Abstract

Penelitian ini merupakan penelitian eksperimental murni dengan tujuan untuk mengetahui gejala toksisitas akut dan lethal dose 50 ekstrak etanol uwi Banggai ungu (Dioscorea alata L.) terhadap tikus putih (Rattus norvegicus). Penelitian ini menggunakan 20 ekor tikus yang dibagi menjadi empat kelompok dan diberikan dosis bertingkat yaitu 1 g/kgBB, 2 g/kgBB, 4 g/kgBB, dan 8 g/kgBB ekstrak etanol uwi Banggai ungu dengan sekali pemberian secara oral kemudian diamati gejala ketoksikan dan jumlah kematian pada tiap hewan uji setelah 24 jam, selanjutnya dilakukan pengamatan bobot badan selama 14 hari tanpa diberikan perlakuan. Hasil penelitian menunjukkan LD50 ekstrak etanol uwi Banggai ungu (Dioscorea alata L.) adalah semu atau bukan LD50 yang sesungguhnya yaitu >8 g/kgBB dan masuk dalam kategori praktis tidak toksik (5-15 g/kgBB). Gejala klinis yang teramati pada tikus yaitu menurunnya aktivitas motorik pada dosis tertinggi dan frekuensi grooming yang meningkat seiring dengan peningkatan dosis.

Abstract

Penelitian ini bertujuan untuk mengetahui aktivitas ekstrak daun rumput bermuda (Cynodon dactylon L. Pers) terhadap penyembuhan luka bakar serta mengetahui konsentrasi yang efektif terhadap penyembuhan luka bakar. Hewan uji yang digunakan pada penelitian ini adalah kelinci jantan (Oryctolagus cuniculus) yang dibagi dalam 5 kelompok uji. Setiap kelinci dibuat luka bakar di bagian punggung dengan diameter 2 cm, kelompok uji I diberi gel Bioplasenton (kontrol positif), kelompok II diberi basis Na CMC (kontrol negatif), kelompok uji III, IV, dan V diberi sediaan ekstrak daun rumput bermuda dengan konsentrasi masing-masing 1%, 3%, dan 5%. Pengukuran luka bakar menggunakan alat jangka sorong dilakukan setiap hari selama 21 hari. Data yang diperoleh dianalisis menggunakan uji One way Anova dan dilanjutkan uji Post hoc Duncan. Hasil statistik menunjukkan bahwa ekstrak daun rumput bermuda memiliki aktivitas dalam penyembuhan luka bakar pada kelinci. Konsentrasi yang efektif adalah konsentrasi 5%.

Abstract

Luka diabetik merupakan luka yang disebabkan oleh infeksi akibat dari tingginya glukosa darah ? 200 mg/dL. Daun Diospyros celebica Bakh., memiliki potensi antibakteri, antidiabetes, antivirus, dan antijamur. Tujuan penelitian ini adalah menentukan aktivitas penyembuhan luka diabetik pada tikus putih yang di induksi aloksan dengan diberikan terapi ekstrak daun eboni. Induksi diabetes dilakukan menggunakan aloksan dosis 140 mg/kgBB intraperitonial. Tikus dilukai pada bagian punggung dengan diameter luka ± 1 cm dan kedalaman 2 mm. Jumlah sampel dalam penelitin ini adalah 25 ekor tikus dibagi menjadi 5 kelompok uji yaitu kelompok 1 diberikan suspensi glibenklamid secara oral dan gel plasebo pada luka, kelompok 2,3 dan 4, kelompok yang diberikan  suspensi glibenklamid secara oral dan gel ekstrak daun eboni konsentrasi 5 %, 10 % dan 15 % pada luka), kelompok 5 (pemberian suspensi Na-CMC 0,5 % secara oral dan gel plasebo pada luka). Pengujian dilakukan selama sepuluh hari dengan dosis dua kali pemberian setiap hari. Pengamatan makroskopik, panjang luka diukur dan dilaporkan sebagai persentase penyembuhan luka. Hasil penelitian menunjukkan bahwa gel ekstrak daun eboni memiliki kemampuan untuk memperbaiki lesi pada luka diabetes pada tikus putih, konsentrasi gel ekstrak daun eboni yang paling efektif untuk memerbaiki lesi luka diabetik adalah konsentrasi 10%, dengan persentase penyembuhan 94,34%. Abstract

