Abstract

Ki lemo barks has been used as traditional spasmolitic (called oils of massage) since a long time by ethnic of Dayak. The odors inhaled is presumed possessing inhibitory on mice locomotor activity. The present research has been done to determine the components in blood plasma of mice after inhalation of essential oil of ki lemo barks, and the possibility of their role in the inhibition on mice locomotor activity.Inhalation of essential oil of ki lemo bark (Litsea cubeba Lour. Pers) at the doses of 0.5 ml inhibit locomotor up to 60,75 %, whereas at the doses of 0.1 ml and 0.3 ml inhibit locomotor activity of 57,44 % and 54,20 %, respectively.Identification and quantification of volatile active compounds in the blood plasma were carried out by GC-MS analysis after half an hour, one hour, and two hours inhalation. The blood plasma of three mice were collected in heparin tube, isolated and concentrated by the C-18 column (100 mg-Seppak Waters) eluted with methanol and bidistilled water (60:40). The major compounds identified were citronellol, citronellal, α-terpineol, and 1,8-cineole. The blood level of those substances was increasing for one hour after inhalation but decreasing after two hours.Key words : essential oil of ki lemo bark (Litsea cubeba Lour. Abstract

Hiperurisemia merupakan suatu kondisi berlebihnya kadar asam urat dalam darah. Hal ini dapat terjadi karena adanya kelebihan produksi asam urat dan kekurangan ekskresi asam urat, yaitu ditandai dengan kadar asam urat yang tinggi (> 7 mg/ dL). Jika hal tersebut terjadi secara terus-menerus maka akan terjadi gout arthritis. Di Arboretum Garut Terdapat banyak tanaman yang dapat digunakan sebagai antihiperurisemia seperti sirsak (Annona muricata L.), kayu manis (Cinnamomum burmanii), temu putih (Curcuma zedoaria), jamblang (Syzigium cumini), kersen (Muntingia calabura L.), pucuk merah (Syzigium myrrtifolium), dan salam (Syzigium polyanthum). Review jurnal ini membahas aktivitas antihiperurisemia beberapa tanaman yang berada di Arboretum Garut. Metode yang digunakan yaitu studi literatur beberapa jurnal yang didapat melalui situs google scholar, sciencedirect, Elsevier, dan sebagainya. Hasil yang didapat dari beberapa tanaman tersebut memiliki aktivitas antihiperurisemia dan tanaman yang memiliki persentase penurunan asam urat terbesar yaitu daun S. polyanthum dengan nilai persentase penurunan asam urat sebanyak 79,35%. Daun S. polyanthum memiliki potensi antihiperurisemia paling baik, sehingga perlu dikembangkan lebih banyak di Arboretum Garut. Kata Kunci: Arboretum, Asam Urat, Gout, Hiperurisemia, Tanaman Obat Hyperuricemia is a condition of excessive levels of uric acid in the blood. This can occur due to an overproduction of uric acid and a lack of uric acid excretion, which is characterized by high uric acid levels (> 7 mg / dL). If it happens continuously, gout arthritis will occur. In Garut Arboretum, there are many plants that can be used as antihyperuricemia such as soursop (Annona muricata L.), cinnamon (Cinnamomum burmanii), white turmeric (Curcuma zedoaria), java plum (Syzigium cumini), cotton candy berry (Muntingia calabura L.), pucuk merah (Syzigium myrrtifolium), and indian baywatch (Syzigium polyanthum). This journal review discusses the antihyperuricemia activity of several plants in Garut Arboretum. The method used is a literature study of several journals obtained from the google scholar site, sciencedirect, Elsevier, etc. The results show that several plants have antihyperuricemia activity and among those plants the S. polyanthum) have the highest reducing effect on blood uric acid with the reducing percentage of 79.35%.  The leaves of S. polyanthum plant have the best potential for antihyperuricemia, so this plant is suggested to be developed in the Garut Arboretum.

Abstract

In the course of our continuing search for novel paralytic compound from Indonesian plants, the methanol extract of the stem bark of Erythrina variegata (Leguminosae) showed significant paralytic activity against the third instar larvae of silkworm (Bombyx mori). The purposes of this research were isolation and structural elucidation of paralytic compound from the stem bark of E. variegata. Using the paralytic activity following the separation, the methanol extract was separated by combination of column chromatography to yield prenylisoflavone, warangalone. The chemical structure of warangalone was identified based on spectroscopic evidence and comparison with the previous reported. The paralytic activity of warangalone showed weak activity against the third instar larvae of silkworm (B. mori).

