Abstract

Generally, cucumber plants have more male flowers than female flowers. The fewer number of female flowers is one of the problems of cucumber production. Flowering regulation can be performed by applying growth regulators and water management. The study aims to determine the effects of paclobutrazol concentrations and watering frequencies on the flowering ratio of cucumber plants. The experiment was arranged in a factorial Randomized Completely Block Design at the Agricultural, Food, and Fishery Extension Center, Seyegan, Yogyakarta. The first factor was the paclobutrazol concentrations consisting of four levels (a control, 0.187 mL.L-1, 0.375 mL.L-1, and 0.562 mL.L-1). Meanwhile, the second factor was the watering frequency consisting of three levels (once a day, once every two days, and once every three days). The observed data were performed using the analysis of variance (ANOVA) and followed with Duncan’s Multiple Range Test at α= 5 %. The paclobutrazol concentrations resulted in a significant effect on all of the observed variables. Meanwhile, watering frequencies showed no significant effect on the male flowers, female flowers and ratio of male to female flowers in cucumber. There was an interaction effect of paclobutrazol concentration and watering frequency on the plant height and flowering age of cucumber plants. The treatment of paclobutrazol at a concentration of 0.375 mL.L-1, decreased of male flowers and increased of female flowers, reducing the ratio of male and female flowers of cucumber plants at various watering frequencies.-1

Abstract

Dalam program perakitan varietas tanaman aglaonema unggul memerlukan beberapa informasi dasar sebagai bahan penentu arah pemuliaan tanaman. Melalui penelitian ini dilakukan karakterisasi berdasarkan morfologi tanaman aglaonema untuk mengetahui karakteristik dari masing-masing tanaman dan mempelajari hubungan kekerabatan genetik antar tanaman aglaonema. Bahan penelitian yaitu 25 tanaman aglaonema antara lain : Pride of Sumatra, Ruby Super Pink, Ruby Kura, Suksom Jaipong, Asri, Nancy,Shinta, Legacy, Tiara, Legacy Putih, Tiara Putih, Kochin Brown, Sexy Pink, Ayu Green, Anjamani, Sartika, Charlie Brown, Snowwhite, Donacarmen, Big Leaf, Krisna Golden, Bidadari, Siam Aurora, Queen of Siam, dan Kanza. Hasil dari karakterisasi morfologi menghasilkan dendrogram dengan koefisien kemiripan sebesar 0,82 terbagi menjadi 2 klaster dan berdasarkan analisis komponen utama (PCA) tanaman aglaonema dapat diklasifikasikan menjadi 3 macam kombinasi warna tunggal, kombinasi dua warna dan kombinasi tiga warna.

Abstract

Produksi kacang hijau di Indonesia pada tahun 2008-2018 cenderung mengalami penurunan. Pada tahun 2018 produksi kacang hijau di Indonesia mencapai 241.334 ton, dan pada tahun 2018 produksi kacang hijau di Indonesia mengalami penurunan menjadi 234.718 ton. Salah satu faktor penting yang mempengaruhi produktivitas kacang hijau yaitu keterbatasan varietas unggul baru, sehingga perlu dilakukan kajian mengenai galur-galur kacang hijau baru sebagai calon varietas unggul baru. Tujuan penelitian yang dilakukan yaitu untuk mengetahui pertumbuhan dan hasil lima aksesi kacang hijau dibandingkan dengan varietas pembanding. Penelitian ini dilaksanakan pada bulan September 2020 – Desember 2020 di Kebun Percobaan Tri Dharma Fakultas Pertanian UGM, Banguntapan, Bantul, Daerah Istimewa Yogyakarta. Penelitian ini menggunakan Rancangan Acak Kelompok Lengkap (RAKL) dengan tiga blok sebagai ulangan. Penelitian ini terdiri dari satu faktor perlakuan yaitu aksesi. Aksesi yang digunakan sebanyak lima (F2, F3.1, F3.2, F3.3, dan F3.4) dan (PK-H, VM-4, dan KTG) sebagai kontrol. Hasil penelitian menunjukkan bahwa pada pertumbuhan vegetatif antara aksesi F2 hingga F3.4 tidak berbeda nyata dengan varietas pembanding yaitu PK-H, VM-4, dan KTG. Pada parameter produktivitas dan indeks panen seluruh aksesi dengan varietas pembanding memiliki nilai yang sama yang menandakan bahwa aksesi F2 hingga F3.4 sudah sama dengan varietas yang sudah banyak dipasarkan dan perlu dikembangkan lebih lanjut.

