Abstract

Penyakit diabetes melitus ditandai oleh hiperglikemia akibat gangguan sekresi insulin atau resistensi insulin, yang dapat menyebabkan komplikasi serius seperti nefropati, retinopati, neuropati, dan penyakit kardiovaskular. Penelitian ini bertujuan untuk meninjau efek antidiabetes dari senyawa bioaktif Sacha Inchi (Plukenetia volubilis L.) dalam pengelolaan diabetes melitus melalui tinjauan sistematis (Systematic Literature Review/SLR) dengan pedoman PRISMA 2020. Berdasarkan analisis terhadap 13 artikel terindeks Scopus dari 2016 hingga 2025, hasil menunjukkan bahwa Sacha Inchi dapat menurunkan kadar glukosa darah melalui mekanisme peningkatan sensitivitas insulin, aktivasi jalur pensinyalan IRS-1  PI3K  Akt, serta perlindungan dan regenerasi sel B pankreas. Selain itu, senyawa bioaktif dalam Sacha Inchi menghambat enzim a glukosidase, a amilase, dan DPP-IV, yang berperan dalam metabolisme glukosa. Aktivitas antioksidan dan antiinflamasi Sacha Inchi juga berkontribusi mengurangi stres oksidatif dan peradangan sistemik, mendukung keseimbangan metabolik, dan kesehatan pankreas. Penelitian menunjukkan bahwa Sacha Inchi berpotensi mengmodulasi mikrobiota usus, yang berperan penting dalam pengaturan metabolisme glukosa dan meningkatkan sensitivitas insulin. Temuan ini mengindikasikan Sacha Inchi sebagai agen fitoterapi alami yang aman dan efektif untuk pengelolaan diabetes melitus. Evaluation of the Potential of Sacha Inchi (Plukenetia volubilis L) in Diabetes Mellitus Disease Therapy: A Systemic Literature Review Abstract Diabetes mellitus is characterized by hyperglycemia due to impaired insulin secretion or insulin resistance, which can lead to serious complications such as nephropathy, retinopathy, neuropathy, and cardiovascular diseases. This study aims to review the antidiabetic effects of bioactive compounds from Sacha Inchi (Plukenetia volubilis L.) in the management of diabetes mellitus through a systematic literature review (SLR) following the PRISMA 2020 guidelines. Based on an analysis of 13 Scopus-indexed articles from 2016 to 2025, the results indicate that Sacha Inchi can lower blood glucose levels through mechanisms such as enhancing insulin sensitivity, activating the IRS-1  PI3K  Akt signaling pathway, and protecting and regenerating pancreatic B-cells. Additionally, the bioactive compounds in Sacha Inchi inhibit enzymes like a-glucosidase, a-amylase, and DPP-IV, which are involved in glucose metabolism. The antioxidant and anti-inflammatory activities of Sacha Inchi also contribute to reducing oxidative stress and systemic inflammation, supporting metabolic balance and pancreatic health. The research shows that Sacha Inchi has the potential to modulate gut microbiota, which plays a crucial role in glucose metabolism regulation and enhancing insulin sensitivity. These findings suggest that Sacha Inchi is a safe and effective natural phytotherapy agent for the management of diabetes mellitus.

