Abstract

This study was aimed to evaluate the metabolite contents and antineuroinflammatory potential of Marsilea crenata Presl. grown under a controlled environmental condition. The antineuroinflammatory test has been carried out in vitro using ethanolic extract of M. crenata leaves on HMC3 microglia cells. An in silico approach was applied to predict the active compounds of the extract. The HMC3 microglia cells were induced with IFNγ to create prolonged inflammatory conditions and then treated with 96% ethanolic extract of the M. crenata leaves of 62.5, 125, and 250 μg/mL. The expression of MHC II was analyzed using the ICC method with the CLSM instrument. Metabolites of the extract were profiled using UPLC-QToF-MS/MS instrument and MassLynx 4.1 software. In silico evaluation was conducted with molecular docking on 3OLS protein using PyRx 0.8 software, and physicochemical properties of the compounds were analyzed using SwissADME webtool. The ethanolic extract of M. crenata leaves could reduce the MHC II expression in HMC3 microglia cells in all concentrations with the values 97.458, 139.574, and 82.128 AU. The result of metabolite profiling found 79 compounds in the extract. In silico evaluation showed that 19 compounds gave agonist interaction toward 3OLS, and three met all parameters of physicochemical analysis. The ethanolic extract of the environmental-controlled growth of M. crenata leaves antineuroinflammatory activity on HMC3 microglia cells. The extract was predicted to contain some phytoestrogen compounds which act as 3OLS agonists.

Abstract

Neurodegenerative disease is one of the problems faced by postmenopausal women due to estrogen deficiency. Phytoestrogen compounds can be used as an alternative treatment for diseases caused by estrogen deficiency by binding to their receptors through the estrogen receptor (ER) dependent pathway. With in silico studies, this study aims to predict how phytoestrogen compounds will stop neurons from dying by using the dependent ER pathway. Genistein, daidzein, glycitein, formononetin, biochanin A, equol, pinoresinol, 4-methoxypinoresinol, eudesmin, α-amyrin, and β-amyrin compounds were prepared with ChemDraw Ultra 12.0. Then their pharmacokinetic and pharmacodynamic properties were examined using SwissADME. Geometry optimization of the compound was performed using Avogadro 1.0.1, and molecular docking of the compound to the ERα (1A52) and ERβ (5TOA) receptors was performed using AutoDock vina (PyRx 0.8). The interaction visualization stage was carried out with Biovia Discover Studio 2021, while the toxicity values of the compounds were analyzed using pkCSM and ProTox II. The results showed that the equol compound met the pharmacokinetic, pharmacodynamic, toxicity criteria, and had similarities with the native ligand 17β-estradiol. Equol compound inhibits neurodegeneration via an ER-dependent pathway by binding to ERα (1A52) and ERβ (5TOA) receptors.

Abstract

Semanggi (Marsilea crenata Presl.) merupakan salah satu tanaman yang telah dimanfaatkan oleh masyarakat Indonesia, khususnya Jawa Timur, sebagai bahan makanan selama beberapa dekade. Marsilea crenata Presl. mengandung senyawa fitoestrogen memiliki struktur dan fungsi mirip estrogen dalam tubuh. Tujuan penelitian melakukan uji sitotoksisitas dan menentukan nilai IC50 dari ekstrak etanol 96 persen daun Marsilea crenata Presl. terhadap sel hFOB 1.19. Metode yang digunakan adalah microtetrazolium (MTT) assay dengan microplate reader. Sel dikultur pada microplate-96 well dan diberikan ekstrak dengan variasi dosis 62,5 ppm; 125 ppm; 250 ppm; 500 ppm; 1000 ppm; 2000 ppm dan 4000 ppm, kemudian ditambahkan reagen MTT 100μL, diinkubasi selama 2-4 jam dan ditambahkan SDS 10 persen sebagai stopper. Pembacaan hasil menggunakan microplate reader. Hasil dari pembacaan tersebut digunakan sebagai perhitungan nilai IC50 yang dapat menunjukkan tingkat sitotoksisitas ektrak etanol 96 persen Marsilea crenata Presl. Nilai IC50 yang didapatkan sebesar 2074,965 μg/ml. Berdasarkan hasil tersebut ekstrak etanol 96 persen Marsilea crenata Presl. dapat dikatakan aman dan tidak bersifat sitotoksik jika dikembangkan menjadi bahan obat.

