are many factors that are associated with PD, including metabolic syndrome (MetS). However, molecular links between MetS and PD for biomarkers is still a challenge. This study aims to determine the molecular links between MetS and PD using an in silico approach. Methods: Data were obtained from Gene Expression Omnibus to identify differentially expressed genes (DEGs) and microRNA (DEMs) in PD and MetS. DEGs were mapped to protein-protein interaction (PPI) networks using the STRING platform. Then, gene ontology (GO) and pathway analysis was performed with EnrichR to reveal specific and overlapped biological processes. Finally, microRNA (miRNA) and mRNA pairs were predicted using TargetScan. Results: A total of 25 DEGs were identified in both MetS and PD. GO and pathway analysis for MetS-PD revealed that these DEGs mainly related to metabolic and cytokine pathways. Examination of miRNA showed that hsa-miR-631 was down-regulated in both MetS and PD. GO and pathway analysis on predicted targets of hsa-miR-631 showed major changes in metabolic pathways. Hsa-miR-631 can target the 3' UTR of ATP2B4 at poorly conserved site, indicating a specific miRNA-mRNA pairing in humans. Conclusion: In patients with MetS and PD, hsa-miR-631 and ATP2B4 are depleted and elevated, respectively. Bioinformatic analysis indicates that ATP2B4 is a target of hsa-miR-631. We demonstrate for the first time that hsa-miR-631/ ATP2B4 pair is potential biomarker for PD due to MetS.

Abstract

Background: Resveratrol (RSV) is a polyphenol with potent antioxidant activity and is abundant in fruits. There has been a lot of scientific evidence regarding the anti-aging effect of RSV. Aging can be induced by UV-B (photoaging) due to the production of reactive oxygen species (ROS) and oxidative stress. This study aimed to test the anti-photoaging activity of RSV on UV-B -induced Caenorhabditis elegans.Materials and methods: C. elegans was cultured at 20˚C in nematode growth medium (NGM) and was subjected to various concentrations of RSV and UV-B. The UV-B light exposure was given on day 0 post-synchronization at a dose of 100 J/m2 using a UV cross-linker. The health span (indicated by pharyngeal pumping rate) and lifespan of worms were observed. The quantification of collagen was performed using a Sircol Collagen assay kit. The mRNA expression level of gcs-1, col-19, hus-1, cep-1, egl-1, and ced-13 was examined by qRT-PCR. Results: UV-B reduced pharyngeal pumping rate, shortened the lifespan, decreased collagen, and increased the expression of apoptosis-related genes (hus-1, cep-1, egl-1, and ced-13). RSV ameliorated these aging phenotypes induced by UV-B. Anti-aging activities of RSV were not observed in the skn-1 loss-of-function strain (VC1772, skn-1(ok2315)), indicating the critical involvement of skn-1 in the mechanism of action of RSV. The activation of skn-1 was shown by elevated skn-1 target gene that play role in glutathione biosynthesis called gcs-1.Conclusion: RSV prevents accelerated aging due to UV-B in C. elegans by enforcing skn-1 signaling pathway and its downstream gcs-1 gene expression.   Keywords: anti-aging, resveratrol, oxidative stress, UV-B

Abstract

Pendahuluan: Penuaan kulit adalah proses kompleks yang melibatkan faktor intrinsik dan ekstrinsik. Berbagai parameter untuk mengevaluasi karakteristik kulit telah diusulkan. Namun, biomarker yang akurat untuk penuaan kulit dan hubungan mekanisme molekuler penuaan kulit masih sangat terbatas. Penelitian ini bertujuan untuk memvalidasi peran mikroRNA (miRNA) dalam regulasi epigenetik penuaan kulit sebagai kandidat biomarker dan target terapi. Metode: Profil ekspresi miRNA kakek dan cucu dari 2 famili Jawa dibandingkan untuk memilih kandidat miRNA yang memiliki perbedaan ekspresi pada jaringan kulit. Selanjutnya, miRNA yang mengalami perubahan ekspresi tersebut dianalisis fungsi dan perannya pada tingkat molekuler dengan pendekatan bioinformatika. Hasil: Hasil profiling menunjukkan miR-4454 yang konsisten mengalami peningkatan ekspresi pada kedua famili yaitu sebesar 3,318 pada famili I dan 9,315 pada famili II. Berdasarkan data dari GeneCards, miR-4454 memiliki ekspresi yang cukup tinggi pada kulit dan diekspresikan pada seluruh komponen sel. Analisis data dari GWAS mengindikasikan bahwa miR-4454 terlibat dalam pengaturan panjang telomer. Analisis gen target dengan menggunakan TargetScanHuman, miRTarBase, miRTargetLink dilanjutkan dengan pengayaan ontologi gen dan jalur molekuler dengan platform EnrichR menunjukkan miR-4454 berperan dalam regulasi produksi IL-12, respons terhadap radiasi, autofagi, metabolisme, FoxO signaling pathway, dan cellular senescence. Gen ataxia telangiectasia mutated (ATM) dan mitogen-activated protein kinase 14 (MAPK14) diduga berperan penting pada regulasi penuaan kulit akibat miR-4454 karena keterlibatannya pada cellular senescence. Simpulan: Pada penuaan kulit, ekspresi miR-4454 yang tinggi berpotensi digunakan sebagai kandidat biomarker dan dasar pengembangan target terapi. Gen ATM dan MAPK14 merupakan dua kandidat target miR-4454.

