Abstract

Oxidative stress induced by the radicals have been known to affect the occurrence of various degenerative disease. As the search for natural antioxidant compounds had been studied the isolation and identification of antioxidant compounds in fern stems (Alsophila glauca J.Sm) using DPPH method (1,1-diphenyl 2-picrylhydrazyl). Fern stem extraction was done by maceration with wasbenzen and metanol. Extracts was obtained by evaporating the wasbenzen solvent with a rotavapor. Material was re-macerated with methanol after all wasbenzen evaporation. The same way was done on  wasbenzen to obtain the metanol extract. These extracts were tested for antioxidant activity using DPPH method with TLC. Active extract was partitioned in 80% methanol. Extracts both soluble and insoluble in methanol 80% were tested their antioxidant activity using DPPH method (TLC). Active extract was fractionated in vacuum liquid chromatography using a mobile phase with different polarity gradient and different concentrations (wasbenzen:chloroform). Active  fraction  was  isolated  by  preparative TLC method and purity of compounds obtained was tested by TLC. Antioxidant activity of isolates obtained were tested by DPPH method using spectrophotometry. Compounds known to had antioxidant activity as radical catcher with IC50 178.4 µg/ml.

Abstract

Kulit buah manggis (Garcinia mangostana L.) memiliki kandungan xanton yang khasiatnya sebagai antioksidan yang baik untuk kulit. Tujuan penelitian ini adalah untuk membuat formulasi sediaan lotion yang akan di uji mutu fisik dari sediaannya. Ekstrak kulit manggis dibuat dengan cara maserasi menggunakan etanol 70% dengan rendemen yang dihasilkan 23.36 %. Konsentrasi yang digunakan pada sediaan lotion ini adalah 3%, 5% dan 7%. Evaluasi uji mutu fisik lotion meliputi pengamatan organoleptis, uji Ph, uji homogenitas, uji daya sebar, uji daya lekat dan identifikasi tipe emulsi. Dari hasil penelitian uji mutu fisik lotion beraroma khas, berwarna coklat muda hingga coklat tua, tekstur halus pada Formulasi 3% dan 5% sedangkan pada formulasi 7% terdapat butiran ekstrak, pH 6-6,5, daya sebar 3-7 cm, daya lekat formulasi 3% 3 menit formulasi 5% dan 7% 5 menit, dan tipe emulsi minyak dalam air.

Abstract

Background: Hylocereus polyrhizus has activities as antimicrobial agent, anti-hypercholesterolemia, anti-diabetic (diabetes mellitus), cardiovascular risk reduction, health supplement, and melanoma cell suppression. The extracts from the peels of H. polyrhizus were able to increase phagocytic ability, cell numbers and leukocytes and to influence relative spleen weights in the formation of body immune system in male rats. The fruit peels contained phenolics, flavonoids, carotenoids, and anthocyanins. This study investigated the active compounds of H. polyrhizus peels, which are able to increase immune system of human body.Materials and method: In vitro assay was applied to examine the active compounds, identified as lupeol, obtained from isolated extract of red dragon fruit for their lymphocyte proliferation and nitric oxide (NO)-producing activities. Lymphocyte proliferation assay was performed with 3-4.5-dimethylthiazol-2-yl)-2.5-diphenyltetrazolium bromide (MTT) method. The cell control was lymphocyte cell suspension in RPMI medium added with phytohaemaglutinine (PHA). The NO measurement was conducted with nitric solvent and Greiss reagent.Results: The ANOVA analysis of the average optical density (OD) of lymphocyte proliferation showed that the addition of isolated lupeol at the concentrations of 6.25, 12.5, 25, 50 and 100 µg/mL were able to improve lymphocyte proliferation and activate the NO production in the rats with treatment of positive control. Conclusion: Isolated lupeol at concentrations of 6.25, 12.5, 25, 50 and 100 µg/mL revealed significant difference with medium control and cell control. It was able to increase effects on lymphocyte proliferation and NO production. Therefore, the lupeol which was isolated might have high potential to be an immunostimulant.Keywords: Hylocereus polyrhizus, lupeol, lymphocyte proliferation, nitric oxide production

