Abstract

HD disease is a common cause of neonatal intestinal obstruction. HD disease is a congenital disorder of the colon characterized by the absence of parasympathetic ganglion cells in the sub mucosal plexus of Meissneri and the mienterikus plexus of Auerbachi. Objective: To design specific primers for amplification of exon 5 in EDNRB gene and test the specificity in silico. To determine the optimum PCR conditions for amplification of exon 5 in EDNRB gene. Methods This type of research is descriptive. The purpose of descriptive research is to provide an overview or description of the EDNRB exon 5 gene primer design and PCR optimization. Electrophoresis results showed the temperature of 56 oC is the right temperature characterized by a single band and looks the thickest and brightest with the appropriate amplicons size of 367 bp, The results for the EDNRB gene primer exon 5 obtained the optimum concentration of 0.8 µM. This is characterized by DNA bands that look clear and thickest. The specific sequence of forward primer is 5'ATTTCA GAGACG GGAAG TGG3' reverse primer is 5'GGCACT CACTCATCATCTTG 3' which produces amplicons with a length of 367 bp. The optimum annealing temperature is 56 oC and the optimum primer concentration is 0.8 µM.

Abstract

EDNRB gene is a gene located on chromosome 13q22, plays a role in the signaling pathway during the development of the ENS intestinal nerves. Mutations in the EDNRB gene in humans are associated with Hirschsprung Disease (HD). Mutations in the EDNRB gene are the second most common cause after the RET gene in cases of HD with a percentage of 5–10%. To detect mutations, PCR products are needed. Primers are one of the basic components of PCR, while optimization of annealing temperature and optimization of primer concentration are factors in the success of primer attachment to the target gene. This study aims to design EDNRB exon 4 gene primers and determine the optimum annealing temperature and primer concentration to amplify the EDNRB gene. Primer design with Geneious Prime, and good primer criteria are analyzed in silico. Optimization of annealing temperature and primer concentration is carried out with gradient PCR and variations in primer concentration. The results of this study obtained the best primers, namely the forward primer 5'- CAGTAAGTGTGGCCTGAAAG-3' and the reverse primer 5'-GTGGAACCGAAGTGACTAGA-3', which amplified the EDNRB gene exon 4. The optimum PCR conditions for amplification of exon 4 of the EDNRB gene were at an annealing temperature of 57.2 ℃ and at a primer concentration of 0.5 μM.

Abstract

The endothelin receptor type B (EDNRB) gene is located on chromosome 13, consisting of 7 exons and 6 introns. The EDNRB protein produced from this gene belongs to the family of G-protein coupled receptors. Mutations in the EDNRB gene play a role in some cases of Hirschsprung's cause, although the frequency is low. This study aims to design primers and test specificity using BLAST primers to determine the specific primer attached to the EDNRB gene. Primers were designed using Geneious Prime, which were then analyzed in silico based on good primer criteria. The results of this research showed that primers made using Geneious Prime produced the best primers, namely forward primer 5'-GAGTTCGGAGCTTTGCCT-3' and reverse 5'-AGCCCACCA TGATTTCA GC-3' and the results of the specificity test showed that the primer specifically attached to chromosome 13 which is the location of the EDNRB gene (13q22) with a product length of 879 bp. Abstract

Long bean (Vigna sinensis) is a plant from the Leguminosae family, which has an important role in the Indonesian economy because it is widely consumed as food. The purpose of this research is to utilize mutation technology in supporting plant breeding to produce superior long bean varieties. The research method carried out was a non-factorial Randomized Group Design (RAK), namely soaking the seeds of long bean plants with colchicine to see the effect of colchicine treatment on chromosomal mutations in long bean plants. The results showed significant differences between colchicine concentrations of 0.02% and 0.03%. At a concentration of 0.02%, the cell nucleus appeared solid red and in the center of the cell, but the number and shape of chromosomes were not clearly visible. This concentration is not optimal for inducing polyploidy. Meanwhile, at a concentration of 0.03%, the chromosomes were more clearly visible than the previous concentration.

Abstract

Islamic values in the subject matter of global warming symptoms for class XI at Senior high school. This research is a continuation of R&D research and development with the ADDIE model using 5 stages namely Analysis, Design, Development, Implementation, and Evaluation. The research instrument used was a practicality test questionnaire and student answer sheets on the e-module. there are 31 students and one educator have participated in this practicality test. The results of data analysis on the average value at the practicality test stage were carried out by educators and students. With the very practical category, the practicality analysis value of educators is 92.65 %, Very practical criteria for data analysis by students yielded 89.09 %. The results of the study concluded that the integrated e-module of Islamic values in the material on global warming symptoms that was developed was very practical to use in the learning process for class XI as an alternative teaching material in the learning processABSTRAK: Penelitian ini bertujuan untuk mengukur tingkat praktikalitas e-modul terintegrasi nilai-nilai islam pada materi gejala pemanasan global untuk kelas XI Sekolah Menegah Atas. Penelitian ini merupakan lanjutan penelitian dan pengembangan R&D dengan model ADDIE menggunakan 5 tahapan yaitu Analisis, Desain, Pengembangan, Implementasi, dan Evaluasi. Instrumen penelitian yang digunakan berupa angket uji praktikalitas dan lembar jawaban peserta didik pada e-modul. terdapat 31 peserta didik dan satu pendidik telah berpartisipasi dalam pengujian kepraktisan ini. Hasil analisis data rata-rata nilai pada tahap uji praktikalitas dilakukan oleh pendidik dan peserta didik. Dengan katerogi sangat praktis diperoleh nilai analisis kepraktisan pendidik sebesar 92,65%, Kriteria sangat praktis analisis data oleh peserta didik menghasilkan sebesar 89,09%,. Hasil penelitian menyimpulkan bahwa e-modul terintegrasi nilai-nilai islam pada materi gejala pemanasan global yang dikembangkan sangat praktis digunakan dalam proses pembelajaran kelas XI sebagai bahan ajar alternative dalam proses pembelajaran.