Abstract

Zinnia elegans Jacq. are found in Indonesia, but in general these plants have a less attractive appearance. Efforts to developed Zinnia elegans Jacq. need to be done so that it can be used as cut flowers besides Chrysanthemum. Method of cultivation have been obtained in producingflower seeds (first year) the next step is testing the quality of the character of the shape of flowers and consumers preference test. It was necessary to be able to ensure that flowers produced have good quality and can have economic value. Experiment was conducted to test consumer preferences for phenotypic characters on Zinnia elegans Jacq. flower plant populations generation M5 and M6 result of X-ray irradiation. Preference test was through questionnaire survey method. Results of the assessment Respondents were then tested using Friedman Test / Two-Way ANOVA by rating (Two-way ANOVA by ranks) with SPSS software. The results showed that the type of pompom flowers was preferred than a single type (single). Respondents were also prefer flowers that are not too high, flower with widediameter, and flowers with more fragrant. Flower color preferences based on analysis of respondents in a row is red, purple, white, and yellow.

Abstract

Colour is an interesting character to be studied in breeding program of ornamental chilli. Colour of plant was affected by the accumulation chlorophyll, carotenoids and anthocyanins that regulated by multiple genes controlling color (gene B, gene cl, gene y + , gene c1 / c2, and gene A). Genes regulating of colour have different dominance degrees thus affecting potential properties inherited in plants. Aim of the research was to determine the alleged dominance of the gene encoding a colour using the colour ratio of female and male parent, and also population of F1 generation. Research materials used are F1 generation and reciprocal accession (A, BR, and CP). The measured parameter was the character of fruit colour using a colour sensor Conica Minotla 400 to get the value of L*a*b* (L: brightness; a: axis extends from green (-a) to red (+a); b: the b axis from blue (- b) to yellow color).Quantitative measurement based on the coordinates L*a* b* showed Chroma and Hue values that describe the color of the fruit. The results showed that the potential value of color brightness ratio of generation F1 and F1R (F1: AxBR, F1R: BRxA) were over dominant. Gene partially dominant shown in cross breeding of accession A and CP (F1: AxCP, F1R: CPxA).

Abstract

Chili is an agricultural commodity used in the field of food and industry, can also be used as an ornamental plant. As an ornamental plant, chilies are able to provide exciting new variations due to the striking color of the fruit. This study was conducted as an attempt to increase the variation genetic of chili especially related to fruit color character. In addition, it is expected to be used to study the pattern of inheritance of the fruit color of chili from the cross. The experiment was conducted in greenhouse of Faculty of Agriculture Gadjah Mada University and the screenhouse of Faculty of Agriculture Veteran National Development University. The research took place from November 2010 until in August 2013. Three introduction chilies were used in this experiment are Pepper Sweet Minimix, Orange Baby and Pepper Hot Razzamatazz and one local cayenne varieties (White Trisula). The results showed that the dominant red color to yellow, brown and orange, except on the crossing MX2 × RWT. The color of ripe fruit is controlled by two loci, although in some crosses shows cytoplasmic inheritance.

Abstract

Orchids (Orchidaceae) are ornamental plants known for their high aesthetic value attributed to the shapes, colours, and fragrances of their flowers. Two types of hybrid orchids with attractive flowers, namely the Phalaenopsis 'OX Queen' orchid and the Dendrobium 'Cheddi Jagan' boast attractive flowers were used in this research, because of the beauty of its flower colour. The objective of this research is to characterise the morphology of flower colour and CHS (Chalcone Synthase) gene content that induces flower colour. The method used in this research analyzing the flower’s colour by using the RHS (Royal Horticultural Society) colour chart and molecular analysis by DNA genomic isolation and PCR amplification of gDNA for CHS gene specific primers. The results showed that purple colour is observed through the RHS, with P. 'OX Queen' coded as Deep Purple Pink (N73A) and D. 'Cheddi Jagan' coded as Strong Reddish Purple (N72C). The CHS gene can be amplified in P. ’OX Queen’ 1,287 bp and D. ’Cheddi jagan’ 3,731 bp. In both orchids, the results of amplification showed CHS motifs with conserved domains PLN03172 and PLN03170. The research results show that there is a significant difference in the morphology of the flowers of orchids. Purple colour is observed through the RHS, with P. 'OX Queen' coded as N73A and D. 'Cheddi Jagan' coded as N73C. The results showed that gDNA can be isolated by using CTAB method according to Murray and Thomson, and the CHS gene can be amplified by using CHS primers, resulting 1200 bp of P. 'OX Queen' and 2500 bp for D. 'Cheddi Jagan'. Through this study, preliminary data is expected to be obtained for future research, which is the formation of variegated flowers through editing the CRISPR/Cas9 genome in the CHS gene. This research is intended to support further studies on the formation of variegated flower patterns in P. 'OX Queen' and D. 'Cheddi Jagan’, focusing on the CHS gene using CRISPR/Cas9 technique.

