Abstract

Abstract

Toona sureni (Blume) Merr., known as Indonesian mahogany, is valued for its wood, while its leaves are used as vegetables and animal feed. However, its medicinal potential remains underexplored. This study evaluated the antibacterial and tyrosinase inhibitory activities of T. sureni leaf extracts from 16-, 27-, and 34-year-old trees. Extraction was performed by maceration using methanol, ethyl acetate, and n-hexane for antibacterial testing; and 30, 70, and 96% ethanol for tyrosinase inhibition. Total alkaloid content was determined using the bromocresol green method, while antibacterial activity was assessed by disc diffusion and MIC, and tyrosinase inhibition by spectrophotometry using kojic acid as a control. It revealed a 3.83% alkaloid content in the 70% ethanolic leaves extract of the 34-year-old tree. The 96% ethanolic extract from 16-year-old leaves exhibited the highest tyrosinase inhibitory activity at 86.66% with an IC50 value of 53.71±1.04 μg/mL. Antibacterial tests revealed that the methanol extract of 16-year-old trees inhibited Staphylococcus aureus (17.73 mm) and Propionibacterium acnes (15.80 mm) at 10,000 μg/mL, with MIC of 1000 and 500 μg/mL, respectively. These results suggest that the extract of 16-year-old tree has both antibacterial and tyrosinase inhibitory effects, with greater potential for cosmetic use, particularly in anti-acne and skin-brightening products.

Abstract

Falah S, Sari NM, Hidayat A. 2018. Decolorization of Remazol Brilliant Blue R by laccase of newly isolated Leiotrametes flavida Strain ZUL62 from Bangka Heath Forest, Indonesia. Biodiversitas 19: 633-639. The use of microbial enzymes seems to be an appropriate approach to neutralize synthetic dyes, providing an eco-friendly and cost-competitive alternative solution for treating industrial effluents. The objective of this research was to select the most potential fungal isolate with high laccase activity to decolorize Remazol Brilliant Blue R (RBBR) synthetic dyes. The isolation of fungal body and sampling of wood decay specimens were conducted in six different locations in heath (kerangas) forest of Bangka Belitung, Indonesia. In the present study, a total of 13 isolates were screened by using indicator plate method and for their enzyme activity. The preliminary screening was done to screen ligninolytic fungi. Then the primary screening using various indicator compounds was conducted to select laccases-producing fungi. The enzyme activity assay was performed to select fungal isolate with the highest activity. We found that the most potential fungi belonged to Leiotrametes flavida Strain ZUL62, which had been confirmed by molecular identification using 5.8S rDNA/ITS analysis. In addition, the laccase from Leiotrametes flavida Strain ZUL62 could decolorize RBBR, exhibiting a high rate of decolorization rate of 62% without any mediator within 24 h of incubation. To our best of knowledge, this study represented the first report about Leiotrametes flavida Strain ZUL62 and its potential laccase enzyme for dyes effluent treatment.

Abstract

Elfirta RR, Falah S, Andrianto D, Lastini T. 2018. Identification of active compounds and antifungal activity of Toona sinensis leaves fractions against wood rot fungi. Biodiversitas 19: 1313-1318. Inhibitory activities of surian or toon (Toona sinensis) leaves fraction origin Sumedang, West Java, Indonesia against wood rot fungi (Ganoderma boninense IPBCC 10.658, Trametes versicolor InaCC F200, and Phanerochaete chrysosporium IPBCC 93.259) were studied. The samples were macerated using methanol and acetone. The results of brine shrimp lethality test (BSLT) showed that the methanol extract had the highest cytotoxicity of LC50 at 29.76 ?g/mL and inhibit the growth of G. boninense (27.78%), T. versicolor (79.26%) and P. chrysosporium (81.11%). The methanol extract was then subsequently fractionated using n-hexane, diethyl ether, and ethyl acetate respectively. The diethyl ether fraction was found to have the highest inhibitory activity against T. versicolor (46.30%) and P. chrysosporium (81.11%). This fraction was further separated using column chromatography and analyzed using thin layer chromatography (TLC), which gave six fractions. Antifungal test exhibited that fraction 5 had the highest antifungal properties against T. versicolor (74.07%) and P. chrysosporium (80.37%). Inhibition of P. chrysosporium resulted in abnormal growth of hyphae morphology as indicated by changes in its growth direction and excessive hyphae branching. Additionally, the results of LC-MS/MS experiment indicated toon leaves fraction 5 which contains N-[2- (D-Glucopyranosyloxy)ethyl]-2-hydroxy-N-[2hydroxy3 (octadecyloxy) propyl] butanamide dan Brucine compounds that were regarded as the antifungal compounds.