Abstract

Piperine is bioactive alkaloid extracted from white pepper (Piper albi Linn) with several beneficial pharmacological activities. It is also known to enhance the bioavailability of several limited solubilities of other compounds such as curcumin and resveratrol. Nevertheless, due to its poorly water-soluble character, piperine and extract of white pepper are less applied among other plant extracts. Therefore, our research focussed on the engineering of white pepper extract standardized to piperine in liposome solution and liposome powder for further preparation of various pharmaceutical dosage forms.Our research in preparation of extracted white pepper in liposome delivery system successfully prepared spheric liposome solution with 98.92%±1.17% and an average size of 398.7 nm. The liposome was also successfully prepared in dry powder form with sucrose as a carrier for liposome protection. The rehydration is successfully forming a spheric shape with a similar profile to the initial liposome solution. This research paves the way for scalable white pepper extract liposome preparation without the utilization of toxic organic solvent, to produce the commercial solvent-free product in the future.Keywords: Piperine, Piper albi Linn, liposome, extract, organic solvent-free, drug delivery system, spray dry

Abstract

Antioxidants are substances that can provide endogenous protection and exogenous oxidative stress by capturing free radicals. Many plants are efficacious as antioxidants, namely plants that contain polyphenols, especially flavonoids, so many are formulated as natural antioxidants. Plants such as Muntingia calabura, Syzygium cumini, Ocimum basilicum, and Eleutherine bulbosa contain polyphenol compounds, especially flavonoids which are efficacious as natural antioxidants. This research aimed to study antioxidant activity derived from some potential plants using the DPPH method by calculating the IC50 value of each plant extract. This research method starts from the determination process to prove the validity of the plants used, the extraction process using the maceration method with 70% ethanol solvent, then the antioxidant activity of extracts from each plant was carried out using the DPPH method. This research starts from the determination process to ensure the correctness of the plants used, then the extraction process is carried out using the maceration method with 70% ethanol solvent. After that the antioxidant activity was determined from the four plants using the DPPH method to see the strongest IC50 value among the four plants. IC50 is the concentration of the sample to inhibit 50% of free radicals. The results of IC50 values from ethanol extract of M. calabura leaves, Syzygium cumini leaves, Ocimum basilicum leaves and Eleutherine bulbosa bulbs, were 18.72; 63,84; 141.59 and 173.15 ppm. Ethanol extract of M. Calabura has a smaller IC50 value of 18.72 ppm which has a very strong and most powerful antioxidant from the ethanol extract of Syzygium cumini, Ocimum basilicum and Eleutherine bulbosa. Keywords : Antioxidant, Muntingia calabura, Syzygium cumini, Ocimum basilicum, Eleutherine bulbosa, DPPH Method

Abstract

Erythrina plants, locally known as “dadap ayam”, are higher plant species and have been used as a folk medicine for treatment of cancer. To prove the effectiveness of the leaves and stem bark of E. variegata as an anti-cancer agent, the assay in this research was focused on in vitro test towards breast cancer cell T47D. In the course of our continuing search for novel anti-cancer agent from Erythrina plants, the methanol extract of the leaves and stem bark of E. variegata showed significant anti-cancer activity against breast cancer cell T47D in vitro using the Sulphorhodamine B (SRB) assay. By using the anti-cancer activity to follow the separations, the methanol extract was separated by combination of column chromatography. The chemical structure of an anti-cancer compounds were determined on the basis of spectroscopic evidence and comparison with the previously reported and identified as an erythrina alkaloid (1) and isoflavonoid (2). Compounds (1-2) showed anti-cancer activity against breast cancer cell T47D used with IC50 of  1.0 and 3.3 µg/mL, respectively. Abstract

During the course of our continuing search for novel anti-malarial compounds from Indonesian plants, the methanol extract of the bark of E. variegata showed significant anti-malarial activity toward Plasmodium falciparum in vitro using the lactate dehydrogenase (LDH) assay. The methanol extract of the bark of E. variegata  was separated by using bioassay-guide fractionation. The ethyl acetate fraction showed the most activity, exhibiting equipotency against both strains of parasite with IC50 of 23.8 µg/mL against 3D7 and 9.3 µg/mL against K1. Furthermore, by using the anti-malarial activity to follow separation, the ethyl acetate fraction was separated by combination of column chromatography to yield an active compound. The chemical structure of active compound was determined on the basis of spectroscopic evidences and comparison with those previously reported and identified as an isoflavonoid, warangalone. The warangalone showed anti-malarial activity against both strains of parasite used with IC50 of 4.8 µg/mL against 3D7 and 3.7 µg/mL against K1.

