Abstract

Spirulina platensis is a bioactive-rich microalga of great potential as a source of natural antioxidant. Natural antioxidants are increasing in demand, because it is considered as more safe than synthetic antioxidants. The objective of this study was to determine the effect of solvent on the potential of natural antioxidant from fresh S. platensis. The antioxidant compound was extracted using either acetone or ethyl acetate by reflux method. The yield of extractions were 1.86% and 3.07% for aceton and ethyl acetate, respectively. Fresh S. platensis extracts were considered as potential antioxidant source with IC50 values 65.89 ppm, and 76.36 ppm when using acetone and ethyl acetate, respectivetly, even though it were still lower compare to the IC50 of positive control quercetin i.e 21.64 ppm. Phytochemical test showed positive results of phenolic compound, triterpenoids, steroid, flavanoid, and saponin. aw value of Spirulina platensis was 0.61 and 0.81 for aceton and ethyl acetate, respectively. It was concluded that different solvent affected the antioxidant activity of S. platensis extract. Keywords: Antioxidants, aceton, ethyl acetate, fresh Spirulina platensis

Abstract

AbstractDunaliella salina is a microalga rich pigment carotenoids that can be used asnatural dyes. The problemfaced for temporative β-carotene stability which is unstable and easily degraded is unstable and easilydegraded. The purpose ofthis researchwas to determine content and stability of β-carotene pigment inD.salina with fixings addition of MgCO3 and NaHCO3 with temperature treatment as factor that influenceβ-carotene pigment degradation during storage condition. The research method used β-carotene pigmentwhich was obtainedbymaceration and adding fixing agent (NaHCO3 0,5% and without fixings addition),and was heated at temperature of (50°, 70° and 90°C), be measured the content of β-carotene pigmentduring storage (observation hour 0, 24, 48, 72, 96, 120 and 144). Data analyzed by ANOVA and HonestlySignificant Different (HSD). The result showed that content of β-carotene pigment at 50°C temperatureshowed that stability and pH value performed slowest degradation after144 hours. While stability and pHvalue at 90°C temperature performed fastest degradation after 96 hours. Based on this study the addition ofNaHCO3 gave more effective in retaining β-carotene degradation compared to that of MgCO3 which showedby high stability in sample NaHCO3 0,5%, temperature 50°C in 144 hours storage length.

Abstract

Fish ball is a perishable food product. One of an alternative method which could be applied to delay the deterioration process was using carrageenan edible coating with the addition of galangal rhizome’s essential oil as an antibacterial agent. The aim of this study was to find out whether the addition of carrageenan edible coating with galangal rhizome’s essential oil could increase the shelf life of the tilapia fish ball during cold storage or not. This study was used experimental laboratories with the completely randomized design used factorial (2x6) two factors include essential oils concentrations (0%, dan 1%) and storage time (0, 3, 6, 9, 12, 15) with three repetitions. The shelf life of the fish ball observed by sensory, TPC, TVBN, pH, Aw, moisture content, and gel strength test. The parametric data were analyzed by the ANOVA, while the non-parametric was using the Kruskal Wallis test. As the result, the fish ball with the 1% treatment was still acceptable until 15 days of storage compared with the control which only lasts for 9 days. The other results were the sensory test was 6.63≤π≤6.95, TPC value was 1.07x105 cfu/g, TVBN value was 30.33±1.68 mgN%, the moisture content was 61.38%±0.47, the Aw was 0.89±0.006, the pH was 6.24±0.0, and the gel strength was 721.19±1.61 g.cm. The treatment of 1% essential oil addition to carrageenan edible coating was significantly different (p<0.05) compared to the control treatment during cold storage to the characteristic of tilapia fish ball.

Abstract

Application of ozone combined with the chilling system on preserving fresh fish has obviously brought about more advantages. This study observed the application of an ozone-slurry ice combined system for maintaining the freshness of two different fishes during storage. The fishes used were red tilapia (Oreochromis niloticus) and short-bodied mackerel (Scomberomorus rastrelliger), using ozone and slurry ice. The experimental design used was factorial using a completely randomized design employed with an ozone concentration of 0 ppm and 3.5 ppm with storage times of 0, 4, 8, 12, and 16 days. The parameters observed included: peroxide value (PV), total volatile base nitrogen (TVBN), total viable count (TVC), and a sensory test. The parametric data were analyzed using ANOVA and followed by least significant difference (LSD), whereas the non-parametric data were analyzed using the Kruskal Wallis test followed by multiple comparison tests. Ozone and slurry ice treatment of different concentrations had a significant (p<0.05) effect on the TVBN, the PV, and the TVC. The interaction of ozone and slurry ice provided no significant  difference (p>0.05) in both samples.A sensory evaluation in both samples showed good correlation with TVC. This study showed that ozone and slurry ice could  maintain the samples freshness during storage.