Abstract

This meta-analysis investigates the effectiveness of the Problem-Based Learning (PBL) model in enhancing students' critical thinking and problem-solving skills in science education. A total of 42 peer-reviewed articles published between 2020 and 2024 were analyzed, covering diverse educational levels and scientific disciplines. The analysis revealed a large average effect size (ES = 0.798; 95% CI: 0.782–0.813), indicating a strong positive impact of PBL. The low standard error (SE = 0.043–0.060) reflects high estimation precision. Statistical tests, including funnel plot symmetry, Trim-and-Fill analysis (p = 0.991), and Egger’s regression test (p = 0.947), confirm the absence of significant publication bias. Moreover, residual heterogeneity was minimal (τ² = 0.000), validating the appropriateness of the fixed-effects model. These findings provide robust empirical evidence that PBL is consistently effective across various contexts in improving students’ 21st-century competencies. The study offers meaningful implications for educators and policymakers in designing science learning strategies that foster higher-order thinking skills.

Abstract

Consumers have the right to correct information about the products they consume. One example of a food product that is vulnerable to counterfeiting is chicken nuggets. To overcome this problem, qPCR can be an effective solution. Previous research has designed primers targeting the chicken ND5 gene. However, the PCR conditions for this primer pair have never been optimized. This research aims to optimize qPCR conditions and carry out chicken nugget authentication trials. Genomic DNA was isolated from five weight variations of chicken meat samples as a qPCR standard curve and three chicken nugget samples. Optimizing the annealing temperature with gradient PCR gave the best results at a temperature of 57.6OC. The results of the authentication test showed that the chicken meat content in TD nuggets was 54.7%, C nuggets 43.8%, and D nuggets 37.3%. This value is still in accordance with the Indonesian National Standard (SNI).

Abstract

Stem cells are emerging as an attractive option because of their intrinsic ability as vectors for drug delivery to diseased tissues. The term stem cells or stem cells was first introduced by a Russian histologist, Alexander Maksimov (1874-1928). The purpose of this study is to describe the research and development of stem cell applications for stroke. This study is a literature study (review article) regarding stem cells (stem cells) in stroke which has been discussed in several scientific journals. From several articles that have been collected in giving stem cells to people with stroke, namely in bone marrow. MSC (Mesenchymal Stem Cell) is a multipotent stem cell from bone marrow and Adult SPM and because of its high applicability. Bone marrow is a rich source of hematopoietic stem cells so it can be applied to stroke patients.

Abstract

Hirschsprung’s Disease (HSCR) is a congenital disorder characterized by the absence of ganglion cells in the intestinal wall, leading to impaired intestinal motility. One of the key genes involved in the pathogenesis of HSCR is the Endothelin Receptor Type B (EDNRB) gene, particularly mutations in exon 6. Detecting these mutations requires specific primers and well-optimized PCR conditions. This study aims to design specific primers for exon 6 of the EDNRB gene using in silico methods and to determine optimal PCR conditions through annealing temperature and primer concentration optimization. Primers were designed based on the EDNRB gene sequence (Accession No. NG_011630.3) using Geneious Prime and validated for specificity using Primer-BLAST. PCR optimization was conducted in vitro using various annealing temperatures 52.8–55.3°C and primer concentrations 0.5–1.1 µM on normal human DNA samples. PCR products were analyzed by 1.5% agarose gel electrophoresis. The primer design for exon 6 of the EDNRB gene yielded the following results: both forward and reverse primers were 20 bp in length, with melting temperatures (Tm) of 58.1°C and 56.1°C, and GC contents of 55% and 50%, respectively. In silico analysis confirmed primer specificity to chromosome 13. Optimal PCR conditions were achieved at an annealing temperature of 55.3°C and a primer concentration of 0.7 µM, yielding a single amplicon of 517 bp.

Abstract

Teratogen berefek membahayakan terhadap perkembangan embrio. Tujuan penelitian ini untuk mengetahui pengaruh teratogen AlCl3 terhadap perkembangan embrio ayam (Gallus gallus domesticus). Metode penelitian ini dilakukan secara eksperimental dengan perlakuan induksi AlCl3 0,2 mg/L dan kontrol (tanpa perlakuan). Pemberian perlakuan diberikan kepada masing-masing 5 butir telur ayam yang kemudian diinkubasi selama 48 jam. Pengamatan preparat embrio ayam menggunakan whole mount method dengan pewarnaan metilen blue dan eosin. Hasil penelitian menunjukkan ukuran embrio pada kelompok perlakuan lebih kecil (0,6 cm) dibandingkan dengan embrio kontrol (1cm). Jumlah somit pada kelompok perlakuan sejumlah 12 pasang sedangkan pada kelompok kontrol 28 pasang somit. Ukuran kepala lebih kecil jika dibandingkan dengan kelompok kontrol. Penelitian ini menunjukkan bahwa AlCl3 dapat menyebabkan malformasi pada embrio ayam, terkait dengan ukuran embrio, jumlah somit, dan ukuran kepala.
