Abstract

The objectives of this experiment were to determine response of epycotyl of nine peanut cultivars against polyethylene glycol (PEG 6000) induced stress under in vitro conditions, effective concentration of PEG to inhibit growth and development of epycotyl, evaluate tolerance of the cultivars against PEG stress, and evaluate changes in total proline content due to PEG stress. Growing epycotyls from peanut seeds (TDK) or from embryo axis (TTK) were planted on liquid MS-0 medium containing PEG 6000 (0%, 5%, 10%, 15%, and 20%). Growth, development, and the tissue damage score of the epycotyl were observed after six weeks. Total content of proline were analyzed for stressed and non stressed epycotyl to determine effect of PEG stress on proline accumulation. Results of the experiment indicated that addition of PEG 6000 in to MS-0 medium inhibited growth and development of peanut epycotyls and increased its total proline content. Addition of PEG 6000 might be used to simulate drought stress under in vitro condition. PEG at 15% concentration was effective for inhibiting growth and development of epycotyl explant. The response of peanut epycotyls against medium containing 15% PEG 6000 might be used as alternative methods for screening peanut tolerance against drought stress. The TDK and TTK might be used as explant, while increased in shoot length (TTK), in leaf number (TDK and TTK), in milted leaf number (TDK and TTK), in root number (TDK) and score of tissue damage (TDK) might be used as criteria for drought tolerance.

Abstract

This study was aimed to evaluate the effectiveness of in vitro selection for identifying PEG insensitive somaclonal variant of peanut somatic embryos (SE). In one of the experiments,  evaluation on responses of four peanut cultivars against selective medium containing polyethylene glycol 6000 (PEG 6000) was conducted and sub-lethal concentration of PEG was determined. In the other experiment, in vitro selection on selective medium containing sub-lethal level of PEG was conducted to identify PEG insensitive SE of peanut. Secondary SE of peanut cv. Badak, Kelinci, Singa, and Zebra were cultured on liquid MS medium supplemented with 16 µM of picloram and 5%, 10%, 15%, or 20% of PEG 6000. Survival of explant, average number of proliferated SE/explant, and total number of proliferated SE after in vitro selection were recorded monthly, up to three months. Sub-lethal level of PEG was defined as one inhibiting more than 95% of the total number of proliferated SE. In vitro selection on medium supplemented with sub-lethal level of PEG 6000 was conducted on at least 4000-5000 SE of peanut cv. Kelinci and Singa. The PEG insensitive SE was identified after subsequent three months of in vitro selection. The results showed that supplementation of PEG 6000 on medium for induction of SE inhibited proliferation of peanut SE. Sub-lethal level was obtained at 15% concentration of PEG 6000. The frequencies of obtaining PEG insensitive SE of peanut cv. Kelinci was 8% to 10%  and for peanut cv. Singa was 10% to 12%. The R0 plants of peanut cv. Kelinci (62 R0 plants) and Singa (48 R0 plants) regenerated from PEG insensitive SE were obtained and grown in the glasshouse to produce R1 and R2 seeds.

Abstract

Penelitian ini bertujuan mendeskripsikan proses pembelajaran biologi di SMA 1 Kandangserang, mengembangkan perangkat pembelajaran dengan pendekatan scientific skill berbasis masalah lingkungan pada limbah produksi tempe-tahu, dan mengevaluasi validitas, efektivitas, serta kepraktisan perangkat pembelajaran tersebut. Metode yang digunakan adalah Research and Development dengan menganalisis kebutuhan kemudian mengembangkan produk dan diuji validitas, efektivitas, dan kepraktisanya hingga diperoleh produk final. Produk yang dikembangkan adalah perangkat pembelajaran dengan pendekatan scientific skill teknologi fermentasi berbasis masalah lingkungan pada limbah produksi tempe-tahu, yaitu meliputi silabus, RPP, bahan ajar, lembar diskusi peserta didik (LDPD), dan lembar penilaian scientific skill. Hasil analisis menunjukkan perangkat pembelajaran sangat valid, efektif, dan praktis diterapkan. Guru hendaknya menerapkan perangkat pembelajaran dengan pendekatan scientific skill teknologi fermentasi berbasis masalah lingkungan pada limbah produksi tempe-tahu. The research aims to describe biology learning process in SMA 1 Kandangserang detaily, expand learning process with scientific skill approach problem based learning, and evaluate the validity, effectivity, and practical the learning equipment. The method in this research is Research and Development, by analyzing the need and then developing a product and find the validity, effectivity, practically until gets the final product. The product that been developed is learning equipment with scientific skill technology of fermentation that principle with surrounding problems, consist of the syllabi, lesson plan, teaching materials, student discussion papers, and scientific skill’s evaluation  paper. The learning equipment is valid, efective and has been used. Learning equipment using scientific skill inenvironment problem based should be applied in making of tempe-tahu by the teacher.