Abstract

Background: Estrogen deficiency can trigger several diseases, one of which is neurodegenerative. Neurodegenerative begins with neuroinflammation, which triggers the activation of microglia cells resulting in a pro-inflammatory activity. Potential and relatively safe therapy to use to overcome it is by using phytoestrogen compounds. Marsilea crenata Presl. is a plant that contains phytoestrogens. Objective: The purpose of this study was to analyze the neuroprotective effect of the water fraction of Marsilea crenata Presl. leaves which was shown by inhibition of neuroinflammation and marked by increased levels of MHC II against HMC3 microglia cells. Methods: IFN-γ is induced into HMC3 microglia cells for 24 hours to cause inflammatory conditions. Marsilea crenata Presl. leaves water fraction was given at a dose of 62,5; 125; and 250 µg / ml. Analysis of the neuroprotective effect of HMC3 microglia cells using the ICC method with the aid of the CLSM instrument. Results: The results of this study indicate that the water fraction of Marsilea crenata Presl. leaves can reduce MHC II expression at concentrations of 125 and 250 μg / ml with values ​​of 465,748 and 460,884 AU at p less than 0,005. Conclusion: This study concludes that the water fraction of Marsilea crenata Presl. has neuroprotective activity shown at doses of 125 and 250 μg/ml which can reduce MHC II expression in HMC3 microglia cells induced by IFN-γ. The ED50 value which has a neuroprotective effect is at a dose of 0,582 μg/ml.

Abstract

Pendahuluan: Prevalensi penyakit neurodegeneratif meningkat tiap tahunnya dan mayoritas dialami perempuan yang mencapai fase menopause. Hal tersebut kemudian mengurangi kualitas hidup perempuan menopause. Penyakit neurodegeneratif pada wanita menopause sering diakibatkan oleh defisiensi estrogen, sehingga perlu adanya pengganti hormon estrogen dalam menjaga homeostasis sistem saraf pusat, salah satunya fitoestrogen. Salah satu tanaman yang mengandung fitoestrogen adalah semanggi (Marsilea crenata Presl.) Tujuan: Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak etanol daun M. crenata terhadap peningkatan aktivitas lokomotor ikan zebra. Metode: Penelitian ini dilakukan dengan induksi rotenon sebagai pengganggu aktivitas lokomotor ikan zebra, melalui pengukuran motilitas. Perlakuan dilakukan dengan dengan memberikan ekstrak etanol daun M. crenata pada dosis 2,5; 5; 10; dan 20 mg/mL untuk mengetahui pengaruh ekstrak terhadap peningkatan aktivitas lokomotor ikan zebra yang telah diinduksi rotenon. Pengamatan dilakukan dengan melihat kuantitas berenang ikan zebra setiap 5 menit pada hari ke 0; 7; 14; 21; dan 28. Kesimpulan: Pemberian ekstrak etanol 96 persen daun M. crenata dapat meningkatkan aktivitas lokomotor yang diketahui melalui peningkatan motilitas ikan zebra secara signifikan pada dosis optimum 2,5 mg/mL pada hampir tiap kelompok hari. Hal ini membuktikan ekstrak daun M. crenata berpotensi dalam mencegah progresifitas penyakit neurodegeneratif.

Abstract

Background: Overweight is the accumulation of excessive fat that can harm health. People who are overweight usually have a size of body anthropometry more than normal, such as the size of the waist circumference (WC), upper arm circumference (UAC), and subcutaneous fat thickness (SFT). Women are more to being overweight, because metabolism in women is lower and body fat is more than men. They attempt to return to normal body weight, by following the fitness clubs, diet and so forth. In this study, researchers used a slimming herbs tea to help restore their body size. Purpose: This study was to determine the effects of slimming herbs tea with dietary adjustments to the results of anthropometric measurements in the group of gymnastic class women. Method: The method used is quasy experimental design with the design of "pre-posttest control group design”. The study was held for 14 days by the number of respondents 14 people. Respondents were divided into 2 groups: control and treatment (consumption of slimming herbs tea). Slimming herbs tea composition consisting of green tea, guazuma extracts, parameria extract, fennel extract and curcuma extract consumed 2 times a day after meals every day. Data were analyzed using SPSS with Paired T-Test. Result: The results showed slimming herbs tea with dietary adjustments can reduce the results of anthropometric measurements such as WC as much as 6,21 cm ,UAC as much as 2,2 cm ,and SFT as much as 2,7 mm. While the reducing measurement in control group, in WC as much as 2,57 cm, UAC as much as 1,6 cm, SFT as much as 1,4 mm. Abstract

Phytoestrogen is a group of compounds that can replace the estrogen function in the body. One of its roles was as anti-neuroinflammatory by inhibiting the microglia M1 polarity activation. Marsilea crenata Presl. is a plant that suspected to contain phytoestrogens. The aim of this research was to determine the metabolite profile of 96% ethanol extract of M. crenata using UPLC-QToF-MS/MS, and prediction the anti-neuroinflammatory activity of compounds with molecular docking. The 100 ppm of 96% ethanol extract in DCM and methanol were injected 5 µl each into the UPLC-QToF-MS/MS, and then analyzed by Masslynx 4.1 software to determine the compounds. The result of metabolite profiling shows a total 59 compounds in both DCM and methanol. Molecular docking was done with Autodock 4.2.6. After being analyzed, there are 3 compounds that are predicted to have activities similar to 17?-estradiol, they are prochlorperazine, 12-Aminododecanoic acid, and 1-methyl-2-[(4-methylpiperazin-1-yl)methyl]benzimidaol-5-amine hydrochloride. The results showed that the three compounds were predicted to be phytoestrogen compounds from M. crenata leaves, which have potential as anti-neuroinflammatory.

