Abstract

Intestinal bacterial species play an essential role in the duck body metabolism and productivity. They are affected by various factors, such as heavy metal contamination. This research aims to study metal emission levels and their correlation with the composition of intestinal bacteria. A total of twenty-five duck samples (Anas platyrhynchos L.) were obtained from Semarang, Temanggung, Magelang, Pati, and Salatiga. Five g of intestinal substances were employed in the isolation 16S rRNA gene V3-V4 region. Data sequence in fastqc format was then analyzed using QIIME. Also, the heavy metals in water, feed, and meat samples were analyzed using inductively coupled plasm-optical emission spectroscopy (ICP-OES). The highest heavy metal contaminant was Hg, with a total concentration reaching 5.55±1.79 ppm of input and 4.42±0.80 ppm in meat. The bacterial composition comprises the four highest phyla, sequentially including Firmicutes, Actinobacteria, Proteobacteria, and Bacteriodetes. The relationship between heavy metals and intestinal bacteria shows a significant value with a coefficient of >0.90. Therefore, there was a correlation between heavy metals and intestinal bacteria composition, suggesting that bacteria are likely to protect ducks from the adverse effects of heavy metals in water and feed.

Abstract

The Women Farmer Group in Semarang was established in 2013, and it has been developing small businesses in the processing of local fresh milk into various snack products. However, the selling value has not reached the optimum results since it faces a problem in packagingquality. Therefore, as one of the goals of Tridarma of Indonesian higher education which have directed the university to care about problems in society, an accompaniment to the business was carried out for improving the quality of product packaging. There were three activities conducted, i.e., training; re-designing; and monitoring. The results of the community service showed that the program was highly effective to enhance the understanding of Women Farmer Group members on the importance of product packaging as a strategy of marketing proved by the results of the average posttest score at 80.55. Product testing to consumers shows that the new packaging could increase consumer buying interest in comparison to the old one. Also, with the new packaging, the non-preservative snacks could have a longer storability compared tothe old packaging. In this community service activity, an empowerment model can be obtained and can be further developed.

Abstract

Tempe known as a traditional fermented food originated from Indonesia. It has a unique flavour and texture. It also contains high protein and usually serves to substitute meat, fish, or egg as a complement to rice. The manufacture process of Tempe is quite complex and mostly, the traditional process has not employed the hygienic standard. In the process of Tempe making, there are two critical stages of the whole process; i.e. soaking of soybeans and solid state fermentation by Rhizopus sp. During the process, foodborne pathogen bacteria such as Escherichia coli could contaminate the product of Tempe. The bacterial contamination could be revealed through culture dependent methods which is costly, laborious, and time consuming. Therefore, the culture-independent method such as polymerase chain reaction using a specific primer could be applied to detect target microorganism to save time and labour. In this study, thirty-one Tempe samples collected from different manufacturers in Semarang, Central Java, Indonesia were analysed by PCR. In order to obtain the bacterial genomic DNA, a modified Chelex 100-Microwave method was employed. The results of DNA extraction showed that the method was an applicable method. It gave high quantity and quality of DNA; therefore, it could be applied in the PCR reaction. The DNA samples were employed in PCR for detection of Escherichia coli using Ecoli706F/R. It was found that 27 out of 31 samples were detected having Escherichia coli contamination showed by the presence of the amplified product size 706 bp. The application of this method could significantly reduce costs and time of analysis in the laboratory. Further response after E. coli were detected could be employed, including investigation of the critical factors in Tempe manufacturing process which allowed E. coli contamination.

Abstract

This study aims to analyze the religious character and logical thinking skills of students after using solar systems teaching materials integrated with Islamic Science. The type of research used is descriptive analysis with a Post-Test-Only with Non-equivalent Groups research design. The research data were obtained from observation sheets, questionnaire sheets, and posttest. The religious character and students’ logical thinking skills of the experiment class students are better than the control class. Students in the experiment class were superior to the control class. The effect of the correlation of the use of teaching materials on the religious character of the students obtained 79.21%, while the effect of the correlation of the use of teaching materials on students’ logical thinking skills was 38.44%. The study concludes that students’ religious character after using solar system teaching materials integrated Islamic science is dominant in the “very good” category and students’ logical thinking skills after the use of solar system teaching materials integrated Islamic science is dominant in the high category.