Abstract

Penuaan kulit semakin mendapat perhatian, namun pemahaman tentang peran senyawa alami sebagai agen anti-aging masih perlu diperdalam. Squalene, senyawa triterpen hidrokarbon alami (C30H50) yang ditemukan dalam minyak hati ikan hiu, minyak zaitun, amaranth, dedak padi, serta mikroba rekayasa genetik, menarik perhatian sebagai bahan aktif anti-aging alami. Kajian ini bertujuan menganalisis kandungan aktif, mekanisme kerja, bentuk konsumsi, dan bukti ilmiah squalene dalam potensi anti-aging melalui pendekatan systematic literature review. Penelitian ini menganalisis 14 artikel dari bidang kosmetologi, dermatologi, dan bioteknologi farmasi (2016-2025), yang sebagian besar merupakan penelitian eksperimental (in vitro, ex vivo, dan in vivo) serta beberapa studi klinis awal. Hasil menunjukkan bahwa squalene bekerja melalui empat mekanisme utama: aktivitas antioksidan (menetralkan ROS, menekan MMP-1, mengaktifkan jalur Nrf2/ARE), antiinflamasi (menghambat NF-KB dan sitokin proinflamasi), fotoprotektif (mencegah peroksidasi lipid akibat UV), dan regeneratif (menstimulasi TGF-B untuk pembentukan kolagen). Formulasi nanoemulsi dan oleogel berbasis squalene efektif meningkatkan kondisi kulit. Squalene nabati, seperti dari minyak zaitun dan amaranth, menunjukkan efektivitas setara dengan sumber hewani, mendukung keberlanjutan dan etika produksi kosmetik. Squalene berpotensi sebagai bahan aktif anti-aging generasi baru yang aman, efektif, dan ramah lingkungan, namun kajian lebih lanjut diperlukan untuk menilai efektivitas jangka panjang dan dosis optimalnya. The Potential of Squalene in Anti-aging: A Literature Review Abstract Skin aging is becoming an increasingly recognized issue, but understanding the role of natural compounds as anti-aging agents still requires further exploration. Squalene, a natural triterpene hydrocarbon (C30H50) found in shark liver oil, olive oil, amaranth, rice bran, and genetically engineered microbes, has gained attention as a natural anti-aging active ingredient. This review aims to analyze the active content, mechanisms of action, consumption forms, and scientific evidence supporting the anti-aging potential of squalene through a systematic literature review approach. The study analyzes 14 articles from the fields of cosmetology, dermatology, and pharmaceutical biotechnology (2016-2025), mostly experimental research (in vitro, ex vivo, and in vivo) supported by some early clinical studies. The results show that squalene works through four main mechanisms: antioxidant activity (neutralizing ROS, inhibiting MMP-1, activating Nrf2/ARE pathway), anti-inflammatory (inhibiting NF-KB and pro-inflammatory cytokines), photoprotective (preventing lipid peroxidation due to UV), and regenerative (stimulating TGF-B for collagen formation). Squalene-based nanoemulsion and oleogel formulations effectively improve skin conditions. Plant-based squalene, such as from olive oil and amaranth, shows comparable effectiveness to animal sources, supporting sustainability and ethical considerations in modern cosmetic production. Overall, squalene has great potential as a new generation of safe, effective, and environmentally friendly anti-aging active ingredients, though further studies are needed to assess long-term effectiveness and optimal dosages.

Abstract

Hiperlipidemia merupakan faktor risiko utama penyakit kardiovaskular dan masih menjadi penyebab morbiditas serta mortalitas yang tinggi. Sacha Inchi (Plukenetia volubilis L.) mengandung asam lemak esensial omega-3 (ALA), omega-6 (LA), omega-9 (OA), serta tokoferol dan fitosterol yang berperan dalam regulasi metabolisme lipid. Penelitian ini bertujuan untuk meninjau secara sistematis bukti ilmiah mengenai potensi Sacha Inchi sebagai agen antihiperlipidemia berdasarkan hasil penelitian in vitro, in vivo, dan uji klinis yang dipublikasikan pada jurnal terindeks Scopus periode 2016–2025. Metode yang digunakan adalah Systematic Literature Review (SLR) dengan mengacu pada pedoman PRISMA 2020 dan menggunakan basis data Scopus. Dari 25 artikel yang ditemukan, sebanyak 17 memenuhi kriteria inklusi. Hasil analisis menunjukkan bahwa konsumsi minyak Sacha Inchi menurunkan kolesterol total, LDL, dan trigliserida serta meningkatkan HDL melalui penghambatan enzim HMG-CoA reduktase, aktivasi PPAR-a, dan modulasi mikrobiota usus. Senyawa tokoferol dan polifenol juga berkontribusi terhadap efek antioksidan dan antiinflamasi melalui penekanan NF-KB, TNF-a, dan IL-6. Dengan demikian, Sacha Inchi memiliki potensi sebagai fitoterapi alami antihiperlipidemia yang aman dan berpotensi mendukung kesehatan kardiovaskular. As an Antihyperlipidemia Agent: Systematic Literature Review Abstract Hyperlipidemia is a major risk factor for cardiovascular disease and is still a cause of high morbidity and mortality. Sacha Inchi (Plukenetia volubilis L.) contains omega-3 (ALA), omega-6 (LA), omega-9 (OA), as well as tocopherols and phytosterols that play a role in the regulation of lipid metabolism. This study aims to systematically review scientific evidence regarding the potential of Sacha Inchi as an antihyperlipidemia agent based on the results of in vitro, in vivo, and clinical trials published in the Scopus indexed journal for the period 2016–2025. The method used is Systematic Literature Review (SLR) with reference to the PRISMA 2020 guidelines and using the Scopus database. Of the 25 articles found, 17 met the inclusion criteria. The results of the analysis showed that the consumption of Sacha Inchi oil lowered total cholesterol, LDL, and triglycerides and increased HDL through inhibition of HMG-CoA reductase enzyme, activation of PPAR-a, and modulation of the gut microbiota. Tocopherol and polyphenol compounds also contribute to antioxidant and anti-inflammatory effects through the suppression of NF-kB, TNF-a, and IL-6. Thus, Sacha Inchi has the potential as a natural phytotherapy for antihyperlipidemia that is safe and has the potential to support cardiovascular health.
