Abstract

DNA based RAPD (random amplified polymorphic DNA) markers have been used extensivelyto study genetic relationships in a number of fruit crops. A wide genetic diversity exists in theJatropha curcas in Indonesia. Three accession of Jatropha curcas were selected to assess geneticrelatedness. Total genomic DNA were extracted and subjected to RAPD analysis using 10 primers.Of these, 27 primers ampified Jetropha curcas genomic DNA . Abstract

Experimental activities is conduct in several stages, that is : somaclonal variation induction incastor oil in vitro culture through the use of PEG and selected plantlet is reproduce in vitro. In vitroselection procedure is done by planting cotyledon explants from castor oil seedlings age 10-14 daysin nutrient medium MS+BAP+IBA. Seed sterilization is done using Tween 20, Benlate, 90% alcohol,50% chloroc and sterile water.After callus is grown, callus will be sub-cultured into nutrient mediumMS+BAP with PEG addition. Selected callus will be transferred into nutrient medium MS + potassiumpantotenat + active coal until it shows root growth.Result of experiment has shown that the higher PEG concentration will suppress thegrowth of castor oil callus. In PEG concentration 15% the mortality rate of castor oil callus (LD) is64,29% and for PEG concentration 20% the mortality rate of castor oil callus is 92,86%. For thenext phase is in vitro selection process through treatment of 15% PEG toward all seven accessions.Result of this experiment has revealed that average mortality rate of callus in 15% PEG treatment is87,64%. HS-49 has shown the lowest percentage rate of response (79%) in 15% PEG treatment,while the highest percentage rate of response occurs for SP-8 (93%). Callus that succeeding growthfrom PEG selection medium will be reproduce in regeneration medium until bud is formed. Buddingconstruct percentage is range between 28,57% (accession HS-49 and SP-38) until 37,03% (accessionSM-35). Average budding construct percentage from selected callus in regeneration medium is31,45%. For the next stage of experiment (second year), those bud will be sub-cultured into rootingmedium until forming root and become complete plantlet ready to acclimatize.

Abstract

Nawala Mangsa is a weather station system owned by the Computer Science Faculty of Narotama University. The system has been installed in several locations and is used to monitor the temperature and relative humidity of the site for research purposes. All information from the remote, currently stored in the server online via an internet connection. In this latest version (ver-2.0), Nawala Mangsa had some updates, including the use of Raspberry Pi 3 boards, adding a sensor to measure sunlight illumination (TSL2561), and replacing temperature and humidity sensor with BME280 so it can also measure air pressure. The using of a board that equipped with Bluetooth version 4.1, which is Bluetooth Low Energy (BLE) ready, make this version of this Nawala Mangsa can also broadcast the data that are read by the sensor through BLE, in addition to sent to the server over the TCP/IP network. By this way, so the weather station data can be directly read by the mobile device that equipped with BLE observer.

Abstract

ABSTRACTThe objective of this research was to obtain genetic relationshipamong 13 Jatropha curcas L. accession plants based on Random Amplified Polymorphic DNA marker. This experiment used 13 accessionsof J. curcas L. potential to have higher seed productivity, including HS-49,SP-16, SP-38, SP-8, SM-33, SP-34, SM-35, IP-1A, IP-1M, IP-1P, IP-2A,IP-2M, and IP-2P. Polymerase Chain Reaction (PCR) was performedusing 10 selected primers of RAPD markers (OPA 2, OPA 9, OPA 13,OPA 15, OPA 18, OPA 19, OPA 20, OPF 8, OPF 10, and OPF 15). PCRproduct was used to determine genetic distance which implemented Un-weighted Pair-Group Method With Arithmetic (UPGMA) procedure andconstructed phylogeny trees using Numerical Taxonomy and MultivariateSystem (NTSYS) software version 1.8. The confidence level of UPGMAwas then tested by Boostrap using WinBoot program. Ten primers used inthis research were able to be applied in genomic DNA of J. curcas L. plantwhich had resulted about four (OPA 19) to ten band numbers (OPA 9)with the band size around 72-1,078 bp. However, OPA 13 primer was notable to give different band size. Genetic relationship analysis has foundtwo main groups, firstly accession plants consisted of HS-49, SP-16, SP-18, SP-8, SM-33, SM-35, and SP-34 (coefficient 0.8). In this group, SP-38clustered with SP-8, and SM-33 with SM-35 (coefficient 0.91). In thesecond group, the accessions consisted of IP-1A, IP-1M, IP-1P, IP-2A, IP-2M, and IP-2P (coefficient 0.78). In this group, accession of IP-1Aclustered with IP-1M (coefficient 0.85), IP-1P with IP-2M (coefficient0.87), and IP-2A with IP-2P (coefficient 0.90). Abstract

