Abstract

Background: Exposure to ultraviolet B (UV-B) rays causes skin aging characterized through increased free radical damaged. To prevent skin aging due to UV-B exposure, it is necessary to additionally provide antioxidants as a measure to inhibit the skin aging process. Bajakah tree (Spatholobus littoralis hassk) is one antioxidants sourche such as phenols, flavonoids, and tannins. The aim was to prove that the administration of Bajakah stem ethanol extract cream inhibits increasing MMP-1 expression and reduction of collagen number in male Wistar rats exposed to UV-B. Methods: A posttest control group study design was coducted in male rats (Rattus norvegicus), Wistar strain, aged 2-3 months, weigh 180-200gram which was divided randomly into two groups. The control group (n = 18) was exposed to UV-B rays and placebo cream (0.2 mg/cm2), while the treatment group (n = 18) was exposed to UV-B rays and Bajakah stem ethanol extract cream 15 % (0.2 mg/cm2). Both creams have aplied twice before and after exposure to UV-B rays (3 times a week, with 840 mJ/cm2) for 4 weeks. Twenty-four hours after the last day intervention, a punch biopsy of skin tissue was prepared for histological examination followed by immunohistochemical assay (for MMP-1) and Sirius red (for collagen) staining. Results: The expression of MMP-1 was significantly higher in the control group (24.3 ± 6.20%) compared to the treatment group (11.5 ± 3.21%), p<0.001. Furthermore, the number of collagen was statistically lower in the control group (63.1 ± 3.94%) compared to the treatment group (82.0 ± 3.02%), p<0.001. Conclusion: The administration of bajakah (Spatholobus littoralis hassk) stem ethanol extract cream inhibited the increasing of MMP-1 expression and the reducing of collagen cells in male Wistar rats (Rattus norvegicus) those were exposed to UV- B.

Abstract

Buah merah (Pandanus conoideus Lam.) secara empiris telah digunakan untuk pengobatan kanker. Buah merah mengandung -carotene dan ?-tocopherol, yang merupakan sumber antioksidan. Penelitian ini bertujuan untuk mengetahui efek antiproliferasi dan ekspresi tumor suppressor gene p53 ekstrak etanol minyak buah merah (EEMBM) pada sel kanker serviks SiHa.Uji proliferasi dan uji ekspresi p53 dilakukan secara in vitro. Uji proliferasi menggunakan enam variasi dosis EEMBM dan dievaluasi dengan metode tryphan blue staining setelah inkubasi selama 24, 48, dan 72 jam. Uji ekspresi tumor supresor gene p53 menggunakan tiga variasi dosis EEMBM dan dievaluasi dengan metode imunohsitokimia setelah inkubasi selama 24 jam.Hasil penelitian menunjukkan bahwa persentase penghambatan proliferasi sel kanker servikss SiHa oleh EEMBM pada dosis tertinggi (0,125 ?g/mL) setelah inkubasi selama 24, 48, dan 72 jam berturut-turut adalah 97,97%; 100,0%; dan 100,0%. Bila dibandingkan dengan kontrol positif (doksorubisin), persentase penghambatan sel kanker pada dosis tertinggi setelah inkubasi 24, 48, dan 72 jam berturut-turut adalah 85,31%; 94,43%; dan 99,82%. Terdapat perbedaan bermakna pada persentase penghambatan proliferasi sel kanker serviks SiHa antar kelompok uji pada masing-masing waktu inkubasi (p<0,001). EEMBM juga meningkatkan ekspresi tumor suppressor gene p53 pada dosis 0,06250; 0,03125; dan 0,01562 ?g/mL berturut-turut sebesar 43,69%; 61,99%; dan 18,14%. Bila dibandingkan dengan kontrol positif, ekspresi p53 pada tiga variasi dosis (3,75; 1,875; 0,9375 ?g/mL) berturut-turut sebesar 42,63%; 41,88%; dan 35,94%. Terdapat perbedaan bermakna pada ekspresi p53 antar-kelompok uji (p<0,001).Kesimpulannya, ekstrak etanol minyak buah merah memiliki efek antiproliferasi dan dapat meningkatkan ekspresi tumor suppressor gene p53 pada sel kanker serviks SiHa in vitro.Kata kunci: Ekstrak etanol minyak buah merah, efek antiproliferasi, ekspresi tumor suppressor gene p53, sel kanker serviks SiHa

