Abstract

Greenhouse ecosystem with more stable abiotic factors could affect the population and diversity of pests and diseases found on foxtail millet (Setaria italica L. Beauv.) compared to their natural ecosystems. We observed and identified pests and diseases in seven genotypes of foxtail millet namely “Toraja”, “ICERI 5”, “ICERI 6”, “Botok 4”, “Botok 10”, “Mauliru”, and “Hambapraing”; these activities are important for the formulation of appropriate integrated pest management techniques. Using plant samples that were at the end of their vegetative phase, we found white mycelia of Fusarium incarnatum (yellowish-white colonies) and F. verticilloides (violetish-pink colonies) covering the ear-tip of the seeds and developed rapidly leading to seed rotten symptoms in “Toraja”, “ICERI 5”, and “ICERI 6”. The disease severity remained constant after these integrated management techniques were put in place. However, abnormalities in leaves leading to failure of panicle emergence occurred in “Botok 4”, “Botok 10”, “Mauliru”, and “Hambapraing”. These were caused by the fungi Penicillium sp. Identical controlling techniques were applied to this incidence and the disease incidence was reduced. Corn leaf aphids (Rhophalosiphum maidis) and rice mealybugs (Brevenia rehi) were recorded as main insect pests with severe attack. The aphid, R. maidis, colonized the stems and were associated with the sooty mold (Capnodium sp.), resulting in wilting. The mealybugs, B. rehi, colonized the flag leaves resulting in leaf rotting. Insecticide and isolating the attacked plants were used as the controlling techniques. Red-mites (Tetranychus urticae) were also detected as indicated by chlorotic spots on the upper part of the leaves. Acaricide was used to reduce its population. The pests and diseases found in the seven genotypes of foxtail millet are commonly known to occur in Poaceae. In general, greenhouse ecosystem for foxtail millet facilitated planting with high population with lower diversity of pests and diseases compared to the open field planting.

Abstract

Foxtail millet is annual grasses with grains that are smaller than those of sorghum, rice, and wheat, and is considered one of the minor economic crops but with nutritional values similar to other major food crops. The development of millet as major cereal crop is considered challenging due to the low quality of its seeds, and studies have been conducted to improve millet’s seed quality. We conducted this study to determine the harvesting criteria of foxtail millet seeds based on the change in color of the panicles. We also wanted to determine the drying treatment, and evaluate the relationship between the position of the seed on the panicles and the seed viability and vigor. The first experiment was arranged based on a completely randomized design with panicle color as the first factor (green, yellow 75%, and brown) and drying time as the second factor (0 hours, 24 hours, 48 hours, 72 hours). The second experiment was arranged in a completely randomized design with seed position as the main factor (base, middle, tip). The study was conducted on two genotypes of millet, i.e. “BOTOK 4” and “BOTOK 10”. The highest seed quality of “BOTOK 4” and “BOTOK 10” genotypes were obtained when the panicles were brown and dried for 72 hours; the seed chlorophyll content was the lowest and water content was 8.88%, with the highest viability and vigor, i.e., germination rate of 88.3%-90%, maximum growth potential of 92%-95.3%, normal sprout dry weight 596-620 mg, vigor index of 33.6% - 21.6%, and growth rate of 18.2%-17.1% etmal-1. The highest seed viability and vigor in “BOTOK 4” genotypes were obtained from the base position, i.e., 78.7% germination rate, maximum growth potential of 83.7%, vigor index of 56.5%, growth speed of 19.6 etmal-1, normal sprout dry weight of 48 mg, radicle length of 3.3 cm) and “BOTOK 10” genotypes from the middle position with 91.5% germination, maximum growth potential of 97.2%, vigor index of 21.7%, growth speed of 17.0% etmal-1, and normal sprout dry weight 61 mg.

