Abstract

Wound healing disorder are often found in people with diabetes mellitus. Low Intensity Pulse Ultrasound (LIPUS) therapy was developed to accelerate the wound healing process. The purpose of this study was to determine the effect of LIPUS on wound closure and angiogenesis after skin excision in rats model diabetes mellitus type 2.Twenty-four Sprague dawley rats that match the inclusion criteria were made into type 2 diabetes mellitus by injecting Nicotinamide and Streptozotocin, then devided into six group: DM3, DM7, DM14, DML3, DML7, DML14, with 4 rats each. All rats were made excision wound with punch biopsy in the back area. DML group receive LIPUS therapy in the wound area (frequency 3 MHz, intensity 0,5 W/cm 2 , duty cycle 20%, duration 3 minutes every day) for three days (DML3), seven days (DML7) and fourteen days (DML14). Wound area was measured and the tissue was staineing with hematoksciline eiosin to observe the density of angiogenesis.Two Way ANOVA used to to analyze the differences in the variable wound area and angiogenesis density, Post Hoc LSD to analyze the differences in variables between groups and the day of observation, showed the area of the wound in the DML was lower than in the DM group significant on day 3 and 7, whereas the density of the angiogenesis in DML group was higher than DM group significant on day 7 and 14. The Pearson test showed a correlation between the wound closure and the angiogenesis density (p = 0,000; r = -0,785). Conclusion: LIPUS therapy accelerate wound closure on day 3 and 7and increase the density of angiogenesis on day 7 and 14.

Abstract

Impaired wound healing is one of the Diabetes mellitus complications. Low-intensity Pulsed Ultrasound (LIPUS) therapy may accelerate the impaired wound healing. The use of LIPUS therapy in the early inflammatory phase can induce mast cell degranulation, and in the proliferative phase it can increase collagen synthesis by fibroblasts. The purpose of this study was to determine the effects of LIPUS therapy on mast cell degranulation and fibroblastexpression in the healing process of punch biopsy wound in rats with type 2 diabetes mellitus. Twenty-four Sprague dawley (n=24) were designed into type 2 diabetes mellitus by injecting Nicotinamide and Streptozotocin, then divided into six groups: diabetes mellitus without LIPUS (DM3, DM7, DM14) and diabetes mellitus with LIPUS (DML3, DML7, DML14), 4 each, and punch biopsy wounds were made on the dorsal skin. The DML group received LIPUS therapy in the wound area (frequency 3 MHz, intensity 0.5 W/cm2, duty cycle 20%, duration 3 minutes every day for 3 days (DML3), 7 days (DML7), and 14 days (DML14). The wounded tissue area was stained with toluidine blue to observe mast cell degranulation and immunohistochemical type HSP-47 to observe fibroblast expression. Two-Way ANOVA and Post Hoc LSD tests were used to determine the differences in mast cell degranulation and fibroblast expression. The results showed that mast cell degranulation and fibroblast expression in the DML group were higher than in the DM group (table 1). Pearson test showed a correlation between mast cell degranulation and fibroblast expression (p=0.00; r= 0.839). LIPUS therapy increases mast cell degranulation and fibroblast expression in type 2 diabetes mellitus rat model. The higher the mast cell degranulation, the higher fibroblast expressions.

