Abstract

Daidzein compounds dynamic and the effect of yam tuber (Pachyrhizus erosus (L)) Urb on myometrium rats model of estrogenic were investigated. Twentyfour of 5 months-old female Sprague Dawley rats were randomly assigned to three groups as followed, control, Daidzein administraton and yam tuber juice administration for 24 days. The administration of yam tuber juice were given by orally using a stomach with gavage tube in the morning. Every 8 h, the rats blood and urine were collected from tail until 24 h. The treatment were continued until 24 days. The result showed that the level of daidzein in blood and urine of rats that administratered by Yam tuber juice is lower than pure daidzein treatment rats every 8 until 24 hours of collection. The results demonstrated that the administration of yam tuber juice significantly myometrium proliferated in rats. It is assumed that genistein and quercetin is estrogenic as found in daidzein so that the miometrium is potentially more effecctive. This study suggest that phytoestrogen compound from Pachyrhizus erosus may offer a potential as natural estrogenic in women.

Abstract

The nutritional content of kian marine worm has never been reported. This study aimed to profile the nutritional content of worms from the Kei Islands, Maluku. Nutritional profiles were tested by proximate assays, atomic absorption spectroscopy (AAS), and High-Performance Liquid Chromatography (HPLC) of amino acids. The proximate analysis resulted in 71.16% of protein, 12.05% of water, 2.63% of carbohydrates, 2.21% of fat, 2.14% of crude fiber and 0.44% of ash. Mineral contents of Kian marine worm were 785.28 mg/kg potassium, 585.14 mg/kg magnesium, 180.98 mg/kg calcium, 0.89 mg/kg sodium and 0.44 mg/kg phosphorus. Kian marine worms contained 9 essential amino acids and 11 essential amino acids with levels of essential amino acids up to 206.94 mg/g and non-essential amino acids was 233.21 mg/g. Nutritional content in kian marine worms can be used as a potential high protein food resource.

Abstract

This study aims to develop a teaching materials of biology ubjects based on problem based learning combined by think pair share models that used in teaching circulation system at the 11th grades of natural science senior high school to improve students' science process skills. The development in this study following steps of 4D model (Define, Design, Develop, and Disseminate) that developed by Thiagarajan. In this development study the teaching material just followed three steps until validation product at Develop phase. The teaching materials that developed in this study consist of syllabi, lesson plans, worksheets, and assessment sheet. Validation assessment was done by two lectures as education expert and content material expert from Universitas Negeri Malang and a teacher as practitioner expert from SMAN 1 Kasiman Bojonegoro. Validation by education expert shows the value of validity is 94.03, the validation by content material expert shows the value of validity is 91.50, and the result of the validation by practitioners shows the value of validity is 95.08. Based on the validation results of the three experts indicate that the teaching materials achieve very valid criteria that’s mean did not need to revision and can be be used in biology learning of circulation system to improve students' science process skills of the 11th grades of natural science senior high school. Abstract

This study describes students' perception of the concept of the integration of Islam and science. The integration of religious knowledge into science learning means the elimination of the paradigm of separating science and religion. This is important because many students do not understand how to connect the scientific theories studied to their own religious beliefs. This research surveys 175 students from 4 Islamic universities in Indonesia. The respondents filled an online questionnaire containing 8 two-tiered questions based on 7 conceptual understanding indicators of the integration of Islam and science. The indicators measure students’ understanding of: (1) Science; (2) The relationship between science and religion (Islam); (3) The effect of science on religion; (4) The influence of Islamic insights on science learning; (5) The role of religion in the development/advancement of science; (6) The conflict between science and religion; (7) The differences in scientific concepts developed by Muslim and non-Muslim scientists. The result shows that the students’ perception on the relationship between Islam and science can be categorized as the perception of integration. They argue that religion has a role in the development of science, and science has a role in supporting the sustainability of life. The students also conceive that a lecturer's Islamic insight can have an influence on the learning process, especially in Biology course.