Abstract

Tanaman bawang dayak (Eleutherine americana (Aubl.) Merr. ex K.Heyne.) secara empirik telah digunakan sebagai obat tradisional. Golongan flavonoid yang terdapat dalam tanaman bawang dayak mungkin dapat mempengaruhi pembentukan sel darah, namun belum tersedia  informasi  mengenai  keamanan  terhadap  profil  hematologi  untuk penggunaan secara  oral  terutama ekstrak  etanol  daun  bawang dayak  dalam  jangka  waktu panjang.  Penelitian  ini bertujuan untuk  mengamati  profil  hematologi  ekstrak etanol  daun  bawang dayak  yang  diberikan  pada  tikus  putih  galur  Wistar  jantan  dan betina  menggunakan  metode  uji  prakilinik  toksisitas  subkronik  28  hari.  Daun bawang dayak diekstrak secara maserasi dengan pelarut etanol 70%.  Profil  hematologi  terdiri  dari  parameter  hemoglobin,  leukosit,  eritrosit, hematokrit, trombosit,  MCV,  MCH,  dan  MCHC.  Hewan  dibagi  menjadi  6  kelompok  yaitu kelompok  kontrol  diberikan  CMC-Na  1%,  ekstrak etanol daun bawang dayak  dosis  100  mg/kgBB, dosis  400  mg/kgBB,  dosis  1000  mg/kgBB,  kelompok  satelit  kontrol diberikan  CMC-Na  1%,  dan   satelit  dosis  1000  mg/kgBB.  Induksi  dan pengamatan  dilakukan  selama  28  hari,  dan  dilanjutkan  14  hari  untuk  kelompok satelit.  Hasil  pengamatan  profil  hematologi  menunjukkan bahwa tidak terdapat perbedaan antara kelompok perlakuan dengan kontrol satelit dalam jumlah maupun kadar hemoglobin, eritrosit, leukosit, hematokrit, trombosit,  MCV,  MCH,  dan  MCHC. Dapat disimpulkan  secara  umum  penggunaan  ekstrak etanol daun bawang dayak tidak  mempengaruhi  profil hematologi hewan uji.

Abstract

Excessive free radical production will lead to oxidative stress. One of the indicators used to determine stress oxidative is malondialdehyde (MDA). Hylocereus polyhizus peel is potential used natural exogenous antioxidant. The aim of this research was to study the effect of n-hexane fraction of H.polyhizus peel on MDA level and to determine the dose that could reduce the MDA level. H.polyhizus peel was macerated with chloroform, fractionated with n-Hexane. Rats were divided into 7 groups : normal, negative, positive control vitamin E (18 mg/ kgbw), positive control quercetin (4 mg/200gbw), H.polyhizus peel extract group treated with dose I (10mg/ 200gbw), dose II (20 mg/200gbw) and dose III (40 mg/200gbw). Each group consist of 4 rats and induced to oxidative stress by 10 minute swimming each day and food restriction for 5 days. Measurement of MDA level are using Spectrophotometer UV-Vis in 532 nm. MDA levels were analyzed using One way Anova and LSD test. The data shown that MDA level at normal, negative, positive vitamin E, positive kuersetin, dose I, II, III group is 0.042; 0.051; 0.034; 0.042; 0.038; 0.039; 0.042 µg/ml. The n-Hexane fraction of red dragon fruit peel was a potent antioxidant because posessing the ability to lowering the MDA level on stress oxidative rats.

Abstract

Production of excess free radicals cause oxidative damage and degenerative diseases. Catalase enzyme is one of the endogenous intracellular antioxidant that can be reduced by oxidative stress conditions. The peel of red dragon fruit (Hylocereus polyrhizus) as a potent exogenous antioxidants could support the internal enzyme activity to prevent free radicals. The aim of this research was to determine the effect of n-hexane fraction of H.polyrhizus’s peel and the dose that could increase activity of catalase. The peel was maserated with chloroform, then fractionated by n-hexane. Stress condition was achieved by five days fasting and swimming for 10 mins/day to test animals. UV-Vis spectrophotometer was used to measure the activity of catalase. The rats were divided into 6 groups: normal control, negative control, dose I (10 mg/200 gBB), dose II (40 mg/200 gBB), positive control quercetin as (4 mg/200 gBB) and vitamin E (17.64 mg/kg). The activity of catalase enzyme in the normal control is 29.4 units/ml; negative control is 7.08 units/ml; dose I is 14.42 units/ml; dose II is 14.79 units/ml’quercetin is 28.18 units/ml; and vitamin E is 71.29 units/ml. Our conclusion is the activity of the catalase enzyme rat under oxidative stress can be increased by the n-hexane fraction of H. polyrhizus.