Abstract

Materi pembelajaran bermuatan SETS dan model pembelajaran PBL dapat digunakan untuk memecahkan permasalahan ekologi dan meningkatkan pemahaman materi ekologi. Penelitian ini bertujuan untuk menentukan validitas, kepraktisan, dan efektivitas bahan ajar serta mendiskripsikan karakteristik bahan ajar yang dikembangkan. Metode  penelitian ini adalah penelitian dan pengembangan (research and development). Desain penelitian uji coba skala luas menggunakan pretest-posttest control group design dan posttest-only control design. Hasil validasi bahan ajar yang dilakukan oleh ahli pendidikan, ahli materi dan praktisi pendidikan dengan rata-rata nilainya 91,67 (amat baik). Kepraktisan bahan ajar menurut siswa sebesar 91,00 (sangat baik) dan menurut guru sebesar 98,50 (sangat baik). Berdasarkan hasil belajar kognitif, sikap dan keterampilan modul yang dikembangkan efektif. Karakteristik modul ekologi hasil pengembangan yaitu materinya diawali dengan permasalahan sehari-hari yang ada disekitar kita. Permasalahan ditutup dengan pertanyaan yang mengarahkan ke pemahaman tentang SETS. Hasil belajar kelas kontrol dan kelas perlakuan terjadi perbedaan yang signifikan, baik pada penilaian kognitif, sikap dan keterampilan. T Learning materials contain SETS and PBL learning model can be used to solve the problems of ecology and to increase the understanding of ecological materials. This research is aimed to determine the validity, practicality, and effectiveness of learning materials and to describe the characteristics of the developed learning materials. The research method is a research and development. Design of large scale trials using a pretest-posttest control group design and posttest-only control design.The results of the validation materials made by education experts, materials expert and practitioners of education have average mark of 91.67. The practicality of materials according to the students of 91.00 and according to the teachers of 98.50. Based on the resuts of cognitive learning, attitudes and skills of developed modules, it works effectively. Characteristics of ecological module from that development is that the material is staryed from daily life problems around us. The issue is closed with a question leading to an understanding of the SETS. The results of study control class and experiment occurs a significant difference, both on the cognitive assessment, attitudes and skills.