Abstract

The aim of this study is to examine the possible use of ion-exchange resin on Dowex AG 50x8 in separating Y-90 from Sr-90 which then could be applied in the basic of manufacture of producing Sr-90/Y-90 generator for the production of radiopharmaceutical cancer therapy. The findings showed that a distributive coefficient of Sr-90 and Y-90 with HCl 6 N solvent showed that distributive coefficient of Sr-90 in cation-exchange resin on Dowex 50x8 was greater than Y-90. Y-90 in HCl 6 N solvent was more easily eluted, while Sr-90 was bound to resin.. In conclusion, resin on Dowex AG 50x8 with HCl 6 N elution solvent might be able to separate Y-90 from Sr-90 as the basis for Sr-90/Y-90 generator.

Abstract

Kecombrang (Etlingera elatior) merupakan tanaman suku Zingiberaceae yang memiliki potensi sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan sitotoksik terhadap larva udang dari ekstrak daun, bunga, dan rimpang kecombrang, serta menetapkan kadar senyawa fenol totalnya. Sampel daun, bunga, dan rimpang kecombrang dimaserasi dengan pelarut etanol 70% selama 3x24 jam dalam suhu ruangan. Ekstrak yang didapat selanjutnya dievaporasi pada suhu 50 0C sehingga menghasilkan ekstrak kental. Uji aktivitas antioksidan dilakukan menggunakan metode peredaman radikal bebas 1,1-difenil-2-pikrihidrazil (DPPH), dan uji sitotoksik menggunakan metode BSLT (Brine Scrimp Lethality Test). Penetapan kadar fenol total dengan standar asam galat menggunakan reagen Folin Ciocalteu yang diukur dengan spektrofotometer pada panjang gelombang 765 nm. Aktivitas antioksidan dari ekstrak daun, bunga, dan rimpang kecombrang ditunjukkan oleh nilai IC50 masing-masing 52,05; 457,54; dan 310,69 ppm. Aktivitas sitotosik yang dinyatakan dalam nilai LC50 dari ketiga bahan tersebut berturut-turut adalah 859,039; 418,022; dan 1261,202 ppm. Kadar senyawa fenol total ekstrak daun, bunga, dan rimpang kecombrang dihitung sebagai asam galat ekuivalen berturut-turut sebesar 0,339% ± 0,006; 0,144% ± 0,024; dan 0,074% ± 0,018. Ekstrak daun kecombrang menunjukkan aktivitas antioksidan yang paling baik dan kadar fenol total paling tinggi dibandingkan ekstrak bunga dan rimpang kecombrang. Ekstrak daun dan bunga kecombrang menunjukkan potensi sebagai agen antikanker karena menunjukkan nilai LC50  1000 ppm.

Abstract

Azadirachta indica, A.,Juss is a medicinal plant that is used traditionally for some disease, especially its leaves to treat a rheumatic diseases and lower blood uric acid levels. This study was carried out to examine antihyperuricemia activity of ethanol extract, water fraction, ethyl acetat fraction and n-hexane fraction of the A. Indica leaves in male mice of Swiss-Webster strain. Extract and fraction doses used were 250 and 500 mg/kg of body weight and the doses of allopurinol as a standard drug was 13 mg/kg of body weight. The tests were conducted on mice suffering from hyperuricemia induced by potassium oxonat at adose of 300 mg/kg of body weight intraperitoneally and chicken liver juice orally. Measurement of blood uric acid levels were performed using an Easy Touch® every hour for 4 hours after being given test preparations. The results showed that the ethanol extract and the fractions lowered blood uric acid levels in the same way as allopurinol did. The n-hexane fractions at the all doses showed the highest activity, followed by ethanol fraction, and water fraction at the dose of 250 mg/kg at the 4th hour. These results illustrated that the A. Indica leaves might be potential to be used as antihyperuricemia. The active compounds which possibly reduce blood uric acid levels in mice are believed to be flavonoid or polyphenolic compounds because they are reported to be able to inhibit the action of the xanthine oxidase enzyme that converts purines into uric acid. Then, the most active fraction, n-hexane fraction, was tested for its inhibitory activity on xanthine oxidase enzyme to determine its mechanism of action. The results showed that the n-hexane fraction, like allopurinol, inhibited uric acid biosynthesis through inhibiting the activity of xanthine oxidase enzyme with the IC50 value of 132 μg/mL lower than that of allopurinol IC50 58.35 μg/mL. Key words: A. indica leaves, antihyperuricemia, chicken liver juice, potassium oxonat, xanthine oxidase.