Abstract

Spirulina platensis is a blue-green microalga which is classified into Cyanobacteria. S. platensis is often used as functional food ingredient because their phycocyanin pigment has antioxidant properties. However, the pigment is of low stability and is sensitive to temperature, pH, oxygen, and humidity. The purpose of this study was to determine the effect of microencapsulation and pH treatment on the stability of phycocyanin S. platensis. The experimental design used in the study was Completely Randomized Factorial Design with 2 factors namely microencapsulation process and different pH values (pH 4 and pH 8). The results showed that the microencapsulation process and pH treatment had different effects (P <0.05) on the stability of phycocyanin. S. platensis with microencapsulation at pH 8 produced smaller phycocyanin degradation (5.32±1.37%), antioxidant degradation (38.12±0.31%), and total color different (TCD) (22.1±0.07) compared to other treatments. While the relative concentration (CR) value of the microencapsulated phycocyanin in S. platensis was 94.68±1.37%, higher than that of other treatments. Microencapsulation at pH 8 stabilized phycocyanin by preventing precipitation of the protein.

Abstract

Kandungan protein Kerang Anadara granosacukup tinggi sehingga rentan sekali mengalami pembusukan. Kendala yang dihadapi dalam penggunaan es adalah bentuk es. Penggunaan bongkahan es yang besar dan kasar serta tajam akan menyebabkan kerusakan fisik. Alternatif yang dapat diterapkan adalah Alat pemecah es . Penelitian ini bertujuan  mengamati kualitas  dan kerusakan protein selama pendinginan dengan membandingkan es curai yang dihancurkan manual dan es curai hasil dari mesin penghancur es. Metode yang dilakukan dengan Pengesan daging kerang dengan es yang dihancurkan manual dengan es hasil mesin penghancur.Sampel disimpan selama 6 hari dan diamati kandungan protein dan TVB-N setiap 3 hari sekali. Rancangan Percobaan menggunakan rancangan faktorial dengan 2 faktor yaitu es yang dihancurkan secara manual dan dihancurkan mesin penghancur es. Hasil dari penelitian ini didapatkan kandungan protein kerang tanpa pengesan dari hari ke-0 14.04±0,71 %; dan hari ke-6 9.27±0.17%. Kandungan protein kerang yang diberi es balok yang dihancurkan secara manual hari ke-0 14,23±0,46%; dan hari ke-6 11,68±0,40%. Kerang yang menggunakan es curai hasil mesin penghancur es hari ke-0 adalah 13,91±0,68%; dan hari ke-6 yaitu 14,28±0,63%. Hasil kandungan TVB-N kerang tanpa pengesan dari hari ke-0 13.11±0,29 mgN/100g; dan hari ke-6 64,97±0.61mgN/100g. Kerang yang diberi es balok yang dihancurkan secara manual hari ke-0 13,00±0,48 mgN/100g; dan  hari ke-6 23,28±0,53 mgN/100g. Kerang menggunakan es curai hasil mesin penghancur es hari ke-0 adalah 13,21±0,66 mgN/100g; dan hari ke-6 yaitu 19.72±0,49 mgN/100g. Protein content Anadara granosa shells are high enough to be prone to decay. The obstacle faced in the use of ice is the form of ice. The use of large, rough and sharp blocks of ice will cause physical damage. The applicable alternative is the Ice Breaker. This study aims to observe the quality and deterioration of proteins during cooling by comparing the manually destroyed ice cubes and ice cultivated from ice-breaking machines. The method is done with the Order of shellfish with the ice that is destroyed manually with ice crushing machine. Samples are stored for 6 days, and observed the content of protein and TVB-N every 3 days. The experimental design used a factorial design with 2 factors: manually destroyed ice and crushed ice crusher. The results of this study found the content of shell protein without impressive from day-0 14.04 ± 0.71%; and 6th day 9.27 ± 0.17%. The content of shellfish protein given ice beam which was manually destroyed on day 0 14.23 ± 0.46%; and the 6th day 11.68 ± 0.40%. The shell that used ice cubes from the 0 th day crusher was 13.91 ± 0.68%; and the 6th day is 14.28 ± 0.63%. Results of TVB-N content of shellfish without impression from day 0 13.11 ± 0.29 mgN / 100g; and day 6 64,97 ± 0.61mgN / 100g. Shellfish with ice blocks manually destroyed on day 0 13.00 ± 0.48 mgN / 100g; and day 6 23.28 ± 0.53 mgN / 100g. The shells using ice cubes from the 0 th day crusher were 13.21 ± 0.66 mgN / 100g; and day 6 is 19.72 ± 0.49 mgN / 100g.