Abstract

BACKGROUND: Tamoxifen (TAM) resistance in patient with breast cancer is the leading cause of mortality among women globally. Cytochrome P450 2D6 (CYP2D6) is involved in the metabolism of TAM, and recently NF-E2-related factor 2 (Nrf2) has recently been found as its regulator. However, the impact of Nrf2-mediated CYP2D6 regulation in the context of breast cancer and TAM resistance are currently unknown. Therefore, this study was conducted to examine the role of CYP2D6 and Nrf2 in breast cancer prognosis. MEDTHODS: The roles of CYP2D6 and Nrf2 were investigated in the T47D breast cancer cell line and T47D-derived TAM-resistant cells by examining the gene expression, cell viability, and transcriptional regulation by quantitative reverse transcription polymerase chain reaction (qRT-PCR), MTT, and reporter gene assay, respectively. Additionally, comprehensive in silico analysis of the transcriptomic and clinical data from The Cancer Genome Atlas database were performed to uncover the prognostic role of CYP2D6 and its regulator in breast cancer patients. RESULTS: CYP2D6 mRNA was low and Nrf2 protein was high in TAM-resistant T47D cells compared to parental cells. Nrf2 knockdown upregulated CYP2D6 mRNA levels and enhanced the cytotoxicity of TAM. Similarly, in silico analysis revealed that low CYP2D6 mRNA and high Nrf2 protein were related to a lower probability of survival. The rs1238662089 within the identified Nrf2-binding site was found to greatly affect CYP2D6 expression levels, indicating its role as predictor for better prognosis. CONCLUSION: This study revealed for the first time that Nrf2 regulates CYP2D6expression in breast cancer and is involved in TAM sensitivity; thus, plays a role in breast cancer patient prognosis.KEYWORDS: breast cancer, CYP2D6, Nrf2, pharmacoepigenetics, SNPs

Abstract

The aim of this study was to determine the expression of the P53 gene and testicular weight of male mice (Mus musculus) given Bulo forest honey and exposed to cigarette smoke. The experimental animals used in this study were 24 male mice which were divided into 4 treatment groups, each group consisting of 6 mice. Mice in group P0 were considered as control group (without treatment), while mice in group 1 (P1) were exposed to 2 cigarettes smoke, while mice in groups 2 (P2) and 3 (P3) were exposed to 2 cigarettes smoke and given Bulo forest honey at a dose of 0.8 ml/day and 0.4 mL/day, respectively. The data obtained were analyzed using one way ANOVA followed by using the Least Significant Difference (LSD) test. The results showed that Bulo forest honey administration did not statistically affect (P0.05) the ratio of testicular weight and P53 gene expression in male mice (Mus musculus) exposed to cigarette smoke for 30 days. It is concluded that the administration of Bulo forest honey at dose of 0.8 mL and 0.4 mL has not been able to increase the testicular weight ratio and reduce the expression of the P53 gene in male mice.

Abstract

virus was isolated from suspected Newcastle disease (ND) chicken in backyard farm at Sibang Village, Badung Regency, Bali Province. The isolates were then propagated and confirmed for NDV serologically. RNA isolation was performed by standard Trizol method. Phylogenetic tree analysis of NP gene (nt1020-1561) and HN gene (nt7019-7754) were performed using sequence of Badung-02/AK/14 and selected NDV strains from GenBank. Based on the NP gene sequence, the newly isolate closely related with other NDV strains belong to genotype VII that previously isolated such as Banjarmasin-010/10, Bali-1/07, and Cockatoo/90 with genetic distance 0.2%, 12.6%, and 18.0% respectively. The genetic distance with LaSota/46 virus (genotype II) is 19.6%. Based on HN gene sequence, genetic distance of the Badung-02/AK/14 with other viruses belong to genotype VII such as Banjarmasin-010/10, Bali-1/07, and Cockatoo/90 are 0.4%, 3.7%, and 4.2% respectively. The genetic distance with LaSota/46 virus (genotype II) is 9.0%. There were no difference between the result of nucleotide and amino acid sequence analysis, both in NP and HN gene (P0.05).