Abstract

ABSTRACT  The problem faced during the new normal period of the Covid-19 pandemic for students is to restore interest in learning that has been done at home. Additional efforts are needed to reduce these concerns, such as mentoring for learning outside of school. Community service in the form of learning assistance is carried out with the aim of providing new experiences, information, and knowledge outside of learning at school. The schools participating in the service activities are schools from Cluster VI Tuksono Village, Sentolo, Kulon Progo, Yogyakarta. This activity involves student assistants from the Anatomy Department, Faculty of Veterinary Medicine, Universitas Gadjah Mada with assistance for a certain period. Implementation of online activities providing materials and discussions that have been adapted to the competencies needed in schools. The material given is an introduction to vertebrates macroscopically and microscopically. Students will be divided into several classes and each has an assistant and a co-lecturer. The activity takes place from September to November 2021. The results of the questionnaire show that this learning assistance activity is very interesting (60%) and very useful (69%) for students and teachers in their schools.  Keywords: Animal, Biology Anatomy; Elementary school; Learning; Online   ABSTRAK  Permasalahan yang dihadapi pada masa normal baru pandemi Covid-19 bagi peserta didik adalah mengembalikan minat belajar yang selama ini dilakukan di rumah. Usaha untuk mengurangi keprihatinan tersebut diperlukan seperti pendampingan belajar luar sekolah. Pengabdian kepada masyarakat berupa pendampingan pembelajaran dilaksanakan dengan tujuan memberikan pengalaman, informasi, dan pengetahuan baru di luar pembelajaran di sekolah. Sekolah yang berpartisipasi dalam kegiatan pengabdian adalah sekolah-sekolah dari Gugus VI Desa Tuksono, Sentolo, Kulon Progo, Yogyakarta. Kegiatan ini melibatkan asisten mahasiswa Departemen Anatomi Fakultas Kedokteran Hewan Universitas Gadjah Mada dengan pendampingan selama periode tertentu. Pelaksanaan kegiatan daring pemberian materi dan diskusi yang sudah disesuaikan dengan kompetensi yang dibutuhkan di sekolah. Materi yang diberikan berupa pengenalan vertebrata secara makroskopik dan mikroskopik. Peserta didik akan dibagi menjadi beberapa kelas dan masing-masing memiliki satu asisten dan dosen pendamping. Kegiatan berlangsung dari bulan September sampai November 2021. Hasil kuisioner menunjukkan kegiatan pendampingan pembelajaran ini sangat menarik (60%) dan sangat bermanfaat (69%) bagi para peserta didik dan guru di sekolahnya. Abstract

Catecholamine mesolimbic pathway (CMP) is a dopamine pathways of the brain that derived from cell bodies in the ventral tegmental area (VTA) to the limbic area i.e. nucleus accumbens, amygdala and hippocampus. In normal conditions, CMP plays a role as controls of motor activity, motivation, emotional and cognitive. Bats are flying mammals which can be potential as natural reservoir of rabies. One of the symptoms of animal rabies is impaired in emotional control that related with limbic system. Kalong kapauk (Pteropus vampyrus) is one of the species of bats that distributed in south Sumatra, Java, Borneo, and spread out to the island of Timor, East Nusa Tenggara. The aim of this research is studying the anatomical structures in CMP of kalong kapauk (Pteropus vampyrus) from Timor island. Four kalong kapauk (Pteropus vampyrus) from Soe-Timor island, East Nusa Tenggara was anaesthetized by using ketamine (20 mg/kg bw) and xylazin (2 mg/kg bw). In deep anesthesia condition, animals were perfused by using physiological saline and after the blood were removed well, the physiological saline were changed to 10% buffered formalin as a fixative. The brain were removed from the cranium, dissected midsagital and processed for histology by paraffin method. The brains were cut in 12 μm thickness and then it stained by using cresyl echt violet and immunohistochemistry by tyrosine hydroxilase antibody. The sections were examined for shape and size of neurons in the VTA and their axonal pathways by light microscope and were documented using a digital camera. The results were analyzed descriptively. The results shows that neurons in the VTA are bipolar and multipolar in shape with the size of 10-32 μm (mean 20.31 ± 4.40 μm) and densities are 15.33 ± 5.71 cells/0,116 mm2. The area of nucleus accumbens, amygdala, and cornua ammonis 3 of hippocampal are TH immunoreactive as the axon terminal. The conclusion of this study is there are catecholaminergic neurons in the VTA that made an area limbic pathways i.e. nucleus accumbens, amygdala, and cornua ammonis 3 of hippocampal.

