Abstract

Rheumatoid arthritis (RA) is a chronic form of persistent inflammation. Meanwhile, Substance P is the most associated neuropeptide in neurogenic inflammation and hyperalgesia commonly found in chronic pain. Substance P act by binding to neurokinin-1 receptor. The present study was conducted to evaluate the effect of neurokinin-1 receptor antagonist (CP-96,345) on Adjuvant Induced Arthritis rat model, induced by Complete Freund's Adjuvant (CFA). The objective is to attenuate neurogenic inflammation which in turn will increase the latency time of hyperalgesia response, decreases neurokinin-1 receptor expression, and inhibits the development of RA in AIA rat model. Rats were intra-articularly injected with CFA 1 hour after the administration of CP-96,345 either by 0.63 µg/gr; 1.25 µg/gr; or 2.5 µg/gr also intra-articularly. Caliper measurements and hot-plate test were performed on day 0, 3, 5, 7, 9, 11, and day 13. Expression of neurokinin-1 receptor in joint tissue were evaluated by immunohistochemistry, and RA progress in joint tissue were observed hystopathologically. CP-96,345 at 2.5 µg/gr significantly increases the latency of hyperalgesia response time on CFA induced rats (p=0.044) and decreased the neurokinin-1 receptor expression in joint tissue (p=0.029) compared to CFA induced rats. There was no significant difference for caliper measurements and RA progress between CFA incduced rats and treated group. Conclusively, CP-96,345 increases the latency of hyperalgesia response time and decreases the NK-1 receptor expression in rat joint but could not inhibit RA progression.

Abstract

Rheumatoid arthritis (RA) is an autoimmune diseases which is characterized by chronic inflammation of the synovial tissue in joints. This research was designed to investigate the effect of alpha lipoic acid as antioxidant on rats with complete freund's adjuvant (CFA)-induced RA by intra articular injection of complete freund's adjuvant (CFA). ALA was administered orally once a day for 7 days at 30, 60 and 120 mg doses a week after CFA injection. The severity of arthritis was evaluated by joint diameter and latency time on thermal stimulation. Joint diameter and latency time on thermal stimulation will measured on day 0, 3, 5, 7, 10, 12 and 14. Measurement of malondialdehyde (MDA) level in plasma was performed using thiobarbituric acid (TBA) method to assess lipid peroxidation. Histology of joint was examined by microscope following hematoxylin-eosin staining. The result showed that treatment with ALA at 30 mg and 60 mg significantly decreased the joint diameter compared to CFA group (p=0.003; p=0.001 respectively) and rat's latency time on thermal stimulation was also significantly increased compared to CFA group (p=0.015; p=0.026 respectively). Measurement of MDA in CFA group and ALA group had no significant difference. Histological staining indicated that the recovery of the synovial membranes of joint in ALA group had no effect. Results indicated that ALA has the effect to suppress the development of inflammation in RA but not through oxidative stress pathway.

Abstract

Endothelial dysfunction is an initial condition of atherosclerosis and other vascular diseases where one of the risk factors is hypercholesterolemia. Blood cholesterol levels is associated with an increase in the production of reactive oxygen species (ROS). The increasing of ROS production can cause increased oxidative stress which in turn resulting in endothelial dysfunction. Alpha lipoic acid (ALA) is one of the antioxidant compound that has been developed and studied. In this study we found that the use of ALA in Rattus norvegicus rats signifficantly lower the total cholesterol levels at dose 60 mg/kgBW (p=0.020). ALA also inhibit the expression of Monocyte Chemoattractant Protein-1 (MCP-1) at dose 60 mg/kgBW (p=0.044) and reduces the formation of Malondialdehyde (MDA) at dose 120 mg/kgBW (p=0.009), which is the initial stage of the atherogenic development and prognosis of events, thus, ALA can reduce the risk of further damage to the endothelium.

Abstract

Highlights Glutamine, an amino acid, can help mitigate the side effects of cisplatin by boosting glutathione (GSH) levels, an antioxidant that counteracts oxidative stress. By reducing AIF protein expression and apoptosis, glutamine may protect kidney cells from cisplatin toxicity, potentially improving the safety of chemotherapy. Background: Cisplatin, a widely used chemotherapy that is effective for many forms of cancer, has nephrotoxic qualities that can reduce renal function. Due to its side effects, the usage of cisplatin is limited by its dose. Glutamine, an amino acid, possesses properties that can counteract the nephrotoxicity of cisplatin. Objective: This study aimed to analyze the effect of intravenous glutamine administration on the decrease of AIF expression and apoptosis in intraglomerular mesangial cells of cisplatin-exposed rats. Material and Method: Samples (n) consisting of 30 rats were divided into 3 groups, with each group containing 10 rats. Group P0 received no treatment, group P1 received an injection of cisplatin on the 7th day, and group P2 received glutamine on the 1st–7th day and an injection of cisplatin on the 7th day. After 72 h, the tissue samples were immunohistochemically processed and then observed under a light microscope with 400x magnification. AIF expression and apoptosis were measured in the Allred/H score. Result: AIF expression results were: P0 = 4,35(2,2-5,4); P1 = 5,9(4,8-6,3); P2 = 5,25(2,4-5,8). The AIF expression of P2 was found to be significantly lower (p = 0.008) than P1. Apoptosis results were: P0 = 5,2(4,7-5,5); P1 = 6,4(6,1-7,1); P2 = 5,7(4,8-5,9). The apoptosis level of P2 was found to be significantly lower (p = <0.0001) than P1. Strong correlations between the decrease in AIF expression and the apoptosis level were found (p = <0.0001). Conclusion: Glutamine contributes to the decreased AIF expression and apoptosis in intraglomerular mesangial cells of cisplatin-exposed rats.