Abstract

Background: Type 2 diabetes mellitus (T2DM) is a disease that has a high prevalence in the world. The development of plants with medicinal potential is an alternative to control blood sugar levels in T2DM disease, such as avocado (Persea americana). Persea americana seeds contain secondary metabolites that have anti-diabetic activity, but their bioavailability is low.Aim: This study aims to review various secondary metabolites in Persea americana seeds that can reduce blood glucose levels in ?-amylase pathway along with the type of potential encapsulation as a delivery system.Review: Secondary metabolites contained in Persea americana seeds which have activity as anti-diabetic are tannin, quercetin, rutin, kaempferol, saponin, triterpenoid, and alkaloid. Each of them has several mechanisms in diabetes, but their role as ?-amylase inhibitor on T2DM be in focus. There are various types of encapsulation that are known to be able to serve as a delivery system for these secondary metabolites. Those encapsulations are SNEDDS, chitosan-alginate nanoparticle, PLGA nanoparticle, lipid carrier, liposome, and polysaccharide-based enteric-coated nanoparticle. All of them showed good results in improving bioavailability.Conclusion: It is known that various secondary metabolites found in Persea americana seeds influence reducing blood glucose levels notably in the ?-amylase pathway. The low bioavailability of secondary metabolites can be improved by several forms of potential encapsulation. Therefore, herbal substances as adjuvant therapy in T2DM might be a viable management option.

Abstract

Introduction: Coronary heart disease is one of the national health problems that gets attention. One of the efforts to develop the treatment of coronary heart disease includes the use of natural ingredients such as soybean seed extract which contains aglycone isoflavones which affect preventing endothelial dysfunction and reducing LDL levels in the blood, where high LDL levels and low HDL levels in the blood are one of the factors. The risk of coronary heart disease. Aglycone isoflavone compounds are non-polar compounds that are difficult to dissolve in water, so they require other ingredients to increase the absorption of aglycone isoflavones, one of which is the Self Microemulsifying Drug Delivery System (SMEDDS) formulation that can help improve solubilization and access drug to lymphatic tissueMethods: This experimental study was conducted with a post-test only approach. The research sample used 35 white rats which were divided into the first group which was given a normal diet, the second group which was given a high-fat diet and the third to fifth groups which were given a high-fat diet and the SMEDDS formulation of soybean seed extract. The SMEDDS formulation of soybean seed extract was administered at 200 mg/mL, 300 mg/mL and 400 mg/mL using a probe for seven days, twenty-one days after rats were induced with coronary heart disease with a high-fat diet. The rat blood was then taken for analysis of LDL and HDL levels. Data were analyzed by SPSS for windows.Results: The results showed that there was no significant difference in LDL and HDL levels from each experimental group (p>0.05). This can be caused by the short period of administration of the SMEDDS formulation and the dosage of the formulation that has not been able to provide clinical effects on experimental animals.Conclusion: The SMEDDS formulation of soybean seed extract did not show the ability to reduce LDL lizards or increase HDL levels in rats induced by coronary heart disease.  Pendahuluan: Penyakit jantung koroner merupakan salah satu masalah kesehatan nasional yang mendapat perhatian. Salah satu upaya pengembangan pengobatan penyakit jantung koroner meliputi pemanfaatan bahan alami seperti ekstrak biji kedelai yang memiliki kandungan isoflavon aglikon yang memiliki efek untuk pencegahan disfungsi endotel serta penurunan kadar LDL dalam darah, dimana tingginya kadar LDL dan rendahnya kadar HDL dalam darah sendiri merupakan salah satu faktor risiko dari kejadian penyakit jantung koroner. Senyawa isoflavon aglikon sendiri adalah senyawa non polar yang sulit untuk terlarut dalam air sehingga memerlukan bahan lain untuk meningkatkan penyerapan isoflavon aglikon, salah satunya adalah formulasi Self Microemulsifying Drug Delivery System (SMEDDS) yang dapat membantu meningkatkan solubilisasi dan akses obat ke jaringan limfatikMetode: Penelitian eksperimental ini dilakukan dengan pendekatan post-test only. Sampel penelitian menggunakan 35 ekor tikus putih yang dibagi menjadi kelompok pertama yang diberikan pakan normal, kelompok kedua yang diberikan pakan tinggi lemak serta kelompok ketiga hingga kelima yang diberikan pakan tinggi lemak dan formulasi SMEDDS ekstrak biji kedelai. Formulasi SMEDDS ekstrak biji kedelai diberikan sebanyak 200mg/mL, 300 mg/mL dan 400 mg/mL dengan menggunakan sonde selama tujuh hari, dua puluh satu hari setelah tikus diinduksi penyakit jantung koroner dengan diet tinggi lemak. Darah tikus kemudia diambil untuk analisis kadar LDL dan HDL. Data dianalisis dengan SPSS untuk windows.Hasil: Hasil penelitian menunjukkan bahwa tidak terdapat perbedaan yang signifikan pada kadar LDL dan HDL dari tiap kelompok percobaan (p>0,05). Hal ini dapat diakibatkan karena jangka waktu pemberian formulasi SMEDDS yang cukup singkat dan dosis formulasi yang belum dapat memberikan efek klinis terhadap hewan cobaSimpulan: Pemberian formulasi SMEDDS ekstrak biji kedelai tidak menunjukkan kemampuan untuk menurunkan kadal LDL ataupun meningkatkan kadar HDL pada tikus yang diinduksi penyakit jantung koroner

