Abstract

AbstractSubclinical mastitis is an inflammation of the udder that often attacks livestock, especially milk-producing livestock. The impact of the subclinical mastitis losses is quite large, including a drastic decrease in the quantity and quality of milk, but no pathological changes were found in cows. The Karya Bhakti Village Unit Cooperative (VUC) is a center for milk production in the Kediri Regency which accommodates dairy cows' milk products from people's farms in the Kediri Regency. The low awareness of farmers in the management of livestock health care and hygiene results in low productivity and quality of milk and the high incidence of subclinical mastitis. This encourages counseling about subclinical mastitis. The socialization method used for counseling were carried out by presenting material about subclinical mastitis with lecture techniques (power point slides), videos about the method of early detection of subclinical mastitis using California Mastitis Test (CMT), conducting demonstrations of testing milk samples using the CMT method and discussion about the material that has been delivered as an indicator of the success of counseling. This series of counseling activities is an effort to reduce the incidence of subclinical mastitis through increasing public knowledge and understanding. This activity also has a positive impact by reducing infectious diseases, especially subclinical mastitis in livestock so that the health of livestock and products from livestock will also be guaranteed both in terms of quality and quantity. Keywords: mastitis, subclinical mastitis, cows, Counseling, CMTAbstrakMastitis subklinis merupakan radang ambing yang sering menyerang hewan ternak khususnya ternak penghasil susu.

Abstract

Background: The coliform bacteria group is a group of bacteria used as an indicator of food sanitation, one of which is in dairy products. Objective: This study aimed to determine the relationship of milking techniques with the number of Escherichia coli in raw milk compared to the bacterial contamination limit set by the National Standardization Agency. Methods: The design of this study was a descriptive study. Raw milk samples were taken from Lucky Farm and several traditional dairy farms in the KUD Kertajaya area, Kediri Regency, East Java. This study uses the most probable number (MPN) test. Results: The results showed 11 (73.3%) of raw milk samples using conventional milking techniques exceeding the maximum limit of E. coli bacterial contamination determined by the National Standardization Agency (< 3 MPN/mL). However, all 15 samples, or 100% of raw milk samples using machine milking techniques, were negative for E. coli and met the maximum bacterial contamination limit requirements. Conclusion: The milking techniques affect E. coli contamination limits in raw milk. The milk samples taken using machine milking techniques had better results with MPN values < 3 MPN/mL than those taken using conventional milking techniques.

Abstract

Escherichia coli is a typical flora of an animals' digestive tract. Based on these details, this study was done on the detection and identification of multidrug-resistant E. coli in cattle in Surabaya abattoirs. Each cattle rectum swab sample was streaked into EMB agar media and followed by Gram staining. The IMViC test was used to confirm the presence of E. coli. Based on morphological culture features, Gram staining, and biochemical testing, the sample examination results revealed that 41 samples (41%) of the 100 cattle rectal swab samples that were isolated were confirmed to be positive for E. coli. The profile of antibiotic resistance from the results of the E. coli resistance test to antibiotics showed that out of a total of 41 isolates of E. coli, 12 isolates (29.27%) were detected as resistant to 1 class of antibiotics tested, while 2 isolates (4.88%) were resistant to 2 classes of antibiotics, and 3 isolates (7.32%) were confirmed to be multidrug resistance (MDR).

Abstract

Vero cell culture as a platform for producing viral vaccines is an established and standardized process in vaccine manufacture. Generally, Vero cell culture requires media as a source of nutrition with serum supplementation to provide growth factors. However, the serum has several disadvantages including batch to batch variation and adventitious agent. Therefore, chemically defined serum-free media (SFM) are formulated by using standardized growth factors. Epidermal Growth Factor (EGF) is one of growth factors that showed adequate mitogenic support in serum-free medium system, especially in Vero cells. In this study, SFM-EGF media was compared with serum supplementation media, namely MEM 10% and MEM 5% FBS supplementation. The cell morphology was observed using an inverted microscope and their proliferation was evaluated using a MTT colorimetric-based assay. Vero E6 and Vero CCL-81 cells morphology did not show any morphological changes. Vero E6 and Vero CCL-81 proliferation in SFM-EGF media on day one to four did not show a significant difference compared to MEM 10% or MEM 5% serum supplementation media. However, the OD values of both Vero E6 and Vero CCL-81 cells given SFM-EGF media produced an average value below MEM 10% but higher than MEM 5% FBS supplementation. As such, this study proved that utilizing SFM-EGF could support the proliferation of Vero E6 and Vero CCL-81 cells.