Abstract

Helicobacter pylori (HP) is a prevalent cause of bacterial infections, particularly chronic gastritis in the human body. The diagnosis of gastritis relied on gastric biopsy examination, where inflammation is characterized by an increase in various inflammatory cells, including macrophage, neutrophil, lymphocyte, or plasma cells. Therefore, this research aimed to determine the profiles of neutrophil, leukocyte, lymphocyte, and neutrophil lymphocyte ratio (NLR) in chronic gastritis patients, distinguishing between those with Helicobacter pylori and non-Helicobacter pylori infection at Dr. Soetomo Hospital in Surabaya. This research adopted a retrospective method, using medical records for chronic gastritis patients at Dr. Soetomo Hospital between 2018 and 2022. The variables analyzed included Helicobacter pylori infections and values of neutrophil, leukocyte, lymphocyte, and NLR cells. The obtained data were subjected to analysis using descriptive statistics. The results showed that the majority of participants were females (61.9%), senior high school graduates (52.38%), and aged 45-59 (pre-elderly) (32.14%). In non-Helicobacter pylori group, the average values of neutrophil, leukocyte, lymphocyte, and NLR cells were 62.29 (SD = 12.17), 8.155 (SD = 4.02), 29.165 (SD = 12.31), and 3.68 (SD=4.865), respectively. In conclusion, females showed a propensity for chronic gastritis with Helicobacter pylori. It was observed that non-Helicobacter pylori chronic gastritis tended to have elevated neutrophil and leukocyte values. Meanwhile, Helicobacter pylori-related chronic gastritis tended to show increased lymphocyte and NLR values due to systemic inflammation. Helicobacter pylori was associated with conditions such as iron deficiency anemia, cardiovascular disease, diabetes, thyroid disease, and malnutrition.

Abstract

Penelitian ini bertujuan untuk meningkatkan pemahaman masyarakat tentang deteksi dini kanker serviks dan evaluasi Pap smear, serta meningkatkan keterampilan pengambilan sampel Pap smear pada penyedia layanan kesehatan di Kecamatan Mawaddah, Kabupaten Mojokerto. Studi deskriptif observasional dengan desain cross sectional dilakukan pada 100 wanita berusia 20-70 tahun dari Rumah Sakit Mawaddah. Penilaian dilakukan sebelum dan sesudah pemaparan materi. 100 sampel Pap smear yang dikumpulkan oleh bidan setempat dievaluasi untuk menilai kecukupan spesimen menggunakan sistem Papanicolaou dan Bethesda. Kegiatan ini secara signifikan meningkatkan pemahaman tentang kanker serviks (8,05% menjadi 82,26%), sementara kegiatan pelatihan tenaga medis meningkatkan pengetahuan mengenai pengambilan sampel Pap smear (51,62% menjadi 100%). 99% dari sampel Pap smear yang dikumpulkan oleh tenaga medis di Kecamatan Mawaddah dinilai memadai dan memuaskan. Program pengabdian masyarakat yang berfokus pada deteksi dini kanker serviks melalui Pap smear, secara efektif meningkatkan pemahaman masyarakat, meningkatkan pengetahuan dan keterampilan pengambilan sampel oleh tenaga medis, yang dapat meningkatkan pula efektifitas layanan Pap smear di Rumah Sakit Mawaddah pada masyarakat.

Abstract

Background: Early detection of cervical cancer through IVA and Pap smear examination is crucial for preventing and reducing cervical cancer mortality. However, IVA and Pap smear coverage in Indonesia, particularly in Mentaras Village, Gresik Regency, remains low. The low knowledge and attitudes of women of childbearing age towards IVA and Pap smear examinations are major obstacles to early detection. Objective: This community service aims to improve the knowledge and attitudes of women of childbearing age in Mentaras Village towards IVA and Pap smear examinations through health socialization. Method: Health socialization was conducted for 23 participants from the PKK mothers' group using PowerPoint presentations and brochures. Evaluation was performed using a post-test questionnaire to measure participants' knowledge and attitudes after the activity. Results: After the socialization, all participants (100%) demonstrated a good level of knowledge, and 91.30% showed a positive attitude towards IVA and Pap smear examinations. These results indicate that the socialization activity positively impacted participants' awareness. Conclusion: Health socialization effectively improved understanding and attitudes towards the early detection of cervical cancer. Regular similar activities are recommended to expand community participation.