Abstract

Tuberculosis in children is a global health problem that decreases the quality of life of children. Based on data from the Indonesian Ministry of Health in 2016, nearly 69.000 children had tuberculosis and the case keeps increasing every year. Moringa oleifera leaf nanoemulsion syrup has immunonutrition and hepatoprotectant effects in children with tuberculosis. Moringa oleifera leaf nanoemulsion syrup contains proteins, micronutrients, and minerals which have a biological role as an immunity agent and prevent toxic effects of tuberculosis drugs. Until now, the use of Moringa oleifera leaf nanoemulsion syrup has been carried out for the immunomodulatory and hepatoprotective aspects. Immunomodulatory and hepatoprotective aspects will be discussed further in this literature review. The sources of articles in this literature review are pubmed.com, ncbi.com, plosone.com, sciencedirect.com, and googleschoolar.com from 2010-2020, except when there is no new research against the article. The authors searched for the keywords: "immunonutrition", "tuberculosis in children", "hepatoprotectant", and "Moringa oleifera". As an immunomodulator, Moringa oleifera leaf nanoemulsion syrup stimulate activation of polimorphonuclear (PMN) cells. As a hepatoprotectant, Moringa oleifera leaf nanoemulsion syrup work by reducing the side effects of conventional tuberculosis drugs such as rifampicin by suppressing the action of cytochrome p450 (CYP1A2 and CYP2B), thus decreases the production of toxic hydrazine which causes liver toxicity in tuberculosis patient. Seeing the various interests in the immunomodulatory and hepatoprotective aspects, Moringa oleifera leaf nanoemulsion syrup can be used as an adjuvant therapy in overcoming tuberculosis in children by stimulating the activation of immunity cell such as PMN, increasing nutrient absorption, and suppressing the action of cytochrome p450 (CYP1A2 and CYP2B).

Abstract

Generally, a person will spend quite a long time using his smartphone. Prolonged use of smartphones can cause musculoskeletal disorders. This event can make the function of the biomotor component decrease. One of the decreased biomotor components is handgrip strength. The role of physiotherapy is needed to educate in avoiding the risk of decreased grip strength. Therefore, this study aims to determine the relationship between smartphone usage duration with handgrip strength in adolescents aged 18-24 years. This research is an analytical observational study using a cross-sectional approach. Sampling using a purposive sampling technique. Fifty women aged 18-24 years who had met the participating criteria were the participants in this study. Handgrip strength was measured using a Camry Handgrip Dynamometer. Smartphone usage duration was obtained from recording the average daily screen time for a month. The average smartphone usage duration the participant's smartphone was 453.88 ± 159.16 minutes. Based on pearson correlation analysis, there is a significant negative relationship weak to moderate strength between smartphone usage duration and handgrip strength (r=-0.51; p=0.00 dominant handgrip, r=-0.33; p=0,02 non-dominant hand grip). Linear regression showed variations in the strength of the dominant handgrip as much as 26.4% and the non-dominant hand grip as much as 11% can be explained by the smartphone usage duration (p<0.05). The conclusion is smartphone usage duration is correlated with a decrease in handgrip strength. The longer using a smartphone, the weaker the grip strength. Abstract

Introduction: Coronavirus disease 2019 (COVID-19) is an infectious disease that caused by SARS-CoV-2 with new cases and mortality rates that are increasing recently. Due to inadequate of health facilities in Indonesia and the absence of a COVID-19 vaccine, some actions are required to reduce the spread of the virus, such as physical distancing. In educational aspect, conventional learning system are replaced by online learning system. This kind of system may cause burnout in medical students. The aim of the study was to describe the burnout incidence during online lectures in medical students in Udayana University during the Covid-19 pandemic. Materials and Methods: A cross sectional study with 175 medical students in Udayana Univerity class of 2018 and 2019 was conducted in this study. The data were collected via google form which includes informed consent and questionnaire which contains burnout scale. Abstract

