Abstract

The incessant socialization of the government regarding health protocols to a new life order has resulted in most Indonesians becoming introspective in maintaining cleanliness. However, some of the community also experienced panic, so that they acted unnaturally by buying up more goods than they needed, including hand sanitizers. This is one of the reason the price of hand sanitizers has increased. That’s why people need to know how to make hand sanitizers so that they are no longer worry about the scarcity of hand sanitizers and can make their own hand sanitizers. The purpose of this activity is to meet the needs of residents for hand sanitizer as an antiseptic with WHO standards. The method used in training in making hand sanitizers in a direct demonstration. Demonstrations are also supported by a PowerPoint and video that is displayed repeatedly with the help of an LCD projector. This was done to repeat the key points of the demonstration sp that a good level of understanding was achieved by the citizens. Then the Citizen representativeswere asked to repeat the demonstration so that the activities of seeingm listening and practicing the knowledge  of making hand sanitizers were completed. Then at the end of the event, an evaluation was carried out using the discussion or question and answer method. The results of the training in making hand sanitizers have helped meet the needs of residents for hand sanitizers, helped residents to be more independent in meeting their needs for hand sanitizers, and increased knowledge and skills of residents regarding making hand sanitizers as WHO standard antiseptics

Abstract

The Highly Pathogenic Avian Influenza (HPAI) H5N1 virus caused extraordinary incidents in several countries, especially Asia, Africa, and Europe. The disease's fatality rate caused by the H5N1 virus reaches 60%. This incident raised concerns about a wider HPAI H5N1 virus pandemic if effective human-to-human transmission occurred. Therefore, the World Health Organization (WHO) calls on all countries to prepare themselves for a pandemic that may occur. One of the strategies in preparation for this pandemic is to develop a vaccine. So far, vaccines are effective in disease prevention efforts because they can work to trigger a natural immune response and induce a memory response against the pathogen in question. Several strategies are needed to develop an effective influenza vaccine in carrying out vaccine development research and in-depth understanding of (i) virology, (ii) virus life cycle, (iii) host immune response to infection, (iv) pathology, and (v) HPAI H5N1 vaccine development strategy. Until now, many HPAI H5N1 vaccines have been developed, from vaccines developed using conventional methods to alternative vaccines using more modern methods. The results of the development of the vaccine are expected to provide provisions for a possible pandemic caused by the HPAI H5N1 virus. Keywords : Influenza, virus, vaccine

Abstract

Human Immunodeficiency Virus/Acquired Immunodeficiency Syndrome (HIV/AIDS) cases continue to increase and are becoming a top global public health problem. When weight loss is more than 10% below the previous body weight, the relative risk of death is going to increase almost 6-fold. A viral load checkup is a test to measure the amount of HIV in the blood. The increase in nutritional needs and tissue catabolism is caused by the increase in viral load in the body. Malnutrition screening is necessary to evaluate nutritional status so that this treatment can be recommended if the patient is malnourished. This research is an analytical observational study with a cross-sectional design. Analysis was carried out on 109 people living with HIV who received treatment at RSUD Dr. M Yunus Bengkulu. The independent variable in this study is the viral load level, while the dependent variable is screening for malnutrition and the nutritional status of people living with HIV/AIDS—data analysis using the Spearman Rank test. The study results showed no correlation between viral load levels and nutritional status with a value of p=0.243 (p>0.05), and there was a correlation between viral load levels and malnutrition screening with p≤001. The viral load of HIV patients at Dr. M Yunus Bengkulu does not correlate with nutritional status, but it correlates with malnutrition screening.

Abstract

Conventional cancer therapies, such as surgery, chemotherapy, and radiation therapy, often produce limited results and negatively affect patients' quality of life. In recent years, immunotherapy based on immune checkpoint inhibitors (ICIs) has emerged as a promising new approach. Immune checkpoint molecules, such as PD-1, PD-L1, and CTLA-4, play a crucial role in suppressing the immune response to tumors, making them primary targets in ICI therapy. This study examines the mechanisms of immune checkpoints, the efficacy of ICI therapy, and the remaining challenges in clinical practice. The review was conducted through a literature review of scientific publications and the latest clinical trial results. The results demonstrate the efficacy of ICIs, such as ipilimumab and pembrolizumab, in improving overall and progression-free survival rates across various types of cancer, particularly melanoma and non-small cell lung cancer. Combining two ICIs has been shown to have higher synergistic effects than monotherapy, as demonstrated in the CheckMate 067 and KEYNOTE-024 clinical trials. Despite its promise, this therapy is not fully effective in all patients due to immune resistance, autoimmune side effects, and the complexity of tumor heterogeneity and the immune microenvironment. Therefore, combination therapy strategies, dose adjustments, schedule modifications, and personalized approaches based on biomarkers are needed to optimize clinical benefits and expand the scope of therapy. In conclusion, ICIs hold great potential for future cancer therapies and are being developed to be more effective and personalized.

Abstract

Background: Genetic and environmental factors influence the onset and progression of type 2 diabetes mellitus (T2DM). Variants affecting peroxisome proliferator activated-receptor gamma (PPAR-γ) and potassium, inwardly rectifying channel subfamily J member 11 (KCNJ11) may alter insulin secretion and sensitivity. This study investigated the association between PPAR-γ and KCNJ11 gene expression and the risk of T2DM among first-generation (F1) offspring of T2DM patients of Lembak ethnicity in Bengkulu, Indonesia. Methods: A cross-sectional observational study was conducted from July to September 2024, recruiting 60 unrelated participants aged 18–40 years, all of whom were exposed to the neron tradition (high-sugar consumption). Gene expression of PPAR-γ and KCNJ11 was determined using real-time quantitative PCR (qRT-PCR).Results: F1 offspring of T2DM patients (n=30) had significantly (p<0.05) higher body weight (66.89±15.62 kg; p=0.008), body mass index (BMI) (23.55±3.46 kg/m2), HbA1C (6.55±1.25), random blood glucose levels (median 131 [75–371] mg/dl), and duration of neron consumption (median 3 [1–12] years) compared with controls (n=30). PPAR-γ expression differed significantly between group (1.60±2.91 vs 4.23±8.54; p=0.009), whereas KCNJ11 expression did not (0.79±0.76 vs 1.37±0.89; p=0.124). Multivariate analysis revealed no correlation between gene expression and the patients characteristic (p>0.05). Linear regression showed 30.4% of PPAR-γ and 45.8% of KCNJ11 variability. Conclusions: PPAR-γ expression is associated with T2DM onset among Lembak F1 offspring, whilst KCNJ11 expression is not. Multiple genetic and environmental factors likely contribute to disease progression. Screening for PPAR-γ expression may support interventions targeting insulin sensitivity and lipid metabolism.
