Abstract

The development of quality shallot seeds from new superior varieties needs to be done because only about 10% of the shallot seed needs are currently covered. However, true shallot seeds (TSS) production from the local varieties, such as the Bima Brebes variety, is still rarely developed and has not been able to produce many split bulbs from TSS. The existence of split bulbs in one plant is preferred by consumers and is beneficial for shallot seedlings. This is related to the number of seeds farmers can use per kilogram of seed obtained. Therefore, this study aimed to select shallots that produce split bulbs from seeds produced from TSS. This research was conducted at Tajur Experimental Farm, Center for Tropical Horticulture Studies IPB University, Bogor. Seedlings were done in beds shaded with transparent plastic that can be opened and closed. The split bulbs produced by the Bima Brebes variety derived from TSS seed sources were still relatively low at 39.69% and significantly different from the comparison varieties at more than 60%. Sanren variety produced split bulbs from TSS seed sources as high as 64.13% and the Maserati variety about 61.25%. Keywords: spit bulb; dry weight; fresh weight; true shallot seed

Abstract

Pepper yellow leaf curl disease (PYLCD) is the primary disease that affects chili plants, resulting in a loss of quality and quantity. The identification of chili plants resistant to PYLCD is a solution to support optimal chili cultivation. The purpose of this study was to estimate genetic parameters to obtain information on the genetic control of chili resistance to PYLCD. The genotypes used in this study consisted of four parental, four hybrid genotypes, and 180 individuals of the F2 population. The results showed that the genotypes "F6074" and "IPBC12" were categorized as resistant, while the genotypes “IPBC5” and “YUNI” were categorized as susceptible. The distribution of resistance level in chili peppers to yellow curly leaf disease tends to follow a ratio of 13:3, with a dominance of susceptible traits. The assumption for this phenomenon is that resistance to PYLCD is controlled by at least two genes. Both genes have dominant and recessive epistatic interactions. Abstract

Eggplant (Solanum melongena L.) is an important horticultural crop widely cultivated worldwide. Stable genetic material and comprehensive information on genetic variability are essential for the development of superior varieties. Previous studies have obtained genetically stable doubled-haploid (DH) lines through anther culture. However, their agronomic performance has not been well identified. Therefore, this study aims to characterize doubled haploid (DH) eggplant lines (Solanum melongena L.) and identify potential candidates for future use as parental lines in hybrid breeding programs. Eleven doubled-haploid lines and three commercial varieties were evaluated using a randomized complete block design. Quantitative data were analyzed using analysis of variance (ANOVA), followed by Tukey’s HSD test at a 5% significance level, and cluster analysis based on phenotypic similarity. The results revealed significant variability among the genotypes for all quantitative characters except days to final harvest. Cluster analysis classified the genotypes into two major groups. Among the evaluated genotypes, Pro-AM1, Pro-AM38, Pro- AM50, and Hit-AM30 exhibited superior yield-related characters and are considered promising parental lines for hybrid development in eggplant. These findings demonstrate substantial genetic variability among the double-haploid eggplant lines and provide valuable resources for selecting superior lines to support future hybrid development.

Abstract

These experiments were aimed to obtain optimum medium for micropropagation of Ruscus. There were two experiments consist of in vitro shoots proliferation, shoot elongation and rooting.  The experiment of shoot proliferation performed by inducing adventitious shoots from explant in the Murashige and Skoog (1962)(MS) basal medium supplemented with combination of plant growth regulators BAP (0.0, 0.5, 1.0, 2.0, 4.0 and 6.0 mg/l) and IAA (0.0, 0.1, 0.2 and 0.4 mg/l). The elongation and rooting of plantlets were induced in the different concentration of the MS basal medium (0.5, 1.0 and 2.0 strength) combined with IBA (0.0, 1.0, 2.0, 3.0 and 4.0 mg/l).  Both experiments were arranged as completely randomized design with 15 replications. Adventitious shoots were produced in all medium supplemented with BAP with or without IAA. However MS medium supplemented with BAP 1 mg/l or 2 mg/l combined with IAA 0.2 mg/l were the best.  The number of adventitious shoots in these medium were 9.2 and 9.4 shoots after 8 weeks cultured respectively.  Increasing concentration of BAP more than 4 mg/l decreased number and size of adventitious shoots.  The plantlets produced in the proliferation medium were then transferring to the next treatments for elongation and rooting.  The best medium for elongation and rooting were medium with half strength of MS with or without IBA. Acclimatization conducted by transferring the rooted plantlets on the medium containing sterilized soil and rice husk charcoal (1:1).  After 4 weeks acclimatization, 60-100 percent of plantlets were survived and growth, depend on treatments.   Key words: Acclimatization, adventitious shoot, micropropagation, rooting, Ruscus hypophyllum L.