Abstract

Asesmen merupakan bagian integral dari proses pembelajaran sains. Aspek-aspek yang diukur dalam sains menentukan strategi pembelajaran dan jenis asesmen. Selama ini, aspek yang diukur masih terbatas pada kemampuan kognitif dan belum mengarah pada penguasaan literasi sains. Tujuan penelitian adalah untuk mengembangkan instrumen asesmen autentik berbasis literasi sains pada materi sistem ekskresi berdasarkan analisis kebutuhan disekolah. Jenis penelitian yang digunakan adalah Research and Development, dengan desain penelitian pretest posttest control group design. Subjek penelitian ujicoba skala kecil adalah 10 siswa anggota KIR SMAN 3Semarang, dan ujicoba skala luas adalah 68 siswa kelas XI IPA SMAN 3 Semarang. Produk penelitian berupa peer assessment, penilaian unjuk kerja, proyek, portofolio, sikap, dan tes. Hasil pengembangan instrumen asesmen autentik berbasis literasi sains menunjukkan bahwa instrumen asesmen valid, reliabel, efektif, dan tingkat kepraktisannya tinggi. Hal ini ditunjukkan oleh rata-rata validitas menunjukkan kriteria sangat tinggi. Keefektivan instrumen ditunjukkan dengan perubahan positif hasil belajar siswa, peningkatan indeks gain berada pada kisaran sedang, dan adanya pengaruh positif penerapan instrumen asesmen terhadap hasil belajar. Kepraktisan instrumen asesmen ditunjukkan oleh adanya respon positif siswa dan guru. Assessment is an integral part of the learning process of science. Measured aspects in determining science learning strategies and types of assessment. During this time,aspects ofthe measured is limited to cognitive abilities and not lead to mastery of science literacy. The research objective is to develop authentic assessment instrument based on material science literacy excretion system based on analysis of school needs.The type of the research was Research and Development, and pretest posttest control group design. The limited-scale subjects were 10 scientific group students of SMAN 3 Semarang, and the implementation-scale subjects were 68 students of Grade XI IPA of SMAN 3 Semarang. Product research is peer assessment, performance assessments, projects, portfolios, attitudes, and test.The research analysis showed that the science literacy based authentic assessment instrument developed in the study was valid, reliable, effective, and practical. This was shown from the very high score of the validity mean. The effectiveness of the instrument could be shown from the positive changes on the students’ study outcomes, with the gain index in medium rate, and the positive impact of the instrument implementation on the students’ study outcomes. The assessment instrument was considered practical based on the positive responses from students and teachers.

Abstract

Guru Biologi Madrasah Aliyah Kota Semarang yang mengajar Prakarya dan Kewirausahaan (PKWU) mengeluhkan semakin menurunnya kreativitas peserta didik. Mereka cenderung mereplikasi atau menduplikasi karya kakak kelas, sehingga nilai kreativitas dan inovasinya rendah. Kecenderungan ini harus segera diminimalkan. Teknik eco-print merupakan salah satu jawaban terhadap permasalahan tersebut karena hasilnya bersifat unik atau ekslusif. Oleh karena itu perlu dikenalkan dan dilatihkan keterampilan teknik eco-print kepada guru-guru Biologi MA Kota Semarang. Tujuan pengabdian pada masyarakat adalah mengenalkan, menambah pengetahuan dan keterampilan teknik eco-print pada guru-guru Biologi MA Kota Semarang. Mitra kegiatan ini adalah MGMP Biologi MA Kota Semarang dan diikuti oleh 10 guru terpilih. Kegiatan pengabdian pada masyarakat diawali dengan pretest, pengenalan tentang teknik eco-print, praktik teknik pounding, praktik teknik steaming, fiksasi warna, diskusi, posttest, dan tugas mandiri. Dari segi pengetahuan diperoleh N-gain= 0,502 (kategori sedang). Semua peserta aktif dalam kegiatan dan mampu menghasilkan eco-print dengan teknik pounding dan steaming menggunakan kain Primisima, dan berhasil membuat totebag eco-print dengan teknik pounding secara mandiri. Hasil tersebut menunjukkan bahwa dari segi pengetahuan teknik eco-print peserta mengalami peningkatan. Dari segi keterampilan semua peserta mampu membuat kain eco-print dengan teknik pounding maupun steaming. Secara mandiri, peserta sudah mampu membuat totebag eco-print dengan teknik pounding. Keterampilan peserta masih harus ditingkatkan dengan secara mandiri atau berkelompok dengan mencoba berbagai variasi untuk melatih kreativitas serta inovasi, sehingga mampu menularkan pengetahuan dan keterampilannya kepada para siswa.