Abstract

Wanita pascamenopause dapat menyebabkan berbagai macam masalah kesehatan, diantaranya adalah osteoporosis yang diakibatkan karena gangguan produksi hormon estrogen dalam tubuh. Salah satu alternatif yang muncul dan berpotensi untuk menggantikan hormon estrogen adalah fitoestrogen. Daun M. crenata diketahui mengandung senyawa fitoestrogen. Tujuan dari penelitian ini adalah untuk mengetahui fraksi n-Butanol daun M. crenata dalam meningkatkan aktivitas osteoblas pada proses pembentukan tulang terhadap sel hFOB 1.19. Penelitian ini menggunakan metode ICC dengan bantuan instrumen CLSM. Penelitian ini dilakukan dengan pemberian fraksi n-Butanol dengan varian konsentrasi62,5; 125; dan 250 ppm, serta pemberian genistein sebagai kontrol positif pada sel hFOB 1.19. Hasil dari penelitian ini menunjukkan bahwa fraksi n-Butanol daun M. crenata memiliki kandungan senyawa estrogenik sebagai agen untuk meningkatkan aktivitas osteoblas melalui penurunan ekspresi ER? bebas sel hFOB 1.19 dan konsentrasi 62,5 ppm sebagai konsentrasi optimal dengan nilai ekspresi 594.108 AU.

Abstract

Tubuh manusia akan mengalami penuaan dan menyebabkan kenaikan resiko penyakit dan pada akhirnya menyebabkan kematian. Fenomena tersebut dapat muncul salah satunya akibat penurunan homeostasis tubuh karena menurunnya hormon estrogen. Salah satu senyawa yang berpotensi mengatasi keadaan tersebut adalah fitoestrogen dari golongan flavonoid. Potensi senyawa flavonoid dengan aktivitas fitoestrogenik, dapat diketahui dengan melakukan pengamatan In silico terhadap Estrogen Receptor ? (ER-?) dengan molecular docking. Tujuan penelitian ini adalah untuk mengetahui mekanisme senyawa fitoestrogen melalui jalur ER-dependent lain seperti ER-?. Tahapan molecular docking dilakukan menggunakan plug-in Autodock Vina dalam PyRX 0.8. Hasilnya diketahui senyawa apigenin, quercetin, kaempferol, katekin, genistein dan daidzein memiliki interaksi agonis terhadap ER-?. Interaksi yang diperoleh dapat menggambarkan bahwa senyawa tersebut mampu memberikan aktivitas estrogenik ketika membentuk ikatan ligan reseptor.

Abstract

Background: Pregnant women who drink herbal medicine can increase the risk of meconium amniotic fluid, and meconium amniotic fluid is a risk factor for asphyxia in newborns. Objectives: Identify three forms of culture in drinking herbal medicine in pregnant women, namely the mother's values and ideas about drinking herbal medicine, herbal drinking activities, and material on the culture of drinking herbal medicine. Methods: This research is a qualitative method with a phenomenological approach. Informants in this study consisted of pregnant women and key informants consisted of families of pregnant women, cadres, ustadzah, midwives, and herbal medicine providers. Results: The cultural forms of drinking herbal medicine include: 1) Values and Ideas. Drinking herbal medicine for pregnant women is still practiced because of their subjective norms and normative beliefs from the surrounding environment. All informants believed that herbal medicine could provide benefits rather than negative effects, and only midwives stated that herbal medicine had negative effects, namely meconium amniotic fluid, and even asphyxia. 2) The activity of drinking herbal medicine. Pregnant women drink herbal medicine from the beginning of pregnancy until delivery. The frequency of taking herbal medicine was various. Religious figures and community leaders have an important role in the culture of drinking herbal medicine. 3) Herbal ingredients. The herbs consumed by each informant were different, there were informants who consumed homemade herbs and bought them at the market. Conclusions: The culture of drinking herbal medicine is still carried out because the local community's belief about the benefits of herbal medicine is still high, and lack of public knowledge about the effects of drinking herbal medicine on pregnant women.