Abstract

Several bacterial pathogens can cause diseases of potato. Ralstonia solanacearum and Erwinia carotovora are those of the world’s most important diseases of potato, especially in tropical climates. This experiment was aimed to obtain putative potato cultivars having good tolerance to bacterial wilt caused by R. solanacearum and soft rot caused by E. carotovora. Two adopted potato cultivars in Indonesia, cv. Atlantic (2n=4X=48) and cv. Granola (2n=4X=48), were used as parents. The experiment was arranged in a Randomized Complete Design. Test for tolerance was perfomed in vitro using BF15 as susceptible control and Solanum stenotonum as tolerance control. The result showed that there was high diversity of tolerance level to both R. solanacearum and E. carotovora. Some clones showed good tolerance level as compared to cv. Atlantic and cv. Granola, but none showed that as compared to S. stenotonum.

Abstract

The improvement of eggplant yield through breeding is crucial. Doubled haploid technology has accelerated the development of varieties. This study utilized doubled haploid anther culture-derived lines (confirmed through flow cytometry and morphology) developed from embryos from a previous study to evaluate the morpho-agronomic performance and yield of doubled haploid eggplant lines. The experiment used a randomized complete block design (RCBD), three replications, and one factor, namely genotype (35 doubled haploid lines, three F1 varieties). Observation was made on plant height, dichotomous height, stem diameter, days to flowering and harvesting, fruit length, fruit diameter, weight per fruit, number of fruits, and fruit yield per plant. The data were analyzed using ANOVA, t-Dunnett, Tukey-Kramer at a 5% level, Pearson correlation coefficient, and selection index. The results showed significant variability in the population of doubled haploid lines. Fruit yield was positively correlated with plant height, dichotomous height, stem diameter, days to flowering, fruit length, and weight per fruit, but negatively correlated with the number of fruits per plant. These variables can be used as selection criteria because of their high heritability and genotypic coefficients of variation. The selection index revealed that the high-yielding doubled haploid lines with desirable morpho-agronomic traits were RS-P2, RS-P6, RS-P9, RS-P14, RS-P18, RS-H19, RS-H20, RS-H23, RS-H27, RS-H3, RS-M31, RS-M32, RS-M33, RS-M34, and RS-M37. The selected lines with high yield and good quality fruit, similar to the commercial hybrid parent, were Hitavi’s derived lines. All selected lines serve as the genetic basis for production improvements and long-term breeding programs for sustainable and productive eggplants that benefit farmers.

Abstract

Authors. 2018. Genetic variability, heritability, correlation, and path analysis in tomato (Solanum lycopersicum) under shading condition. Biodiversitas 19: 1527-1531. Information on genetic variability, heritability and character association between quantitative characters with yield are crucial in crop improvement. Eighteen genotypes of tomato were evaluated to study the quantitative genetic of yield and various yield attributing character under shading condition at Pasir Kuda Station, Bogor Agriculture University, West Java, Indonesia from August 2016 until January 2017. The result showed that plant height, dichotomous height, fruit weight, fruit length, fruit diameter, number of fruit per plant, and the fruit set had broad genetic variability and high heritability. Characters with broad genetic variability and high heritability can be used as sources in shading tolerance tomato improvement. Fruit weight and fruit number per plant had significant positive correlation coefficient and direct positive effect on fruit yield per plant. It is, therefore, recommended that fruit weight and fruit of number per plant should be given due importance in selection to develop shading tolerance variety in tomato