Abstract

Pteropus vampyrus is one of the animal species that plays an important role in the ecosystem by spreading the seeds of the edible fruits. Feed variations on each animal greatly affect the digestive tract. This study was aimed to determine the anatomical structure of the large intestine. Five adult large flying foxes weighing 500-800 g were used as the sample in this study. Large intestines were embedded in paraffin and stained with H & E method. H & E staining resulted in the presence of intestinal villi on colon and rectum.

Abstract

Objective: In Parkinson's disease (PD) patients, there is a disruption in the function of catecholaminergic neurons in Enteric nervous system (SSE) with some symtoms: constipations and diarrhea. Paraquat dichloride (PQ) is a neurotoxic herbicide which is thought to induce PD. This study aims to study the histological features of neurons in the enteric nervous system of small and large intestines injected with PQ.Methods: Ten rats were divided into 2 groups of 5 each. The control group was injected with distilled water and the treatment group was injected with PQ 7 mg/kg BW. The injection was given intraperitoneally, twice a week for 3 weeks with a volume of 1 ml/injection. Small intestine and large intestine were collected and processed for histological preparations in paraffin incisions, then stained with cresyl violet and immunohistochemistry using tyrosine hydroxylase antibody as a marker of catecholaminergic neurons. Intestinal histological preparations were observed under light microscope and analyzed descriptively.Results: Neurons in the small intestine and large intestine of normal group rats were observed normal, while in the treatment group some neurons were normal, but some of them became degeneration in the form of chromatolysis, also necrosis which was characterized by damage of cell membranes, karyolysis, loss of most of the Nissl bodies, and decreased numbers of catecholaminergic neurons.Conclusions: Paraquat dichloride cause changes in enteric nervous system’s neuron structures in the form of degeneration, necrosis, and a decrease in the number of catecholaminergic neurons in the small intestine and large intestine.

Abstract

Diabetes mellitus is a disease related to hyperglycemia and insulin resistance that can lead to the outcome of chronic liver diseases such as nonalcoholic fatty liver disease (NAFLD). Red betel leaves are known as traditional plants that have anti-hyperglycemic potential. This study aimed to investigate the effect of ethanolic extract of red betel leaves nanoparticle (RbL-Nps) on the liver and hepatic insulin receptor’s (INSR) expression in diabetic rats. Thirty rats were included in this study and further divided into five groups containing six rats each. Group I (GI) comprised of the normal rats; while group II (GII), III (GIII), IV (GIV) and V (GV) comprised of diabetic rats induced by streptozotocin (STZ) at dose of 45 mg/kg bw and nicotinamide (NA) at dose of 110 mg/kg bw, intraperitoneally. Group I and II were treated with 0,5% Na-CMC orally.  Group III, IV and V were given the oral administration of RbL-Nps at the doses 30, 60, and 90 mg/kg bw diluted in 0,5% Na-CMC, respectively. All groups were treated once daily and subsequently euthanized after 28 days. Liver tissues were collected for immunohistochemistry method to see the INSR expression and haematoxylin-eosin (HE) staining. Result in this study revealed that INSR expression on the GI, GIII and GIV were significantly higher compared to that on the GII (p < 0.05). On the other hand, there were no significant differences on the INSR expression between GV and GII (p > 0.05). Histologically, liver tissues retrieved from GII showed severe vacuolic and necrotic hepatocytes with dilatated sinusoid. Mild vacuolic and necrotic hepatocytes were observed from GV. There were no pathological changes observed in the liver tissues retrieved from GI, GII, and GIV. We concluded that RbL-Nps improved the liver condition of diabetic rats at doses of 30 and 60 mg/kg bw, but not at doses of 90 mg/kg bw.

