Abstract

Asthma is a disease defined as chronic inflammation of the airways which is characterized by recurrent coughing and shortness of breath. Dermatoglyphs are the appearance of tendrils and tendril patterns on the fingers, palms, toes and soles. Dermatoglyph is used as an aid in the detection of a number of diseases that have a strong heritable basis and is also used to detect abnormalities. This study aims to analyze and compare the patterns of dermatoglyphics in asthma sufferers and non-asthmatics on the fingertips. This type of research is descriptive research with cross sectional sampling method. Samples were taken from the student population of the Faculty of Mathematics and Natural Sciences, Padang State University as many as 100 samples of fingerprint patterns. The results obtained showed that there was a significant difference in the number of arch fingerprint patterns of asthmatics compared to normal individuals. Seeing this significant difference in numbers indicates a possible link between fingerprint patterns and asthma phenotypes.

Abstract

Dermatoglyphics is a science that studies fingerprint patterns that can be inherited genetically. Fingerprint patterns can be divided into three patterns, arch, loop, and whorl. Observation of fingerprints is used to determine fingerprint patterns and the relationship between these patterns and the fields of health, education, and criminology. Left-handed is a term for people who predominantly use their left hand as their main hand to carry out daily activities. This research aims to determine the relationship between fingerprint patterns and individuals who tend to carry out activities with the left side of their body (left-handed). The type of research carried out was descriptive research with a cross-sectional sample collection method. The sample was taken from the 2019-2022 student population of the Faculty of Mathematics and Natural Sciences, Padang State University, totaling 100 individuals. The samples were taken in November and December. This research was carried out at the Genetics and Biotechnology Laboratory, Faculty of Mathematics and Natural Sciences, Padang State University. The results of this study show that the dominant fingerprint pattern found in the left-handed individual phenotype is 16 fingerprint patterns, while normal individuals have 5 arch fingerprint patterns. This shows that there is a link between the phenotype of left-handed individuals and dermatoglyphics.

Abstract

The mitochondrial genome is useful for identification because the mutation rate of mtDNA is higher than nDNA. Previous studies have carried out many analyses on the mitochondrial genome of several species, the Chaetodontidae group. Fish in this group are biological indicator fish of coral reefs, so their molecular characteristics must be enriched to conserve these fish. This article aims to review several forms of the mitochondrial genome of the Chaetodontidae group of species, including: Chaetodon speculum, Chaetodon octofasciatus, Chaetodon wiebeli, and Heniochus chrysostomus. Data were collected qualitatively by reviewing previous research articles related to the Chaetodontidae mitochondrial genome. The review focused on the characteristics of each species' mitochondrial genome, including length, overall composition, tRNA, and non-coding region. Each species exhibits various features of the mitochondrial genome and is mapped in its phylogenetic analysis.

Abstract

Science learning requires direct experience in order to develop the ability to explore and understand the natural surroundings scientifically. So far in elementary school at Bukittinggi, the process of learning science only memorizes facts, principles or theories. In science learning, students are not involved in finding or applying their own ideas and they do not instill the values contained in the learning. The science learning process in classroom is not yet fully in accordance with the demands of the curriculum. The teacher provides more material in the textbook without relating it to students' daily problems that are in accordance with the material, thus causing less interesting in learning. The teacher also has not actively involved all students in learning and emphasizes learning only on students who are smarter. The purpose of this study was to increase the activity and learning outcomes of science learning by using a problem-based learning model in SDN 11 Aur Kuning Bukittinggi, West Sumatra. This classroom action research was conducted in 2 cycles with the research subjects of grade IV students with totalling 25 people. The research data were collected through observation and tests. The effectiveness of actions in each cycle from the results of observations and tests is described and then reflected to take corrective action in the next cycle. In science learning, students' learning activities were observed, namely: visual activities, oral activities, writing activities, motoric activities and emotional activities. The research findings show that the problem-based learning model can increase student activity and learning outcomes.

Abstract

The village of Muara Sikabaluan in the Mentawai Islands faces various challenges in education, economy, and the environment. A community empowerment program based on the circular economy has been implemented to enhance the capacity for processing agricultural and marine products, such as bananas, taro, cassava, and fish. Training in product processing and online and offline marketing was provided to improve the knowledge and skills of residents, especially housewives. The program also promotes the establishment of a village business and economic center as a hub for business development and community welfare improvement. Pretest and posttest results showed significant increases in residents' knowledge and skills related to product processing and the application of circular economy principles. With a participatory and sustainable approach, Muara Sikabaluan Village is expected to become a model for sustainable village development.