Hantap (Sterculia coccinea Jack) has been used traditionally for various health issues, including cancer treatment. The therapeutic effects of natural ingredients are often attributed to their chemical constituents. This study aimed to analyze the phytochemical contents and cytotoxic activities of S. coccinea leaves on HeLa and MCF-7 cancer cell lines. The quantitative phytochemical analysis was carried out following standard laboratory procedures. Phytochemical compounds were identified using LC-MS/MS QTOF. The MTT assay PrestoBlue™ Cell Viability Reagent test method was used to test cytotoxic activity in the cell culture. Extraction was carried out by the maceration method using 96% ethanol as solvent. The quantitative analysis revealed that tannins were the major phytochemical constituent in the highest percentage of 72.16%, followed by alkaloids, flavonoids, and steroids, with values of 30.80, 28.66, and 2.85%, respectively. Saponins were present in the lowest percentage of 1.15%. The ethanolic extract exhibited moderate cytotoxicity on HeLa and MCF-7 cells with IC50 values of 591.00 and 578.10 µg/mL, respectively. Identification using LC-MS/MS showed the suspected compounds 5,7-dihydroxy-3-(4'-hydroxybenzyl)chromone as homoisoflavanones and kaempferide-3-O-α-L-rhamnosyl-7-O-α-L-rhamnoside from flavonol triglycosides. These results may contribute to the study on the use of leaves extract of S. coccinea for developing a chemoprevention agent.

Abstract

Penyakit degeneratif merupakan kondisi kesehatan yang menyebabkan jaringan atau organ memburuk dari waktu ke waktu berkaitan dengan penuaan, atau memburuk selama proses penuaan. Hipertensi dan Diabetes Mellitus termasuk penyakit degeneratif dengan morbiditas dan mortalitas tinggi. Pengetahuan mengenai penyakit ini masih kurang terutama masyarakat di pedesaan yang terbatas untuk akses informasi dan layanan kesehatan. Oleh karena itu kegiatan pengabdian ini bertujuan untuk memberikan edukasi kepada masyarakat desa Apal, kecamatan Liang, Banggai Kepulauan tentang penata laksanaan penyakit degeneratif khususnya hipertensi dan DM serta melakukan skrining kesehatan dengan pengukuran tekanan darah dan kadar gula darah (KGD) sewaktu sebagai upaya peningkatan kesadaran masyarakat untuk menjaga kesehatan. Metode yang digunakan adalah penyuluhan, pengukuran tekanan darah dan KGD sewaktu serta konsultasi kesehatan. Hasil skrining menunjukkan bahwa 68% peserta mengalami hipertensi stage 1 (12%) dan stage 2 (56%) serta prehipertensi 32%, sedangkan hasil pemeriksaan KGD menunjukkan 76% memiliki KGD normal, 24% masuk batas tinggi (6%) dan tinggi (18%). Berdasarkan hasil pengabdian yang dilakukan menunjukkan adanya peningkatan pengetahuan dan kesadaran masyarakat di desa Apal mengenai penyakit hipertensi dan DM.

Abstract

The risk of various degenerative diseases is more experienced by older adults than young people. One of the most common health problems among the elderly is hypertension. Delays in treating hypertensive patients are caused by the majority of patients only coming to health facilities after experiencing complications and lack of access to public health services. The incidence of dyslipidemia and diabetes mellitus (DM) also continues to grow, which are risk factors for coronary heart disease. For this reason, promotive and preventive efforts for the elderly group are significant through increasing knowledge and understanding of degenerative diseases. The community service partner is the Banpres Pololo Community Health Center, one of the community health centers in Sigi Regency with high cases of degenerative diseases, and the target of community service is the elderly in Uenuni Palolo village. Implementing the activity was health education about hypertension, DM, and dyslipidemia, health screening, and health counseling. The screening results found that 94.12% of older adults suffered from hypertension, 17.65% suffered from DM, and 52.94% suffered from hypercholesterolemia. Health counseling results show that most older adults did not understand the importance of early detection of degenerative diseases. Therefore, health education and counseling for the community still need to be improved.