Abstract

Identification of post-harvest physiological deterioration (PPD) tolerance in cassava is crucial, as PPD significantly hampers the cassava tuberous root industry by shortening storage periods post-harvest and diminishing product quality. Characteristics linked to PPD tolerance encompass high carotenoid levels and low dry matter content (DMC). This study aimed to evaluate the PPD responses of six yellow fleshed and ten white-fleshed cassava tuberous roots and determine the source of PPD tolerance. PPD and DMC assessments were conducted using standard methods at three storage periods: 2, 5, and 10 days after harvest (DAH). The k-means clustering analysis revealed six clusters, each corresponding to distinct PPD symptom patterns and tolerance statuses. Cluster 1, comprising three yellow-fleshed and three white-fleshed cassava cultivars, demonstrated PPD tolerance with minimal symptoms up to 5 DAH. Clusters 2 and 3 exhibited a moderate PPD response with elevated symptoms at 5 DAH, comprising three yellow-fleshed and three white-fleshed cassava cultivars. Clusters 4 to 6 displayed a sensitive response to PPD, showcasing a significant increase in symptoms at 5 and 10 DAH, with four white-fleshed cassava cultivars identified within this cluster. These findings underscored the presence of PPD tolerance in both yellow-fleshed and white-fleshed cassava tuberous roots. The correlation between PPD and DMC was significant only at 2 DAH, displaying a moderate positive correlation. Consequently, this study identified three cultivars, “Carvita-25”, “Manggu”, and ““Ubi Kuning””, with high DMC and low PPD incidence at 5 DAH, suggesting their suitability for further breeding programs.

Abstract

ABSTRACTThe major constraint of rice cultivation in the flood-prone area is the lack of tolerant varieties. A rapid screening method have to be developed under controlled environments, such as a green house, to improve the effectiveness and efficiency of the selection under submergence condition. The aim of this research was to study the correlation beetween selection methods in greenhouse and in the field. This experiment was conducted at a green house of Muara Experimental Station, Bogor in 2011/2012. The experimental designs used were randomized complete design for greenhouse experiment and randomized complete block design for field experiment, with three replicates respectively. There were four sub-experiments in the greenhouse experiment, grouped based on seedling age (10 days and 35 days after seeding) and duration of submergence (10 days and 14 days). Those sub-experiments in the green house were compared with the experiment in the field. The results showed that there was strong correlation between recovery percentage in the green house and percentage recovery in the field. The same correlation indeed happen between recovery percentage in the green house to grain yield. Complete submergence at 10-day-old seedlings in green house could be developed as rapid selection method for submergence tolerant rice.Keywords: aciotic stress, correlation, flooding, Oryza sativa

Abstract

ABSTRACT Drought stress is one of the major limiting factors for plant growth and production. Therefore, the mechanism of drought tolerance is important to be studied. Eggplant (Solanum spp.) is relatively tolerant to drought stress compared to other member of Solanaceae family. The objective of this study was to study the mechanism of drought tolerance in eggplant related to accumulation pattern of proline and polyamines. Seven eggplant accessions were drought stressed for 21 days and rewatered for the following next 7 days. Drought stress treatment significantly decreased the soil water content and reduced plant height of all accessions. Eggplant responded the drought stress by reducing stomatal density and stomatal opening to reduce transpiration rate. Drought stress also induced proline and polyamine (PA) accumulation in the leaf tissues. These accumulations started at 14-days after drought stress period when soil water content less than 20% and reached their peaks at 21 days after drought stress. The highest level of proline (134.70 μmol g-1) and that of putrescine (20.836 ng g-1)  could  be used as indicators for eggplant drought stress condition. Keywords: physiological responses, polyamine (PA), proline, putrescine, stomata

Abstract

ABSTRACTThe limited fertile land and increasing flood intensity as a result of global climate changes become serious constraints in the sustainability of national rice production. Improvement of rice varieties, especially for rice cultivation in swampy area is necessary. The objective of this research were to evaluate responses and productivity of rice genotypes to stagnant flooding stress which occurs on swampy area. A total of 22 genotypes had been tested, included Tapus, IR42, and IR64 as check varieties. Water level at stagnant flooding environment was 50-60 cm and  started from 35 days after planting until harvest. Research was conducted in wet season of 2011/2012 at Babakan Experimental Farm, Bogor Agricultural University. The experiment used randomized block design with three replicates. The results showed that stagnant flooding stress caused an increase in plant height, flowering and maturity date, number of unfilled grain, and stem elongation ability. On the other hand, the number of productive tiller and the number of filled grain decreased. The number of filled grain positively correlated with grain yield (r = 0.74**), but stem elongation ability didn’t have strong correlation with grain yield (r = -0.29). Genotypes IPB107-F-5-1-1 and IPB107-F-82-2-1 showed the highest productivity, 5.47 ton ha-1 and 5.80 ton ha-1,, respectively, at which productivity decreased up to 20% compared to optimum condition. Keywords: basin swampy rice, stagnant flooding