Indoor agriculture has begun in urban areas. With the narrowness of land and the model of vertical house development, makes this method of indoor agriculture has become a trend in several big cities in the world. Meanwhile, the one that is always needed by every plant is photosynthesis, and every natural photosynthesis of plants continually requires abiotic components of visible light from sunlight. That's why the indoor agriculture requires a replacement source of the sun with artificial sunlight. We can make this artificial sunlight from several light sources, such as incandescent lamps, compact fluorescent lamps (CFL), or the latest with Light Emitting Diode (LED). In this paper, we measured the intensity of light generated from several LEDs with some radiation distance to obtain the optimal energy for plants photosynthesis.

Abstract

ABSTRAKPenelitian ini dilakukan untuk memanfaatkan suatu tanaman air Azolla sp. yang memiliki mikrosimbion mikroalga (ganggang) Anabaena azollae., yang dikembangkan sebagai formula imbuhan pakan (feed additive) pada pakan ayam pedaging (broiler). Formula imbuhan pakan ini adalah untuk pakan komplit ayam pedaging, baik fase starter maupun finisher. Hasil penelitian menunjukkan bahwa pemberian imbuhan pakan Azolla sp. yang bersimbiosis dengan A. azollae pada pakan jadi merk Malindo yang diujikan untuk ayam pedaging strain Hubbard, baik fase awal (starter) dan kemudian fase akhir (finisher) memberikan performa Pertambahan Bobot Badan Harian (PBBH) ayam broiler strain Hubbard sebesar 52,42 gram/ekor. Sedangkan pencapaian bobot badan akhir rata-rata sebesar 1,880.95 gram/ekor. Baik pertambahan bobot badan harian maupun pencapaian bobot badan akhir secara umum masih dalam kisaran sebagaimana disarankan, demikian pula konsumsi pakannya. Tetapi, kandungan kolestrol daging broiler perlakuan menunjukkan penurunan hingga perlakuan level 2 %. Berdasarkan hasil penelitian maka dapat disimpulkan bahwa Azolla sp. yang bersimbiosis dengan mikroalga A. Azollae dapat digunakan sebagai imbuhan pakan pada pakan komplit ayam pedaging dengan tidak memberikanefek terhadap penurunan palatabilitas maupun metabolisme, dan juga sumber bahan pakan ini merupakan non konvensional, sehingga hal ini merupakan potensi yang dapat dikembangkan untuk masa mendatangkan dengan meningkatkan efisiensi pakan dan kemampuan produksi ayam broiler.Kata kunci : Azolla sp.; Anabaena azollae; broiler; Hubbard; imbuhan pakan

Abstract

Jarak pagar (Jatropha curcas L.) merupakan salah satu komoditi perkebunan tropis yang memiliki potensi sebagai  pengganti bahan bakar minyak. Biji jarak pagar mengandung kadar minyak cukup tinggi dan sangat potensial sebagai bahan baku biodiesel.  Kegiatan penelitian ini bertujuan mendapatkan informasi tentang tingkat produksi biji, kualitas crude jatropha oil dan biodiesel  J.curcas Linn. sebagai sumber biofuels. Penelitian dilaksanakan di Desa Kedung Pengaron Pasuruan dan Laboratorium Teknologi Minyak Nabati Universitas Muhammadiayh Malang. Penelitian menggunakan Rancangan Acak Kelompok empat ulangan. Hasil pengamatan menunjukkan bahwa genotip J.curcas berpengaruh secara signifikan terhadap rerata jumlah buah per tanaman, jumlah biji per tanaman, berat kering biji per tanaman dan kadar minyak biji. Jumlah buah per tanaman dan berat kering biji per tanaman yang dihasilkan genotip JCUMM (JCUMM-5, JCUMM-6, JCUMM-7, JCUMM-18) lebih tinggi daripada kontrol IP3A dan IP3P. Jumlah biji per tanaman berkisar 91,3 (IP3P) hingga 236,6 (JCUMM-5). Berat  kering 100 biji dari berbagai genotip J.curcas berkisar antara 64,1 g (JCUMM-6) hingga 69,7 g (JCUMM-5) sedangkan kadar minyak biji berkisar antara 39,5% (JCUMM-6) hingga 57,7% (JCUMM-7). Hasil uji esterifikasi menunjukkan hasil Free Fatty Acid dari crude jatropha oil berkisar antara 0,2% (JCUMM-18) hingga 1,97% (IP3A). Seluruh genotip memiliki kadar FFA kurang dari 2% sehingga masih layak dipergunakan sebagai bahan biodiesel. Nilai bilangan asam yang diperoleh antara 2,9 mg-KOH/g (JCUMM-6) hingga 6,40 mg-KOH/g (IP3A). Sedangkan bilangan Iodine diperoleh hasil antara 94,5 g I2/100g (JCUMM-5) hingga 102,4 g I2/100g (JCUMM-6)