Abstract

Science education in schools faces various challenges, such as the dominance of teacher-centered lecture methods, a lack of integration between theory and practice, and limited learning facilities. These issues hinder students from developing critical thinking, collaboration, and adaptability to modern demands. This study aims to develop a Problem-Project Based Learning (P2BL) model as a solution to address these challenges. The model integrates the strengths of Problem-Based Learning (PBL) and Project-Based Learning (PjBL) to create contextual, practical learning experiences that encourage critical thinking among students. A Research and Development (R&D) approach with the ADDIE framework is employed to ensure the systematic development of the model. The P2BL model consists of five main components: syntax, reaction principles, social system, support system, and instructional as well as accompanying impacts. The syntax of this model includes stages such as problem definition, experimentation, planning, presentation, and evaluation, supported by multi-directional interactions between instructors and students, as well as collaboration among students. The research findings indicate that implementing the P2BL model enhances academic abilities, practical skills, and the development of attitudes such as teamwork, responsibility, and time management. With its relevance to the demands of the Industrial Revolution 4.0 era, the P2BL model offers a more interactive, effective, and adaptive learning alternative to improve the quality of Science education in schools.

Abstract

Validity refers to the degree or level of accuracy of a research instrument used to measure a variable and serves as an essential component in the development of instruments. This study aims to determine the construct and content validity of the science process skills (SPS) test instrument for the Science Learning course in the MI/SD Teacher Education Program at the State Islamic University of Sheikh Ali Hasan Ahmad Addary, Padangsidimpuan. The research method employed is a descriptive quantitative-qualitative approach, with data collected using observation sheets for the assessment of the SPS test instrument by two validators. The data from the validators' assessments were analyzed using Gregory's Test formula to determine the instrument's validity level. The results show that 13 SPS test items received validator assessments in the range of scores 3-4, categorized as highly relevant, with an overall validity score of 1 (one). Therefore, it can be concluded that the 13 test items of the science process skills instrument on the topic of temperature and heat possess a very high level of validity.

Abstract

HD disease is a common cause of neonatal intestinal obstruction. HD disease is a congenital disorder of the colon characterized by the absence of parasympathetic ganglion cells in the sub mucosal plexus of Meissneri and the mienterikus plexus of Auerbachi. Objective: To design specific primers for amplification of exon 5 in EDNRB gene and test the specificity in silico. To determine the optimum PCR conditions for amplification of exon 5 in EDNRB gene. Methods This type of research is descriptive. The purpose of descriptive research is to provide an overview or description of the EDNRB exon 5 gene primer design and PCR optimization. Electrophoresis results showed the temperature of 56 oC is the right temperature characterized by a single band and looks the thickest and brightest with the appropriate amplicons size of 367 bp, The results for the EDNRB gene primer exon 5 obtained the optimum concentration of 0.8 µM. This is characterized by DNA bands that look clear and thickest. The specific sequence of forward primer is 5'ATTTCA GAGACG GGAAG TGG3' reverse primer is 5'GGCACT CACTCATCATCTTG 3' which produces amplicons with a length of 367 bp. The optimum annealing temperature is 56 oC and the optimum primer concentration is 0.8 µM.

Abstract

EDNRB gene is a gene located on chromosome 13q22, plays a role in the signaling pathway during the development of the ENS intestinal nerves. Mutations in the EDNRB gene in humans are associated with Hirschsprung Disease (HD). Mutations in the EDNRB gene are the second most common cause after the RET gene in cases of HD with a percentage of 5–10%. To detect mutations, PCR products are needed. Primers are one of the basic components of PCR, while optimization of annealing temperature and optimization of primer concentration are factors in the success of primer attachment to the target gene. This study aims to design EDNRB exon 4 gene primers and determine the optimum annealing temperature and primer concentration to amplify the EDNRB gene. Primer design with Geneious Prime, and good primer criteria are analyzed in silico. Optimization of annealing temperature and primer concentration is carried out with gradient PCR and variations in primer concentration. The results of this study obtained the best primers, namely the forward primer 5'- CAGTAAGTGTGGCCTGAAAG-3' and the reverse primer 5'-GTGGAACCGAAGTGACTAGA-3', which amplified the EDNRB gene exon 4. The optimum PCR conditions for amplification of exon 4 of the EDNRB gene were at an annealing temperature of 57.2 ℃ and at a primer concentration of 0.5 μM.

Abstract

The endothelin receptor type B (EDNRB) gene is located on chromosome 13, consisting of 7 exons and 6 introns. The EDNRB protein produced from this gene belongs to the family of G-protein coupled receptors. Mutations in the EDNRB gene play a role in some cases of Hirschsprung's cause, although the frequency is low. This study aims to design primers and test specificity using BLAST primers to determine the specific primer attached to the EDNRB gene. Primers were designed using Geneious Prime, which were then analyzed in silico based on good primer criteria. The results of this research showed that primers made using Geneious Prime produced the best primers, namely forward primer 5'-GAGTTCGGAGCTTTGCCT-3' and reverse 5'-AGCCCACCA TGATTTCA GC-3' and the results of the specificity test showed that the primer specifically attached to chromosome 13 which is the location of the EDNRB gene (13q22) with a product length of 879 bp.