Abstract

Acne can be caused by Staphylococcus epidermidis and Propionibacterium acnes. By using antibiotic for the acne theraphy in a long term period can cause resistance, organ detriment and imunohypersensitivity. Red dragon fruit’s peel (Hylocereus polyrhizus) is one of natural materials which can be used for alternative antibacterial. This study aims to determine the antibacterial activity of n-Hexane fraction of red dragon fruit’s peel against S. epidermidis dan P. acnes by the method of disc diffusion Kirby-Bauer. Red dragon fruit’s peel powder macerated within chloroform, and macerate then fractionated with n-Hexane. Phytochemical screening result red dragon fruit’s peel contains terpenoid and alkaloid. Clindamycin 4µg/disk were used for positive control while DMSO 10% were used for negative control. Data analyzed by using R-Commander program 3.0.3 version.This research show that n-Hexane fraction of red dragon fruit’s peel have antibacterial activity against P. acnes where the averagezone of inhibition obtained from the concentrations from 20; 40; and 80 mg/mL in a row were 9 mm; 10,25 mm; and 10,5 mm. In while, for S epidermidis n-hexane fraction of red dragon fruit’s peel didn’t have antibacterial activity.

Abstract

Free radical from cigarrete smoke cause lipid peroxidation that could be identified by increasing levels of Malondialdehida (MDA). Premna cordifolia leaves known having antioxidant activity in in vitro study. This study aimed to know the effect of P. cordifolia leaves extract on the levels of MDA after exposure to cigarette smoke. The simplicia extracted by maseration using 70% ethanol. Twenty four male wistar rats were divided into six groups, i.e. : control normal, negative control, positive control received cigarette smoke exposure and vitamin E (18mg/kg BW) and three groups received cigarette smoke exposure and P. cordifolia leaves extract with doses 200; 400; 600 mg/kg BW. All rats were treated for 14 days. Measurement of MDA level using Spectrophotometer UV-Vis in 532,2 nm. MDA levels in the normal group was 0.0094±0.080 ppm; negative group was 13,733±2,829 ppm; positive group was 0,051±0,045 ppm; dose 200; 400; 600 mg/kg BW were 9,142±2,364; 0,120±0,019; 0,053±0,041 ppm. The MDA level was analyzed by the Kruskal-Wallis test and following by Mann-Whitney test. The result showed that MDA level of group receiving P. cordifolia leaves extract significantly (p < 0,05) lower than negative control group. These result show P. cordifolia leaves extract can reducing the MDA level on the rats that exposed by cigarette smoke.

Abstract

The excessive production of free radicals causes oxidative damage and degenerative diseases. Whereas, catalase is a macromolecule with a specific activity, but Malondialdehyde (MDA) is a small molecule with no activity. The peels of red dragon fruit (H. polyrhizus) are potential to be natural exogenous antioxidants. This research aimed to determine the effects of methanol fraction from H. polyrhizus peels on catalase activity and MDA level. The peel was macerated using chloroform and fractionated using methanol. The stress condition was done through five-day fasting and ten-minute swimming per day. The activity of catalase and MDA was measured using UV-Vis spectrophotometer. Thirty five rats were divided into 7 groups: normal, negative, dose I (5 mg/200gBW), dose II (10 mg/200gBW), dose III (20 mg/200gBW), positive control (quercetin) (4 mg/200gBW) and vitamin E (17.64 mg/kg). The activity of catalase in the normal, negative, vitamin E, and quercetin, dose I, II, and II respectively were: 28.37; 7.8; 24.85; 34.33; 59.92; 28.18; and 70.85 Unit/mL. The average MDA level in the groups respectively were: 0.042; 0.051; 0.034; 0.042; 0.037; 0.033; 0.030 μg/mL. The result of One Way ANOVA test showed that the catalase activity in dose III was significantly different than normal group; the MDA levels in all groups were significantly different than normal group. This research revealed that methanol fraction from H. polyrhizus peels is a potent antioxidant for its capacity to increase the activity of catalase and reduce the MDA level in rats with oxidative stress.

Abstract

The drying process can affect the content of seconder metabolites such as flavonoids and phenols in plants. The content of flavonoids and phenols plays an important role in the antioxidant activity of a plant. This study aims to determine the effect of drying methods using sunlight and oven on antioxidant activity, total flavonoids, and total phenols. Measurement of antioxidant activity using the DPPH method with vitamin C as a standard, total flavonoid levels using quercetin as a standard, and total phenol using gallic acid as a standard were measured using a UV-Vis spectrophotometer. The results of antioxidant activity (IC50)by sun drying and oven respectively were 123 ppm and 152 ppm, total flavonoid content by sun drying and oven respectively were 22.5% and 20.698%, and total phenol content by sun drying and oven respectively amounted to 37.35% and 36.648%. The conclusion is that the sun-drying method has antioxidant activity, total flavonoids, and total phenol levels which are greater than the oven-drying method.
