Abstract

Free radical from cigarrete smoke can cause lipid peroxidation that could lead to tissue damage. Tissue damage can be identified by increased levels of MDA (Malondialdehyde). Leavesof buas-buas (Premna cordifolia. Linn) has an antioxidant activity in invitro study. This study aimed to analyze the effect of P. cordifolia leaves extract on the levels of malondialdehyde (MDA) after exposure to cigarette smoke. The simplicia extracted by maseration using 70% ethanol. Twenty four  wistar strain of Rattus novergicus were divided into six groups , i.e. : normal group without any treatment, positive control group with cigarette smoke exposure and vitamin e treatment, negative control groupwith only cigarette smoke exposure, treatment groupwith cigarette smoke exposure and ethanolic extract from P. cordifolialeaves with doses 200; 400; 600 mg/kg of body weight/day. All rats were treated for 14 days using 3 cigarette non filetered each day. After 14 days, plasma MDA was measured in this research. Measurement of plasma MDA are using Spectrophotometer UV-Vis in 532.2 nm. MDA level in this study was analyzed by the Kruskal-Wallis and Mann-Whitney test. The result showed that plasma MDA level of groupreceived ethanol extract of P. cordifolia leaves doses at 600 mg/kg of body weight/day significantly (p<0.05)lower than negative control group and not significantly differ (p<0.05) to normal group and positive control group.

Abstract

Malondialdehyde (MDA) is a biomarker of lipid peroxidation that is closely correlated with level of oxidative stress. The aim of this study was to know ethanol extract activities from E. americana leaves against blood plasma MDA in male Wistar rats exposed by cigarette smoke using tiobarbituric acid test. The simplicia extracted by maseration using 70% ethanol. Twenty four rats were divided into six groups, i.e. : a normal group, an exposed cigarette smoke (negative) control group, a positive control group and three treatment groups. One normal group received carboxyl methyl cellulose and another groups exposed cigarette smoke during 14 days. Positive control group received vitamin E (18mg/kg BW), and another groups received ethanol extract of E. americana leaves doses 180; 90; 45mg/kg BW, respectively. The MDA level was analysed by spectrophotometer UV-Visible in 532,2 nm. The MDA level statically analysis by Kruskal-Wallis test and following by Mann-Whitney test. The mean MDA levels of normal group, negative control group, positive control group and groups received ethanol extract of E. americana leaves doses 180; 90; 45mg/kg BW were 0,11±0,09; 13,74±2,84; 0,06±0,03; 10,88±2,19; 0,13±0,03; 0,03±0,02 ppm. The mean MDA levels of groups received ethanol extract of E. americana leaves doses at 180; 90mg/kg BW significantly (p<0,05) lower than negative control group. These results show the beneficial effect of ethanol extract of E. americana leaves in the MDA level reducing on the rats that exposed by cigarette.

Abstract

Exposure to cigarette smoke is one of the exogenous free radicals. Exposure to cigarette smoke is associated with lung inflammation, oxidative stress and lipid peroxidation. Compounds that can inhibit the oxidation reaction by binding free radicals known as antioxidants. Eleutherine americana Merr. leaves has an antioxidant activity in invitro study. The aim of this work was to study E.americana leaves as a natural antioxidant that reduce the level of lung damage from exposure to cigarette smoke in male wistar rats and study effective dose of E.americana leaves to reduce the level of lung damage.  The leaves of E.americana extracted by maceration method for 6 days using ethanol 70%. Rats were divided into six groups of four animals each : exposed to ambient air (GroupA), exposed to cigarette smoke (Group B), exposed to cigarette smoke and treated with ethanol extract of E.americana leaves at dose 45mg/kg bw, 90mg/kg bw, 180mg/kg bw (Group C,D,E), exposed to cigarette smoke and treated with vitamin E dose 18mg/kg bb(Group F). All rats were treated for 14 days using 3 cigarettes non filtered each day by using a smoking chamber. After 14 days rats were killed. Right lungs were removed for histopathology. The results showed that dose 45mg/kg bw, 90mg/kg bw, 180mg/kg bw can reduce the level of lung damage to the enlarge of alveolar, thickening of the alveolar wall and inflammatory cell infiltration. Dose 180mg/kg bw as an effective dose to reduce the level of lung damage that compare to normal control.