Abstract

Objective: This study marked histological features of small intestine neurons in the type-2 diabetes mellitus (T2DM) rat model treated with ethanolic extract red betel leaf nanoparticle (EERbLNp)Methods: Thirty male Wistar rats were allotted into five groups (six rats each). Group I is nondiabetic control; group II is streptozotocin-nicotinamide (STZ-NA)-induced T2DM; group III-V are the STZ-NA-induced T2DM treated daily per oral with EERbL-Np at the doses 30, 60, and 90 mg/kg, respectively, within 28days. The duodenum, jejunum, and ileum were collected for routine histological staining and cresyl violet special staining to evaluate the feature of neurons in the Meissner (submucosa) and Auerbach (muscular) plexus. Descriptive statistical analysis ANOVA and Tukey HSD were used to compare neuron indexes among groups.Results: The necrotic neuron index in the duodenal and ileal Auerbach plexus, including the degenerative neuron index in the jejunal Auerbach plexus, were significantly decreased with the EERbL-Np treatment at the dosages 60 and 90mg/kg.Conclusions: The 60 mg/kg EERbL-Np administration in the T2DM model may provide a neuroprotectant candidate in duodenal and ileal neuropathy. Dosage of 90mg/kg EERbL-Np is also promising in jejunal neuropathy treatment.

Abstract

Penelitian ini bertujuan mengetahui ekspresi insulin pada pankreas mencit (Mus musculus) yang diinduksi streptozotocin berulang dengan pewarnaan imunohistokimia yang berguna sebagai hewan model diabetes melitus. Tiga puluh ekor mencit jantan galur Balb-C, umur 12-14 minggu dengan bobot badan 30-40 g dikelompokkan menjadi 2 kelompok perlakuan, masing-masing kelompok terdiri atas 15 ekor. Kelompok 1 (K1) diberikan pelarut streptozotocin, sedangkan kelompok 2 (K2) diberikan streptozotocin dengan dosis 40 mg/kg bobot badan dalam 50 mM natrium sitrat bufer pH 4,5 secara intraperitoneal sebanyak 0,5 ml selama 5 hari berturut-turut. Hewan percobaan dari masing-masing kelompok dieutanasia sebanyak 2 ekor pada hari ke-0, 7, 14, 21, dan 28 setelah perlakuan, selanjutnya mencit diperfusi dan dinekropsi untuk mengambil jaringan pankreas sebagai sampel pemeriksaan imunohistokimia dengan metode streptavidin peroksidase menggunakan antibodi mouse anti-insulin (1:300). Berdasarkan uji statistik menggunakan analisis varian, ekspresi insulin pada sel beta Langerhans pankreas K1 lebih tinggi dibandingkan K2 (P0,05). Waktu pengamatan dan interaksi antara kelompok dan waktu pengamatan menunjukkan perbedaan yang signifikan (P0,05). Induksi dosis rendah streptozotocin secara berulang dapat menurunkan jumlah ekspresi sel beta Langerhans pankreas yang imunoreaktif terhadap insulin.

Abstract

The study was conducted to investigate the effects of caffeine treatment during organogenesis period to the fetal birth weight, using rat (Rattus norvegicus) as the animal model. Thirty-six primipararat obtained from Unit Pengembangan Hewan Percobaan, Gadjah Mada University (UPHP-GMU), 3 month old, 165-200 g body weight, were divided into 6 groups, consisted of 6 rats each. Six of the ratshave been selected based on the estrous cycles, and only rat with regular estrous were use for theexperiment. The rat then were mated, and during day 6-14 of the pregnancies were treated orally withcaffeine diluted in aquadest in dosage: placebo (1 cc aquadest) for group I (control), and 5.4, 10.8, 16.2,21.6, and 27 g/200 g body weight/day for treatment groups II-VI respectively. The pregnant rat bodyweights were determined at day 6 of pregnancies for calculating the caffeine treatment dosages. At day 20thof the pregnancies all of the pregnant rats were caesarotomized, and all of the fetuses were removed and weighed. The results showed that all of the treatment groups have significantly lower birth weightcompare to the groups control group. More over, fetal obtained from the treatment groups also showedserious subcutaneous hemorrhagic.Keywords: organogenesis, Rattus norvegicus, birth weight