Abstract

Lemongrass (Cymbopogon nardus (L.) Randle) plantlets are interesting to be used as tissue culture-based souvenir. The success of plantlet propagation through photomyxotrophic culture is influenced by several factors, including sucrose concentration and light intensity. The research aimed to analyze the effect of sucrose concentration and light intensity on the growth of lemongrass plantlets. This experimental research used randomized block factorial design with two factors, namely sucrose concentration (0 g/l, 10 g/l, 20 g/l, 30 g/l) and light intensity (2000 lux, 3000 lux, 4000 lux, 5000 lux). The variables observed were some parameters of growth and development of plantlet. The data were analyzed with two-way Analyses of Variance and Duncan Multiple Range Test. The results showed that sucrose concentration had significant effect on the number of shoots and leaves of plantlet. The light intensity and the interaction of both factors had also significant effect on the number of shoots and leaves. The most optimal treatments were sucrose of 20 g/l and light intensity of 3000 lux.  The results of this study innovate the tissue culture technique using photomyxotrophic system in order to produce sterile lemongrass plantlets in large quantities.

Abstract

issue cultures that have passed callus phase can induce somaclonal variation, of which the intensity was influenced by the addition of selective agents to culture media. Somaclonal variations of peanut plant obtained from in-vitro cultured and in-vitro selected somatic embryos using PEG was not yet understood. The objectives of this research were 1) to identify the qualitative variants of peanut plant var. Kelinci obtained from in-vitro cultured and in-vitro selected somatic embryos using PEG, 2) to estimate the factors that control the qualitative variants. The non-selected and the selected variant somatic embryos of peanut were germinated and cultured. From fertile R0 lines, sufficient number of R1 and R2 progenies were grown for evaluation. The results showed that phenotypic variations of qualitative characters can be observed among R0, R1 and R2 generations of somaclonal lines. Variant phenotypes of qualitative characters can be observed, these include wide branching, excessive branching, leaf variegation, leaflet number abnormality, leaf pointed tip, ‘rosette’ leaf, complete sterility and partial sterility. The data indicated that wide branching, excessive branching, leaflet number abnormality, partial sterility and complete sterility were genetically controlled, while variant phenotypes of ‘rosette‘ leaf, leaf variegation, and leaf pointed tip were epigenetically controlled. Abstract

Feronia limonia (L.) Swingle better known as wood apple is an identity plant of Rembang Regency, Central Java Province, Indonesia. However, the population is very limited. In order to identify the conservation efforts, this ethnobotany study was aimed to determine traditional knowledge on the existence, economic value, utility, breeding technique and conservation of wood apple. The survey was conducted on a total of 102 local people from 10 sub-districts which were determined by proportionally stratified random sampling. Data were collected by questionnaires and individual interviews and then were analyzed descriptively. The results showed that all of respondents knew the figure of wood apple, but only 79.50% understood its status as an identity plant. Most of respondents (96%) stated that the fruits have low economic value, 80.39% respondents have utilized ripe fruit as ingredients of beverages and stems as firewood. This utilization was only equal to 29.72% compared to the potential usability based on research results. It was concluded that the traditional knowledge about the existence, economic value, and breeding technique were good and on the contrary, the knowledge about the plant’s usability was bad. The results will form a basic knowledge for in-depth studies to provide a government policy of Rembang Regency in order to develop an efficient strategy of management and conservation of wood apple as plant identity.

Abstract

The population of Mesua ferrea L. in Banyumas Regency, Central of Java Province is very low. There are some factors affecting the quantity of plant population, particularly traditional knowledge of local community about the plant. The aim of this study was to describe the knowledge of Banyumas community about the existence, economic value, usefulness, and propagation technique of M. ferrea. The study was done by field survey on a total of 146 residents from 10 districts which were determined by proportionally stratified random sampling. Data obtained from questionnaires and interviews were analyzed by percentage descriptive. The results revealed that only 9% of respondents were familiar with the M. ferrea and knew its status as plant identity of Banyumas Regency. Most of the respondents did not understand the economic value, only 8% of respondents understood the usefulness, 2% of respondents had utilized the timber as craft material, and 3% understood the propagation technique. The utilization was very low, equal to 14.45% compared to the potential usefulness. Most of the respondents (98%) thought that conservation efforts should be undertaken. This study found that the traditional knowledge of M. ferrea in Banyumas was low. This result should be used as a basic information for developing conservation program of M. ferrea.