Abstract

Fish sausage is a processed fish product in the form of an emulsion, so that the stability of the emulsion is one of the most important factors for the success of the product. The aim of this study is to find out the effect of kappa carrageenan as a hydrocolloid in increasing the emulsion stability and determining the best concentration of kappa carrageenan based on the value of emulsion stability and the characteristics of milkfish sausage. The research method used was experimental laboratories with a completely randomized one-factor design using ANOVA, which was then continued with the HSD test. Organoleptic value data were analyzed using the Kruskal-Wallis and Mann-Whitney tests. The concentrations of kappa carrageenan used were 0%;0.5%;1%;1.5 with 3 repetitions. The results of the study show that kappa carrageenan addition has a significant effect (p<0.05) on the value of emulsion stability, sensory, moisture content, gel strength, and water holding capacity. The concentration of 1% kappa carrageenan (P2) was the best concentration with quality criteria: sensory of 8.14<µ<8.32, moisture content of 63.02%, water holding capacity of 55%, gel strength of 1200.14 g.cm, and emulsion stability of 83.23. Compared to other treatments, fish sausage with the addition of 1% kappa carrageenan has the brightest appearance according to the specific color of the product, has a chewy texture and solid but not hard, and has the highest value of emulsion stability.

Abstract

Edible film merupakan suatu lapisan tipis yang dapat melindungi bahan pangan untuk menjaga kualitas produk. Bawang putih mengandung senyawa allin yang merupakan senyawa antibakteri jika ditambahkan pada edible film karagenan. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh penambahan ekstrak bawang putih sebagai antibakteri dan karakteristik edible film karagenan. Metode penelitian yang digunakan bersifat eksperimental laboratories dengan rancangan percobaan Rancangan Acak Lengkap (RAL) dengan parameter uji konsentrasi ekstrak bawang putih yang berbeda. Data dianalisis menggunakan ANOVA dan dilakukan uji lanjut Beda Nyata Jujur (BNJ). Hasil penelitian menunjukan bahwa ekstrak bawang putih berpengaruh terhadap aktivitas antibakteri dan karakteristik edible film karagenan. Hasil yang diperoleh dari perlakuan penambahan 7,5% ekstrak bawang putih sebagai antibakteri dapat dilihat zona hambatnya terhadap bakteri S. aureus sebesar: 3,00±0,100  mm dan bakteri E. coli: 2,18±0,07 mm sedangkan nilai karakteristik ketebalan: 0,17±0,003 mm, laju transmisi uap air: 6,49±0,42 g/m2/jam, kuat tarik: 13,88±0,29 MPa, persen pemanjangan: 14,75±0,09%. Kesimpulan menunjukkan bahwa penambahan ekstrak bawang putih pada edible film karagenan memberikan pengaruh nyata (P<0,05) terhadap aktivitas antibakteri dan karakteristik edible film karagenan. Edible film is a thin layer which able to protect the food and keep its quality.Garlic contains Allin compound which worked as antibacterial if added into carrageenan edible film. This study aimed to determine the effect of garlic extract addition as antibacterial and the characteristic of carrageenan edible film. The method was experimental laboratories with Completely Randomized Design with different concentration of garlic extract as the testing parameters. Data were analyzed using ANOVA then tested using Honestly Significance Difference (HSD) to determine the significance value of the sample. The result showed that the garlic extract was significant difference toward the antibacterial activities and the characteristic of carrageenan edible film. The result obtained from 7.5% garlic extract addition as antibacterial was the inhibition zone against S. aureus and E. coli of 3.00±0.100 mm and 2.18±0.07 mm; respectively. The other parameters such as thickness 0.17±0.003 mm, the water vapor transmission rate 6.49±0.42 g/m2/hour, the tensile strength 13.88±0.29 MPa, and the percent elongation 14.75±0.09%. The result indicated that the addition of garlic extract was significant difference (P<0,05) toward the antibacterial activities and characteristic of carrageenan edible film.