Abstract

Febriyani E, Falah S, Andrianto D, Lastini T. 2018. Identification of active compounds and anti-acne activity from extracts and fractions of surian (Toona sinensis) leaves planted in Sumedang, West Java, Indonesia. Biodiversitas 19: 1406-1412. Surian or toon (Toona sinensis Merr) leave is one of the herbs containing many bioactive compounds as potential sources of anti-acne agents. The aim of this work is to identify and evaluate the anti-acne activity of Surian leaves extract and fractions against acne bacteria. Crude extracts of Surian leaves from various solvents (methanol, ethanol, and ethyl acetate) were screened for their antimicrobial activity using disc diffusion. The results showed that ethyl acetate extract demonstrated the highest inhibition against Propionibacterium acne (49.93%), Staphylococcus epidermidis (57.14%), Staphylococcus aureus (18.93%), and Eschericia coli (82.03%). The results of Brine Shrimp Lethality Test (BSLT) showed that the methanol extract had the highest cytotoxicity of LC50 at 29.76 ?g/mL. The extract was then fractionated using column chromatography, resulted in 6 fractions. The fraction 3 showed the best antibacterial effect against P. acne (28.27%), S. epidermidis (7.38%) and S. aureus (10.38%). Minimum inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) test were carried out using liquid dilution, whereas dimethyl sulfoxide (DMSO) and Clindamycin were used as a negative and positive control, respectively. The MIC and MBC test assays of fraction 3 varied for different bacteria P. acne bacteria (5000 and 5000 ppm), S. aureus (1250 and 10000 ppm), E. coli (2500 and 5000 ppm), while for S. epidermidis the fraction 3 was able to inhibit with MIC value of 5000 ppm. Based on microscopic observation using Scanning Electron Microscope (SEM), the fraction 3 at the dose of 5000 ppm could promote the cellular destruction of P. acne. Further experiment using Liquid Chromatography-Tendem Mass Spectrometry (LC-MS/MS) showed various compounds associated with the anti-acne activity of the fraction 3, such as pheophorbide a, pheophytin a and dihomo-?-linolenic acid.

Abstract. Faulina SA, Irfani M, Falah S, Hidayat A, Iswanto AH. 2020. Short Communication: The higher laccase enzyme producer, Cerrena sp. BMd. TA.1, isolated from Gunung Rinjani National Park, West Nusa Tenggara, Indonesia. Biodiversitas 21: 3837-3842. Lies in the Wallace line, Gunung Rinjani National Park (GRNP) offers unique biodiversity, fungi included. Fungal enzymes have been unceasingly searched and studied for various applications, particularly for biodegradation. Fungal laccase enzyme showed prospective environmental-friendly approach in treating industrial effluent, remazol brilliant blue R (RBBR) which is used as a synthetic dye. This study aimed to explore the laccase-producing fungi from the GNRP, as well as investigate their ability in decolorizing RBBR. The study discovered that the most prospective fungi isolate, molecularly identified as Cerrena sp. BMd.TA.1, produced a high level of laccase (> 2300 U mL-1) and manganese peroxidase (MnP, 300 U mL-1). In the application of this isolate, the laccase showed as predominant enzyme in RBBR decolorization process and the RBBR could be decolorized more than 80% at 24 h reaction. It suggested that Cerrena sp BMd.TA.1 isolate is highly potential as laccase enzyme producer and may be considered for further investigations in its applications for biodegradation, especially of dyes effluent.