Abstract

This study was conducted to describe students’ perceptions about the genetic course, laboratory experiment, laboratory facilities, and also students’expectations about the development of the virtual laboratory as an alternative experiment in Genetics Courses. A descriptive quantitative method was used in this study. The participants involved were 49 students enrolling in a genetics course at a public university in Banten, Indonesia. Data were collected through a set of questionnaires. To analyze the data,we employed a descriptive quantitative approach in the form of a percentage. The results showed that the genetic course was considered difficult by the students. The reason is that the material is abstract, and the source of learning is difficult to understand. The most commonly used learning media in the lecture is Power Point.Learning strategies are oftenused,i.e.,the discussions and laboratory work in the laboratory. Students need interesting, communicative, and interactive learning media. The student response is enthusiastic about virtual laboratory development. The expected virtual laboratory criteria are can be used offline, looks interesting, easy to operate, and containsaudio-visual.

Abstract

Evolutionary  course materials in the Biology Department, State University of Padang inadequate to guide students to think critically, giving rise to various problems. One of the problems that arise are student learning outcomes at the course of evolution tend to be low, because the students are not familiar with the assessment instruments developed higher-order thinking skills. Problem thinking skills of students that is low, overcome by applying authentic asessment in the textbook used. Qualitative test goal is to produce an authentic assessment of molecular evolution in the textbook is good, and can improve higher-order thinking skills of students. Qualitative test is one of the stages of research development, adapted from the model of the development of 4D-models Thiagarajan, ie develop stages. Qualitative test conducted on students of the Department of Biology State University of Malang, because the student has more accustomed to using assessment that guide higher-order thinking skills, so the results will be better. Qualitative assessment test showed that there were several assessment instruments that need to be revised, in accordance with the values obtained by the students on the posttest. Based on these results, it can be said that the authentic assessment in the textbook of molecular evolution has reached the category of good and fit for use in the teaching of Evolution in Higher Education.Kata kunci: uji kualitatif, asesmen autentik, buku ajar evolusi molekuler

Abstract

Perubahan pola konsumsi masyarakat yang sadar akan pentingnya gizi dan kesehatan, menyebabkan konsumsi ayam ras pedaging cenderung meningkat. Ayam ras pedaging yang paling banyak dikonsumsi adalah jenis Ayam Broiler. Salah satu kandungan tertinggi pada daging ayam broiler selain lemak adalah protein. Daging ayam mudah mengalami penurunan kualitas selama penyimpanan. Penelitian ini bertujuan untuk mengetahui pengaruh waktu inkubasi terhadap konsentrasi protein jaringan otot ayam broiler serta mengetahui profil protein jaringan otot ayam broiler pada waktu inkubasi yang berbeda. Berdasarkan hasil uji Anava, waktu inkubasi berpengaruh terhadap konsentrasi protein jaringan otot ayam broiler. Hasil uji Post-Hoc Duncan menunjukkan jaringan otot Ayam Broiler yang disimpan mulai 1 jam, 3 jam, 5 jam, 7 jam, 9 jam dan 11 jam konsentrasi proteinnya menurun dan berbeda nyata dibandingkan dengan inkubasi 0 jam. Visualisasi protein memperlihatkan adanya perbedaan ketebalan pita protein setelah jaringan otot ayam disimpan selama 1 jam hingga 11 jam, adanya pita protein dengan BM 53 kDa yang hilang setelah jaringan otot ayam disimpan selama 11 jam serta adanya pita protein dengan BM 60 kDa yang muncul setelah jaringan otot disimpan selama 7 jam.

Abstract

Penelitian ini bertujuan untuk 1) menghasilkan modul berbasis penelitian analisis aktivitas fosforilasi protein membran spermatozoa kambing; 2) untuk mengetahui aktivitas fosforilasi protein membran spermatozoa kambing PE dan Boer. Penelitian ini merupakan penelitian pengembangan yang menggunakan model pengembangan Borg & Gall (1983) yang dibatasi sampai lima tahap yaitu, research and information collecting, planning, develop preliminary form of product, preliminary field testing, dan main product revision. Analisis aktivitas fosforilasi protein membran spermatozoa dilakukan dengan beberapa prosedur laboratorium yaitu, isolasi protein, elektroforesis, elektroelusi protein 43 kDa, dan uji aktivitas fosforilasi. Metode penelitian ini adalah deskriptif kuantitatif yang menggunakan instrumen berupa lembar validasi. Hasil validasi menunjukkan bahwa persentase kevalidan modul dari ahli materi sebesar 95,40%, dari ahli bahan ajar sebesar 99,49% dan dari uji coba kelompok kecil sebesar 86,35%. Keseluruhan persentase yang diperoleh termasuk ke dalam kategori valid. Hasil penelitian mengenai aktivitas fosforilasi protein 43 kDa menunjukkan bahwa jumlah ATP yang difosforilasi pada protein membran spermatozoa kambing PE sebesar 179 ppm, sedangkan pada kambing Boer sebesar 159,5 ppm. Unit aktivitas fosforilasi protein 43 kDa pada kambing PE sebesar 69,29 x 10-2 µmol/mL.menit, sedangkan pada kambing Boer sebesar 61,61 x 10-2µmol/mL.menit.Keywords       : Modul berbasis penelitian, aktivitas fosforilasi, teknik analisis biologi molekuler