Abstract

 Permen jelly merupakan permen yang memiliki tekstur kenyal. Tekstur ini terbentuk karena adanya pembentuk gel yaitu gelatin. Penggunaan gelatin dalam permen jelly masih diperdebatkan status kehalalannya, karena adanya unsur babi dalam gelatin. Sementara itu, sumber alternatif lain yang memberikan tekstur kenyal di permen jelly adalah karagenan, tetapi belum jelas sejauh mana dapat menggantikan gelatin. Tujuan dari penelitian ini adalah mengetahui pengaruh iota karagenan yang diekstrak dari Eucheuma spinosum terhadap sifat fisik dan kimia permen jelly dan formulasi terbaik dari permen jelly. Bahan yang digunakan dalam penelitian ini adalah gelatin dan tepung iota karagenan yang diekstrak dari rumput laut E. spinosum. Metode penelitian yang digunakan yaitu experimental laboratories dengan desain percobaan rancangan acak lengkap (RAL) dengan 4 perlakuan yang berbeda yaitu Gelatin : Iota Karagenan = 8% : 0% (A), Gelatin : Iota Karagenan = 0% : 8% (B ), Gelatin : Iota Karagenan = 6% : 2% (C), dan Gelatin : Iota Karagenan = 2% : 6% (D). Hasil dari penelitian ini didapatkan permen jelly terbaik sebagai alternatif pengganti gelatin yaitu perlakuan B dengan kekuatan gel 1.109,27 g.cm2 dan nilai hedonik sebesar 6,84 ≤ µ ≤ 7,52. Kadar air semua perlakuan belum memenuhi standar SNI (20%). Kadar abu semua perlakuan memenuhi standar SNI (3%). Untuk aw dan pH produk tersebut berkisar 0,84–0,89 dan 3–4 . Jelly candy which has chewy texture. This texture was formed by the gelling agent of gelatin. The use of gelatin in jelly candy still debated of halal status, because there were elements of pork in gelatin. While other alternative source which gives chewy texture in jelly candy was carrageenan, but carrageenan were used in making jelly candy was not extent to be replace of gelatin. The aim of this study was to determine the effect of iota carrageenan extracted from Eucheuma spinosum to physical and chemical characteristics of jelly candy and the best formulation of jelly candy. The materials used in the research were gelatin and powder of iota carrageenan extracted from seaweed E. spinosum. The method used was experimental laboratories by using completely randomized design (CRD) with 4 different treatments was Gelatin : Iota carrageenan = 8% : 0% (A), Gelatin : Iota carrageenan = 0% : 8% (B ), Gelatin : Iota carrageenan = 6% : 2% (C), and Gelatin : Iota carrageenan = 2% : 6% (D). The result showed that the best jelly candy as an alternative to gelatin performed by treatment B with gel strength of 1109,27 g.cm2 and the results for sensory was 6,84 ≤ µ ≤ 7,52. The water content of all treatment did not comply with ISO standard (20 %). Ash content of all treatment comply with ISO standard (3 %). For aw and pH of the products were ranged from 0.84–0.89 and 3–4, respectively.

Abstract

Bagi seorang muslim status halal suatu produk non-pangan salah satunya kosmetik maupun obat-obatan sangat mutlak yang harus dipenuhi. Produk yang beredar dipasaran harus terbebas dari kandungan babi atau bahan lain yang menyebabkan produk tersebut tidak lagi halal. Metode Real Time Polymerase Chain Reaction (RT-PCR) yang dapat mendeteksi kandungan babi maupun alkohol untuk dapat memastikan kandungan tersebut terbebas dari cemaran babi. Dalam penelitian ini, telah dilakukan analisis kemungkinan adanya kandungan unsur babi dari ke empat produk non-pangan seperti kapsul, kuas roti, day cream dan sabun kecantikan yang dianalisis menggunakan RT-PCR.Penelitian ini dibagi menjadi dua tahap, tahap awal dilakukan ektraksi DNA dan tahap kedua dilakukan analisis RT-PCR. Didapatkan pada tahap awal ekstraksi DNA, hasil pengukuran dan kemurnian dengan nilai konsentrasi kapsul 2,9 ng/µI; kuas roti sebesar 3,4 ng/µI; day cream sebesar 2,4 ng/µI; dan sampel sabun kecantikan sebesar 2,2 ng/µI. Pada tahap kedua, secara keseluruhan dari ke empat produk yang dipilih secara acak sebagai objek penelitian produk kapsul terbebas dari kandungan babi, kecuali pada produk kuas roti sebesar 3,15%; day cream sebesar 3,52%; dan sabun kecantikan sebesar 124,83% positif tercemar kandungan babi.
