Abstract

The aims of this study is to determine the effect of maturity level and location of the farms to the number of coliform bacteria in bali cattle feces in Nusa Penida. 24 samples of bali cattle’s feces were used in this research. This study used a Randomized Complete Block Design (RCBD) of 2x3 factorial consist of two location of altitude plateu (lowland and highland) and three maturity level (calves, heifers, and adult). The sample was grown on Nutrient Agar using the casting method. The obtained data were tested by analysis of variance followed by Least Significant Difference (LSD) Test. The results showed that the number of coliform bacteria in bali cattle was significantly different at maturity level and location of the farm. Number of coliform bacteria in bali cattle of the lowland was (138.66 x 107CFU/g), which is higher than bali cattle on the high land (102 x 107CFU/g). Test results of LSD Test showed the number of coliform cattle calf is 43,25 x 107 CFU/g is lower compare to heifers 124 x 107 CFU/g and adults 193,75 x 107 CFU/g. The number of coliform bacteria in feces of heifers cattle is lower than the number of coliform bacteria in feces of adult cattle.

Abstract

The protein is one of the meat component. The exestence of the protein and amino acid willdetermine the charateristic and the quality of meat. The aim of the reseach is to know thecharacteristic of protein bands, composition and consentration of amino acid in bali and wagyubeef after boiled. This research was used bali and wagyu beef wich taken from femoralis biceps muscle. Meat size of 1x1x1 cm was boiled for 15 and 30 minutes, five g of each beef (bali andwagyu) extracted, subsequently analized using the SDS-PAGE (sodium dodecyl sulfatpolyacrilamide gel electrophoresis) method, in order to know the bands of  protein. While theHPLC (high performance liquid chromatography) metode was used to analize the amino acid. Theresults showed that boiled for 15 minutes of bali and wagyu beef appear five bands of protein bothbali and wagyu beef. While boiling for 30 minutes show four protein bands for bali beef, and just 3three protein bands for wagyu beef respectively. Results of HPLC showed both bali and wagyubeef contains nine types of essensial amino acid and six types of non-essensial amino acid. After 30minutes of boiled, the essensial amino acid decreases as much as 56,65% for bali beef and 27,37%for wagyu beef. On the other hands, the consentration of non-essensial amino acid for bali beefdecreases as much as 63,05%  and wagyu beef 67,17% respectively.

Abstract

Nusa Penida is a pure breeding area of bali cattle, in which the cattle are mainly kept in conventional maintenance system and potentially infected by parasite, especially gastrointestinal nematodes. This study aims were to determine the prevalence and type of gastrointestinal nematodes in bali cattle breeders in Nusa Penida. Fecal samples were taken from 50 bali cattle breeders kept in cages (simantri) and another 50 samples were from cattle not kept in cage. The floating method was used for morphological examination and prevalence, the data was analyzed with descriptive analysis. The results showed, the prevalence of bovine gastrointestinal nematodes in Nusa Penida was 25%. The prevalence of nematode infection in bali cattle that kept cages was lower compared to the cattle that were not kept in cage. Strongyloides papillosus and Capillaria bovis were the gastrointestinal nematodes found in the infected cattle.

Abstract

The research has been conducted to find out the infection of coccidia and strongyloides in bali cattle, after the administration of minerals in varying amount. The examined samples were 24 feces of bali cattle, which were divided into four treatment groups: without mineral administration (control group), mineral administration of 2.5 gram/head/day; 5.0 gram/ head/day; and 7.5 grams/head/day. After three months administration, examination of coccidia and strongyloides infection was performed using sedimentation concentration and floatation method with saturated salt substance. The data obtained were nominal data which analyzed by using chi square test. The results showed that faecal examination in the bali cattle that were not given mineral treatment (control) was found ookista coccidia and larvae strongyloides. However, no coccidia and strongyloides infection in bali cattle found in the minerals administration (2.5, 5.0, 7.5 gr/d) groups.

Abstract

This study aims to determine about the histology structure and the morphometry white blood cells agranulocytes (lymphocytes and monocytes) of Bali cattle in Nusa Penida. A sample of blood from 50 female calves, had been taken through a jugular vein. The blood samples were fixed and colored by using Giemza staining method. The morphometry measurement was performed with was done under Axio Zeiss Imager 2 microscope with 1000x magnification. The results of the measurements were analyzed descriptively quantitatively, while the histology description was analyzed by qualitative descriptive. The results showed average mean lymphocyte diameter (9.22 ± 0.73µm) was smaller than monocytes (12.48 ± 1.73 µm). The existence of the lymphocyte and monocytes histologic structure difference which are found in the nucleus. The lymphocyte nucleus was round and filled the cytoplasm, whereas the monocyte nucleus formed a curve on one or two sides, thus, it did not fill the cytoplasm.