Abstract

Aging terjadi akibat penurunan progresif kapasitas metabolisme seluler dan aktivitas proliferasi yang disertai dengan hilangnya potensi ekspresi gen, akumulasi mutasi somatik, serta struktur dan fungsi elastisitas kulit berkurang Salah satu upaya yang sering dilakukan untuk menekan terjadinya aging adalah penggunaan kosmetik berbahan kimia yang tanpa disadari justru berdampak buruk bagi kesehatan misalnya dapat menimbulkan kanker. Untuk mengatasi masalah tersebut maka solusi yang dapat dilakukan adalah melalui penggunaan bahan alami yang lebih aman bagi tubuh. Berdasarkan laporan dari penelitian sebelumnya dapat diketahui bahwa tumbuhan Morinda citrifolia memiliki potensi sebagai anti-aging sebab memiliki senyawa alami yaitu berupa senyawa scopoletin. Penelitian ini bertujuan untuk memprediksi apakah senyawa scopoletin dapat menghambat aging pada manusia dengan menggunakan metode reverse docking. Penelitian ini diawali dengan pemodelan struktur 3D senyawa alami scopoletin menggunakandatabase PubChem. Struktur 3D tersebut digunakan untuk menentukan protein target pada manusia dengan WebServer Swiss Target Prediction, PharmMapper, dan SuperPred. Dari tahapan ini diperoleh Carbonic Anhydrase 1 (CA1) dan Carbonic Anhydrase 2 (CA2) sebagai protein protein target pada manusia. Setelah dilakukan Protein Targets Profiling dan Prediksi Protein-protein Network menggunakan String-db menunjukkan bahwa protein target pada manusia untuk Scopoletin adalah CA1 yang berperan pada proses skin aging.

Abstract

Peranan hormon dalam metamorfosis meliputi proses pengelupasan kulit larva, dan pembentukan pupa pada serangga holometabola. Hormon Juvenil menjadi salah satu kunci hormon yang berperan dalam mengatur aktivitas pertumbuhan serangga.Tujuan dari penelitian ini adalah untuk melakukan isolasi 10kDa yang merupakan hormon juvenil sebagai pengatur utama hormon pertumbuhan dan prekursor utama diapause hormon.Penelitian ini merupakan penelitian eksploratif. Materi hormon 10kDa diisolasi dari hemolympha ulat sutera (Bombyx mori L.) fase Instar 3, instar 4, instar 5, serta fase pupa menggunakan phosphate buffered saline kemudian di running menggunakan SDS-PAGE dan dikoleksi dengan elektroelusi. Karkterisasi hormon 10 kDa pada instar 3, 4, 5 dan pupa meliputi penentuan berat molekul melalui SDS-PAGE dan kandungan protein dengan metode nanodrop spectrophotometer.Hasil running isolat hormon menggunakan metode SDS-PAGE diperoleh pita protein hormon yang terdiri atas 5 pita dengan berat molekul 62,42; 39,10; 30,94; 15,34; dan 10,3 kDa. Kadar Isolat protein hormon 10 kDa diperoleh dengan purifikasi menggunakan metode elektroelusi. Kandungan protein dalam isolate protein hormon 10 kDa pada instar 3 sebesar sebesar 0,121 mg/ml, instar 4 sebesar 0,012 mg/ml, dan instar 5 sebesar 0,117 mg/ml.
