Abstract

Growth performance, survival rate, productivity, and carcass analysis were examined in the whole grow-out production system for Pacific white shrimp Litopenaeus vannamei fed four commercial diets over 15 weeks. Shrimp were held in twelve hapas nets with a size of 2 x 2 x 1 m and installed within the commercial ponds. The shrimp were stocked with the density of 100 shrimp m-2 per net in a completely randomized design (CRD). All feed used in this study was characterized with high protein levels within the range of 40 – 42% labeled as HP A and B; and medium protein levels (30 – 35%), labeled as MP A and B. Based on the proximate and nutritional profile analysis of the diet, the nutritional profile of HP B, MP A and MP B written on the feed bags showed an inconsistent results compared to the results of the test.  Shrimp fed the highest protein level had higher final body weight; feed conversion ratio, thermal growth coefficient, survival rate and average daily growth compared to MP B. Shrimp fed higher protein level also exhibited the lowest feed conversion ratio compared to the group of shrimp fed with medium protein. The results obtained in this study indicated that with small difference in growth rates, especially between HP groups compared with MP A, there is still a chance to re-formulate the diet to produce a cost-effective diet that still fulfill the specific nutrient requirement of the shrimp. Feed did not influence the nutritional deposition of the whole body of shrimp, but it seemed due to the inappropriate drying techniques Keywords: Protein level, growth, carcass, Litopenaeus vannamei, specific nutrient

Abstract

Produksi HPI skala pilot menghasilkan HPI yang kaya asam amino esensial dan memiliki aktivitas antioksidan, sehingga berpotensi digunakan sebagai ingridient pangan fungsional.KATA KUNCI: AbstractFish protein hydrolysate (FPH) is a product of the hydrolysis reaction of peptide bonds resulting in the release of short active peptides and more easily absorbed. Fish protein hydrolyzate (HPI) is a product of the hydrolysis reaction of peptide bonds resulting in the release of short active peptides that are more easily absorbed. Snapper is an abundant marine fishery commodity in Indonesia. The aim of this study was to determine the physicochemical characteristics and bioactive capabilities of HPI from red snapper (Lutjanus sp.) on a pilot scale (120 kg biofermenter). Raw material and HPI testing includes organoleptic tests, TVB tests, ALT tests, proximate tests; HPI production uses the alcalase enzyme 20,000 U/kg substrate at a temperature of 55°C with a hydrolysis time of 0 to 8 hours. Tests for the degree of hydrolysis and antioxidant activity tests using the DPPH method are carried out every hour. The results showed that the hydrolysis time began to stabilize at the 6th to the 8th hour. The value of DH HPI at the 6th hour was 13.3% and antioxidant inhibition was 6.75%. The raw material is fresh fish with a TVB of 12.89%; protein content 21.41%; fat 4.66%; water 72.36%; ash 0.95%. The resulting HPI has a protein content of 19.20%; fat 0.15%; water 0.06%; ash 1.15%. HPI yield 15.75% of raw materials. Total microbes and organoleptics meet SNI standards. The highest amino acid content of raw materials and HPI is glutamic acid with values of 3.3% and 3.4% respectively. The lowest amino acid content in raw materials and HPI is histidine with values of 0.43% and 0.52% respectively. Pilot scale HPI production produces HPI which is rich in essential amino acids and has bioactive antioxidant capabilities, so it has the potential to be used as a functional food ingredient.

Abstract

The commercially important red seaweed Kappaphycus alvarezii is extensively cultivated for carrageenan production. Despite its economic value, large-scale reproduction and genetic enhancement remain limited due to its low regeneration potential. This study aimed to optimize plant growth regulator (PGR) concentrations for efficient callus induction in K. alvarezii. A completely randomized design was employed, comprising five treatments with varying concentrations of indole-3-acetic acid (IAA) and 6-benzylaminopurine (BAP), along with a control lacking PGRs. A total of 180 explants from meristematic tissues of acclimatized thalli were cultured (30 explants per treatment). The highest callus induction rate (88%) was achieved with 1.50 mg/L IAA and 5 mg/L BAP (Treatment F), with visible callus formation beginning around day 38. A progressive color change from brown to white was observed, indicating active cellular proliferation. Other treatments exhibited lower induction rates, ranging from 0% (control) to 61% (Treatment D). These findings underscore the critical influence of auxin–cytokinin interactions on callogenesis and offer an optimized hormonal regime for improving in vitro culture efficiency. The established protocol provides a valuable platform for future large-scale propagation and genetic improvement strategies in K. alvarezii, contributing to the advancement of seaweed biotechnology.

Abstract

Cultivation of the seaweed Gracilaria verrucosa is necessary as a preventive measure against overexploitation. One applicable method is tissue culture, which requires special attention to nutrient needs to support accelerated growth. These nutrients can be provided through fertilization. One commonly used chemical fertilizer in seaweed tissue culture is Provasoli’s Enriched Seawater (PES). This study aims to determine the optimal PES fertilizer dosage for the production of G. verrucosa seedlings through tissue culture using a vegetative propagation method. This study used a completely randomized design with analysis of variance (ANOVA) consisting of four treatments of different PES fertilizer doses with three replicates for each treatment. The results showed that fertilizer dose variation did not significantly affect the absolute growth of explant weight, but tended to increase the number of growth points. Growth points began to increase in the second week for all treatments, and by the fourth week, the number of growth points at a 1.5% dosage showed a significant difference compared to other doses. The application of PES fertilizer in G. verrucosa seedling production through tissue culture with vegetative propagation indicates that different fertilizer doses do not affect explant growth but significantly influence the increase in growth points by the fourth week