Abstract

Fish oil has omega-3 and omega-6 fatty acids, which provide health benefits such as prevention of an increase in total cholesterol, triglycerides, and a decrease in blood HDL, as well as being anti-inflammatory agents in humans. The present study was conducted to determine the antihyperlipidemic and anti-inflammatory activities of eel fish (Anguilla marmorata [Q.] Gaimard) oil extracts on white rats (Rattus norvegicus). To test hyperlipidemia, 25 rats were divided into 5 groups and treated with; 1.5 % PEG 400 (negative control), 20 mg simvastatin (positive control), test doses of 18, 36, and 72 mg/kg. The treatments were carried out for 39 days. In investigating the anti-inflammatory activity, 25 white rats were divided into 5 groups; negative control, positive control, three-dose treatments of 22, 44, and 88 mg/200 g. Blood cholesterol levels were measured and percent inflammation was determined. The results showed that the eel oil extracts led to a decreased of total cholesterol levels 49.60 %, triglycerides 31.15 %, and HDL 14.3 %. The percentage of inflammation values ranged from 31.73 to 54.48 %. The findings indicated that the most effective doses for the antihyperlipidemic effect were 72 mg/ kg, while 22 mg/200 g for the anti-inflammatory activity.

Abstract

Background: Standardizing herbal medicines is crucial for assessing their quality and effectiveness. Objectives: This study aims to determine non-specific and specific standardization parameters for medicinal plants. Non-specific parameters include drying shrinkage, water content, total ash content, acid insoluble ash content, total bacteria, total mold, and metal limits (Pb, Hg, and Cd). Specific parameters involve organoleptic examination, identification of chemical content in extracts, dissolved compounds in specific solvents, total flavonoid content, and qualitative testing of anthocyanin content in the extract. Materials and Methods: An experimental laboratory approach was employed to extract Dioscorea alata L using 96% ethanol, which was later concentrated through a rotary evaporator to obtain a viscous extract. Specific and non-specific parameters were determined using standardized techniques. Results: The extract yielded 3.32% of the initial substance. Drying shrinkage was 19.7769±1.2682%, and specific gravity was 0.6192±0.1476 gram/ml for 5% m/v and 0.5101±3.9591 gram/ml for 10% m/v. Ash content was 0.6793%, with acid insoluble ash content at 0.1895%. Heavy metal contamination for Hg was 0.47 ppm, but undetected for Pb & Cd). The total plate number was 27 x 103 colonies/g, and negative Escherichia coli contamination. The mold count was 39 x 101 colonies/g. Additional specific parameters included the organoleptic properties of the viscous extract (purple color, characteristic odor, and sweet taste), water-soluble compound content (15.63%), ethanol-soluble compound content (14.73%), and a flavonoid content of 76.669 mg EQ/100 mg. Conclusions: The ethanol extract of Dioscorea alata complies with the standard requirements established by the Food and Drug Supervisory Agency. It presents as a viscous extract with a distinct odor, purple-black color, and sweet taste. The extract contains alkaloids, flavonoids, saponins, tannins, steroids, and phenolic compounds, with a total flavonoid content of 76.669 mg. Furthermore, it tested positive for anthocyanins.

Abstract

Dyslipidemia is a medical condition caused by a lipid metabolism disorder in the body. In the early stages, dyslipidemia often shows no symptoms or complaints, so many sufferers are unaware of their condition. However, examinations of total cholesterol, low-density lipoprotein (LDL), and triglyceride levels often show elevated results in people with dyslipidemia. Public awareness of the dangers of dyslipidemia remains low, particularly among the elderly. To increase understanding and awareness of dyslipidemia, a community service activity was conducted in Pana Village. This activity began with cholesterol level checks. Following this, a presentation was given on the dangers of dyslipidemia, especially among the elderly, and the importance of maintaining a healthy lifestyle to prevent risks of heart disease, stroke, and even death. This counseling session focused on lifestyle changes that can help control cholesterol levels, such as regulating diet, exercising regularly, and avoiding unhealthy habits like smoking. Out of 95 participants who underwent cholesterol testing, the results showed that 46 elderly individuals (43%) had cholesterol levels below 200 mg/dl, which is still considered within normal limits. However, 54 elderly individuals (57.3%) had cholesterol levels above 200 mg/dl, indicating a high prevalence of dyslipidemia in this age group. These findings emphasize the importance of health education for the elderly, so they become more aware of the risks of dyslipidemia and are motivated to adopt a healthier lifestyle to prevent serious complications.