Abstract

Inappropiate antibiotic use was one of the main reason that caused pathogenic microorganism resistance, leads to ineffective treatment. One of those resistance bacteria is Staphylococcus aureus. The research for discovering new antibiotics can help with this issue. The peel of red dragon fruit (Hylocereus polyrhizus) is one of the plants that has antibacterial compounds. The aim of this research is to prove and determine the minimum inhibitory concentration (MIC) value of n-hexane fraction of red dragon fruit’s peel againts Staphylococcus aureus ATCC 25923. The peel was maserated with chloroform, then fractinated by n-hexane. Futhermore, the antibacterial properties of the fraction was tested using Disc Diffusion (Kirby-Bauer) methode with 40, 20 dan 10 mg/mL concentrations. Ampisilin used as control positive. Based on the phytochemical screening of the fraction, the n-hexane fraction of the dragon fruit’s peel contain triterphenoid, steroid and alkaloid. Clear zones produced by 40 mg/mL is 12,80 mm and for 20 mg/mL is 11,17 mm. Wherea concentration 10 mg/mL didn’t show any inhibitory zone. According to this reasearch, the MIC value of the fraction is  20 mg/mL.

Abstract

Use and demand of traditional medicinal plants currently growing in the direction that research in traditional medicines have also increased. One of the plants that is efficacious as a medicine is bawang mekah (Eleutherine americana Merr.). This plant is usually used by people as a traditional medicine to treat various diseases. This study aims to determine the antioxidant activity of bawang mekah leaves. Antioxidant activity assays performed using the DPPH (2,2-diphenyl-1-picrylhydrazyl) that begins with the extraction by maceration. Dry leaves of bawang mekah (150 g) was soaked with ethanol 70% for 3x24 hours at room temperature. The liquid extract obtained is evaporated by rotary evaporator and waterbath till viscous extracts is obtained. Then to extract performed phytochemical screening and preliminary test with DPPH method by thin layer chromatography (TLC) with a mobile phase BAA (4:1:5). The antioxidant activity was measured using a UV-Vis spectrophotometer and compared with vitamin C. The results of the phytochemical screening showed the extract contains flavonoids, saponins, phenols and tannins. In preliminary tests using KLT, gained 3 spots visualized with UV light 366 nm and sprayed with 0.2% DPPH. Third spot showed changes color to yellow with purple background that indicating the extract positive has antioxidant activity. The results of spectrophotometric measurements showed that the extract has the IC50 at 31.97437 ug/ml, whereas vitamin C had a lower IC50 value (3.90186 ug/ml).

Abstract

Oxidative stress induced by the radicals have been known to affect the occurrence of various degenerative disease. In order to search natural antioxidant compounds, it had been studied the isolation and identification of antioxidant compounds in fern stems (Alsophila glauca J.Sm) using DPPH method (1,1-diphenyl-2-picrylhydrazyl). Fern stem extraction was done by maceration with wasbenzen and metanol. Extracts was obtained by evaporating the wasbenzen solvent with a rotavapor. Material was re-macerated with methanol after wasbenzen evaporation. The same way was done to obtain the metanol extract. These extracts were tested for antioxidant activity using DPPH method with TLC. Active extract was partitioned in 80% methanol. Both 80% methanol soluble and insoluble extract were tested their antioxidant activity using DPPH method (TLC). Active extract was fractionated in vacuum liquid chromatography using a mobile phase with different polarity gradient and different concentrations (wasbenzen:chloroform). Active fraction was isolated by preparative TLC method and purity of obtained compounds was tested by TLC. Antioxidant activity of obtained isolates was tested by DPPH method using spectrophotometry. Compounds known to had antioxidant activity as radical catcher with IC50 178.4µg/mL.

