Abstract

Currently, the effort to invent ideal natural male contraceptives still faces many obstacles, especially due to the fact that ideal contraceptives should meet certain requirements such as: be able to cause total azoospermia; easily used; cause no untoward or toxic effects; not disturbing libido or sexual behavior; and be reversible. As male contraceptives are very limited in types, efforts to develop ideal male contraceptive drugs is crucial; particularly the effort to find alternative effective substances derived from natural resources. The purpose of this study is to investigate whether of young papaya seeds extract has a to inhibit the testosterone. The study used 30 male mice strain Balb-C which were allocated by simple random sampling into two groups: Control Group (P0) consisted of 17 male mice and Treatment Group 1 (P1) consisted of 17 male mice. After 36 days of treatment, control group and treatment group were taken for their blood. The blood was taken for testosterone hormone test using Radioimmunoassay (RIA) technique and the scrotum was taken for obtaining the Leydig cells. The result of t-test analysis showed that the level of testosterone in control group was 104,27 ± 25,79 ng/dl and Leydig cells 129,41±30,92, in treatment group 91,76 ± 30,45 ng/dl and Leydig cells 122,94±20,85.  The difference between control group and treatment group was the decrease of testosterone and Leydig cells. Young papaya seeds extract reduces testosterone and Leydig cells insignificantly.Keywords: Papaya seeds extract, Testosterone hormone, Leydig cel, Male Mice balb/C

Abstract

Background: Caffeine, a methylxanthine derivate, had a role as a phosphodiesterase inhibitor, thereby inhibiting the break down of cAMP and increasing its concentration inside the cell. Aim: This study aimed to assess the effect of caffeine in EBSS and evaluated the increase in motility and the acrosomal reaction of spermatozoa using the swim-up method. Method: This was an experimental study using a pre-post test control group design. This study was carried out at the Sexology and Andrology Clinic, Sanglah Hospital in Denpasar Bali. The samples were sperm specimens of eighteen infertile male or volunteers aged 20-40 years. They were divided into two groups: treatment group (caffeine + EBSS) and control group (EBSS). The data were analyzed statistically by normality test (Kolmogorov – Smirnov Goodness of Fit Test), homogeneity test and paired student’s t test. Result: The caffeine addition in EBSS medium increased the sperm motility significantly from 71.94% to 80.28% (p<0.05) and the acrosomal reaction from 83.33% to 88.89% (p<0.05). Conclusion: The addition of caffeine in EBSS medium increased the motility and the acrosomal reaction of the human sperm.Keywords: Caffeine, EBSS, Sperm motility, Acrosomal reaction, Spermatozoa.

Abstract

Objective: The purpose of this study was to prove the water extract of purple sweet potatoes can increase superoxide dismutase (SOD) gene expression and catalase (CAT) and decrease malondialdehyde (MDA) in vital organs of diabetic rats. Method: This study was randomized experimental design with posttest only control group design. Sample divided into 2 groups: control and treatment groups, respectively 18 diabetic Wistar rats. The control group will be given streptozotocin. The treatment group was given streptozotocin and water extract of purple sweet potato 4 cc/day. This treatment is carried out for 2 months. Result: Water extract of purple sweet potato was proven significantly increased mRNA expression of aorta and kidney SOD but decreased SOD mRNA expression in the liver (p <0.05). This water extract also increases SOD mRNA expression in cardiac but not statistically significant (p> 0.05). Water extract of purple sweet potatoes also found significantly increased renal and cardiac expression of CAT mRNA (P <0.05), but not statistically significant (p> 0.05). Conclusion: Purple sweet potatoes extract to decrease the expression of liver CAT mRNA non-significantly, and decrease MDA in liver and kidney of diabetic Wistar rats significantly (P <0.05).