Abstract

ABSTRACTHigh genetic variability is one of important determinants in successful breeding of clonally propagated crops such as cassava. Induced mutation, including mutagenesis using gamma irradiation, is one strategy to increase genetic variability. The objective of this research was to obtain information of lethal doses (LD20-LD50) from five cassava genotypes (Jame-jame, Ratim, UJ-5, Malang-4, and Adira-4), to analyze the variability, and to obtain potentially high yielding cassava mutant candidates. Cuttings of several cassava genotypes were irradiated by 0, 15, 30, 45, and 60 Gy gamma rays. The results showed that genotype Malang-4 had the lowest LD20 (7.53 Gy) and LD50 (18.47 Gy), while genotype Jame-jame had the highest LD20 (24.94 Gy) and LD50 (33.24 Gy). The highest phenotype variability was obtained in the cassava population irradiated by 15-30 Gy gamma rays. The highest phenotype variability was also determined by the source of tissue, i.e., cuttings from the middle stem resulted in the highest phenotype variability. Abstract

The genetic transformation via pollen-tube pathway in jatropha is the first alternative method that was applied in this plant. The objective of the research was to study fruit set and germination of three genotypes of jatropha following direct transformations via pollen-tube pathways. The research was conducted during April 2014 until January 2015 at jatropa’s experimental field, Pasuruan, and at green house of ICABIOGRAD, Bogor. Three genotypes of jatropha i.e., IP3A, IP3P and JcUMM18 were used. In the first experiment, split plot design was used where 3 levels of DNA plasmid concentration as a main plot and 5 levels of stigma-drip time of DNA plasmid as a subplot. In the second experiment randomized block design was used with single factor consisting of 15 combinations of concentration and stigma-drip time of DNA plasmid as treatments and control. The results demonstrated that interaction between concentration and application time of DNA plasmid did not significantly affect fruit and seeds formation of three Jatropha genotypes. Combination of DNA plasmid concentration with time of stigma-drip had significant effect on seed germination rate of IP3A genotype, but not significant on the other variables. The concentration of 0.05-0.5 µg µL-1 and application time of DNA plasmid at 1-10 hours after pollination could be applied on jatropha genetic transformation via pollen-tube pathways.Keywords: DNA plasmid, Jatropha curcas, pCAMBIA1301, pollen-tube, stigma-drip

Abstract

The genetic transformation via pollen-tube pathway is an alternative method to overcome the constraints imposed by genotype specificity in transformation and regeneration in jatropha (Jatropha curcas L.) tissue culture. Therefore, it is necessary to establish important parameters for efficient genetic transformation of jatropha via pollen-tube pathway. The objective of the research was to study the efficiency of direct transformation of jatropha via pollen-tube pathway based on histochemical and molecular analysis. Solution of purified pCAMBIA1301 DNA plasmid carrying a hptII marker gene and a gus reporter gene with concentration level of 0.05, 0.25, 0.50 µg µl-1 were applied to stigma of flowers at 1, 2, 4, 7, 10 h after pollination. Seedling of IP3A, IP3P and JcUMM18 jatropha’s genotypes derived from 15 combination treatments of plasmid DNA concentration and application time, also wild type was subjected to histochemical and molecular analyses. Based on those analyses, the efficiency of transformation via pollen-tube pathway of three jatropha genotypes ranged from 1.5-16.7%. PCR analysis showed that a number of positive plants were identified by using specific primers hptII and gus, i.e. 1-3 and 3-7 plants of the 15 combined treatments, respectively. It indicated that the transformation efficiency via the pollen-tube pathway varied in each jatropha genotype.Keywords: Jatropha curcas L., pCAMBIA1301, plasmid DNA, stigma-drip

Abstract

ABSTRACTIndonesia is one of the largest wheat importers. Suitable environmental condition for wheat needs to be studied if wheat is going to be widely cultivated in Indonesia. The adaptability of wheat grown in various climates and altitudes is one of the important aspects. The objective of this experiment was to study the quantitative and morphological character of wheat grown in middle land (540 m asl) in Bogor, West Java. The experiment was arranged in randomized complete block design with three replications. Wheat genotypes used were three national varieties (Nias, Selayar, and Dewata), four new improved varieties (Guri 3 Agritan, Guri 4 Agritan, Guri 5 Agritan, and Guri 6 Unand), and one introduced genotype (SBD). Data were collected for several quantitative variables and seventeen morphological characters based on UPOV descriptor. The result showed that wheat growth was restricted in Bogor. Genotype determined plant height, leaf number, ear length, root length, number of spikelet, harvest time, seed weight, number of tillers, and plant biomass. Based on ear length, grain weight, and plant biomass, Guri 3 Agritan had the highest production than the other genotypes.Keywords: diversity, genetic relationship, high temperature, introduced genotype, phylogenetic