Abstract

This research aims to determine the feasibility and characteristics of the guided inquiry learning model assisted by a simple terrarium on students' concept understanding and environmental awareness in the global warming topic of study. The research and development (RnD) methodby analysis, design, development, implementation, and evaluation (ADDIE) was performed. The data collection instruments used were observation sheets, expert validation sheets, and questions. The teaching and learning instruments were declared feasible and ready for application with a few revisions based on the validation results. The n-gain analysis of increasing students' conceptual understanding was in the high category at 0.76. The percentage of observations regarding environmental awareness increased at each successive meeting. The first meeting was 39.06%, the second meeting was 63.12%, and the third meeting was 89.37%. In conclusion, the guided inquiry learning model assisted by a simple terrarium is feasible to improve students' conceptual understanding and environmental virtue ethics.

Abstract

The Mx gene is a potential candidate as a genetic resistance marker to the avian influenza virus in chickens. The purpose of this study is to identify the potential of Mx gene as a marker of resistance to avian influenza viruses in various breeds of chicken. The study used an exploratory design using three common chicken breeds. The Kampong chicken, the Hy-Line Brown laying hen strain, and the White Leghorn broiler strain were collected from the local farms, and blood samples were drawn from each. The DNA of each sample was amplified by a pair of Mx gene primers (NE-F2/F and NE-R2/R). The polymerase chain reaction (PCR) product of Mx gene fragment was cut using restriction enzymes RsaI to determine the genotype of the Mx gene. The PCR-RFLP analysis in this study showed that 100% of the broilers were GG genotype. In laying hens, 80% were AG genotype, 10% GG genotype, and 10% of the sample was not successfully analyzed. In the Kampong chicken, 68.4% were GG genotype, and 31.6% were AG genotype. The results showed that DNA mutation identification can be applied to determine the genotype chicken against virus. The genotypes of Mx genes in chickens indicates avian influenza virus resistance; whereas, the chicken phenotype does not suggest their level of resistance.

Abstract

In this study, twenty local durian accessions obtained from Central Java in situ collection were characterized using the morphological characteristics and the restriction patterns which generated from the region spanning the internal transcribed spacers ITS LEU and ITS 4. Morphological characteristics of durian leaf, stem, tree, and fruit showed variations for the different accessions, whereas polymerase chain reaction (PCR) products of ribosomal DNA region showed a low length of variation. The size of the PCR products and the restriction analyses with the restriction endonucleases Bsp1431yielded a restriction pattern for each accessions. The results of this study can be utilized by local durian farmers as a preliminary reference for durian propagation. The data obtained need to be supported by further research using the other molecular markers to obtain more accurate data. Abstract

Phylogenetic or population genetic studies have been focused on finding genetic variations in mitochondrial DNA (mtDNA) because mutations in this region contained five times higher than other genetic materials. The aim of this research was to generate PCR-RFLP D-loop mtDNA profiles of some Indonesian domestic waterfowl. The samples were 35 local ducks, 5 muscovy duck, and 5 geese. These local ducks included on seven types of Javanese ducks in Central Java, i.e. Magelang duck, Peking duck, Pengging duck, Tegal Branjangan duck, Tegal Jarakan duck, Tegal Blorong duck, and Tegal Lemahan duck. PCR technique was used to amplify the D-loop genes, and RFLP analysis of the D-loop was performed with AluIand RsaI. The results show that muscovy duck has no restriction sites in D-loop gene. Moreover, goose has a restriction site of AluI in D-loop gene at 550 bp and 187 bp; and it has no restriction site of RsaI. The analysis of RsaI-RFLP in mtDNA D-loop region of ducks resulted in 1 restriction fragment pattern which can be applied in the identification of duck to differentiate it from goose and muscovy duck. PCR-RFLP analysis duck D-loop mtDNA using AluI and RsaI restriction enzyme resulted in 3 combinations of restriction fragment pattern shown in 3 haplotypes (A, B, and C). Genetic diversity of Indonesian domestic waterfowl population could be identified by using PCR-RFLP analysis on mtDNA D-loop region. So far, there are few results of research on Indonesian domestic waterfowl genetic characteristics based on PCR-RFLP mtDNA D-loop marker. Also, this research has a good contribution on genetic characterization of local duck. The characterization is an important aspect to maintain breed quality and manage the genetic resources of Indonesian germplasm and in facilitating the breeding program.