Abstract

Semangka triploid merupakan hasil pemuliaan tanaman dengan melakukan penggandaan kromosom pada salah satu tetuanya. Semangka triploid umumnya bersifat tidak memiliki biji namun masih sering dijumpai adanya peristiwa terbentuknya putatif biji normal dengan kulit biji yang berwarna hitam. Penelitian ini bertujuan untuk mengidentifikasi keseragaman genetik tanaman tetua betina, tetua jantan, dan tanaman F1 berdasarkan level ploidi dan karakter morfologi. Penelitian ini dilaksanakan di Dusun Maron, Desa Pujon Lor, Kecamatan Pujon, Kabupaten Malang mulai Agustus 2023 hingga Januari 2024 dengan menggunakan 1 galur tetua betina, 9 galur tetua jantan dan9 galur tanaman F1 sebagai bahan penelitian. Pengujian dilakukan dengan mengamati level ploidi menggunakan flowcytometry serta karakter kualitatif dan karakter kuantitatif. Data karakter kuantitatif dianalisis menggunakan analisis menggunakan uji T dengan taraf α=5%. Hasil penelitian menunjukkan bahwa galur tanaman F1 menunjukkan adanya variasi level ploidi yaitu triploid (3x) dan tetraploid (4x). Hal ini disebabkan oleh galur betina yang digunakan untuk menghasilkan F1 triploid menunjukkan adanya variasi karakter kualitatif sehingga secara genetic belum homozigot.

Abstract

Pepaya gunung (Vasconcellea pubescens A.DC.) merupakan salah satu spesies anggota Suku Caricaceae yang memiliki nilai ekonomi dan agronomi, terutama karena kemampuannya tumbuh di dataran tinggi dan potensinya sebagai bahan baku obat. Tanaman ini diketahui memiliki tiga jenis kelamin, yaitu jantan dioecious, betina dioecious, dan monoecious. Stomata berperan dalam proses fisiologis seperti fotosintesis, transpirasi, dan pertukaran gas. Variasi dalam karakter stomata, seperti bentuk dan ukurannya menunjukkan diferensiasi fisiologis antar individu. Penelitian ini bertujuan untuk mengkaji karakteristik stomata pada tiga jenis kelamin pepaya gunung menggunakan Scanning Electron Microscope (SEM). Analisis dilakukan untuk mengidentifikasi perbedaan karakter stomata yang meliputi bentuk dan ukuran pada permukaan daun tiap-tiap jenis kelamin. Sampel daun diambil dari tanaman yang telah teridentifikasi jenis kelaminnya, kemudian dipreparasi dan diamati menggunakan SEM untuk memperoleh citra dengan resolusi tinggi. Hasil pengamatan menunjukkan adanya variasi pada karakteristik stomata antar jenis kelamin. Tanaman jantan dioecious cenderung memiliki stomata dengan ukuran paling besar dengan rerata 41,09 ± 18,50 x 29,41 ± 16,34 µm sedangkan tanaman betina dioecious menunjukkan ukuran stomata yang lebih kecil dengan rerata 28,08 ± 39,16 x 19,44 ± 26,57 µm. Adapun tanaman monoecious menampilkan ciri-ciri morfologi stomata yang bersifat intermediat dengan rerata 24,06 ± 8,22 x 20,30 ± 8,81 µm. Temuan tersebut memberikan pemahaman mikromorfologis terkait diferensiasi kelamin pada V. pubescens serta potensi penggunaan karakter stomata sebagai penanda awal identifikasi jenis kelamin tanaman

Abstract

Indonesia has been trying to devolop tropical wheat in regard to decrease import of wheat. Therofore, some tropical wheat varieties have been introduced into Indonesia. This research aims to evaluate the yield stability of some varieties which introduced into Indonesia. Five varieties of wheat , nantely Dll/R 162, DlyR 195, HD 2189, HUII/ 234, and Nias as check variety, were grown at three different environments. Field experiment were arranged according to Randonrizecl Completely Block Design procedure with three replications. The result showed that based on the stability parameter of Eberhart and Russel, HUII/ 234 is stable cultivar in yield and yield components, although its yield is Erite anall. The other two arltivars, nantely Dll/ll 195 and Nias tend to adapt in favorable environment. Meanwhile, HD 2lB9 and DIYR 162 are suitable varieties for less favorable environment,