Background: Development is developing human maturity and psychological processes that cannot be measured, such as increasing skills. The development speed in children is influenced by various factors such as the environment, health, nutrition, stimulation, and the role of parents. Fine motor development can be improved by applying the learning process and playing with interesting media, so children do not get bored easily. The media should be educative, like a puzzle game. Physiotherapy plays a role in preventing and reducing further complications that do not occur in children who experience fine motor delays. This research aims to determine the effect of giving puzzle games on fine motor development in preschool children. Methods: The method applied is a literature study by looking for theoretical references related to the research carried out. This literature review uses secondary data and information from journals collected from the Google Scholar, PubMed, and ScienceDirect databases over the last ten years as data sources. Results: Four studies discuss the impact of puzzle games on the fine motor development of preschool children. According to all journals, puzzle games impact the fine motor development of preschool-aged children. On average, about 25 children participate in each study. Conclusion: The findings obtained based on the literature that has been reviewed and the discussion state that giving puzzle games affects fine motor development in preschool children. The findings of this research can be used as a basis for puzzle therapy in children and are expected to be used to improve children's fine motor development through other games. There isn’t any clinical implication from this literature review.

Abstract

Background: College students are one of the groups that have a high risk of experiencing sleep deprivation and sleep disorders. In medical students, this is due to the high duration and intensity of education. Poor sleep quality causes impaired cognitive function, one of which is decreased reaction time. Students with faster reaction times have better cognitive status and academic performance. This study aims to determine how the relationship of sleep quality to reaction time with the type of simple reaction time (SRT) and choice reaction time (CRT) experiment in physiotherapy student at faculty of medicine, university of udayana.Methods: The cross-sectional study was conducted with simple random sampling technique. 45 students participated in this study. Sleep quality was measured with Pittsburgh Sleep Quality Index (PSQI) and reaction time using Deary-Liewald reaction time task with SRT and CRT types of experiment. Bivariate analysis was performed using the Spearman rho test.Result: 21 people (46.7%) had good sleep quality and 24 people (53.3%) had poor sleep quality. The results showed that there was a relationship between sleep quality and reaction time using CRT (p = 0.003) and there was no relationship between sleep quality and reaction time using SRT (p = 0.084).Conclusion: Poor sleep quality resulted in decreased reaction time with the CRT experiment type. The authors suggests research with different methods and populations, also add more control variables.

Abstract

Pendahuluan: Pada masa pandemi Covid-19, penggunaan berbagai alat digital semakin meningkat. Hal itu berdampak pada peningkatan intensitas penggunaan gawai, contohnya pada mahasiswa yang melakukan perkuliahan daring. Intensitas yang tinggi dalam menggunakan gawai berisiko menyebabkan masalah kesehatan, salah satunya disabilitas leher. Disabilitas leher dapat disebabkan oleh posisi buruk yang monoton saat menggunakan gawai dalam jangka waktu yang lama. Penelitian ini bertujuan mengetahui hubungan intensitas penggunaan gawai dengan tingkat disabilitas leher pada mahasiswa Program Studi Sarjana Fisioterapi Fakultas Kedokteran Universitas Udayana. Metode: Penelitian ini menggunakan pendekatan kuantitatif dengan desain penelitian korelasional. Jumlah subjek penelitian ini adalah 75 mahasiswa berusia 18 sampai 22 tahun yang dipilih melalui proses simple random sampling. Data diperoleh dari interpretasi intensitas penggunaan gawai menggunakan kuesioner skala intensitas penggunaan gawai dan tingkat disabilitas leher menggunakan kuesioner neck disability index (NDI). Kedua variabel tersebut dianalisis menggunakan chi-square test. Hasil: Mahasiswa yang menjadi subjek mayoritas menggunakan gawai dalam intensitas tinggi dan sangat tinggi yaitu sebanyak 73 orang dengan 41 orang diantaranya mengeluhkan disabilitas ringan dan 5 orang disabilitas sedang, sedangkan sisanya tidak mengalami disabilitas leher. Hasil chi-square test yang diperoleh yaitu nilai p=0,002 sehingga p<0,05 yang berarti Ho ditolak dan Ha diterima. Simpulan: Dapat disimpulkan bahwa terdapat hubungan antara intensitas penggunaan gawai dengan tingkat disabilitas leher pada mahasiswa Program Studi Sarjana Fisioterapi Fakultas Kedokteran Universitas Udayana. Hasil penelitian ini diharapkan dapat menjadi acuan preventif megenai intensitas penggunaan gawai agar dapat meminimalisir risiko terjadinya disabilitas leher.