Abstract

The infectious diseases can be caused by the pathogenic bacteria among Staphylococcus aureus and Escherichia coli. One of the plants that can be used as antibacterial is essential oils from Pontianak orange peels (Citrus nobilis Lour. var. microcarpa). This research aims to determine the antibacterial activity essential oils Pontianak orange peels againts the pathogenic bacteria by disc diffusion method. This research carried out by using the experimental Completely Randomized Design (CRD) Factorial and outcome data were analyzed using the program CoStat Two Way ANOVA method and continued by using LSD (Least Significant Different). The results showed that the essential oils from Pontianak orange peels contain flavonoids, saponins, and triterpenoids. The essential oils from Pontianak orange peels have antibacterial activity againts Escherichia coli and Staphylococcus aureus, where the average zone of inhibition obtained from the concentrations from 0,5; 1,5; and 2,5 mg/mL respectively are 15; 16; and 19 mm and 16,33; 18; and 21 mm. The concentration that gives the biggest inhibition zone is 2,5 mg/mL.

Abstract

Free radical induced oxidative stress that influences the occurrence of various degenerative diseases such as cancer, coronary heart disease and premature aging. In the case that body's antioxidant defense system does not have excessive antioxidants, additional natural antioxidant via food or other nutrients intake is needed. Stems of ferns are known to contain phenolic compounds that are known to have antioxidant activity. A study has been carried out to determine antioxidant potential of stems of ferns of western Borneo origin compared to quercetin as positive control. Initially, material was macerated gradually with wasbenzen followed by methanol. The extract obtained was filtered and evaporated to dryness to give dried wasbenzen extract and methanol extract respectively. Antioxidant properties of extracts were performed by DPPH on TLC. Active (wasbenzen) extract was then partitioned with 80% methanol to give soluble and insoluble fractions. Active (insoluble) fraction was further fractionated by vacuum liquid chromatography using a gradient mobile phase with increasing polarity (wasbezen: chloroform). Fractions obtained were tested antioxidant activity, and isolation was done by preparative TLC. The purity of isolated antioxidant compound was confirmed by TLC, and antioxidant potential was measured spectroscopically with DPPH method. This compound displayed antioxidant activity, but its value was lower than that of quercetin used as a positive control (IC50 178.4 v / s 2.17 microg / mL)

Futhermore, the antibacterial properties of the fraction was tested using Disc Diffusion (Kirby-Bauer) methode with 20 and 40 mg/mL concentrations. Ampicillin used as control positive, and Dymetil sulfoxida (DMSO) used as control negative . Based on the phytochemical screening of the fraction, the n-hexane fraction of the dragon fruit’s peel contain terpenoid and alkaloid. Clear zones produced by 40 mg/mL is 12.80±1,69 mm and for 20 mg/mL is 11.17±1,11 mm.

Abstract

Siam citrus peel (Citrus nobilis L. var. Microcarpa) is a plant derived from Sambas Regency, West Kalimantan Province. Bioavailability of herbal active compounds can be enhanced by formulating extract into nanoparticle. The polymer used was chitosan with crosslinker Na-TPP. Stirring speed and stirring time play an important role to produce small particle size in forming nanoparticle using ionic gelation method. Enhancement of stirring speed and stirring time could reduce particle size. Nanoparticles were prepared using ionic gelation method by mixing Na-TPP, extract and chitosan (1:1:6) with varying the stirring speed 500 rpm, 1000 rpm, 1500 rpm and stirring time 1 hrs, 2 hrs, 3 hrs. The particle size of nanoparticle was found to be 85.3 nm at 1000 rpm of stirring speed and 3 hrs of stirring times, with polidispersity index 0.287, zeta potential +32.37 mV and entrapment efficiency 87.12 %.