Abstract

Indonesia merupakan negara dengan potensi kopi terbesar ke empat di dunia, selain brazil, Vietnam dan kolombia, berdasarkan data yang dihimpun oleh BPS 2019 menyatakan bahwa produksi kopi meningkat tiap tahunnya dan pada tahun 2019 mencapai 729,1 ribu ton dengan nilai ekspor produk kopi olahan senilai USD 610,89 Juta. Nilai tambah kopi bisa dikembangkan dari produk hilir dimulai dari proses budidaya yang dihasilkan di pedesaan kemudian dikonversi menjadi kopi bubuk skala industri. Diperlukan alternative solusi pendampingan produk unggulan desa yaitu kopi agar dapat menjadikan kopi dipedesaan berkembang melalui agrowisata. Hal ini  memberikan nilai tambah kopi dan wawasan terhadap penikmat kopi. Pengembangan agrotourism dikawasan kopi juga menjadikan konsep ini sebagai panggung transaksi kopi. Desa kaumrejo merupakan desa pada kawasan wisata namun pengembangan produk unggulan desa belum optimal salah satunya kopi. Produksi kopi di desa kaum rejo sangat berpotensi yaitu dengan luas lahan produksi 145 ha dan hasil produksi sebesar 29 ton/ha. Hal ini mungkin tidak banyak diketahui bahwa kopi ngantang tidak kalah dengan buah durian yang merupakan produk unggulan daerah. Kopi yang terdapat disana sekitar 70 persen merupakan varietas robusta, dan 30 persen lainnya adalah Arabica, excels dan liberika. Oleh karena itu diperlukan pendampingan para petani kopi agar dapat menjadikan kopi produk unggulan daerah selain durian melalui pengembangan agrotourism di desa kaum rejo, agrowisata pada prisnsipnya merupakan integrase antara sistem pertanian dan agro industry ke dalam pawisata yang memiliki daya Tarik bagi wisatanwan. Kawasan agrowisata pada perkebunan kopi perlu dirancang beberapa zonasi yang terhubung secara sistemik membentuk agregat kesatuan. adapun metode pelaksanaan tersebut dimulai dari sosialisasi, pelatihan dan pendampingan serta evaluasi program yang di berikan untuk pengembangan agrotourism kopi berbasis literasi tematik dan deversivikasi produk unggulan daerah di desa kaumrejo.

Abstract

This community service program aims to enhance the understanding of Vocational High School (SMK) students in sustainable agriculture, particularly in producing bacterial consortiums of Biofertilizer and Bioprotectant. Situation analysis reveals that SMK students face challenges in accessing training, limited resources and facilities, and a lack of awareness regarding the importance of sustainable farming practices. The program presents strategic solutions, beginning with developing a comprehensive learning module on sustainable agriculture and utilizing bacterial consortiums, biofertilizers, and bioprotectants. This module is designed to assist students in comprehending the concepts, benefits, and practical applications of sustainable farming. The solution also includes practical field training, especially in the propagation of bacterial consortiums, utilizing IoT temperature sensor DS18B20 to monitor the propagation process. The anticipated outcome of this program is an improvement in sustainable agricultural literacy among SMK students. The expectation is that they will better understand the concepts of sustainable agriculture and the implementation of bacterial consortiums, biofertilizers, and bioprotectants. Additionally, students are expected to acquire practical skills in the propagation of bacterial consortiums. Thus, the program aims to positively contribute to developing students' skills and encourage active participation in sustainable agriculture.