Abstract

Teknik PCR mempunyai kemampuan yang sensitif untuk deteksi keberadaan daging babi di dalam daging segar maupun produk olahan yang dicampur dengan bahan lain. Tujuan penelitian ini untuk mengetahui apakah produk bakso yang dijajakan di pusat kota Salatiga mengandung daging babi. Teknik stratified random sampling digunakan untuk mengambil sampel bakso yang dijajakan 13 warung bakso dari 25 warung bakso yang tersebar di pusat Kota Salatiga. Isolasi dan purifikasi DNA sampel bakso, daging sapi, dan daging babi menggunakan metode isolasi DNA jaringan hewan. DNA hasil isolasi dilanjutkan proses PCR menggunakan primer p14 untuk mengamplifikasi lokus PRE-1 pada genom babi. Proses amplifikasi DNA dengan program denaturasi awal pada suhu 93 °C selama 2 menit, diikuti 45 siklus terdiri atas denaturasi 93 °C selama 1 menit, annealing 62 °C selama 30 detik, ekstensi 72 °C selama 1 menit, kemudian diakhiri ekstensi 72 °C selama 2 menit. Produk PCR yang diharapkan muncul berukuran 481bp. Hasil elektroforesis gel agarose 1,2% pada produk PCR menunjukkan adanya pita DNA spesifik berukuran 481 bp pada daging babi dan sampel bakso nomor tiga belas, sehingga disimpulkan warung bakso nomor tiga belas produk baksonya mengandung daging babi.PCR technique has the ability to be sensitive to the detection of the presence of pork in fresh meat and processed products are mixed with other materials. The aim of this research to determine whether the product meatballs are sold in downtown Salatiga containing pork. Stratified random sampling technique is used to take samples of meatballs stall which sold 13 of the 25 meatballs stalls in the Salatiga City centre. Isolation and purification of DNA samples of meatballs, beef, and pork using DNA isolation method of animal tissue. DNA isolation results continue the process of PCR using primers to amplify p14 locus PRE-1 in the pig genome. DNA amplification process with initial denaturation program at a temperature of 93 °C for 2 min, followed by 45 cycles consisting of denaturation 93 °C for 1 min, annealing 62 °C for 30 seconds, extension 72 °C for 1 minute, then topped extension 72 °C for 2 minutes. PCR products were expected to appear sized 481bp. Results of a 1.2% agarose gel electrophoresis of PCR products indicate a specific DNA band sized 481 bp on pork and meatball sample number thirteen, so it concluded meatball stall number thirteen baksonya products containing pork.

Abstract

Crude proteases were extracted from Euphorbiaceae family, i.e. E. milii var imperata, E. trigona, and E. maculata. Among those three crude proteases, the activity of protease from E. trigona was the highest (812.50 U/ml), whereas E. milii and E. maculata crude proteases activity were 298.60 U/ml and 95.80 U/ml, respectively. E. maculata protein concentration was the highest among those three crude enzymes (1.206 mg/ml). The optimum pH and temperature of the enzymes were pH 7.0, pH 6.0, pH 6.5 and 60 °C, 50 °C, and 50 °C, respectively. Crude protease from E. milii var imperata, E. trigona, and E. maculata retained proteolytic activity over a wide range of pH (5.0–9.0) and temperature (up to 65 °C) with casein as substrate. All crude proteases showed milk clotting activity ranged from 0.58 U/ml to 1.01 U/ml. Thus, these crude proteases are potential to be applied in dairy industries. However, further study on enzyme purification and characterization are necessary to obtain high purity of proteases before its application.Protease kasar berhasil diekstrak dari tanaman family Euphorbiaceae, yaitu E. milii var imperata, E. trigona, dan E. maculata. Diantara ketiga protease tersebut, aktivitas protease tertinggi diperoleh dari E. trigona (812,50 U/ml), sedangkan aktivitas protease dari E. milii dan E. maculata adalah 298,60 U/ml dan 95,80 U/ml, berturut-turut. Konsentrasi total protein tertinggi terdapat pada protease kasar E. maculata (1,206 mg/ml). pH dan suhu optimum ketiga enzim tersebut adalah pH 7.0, pH 6.0, pH 6.5 dan suhu 60 °C, 50 °C, and 50 °C, berturut-turut. Protease kasar dari E. milii var imperata, E. trigona, dan E. maculata menunjukkan aktivitas proteolitik pada rentang pH 5.0–9.0 dan rentang suhu sampai 65 °C menggunakan kasein sebagai substrat. Semua protease kasar menunjukkan aktivitas penggumpalan susu dengan rentang dari 0,58 U/ml sampai 1,01 U/ml. Berdasarkan hasil yang diperoleh, protease kasar dari ketiga jenis tanaman ini berpotensi untuk diaplikasikan dalam industri olahan susu. Meskipun demikian, studi lanjut mengenai purifikasi dan karakterisasi sangat diperlukan untuk memperoleh protease murni sebelum aplikasi dalam industri makanan, khususnya pada industri olahan susu.