Abstract

Abstract. Herlina L, Reflinur, Sobir, Maharijaya A, Wiyono S. 2019. The genetic diversity and population structure of shallots (Allium cepa var. aggregatum) in Indonesia based on R gene-derived markers. Biodiversitas 20: 696-703. Molecular markers are very useful for determining plant genetic diversity, especially when they are associated with useful traits. In shallots, type of markers still very limited. Therefore, development of new molecular markers is challenging if the future demand for this crop is to be met. The present study reports the genetic diversity analysis of 36 accessions of shallots using molecular markers derived from partial R gene sequences. Six polymorphic R gene-derived markers (Acepa-1, Acepa-2, Acepa-3, Acepa-4, Acepa-5, and Acepa-6) were developed and applied to analyze the genetic diversity of shallots in Indonesia. These loci produced a total of 963 alleles with an average of 0.51 + 0.77 alleles per marker. The polymorphic information content value ranged from 0.2784 to 0.5236 with an average of 0.36015. The markers were able to differentiate the shallot genotypes into three major clusters, suggesting that shallots are characterized by poor levels of genetic diversity in Indonesia. After further validation, the markers will be very useful, serving as tools to support the breeding of disease-resistant shallot varieties.

Abstract. Sahid ZD, Syukur M, Maharijaya A. 2020. Genetic diversity of capsaicin content, quantitative, and yield component in chili (Capsicum annuum) and their F1 hybrid. Biodiversitas 21: 2251-2257. Chili (Capsicum annuum L.) is one of the horticultural plants that have many benefits. The benefit of chili was determined by pungency level of its fruit. Pungency level of the chili is due to the capsaicin content in fruit. Information about the genetic diversity of capsaicin is still rarely available. The aims of this study were to obtain diversity information on quantitative, yield component, and capsaicin content, and to analyze the correlation among chili genotypes based on their morphological characters. This study used Randomized Complete Block Design with three replications. The genetic material used in this study consisted of 21 genotypes consisting of 6 genotypes of chili elders and 15 hybrid F1 genotypes resulting from their crossing. Six genotypes of the chili parents are C5, F6074, F9160291, Yuni, Bara, and Giant. 15 hybrid F1 genotypes used in this study are C5 x Bara, C5 x F6074, C5 x Yuni, C5 x Giant, C5 x F9160291, Bara x F6074, Bara x Yuni, Bara x Giant, Bara x F9160291, F6074 x Yuni, F6074 x Giant, F6074 x F9160291, Yuni x Giant, Yuni x F9160291, and Giant x F9160291. The observation was made on the variables of quantitative, yield, and capsaicin components on chili. The results showed that the highest capsaicin content only was found in Bara x F9160291. The results of scatterplot analysis showed that the highest capsaicin and yield component was found in BaraxF6074 and C5 x Yuni genotype. The results of cluster analysis showed that chili was clustered into three color groups. The character of capsaicin content is negatively correlated and very different from fruit weight, fruit diameter, fruit length, thick fruit flesh, total amount of fruit per plant, and fruit weight per plant.

Abstract

Abstract. Laisina JKJ, Maharijaya A, Sobir, Purwito A. 2021. Drought adaptive prediction in potato (Solanum tuberosum) using in vitro and in vivo approaches. Biodiversitas 22: 537-545. Prediction of drought adaptive potatoes requires selection indicators. This study aimed to determine in vitro selection indicators for obtaining drought adaptive potato genotypes in vitro and in vivo. The in vitro study was performed using single-node explants from seven genotypes cultured for six weeks on MS medium added with 0.2 M sorbitol to achieve the research aims. The cultures were partially observed and followed by watering the in vitro tubing medium. For the in vivo study, drought stress was applied for 21 days when the plants reached 25 days after planting (DAP) and for 14 days when the plants reached 50 DAP, then plants were irrigated again until harvesting. The results showed that plant height and root length can be used as selection indicators because they had a relative decrease below 50%, a close relationship and had the same variance with the in vitro number of tubers, in vitro tuber fresh weight and in vivo tuber fresh weight in drought stress at 25 DAP. These characters were determinant characters for the clustering pattern in the heat map cluster analysis. Based on these characters, SSI analysis was also performed and it was found that the PKHT4 was more adaptive than the PKHT6 in vitro, but the two genotypes were classified in the medium adaptive in vivo. In conclusion, plant height and root length were determined as the in vitro selection indicators in predicting drought adaptive genotypes in vivo at the plant age of 25 DAP.