Abstract

This study aimed to determine the breeders' knowledge level on cattle estrous  period in buntu batu district, enrekang regency. The type of research used is descriptive quantitative research, which is a type of research that describes the level of knowledge of breeders in knowing the breeding period of female cows in Buntu Batu District, Enrekang Regency. This research was conducted in Buntu Batu District, Enrekang Regency, South Sulawesi from January to April 2022. The population in this study were all cattle breeders in Buntu Batu District, Enrekang Regency. The sample in this study totaled 92 respondents in Buntu Batu District. Data collection consisted of primary and secondary data. Primary data was obtained by direct observation. While secondary data was taken from the Enrekang Livestock Service Office. Data were analyzed using descriptive methods for the overall data in presenting frequency distribution tables. The results of the research analyzed the level of knowledge of breeders on the breeding period of cattle is enoug with a total of 41.3%.Keywords : Cattle, Estrous, Knowledge Of Breeders, Enrekang

Abstract

African Swine Fever (ASF) is a disease of pigs caused by African Swine Fever virus of the genus Asfvirus and family Asfarviridae. ASF virus has a limited host range and is only capable of affecting pigs. The disease affects all breeds and ages of pigs, although domestic pigs are the most susceptible. The disease is not zoonotic but causes economic losses on pig farms. ASF disease has various forms, ranging from per acute, acute, sub acute and chronic. Pigs infected with ASF will show common clinical symptoms such as high fever, cyanosis, haemorrhage, anorexia and ataxia. The spread of the disease is very rapid with a high mortality rate of more than 90%. The disease can spread through direct, indirect and vector contact. Prevention and control measures for ASF include strict biosecurity, reducing direct contact with contaminated feed or equipment such as feeding contaminated feed, quarantine, restricting pig traffic, depopulation of pigs sick with ASF, vector control, area closure, disposal and decontamination. Keywords:. Afrikan Swine Fever, Pig, Virus INTISARIAfrican Swine Fever (ASF) merupakan salah satu penyakit pada ternak babi yang disebabkan oleh virus African Swine Fever dari genus Asfvirus dan family Asfarviridae. Virus ASF memiliki jangkauan inang yang terbatas dan hanya mampu menyerang hewan babi. Virus ini menyerang semua ras dan umur babi walaupun babi peliharaan (domestik) menjadi jenis yang paling peka terhadap virus tersebut. Penyakit ini tidak bersifat zoonosis namun memberikan kerugian ekonomi bagi peternakan babi. Penyakit ASF memiliki bentuk penyakit yang bervariasi, mulai dari per akut, akut, sub akut dan kronis. Babi yang terinfeksi ASF akan menunjukkan gejala klinis umum seperti demam tinggi, sianosis, hemoragi, anoreksia dan ataksia. Penyebaran penyakit ini sangat cepat dengan tingkat mortalitas yang tinggi sampai mencapai 90% lebih. Penyakit ini dapat meyebar melalui kontak langsung, tidak langsung dan vektor. Untuk tindakan pencegahan dan pengendalian penyakit ASF dapat dilakukan dengan cara biosekuriti yang ketat, mengurangi kontak langsung dengan pakan atau perlatan yang terkontaminasi seperti swill feeding, karantina, membatasi lalu lintas babi, depopulasi babi yang sakit ASF, pengendalian vektor, penutupan daerah, disposal dan dekontaminasi.Kata Kunci :  African Swine Fever, Babi, Virus

Aging can be induced by UV-B and ischemia-reperfusion (I/R) due to the production of reactive oxygen species (ROS) and oxidative stress. This study aimed to test the anti-aging activity of RSV on UV-B- and I/R-induced Caenorhabditis elegans. The results showed that UV-B and I/R reduced the healthspan (pharyngeal pumping rate) and shorten the lifespan of wild-type worms. RSV ameliorated the aging phenotypes induced by UV-B and I/R. Anti-aging activities of RSV were not observed in skn-1 loss-of-function strain, indicating the critical involvement of SKN-1 in the mechanism of action of RSV. The expression of the putative SKN-1 target gene, gcs-1, indicated that RSV activated the SKN-1 pathway. It can be concluded that resveratrol can prevent aging due to extrinsic factors through SKN-1. Telah banyak bukti ilmiah mengenai efek anti-aging dari RSV. Penuaan dapat diinduksi oleh sinar UV-B dan iskemia-reperfusi (I/R) karena produksi reactive oxygen species (ROS) dan menyebabkan stres oksidatif. Tujuan dari penelitian ini adalah untuk menguji efek anti-aging RSV pada nematoda Caenorhabditis elegans yang diinduksi sinar UV-B dan I/R. Hasil menunjukkan bahwa paparan sinar UV-B dan perlakuan I/R dapat menurunkan healthspan (laju pompa faring) dan memperpendek lifespan dari nematoda C. elegans strain wild-type. Pemberian RSV dapat meningkatkan rerata healthspan dan median lifespan pada nematoda yang diinduksi UV dan I/R. Efek ini tidak teramati pada strain skn-1 (loss-of-function mutant), yang mengindikasikan pentingnya SKN-1 dalam mekanisme kerja RSV. Pemeriksaan ekspresi gen gcs-1, target gen utama dari SKN-1, mengindikasikan bahwa RSV dapat mengaktifkan jalur SKN-1. Dapat disimpulkan bahwa resveratrol dapat mencegah penuaan akibat faktor ekstrinsik melalui SKN-1. Kata Kunci:         anti-aging, iskemia-reperfusi, resveratrol, stress oksidatif, ultraviolet B
