Abstract

Tujuan dari penelitian ini adalah menentukan kadar genistein dan aktivitas hambatan dari ekstrak kedelai terfermentasi dan non-fermentasi. Kedelai difermentasi oleh Aspergillus oryzae selama 4 hari dan dimonitor kadar genistein serta aktivitas hambatan tirosinase setiap hari. Kadar genistein ditentukan menggunakan densitometer-KLT sedangkan aktivitas hambatan tirosinase dievaluasi menggunakan metode spektrofotometri. Hasil penelitian menunjukkan bahwa kadar genistein meningkat pada saat fermentasi hari ke-1 sampai ke-3 (45,645-1256,042 μg/g ekstrak) tetapi pada hari ke-4 kadarnya menurun (54,373 μg/g ekstrak). Aktivitas hambatan tirosinase juga mengalami peningkatan pada saat fermentasi hari ke-1 sampai ke-3 (dilihat pada nilai IC50) dan menurun saat fermentasi hari ke-4. Disimpulkan bahwa kadar genistein berkaitan dengan aktivitas hambatan tirosinase (R2 = 0,949).

Abstract

The leaf of Guazuma ulmifolia has been used traditionally for antiobesity. The activity of antiobesity was affected by the content of bioactive compounds. Extraction is the primary step to obtain bioactive compounds from plant material. The method and solvent used for extraction are crucial factors to produce extracts that have a high amount of active compounds. This study aims to determine the total phenolic and total flavonoids content from ethanolic extracts, water extract, and infusions of G. ulmifolia leaf and to evaluate the antioxidant and antilipase activity. Folin-Ciocalteu method was used to determine the phenolic content, while flavonoid content determination was done using aluminium chloride colorimetric assay. The antioxidant activity was done using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, and the antilipase activity was quantified using p-nitrophenol release from p-nitrophenyl butyrate (p-NPB) substrate-colorimetric assay. The result of G. ulmifolia leaf extraction showed that the highest yield was obtained from water extraction (10.50%). Whereas, the ethanolic extract was showed the highest total phenolic content (67.761±1.811 mg GAE/g extract) and the highest total flavonoid content (124.643 ± 1.033 mg QE/g extract). The same extract also exhibited the highest antioxidant activity (IC50 = 6.544 ± 0.271 µg/mL) and antilipase activity (IC50 = 307.280 ± 21.430 µg/mL).

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengevaluasi kemampuan antioksidan dari ekstrak air buah salak Salacca zalacca (Gaertn.) Voss varian gula pasir melalui aktivitas penangkapan radikal bebas 1,1-difenil-2-pikrilhidrazil (DPPH). Harga Inhibition Concentration (IC50) dari ekstrak buah salak yang menunjukkan kemampuan suatu bahan dalam menghambat 50% dari total 100% radikal DPPH sebesar 40,89±6,35 μg/mL. Sedangkan kontrol positif kuersetin memiliki harga IC50 sebesar 8,79±0,90 μg/mL. Berdasarkan harga IC50, dapat disimpulkan bahwa ekstrak buah salak varian gula pasir menunjukkan aktivitas antioksidan yang sangat kuat. Skrining fitokimia membuktikan adanya kandungan golongan senyawa polifenol yang diduga bertanggung jawab atas aktivitas antioksidannya yang tinggi. Golongan senyawa ini dapat melindungi dari radikal bebas sebagai penginduksi kerusakan biomolekul. Kata kunci: Salacca zalacca (Gaertn.) Voss, gula pasir, antioksidan, DPPH, polifenol. ABSTRACT The aim of this study was to evaluate antioxidant capacity of gula pasir variant of snake fruit (Salacca zalacca (Gaertn.) Voss) water extract using free radical scavenging activity of the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH). The inhibition concentration (IC50) sufficient to elicit 50% of a maximum effect estimated in 100% was 40.89±6.35 μg/mL for the DPPH radical scavenging activity. While the positive control, quercetin, had IC50 value of 8.79±0.90 μg/mL. Based on the IC50 value, we concluded that gula pasir variant of snake fruit extract exhibited a very strong antioxidant activity. The phytochemical screening revealed the presence of polyphenol which could be responsible for the high antioxidant activity. Abstract

Kemuning (Murraya paniculata) belongs to the Rutaceae family, it grows a lot in tropical areas such as Indonesia. Traditionally, Kemuning leaves are used for weight loss preparations and are believed to contain compounds that can inhibit pancreatic lipase, including flavonoids. This study aimed to reveal the antilipase activity of Kemuning leaves extract and its fractions. The extraction of Kemuning leaves was carried out by maceration with 70% ethanol as solvent. The vacuum liquid chromatography method was used to separate the secondary metabolites in the extract with the stationary phase of silica gel and the eluent mixture of n-hexane, ethyl acetate, and methanol in a gradient manner. Meanwhile, the antilipase activity was tested by colorimetric method, with p-nitrophenyl butyrate as a substrate and orlistat as a positive control. The extract obtained was 19.94% of the weight of the simplicia. The separated extract was collected in 20 flasks and then combined based on the Rf value of the spots observed on thin layer chromatography (TLC) producing 8 fractions (fractions A-H). The percentage yields of fractions A-H were 1.20, 1.33, 1.80, 3.92, 3.57, 4.20, 16.85, and 14.47%, respectively. Antilipase activity was indicated by IC50 values, which were 17.603 ± 1.660 and 108.554 ± 1.660 µg/mL for orlistat and extract, respectively. The antilipase activities of fractions A-H were 130.19 ± 2.90, 119.52 ± 1.95, 96.95 ± 1.81, 91.65 ± 0.78, 95.30 ± 1.46, 114.76 ± 2.87, 127.07 ± 3.56, and 134,04 ± 3.16 µg/mL, respectively. Based on these results, it can be concluded that the extract and its fractions exhibit very strong antilipase activity. Keywords: Murraya paniculata; extract; fraction; antilipase

Abstract

Long-term use of doxorubicin as cancer chemotherapeutic agent would cause tissue toxicity, including cardiotoxicity. Arcangelisia flava is suggested to have cardio protective effect. This study aimed to determine the effect of chloroform extract of A. flava leaves on cardio histopathology of doxorubicin-treated Wistar white male rats. Wistar male rats were divided into four groups (1) control group; (2) doxorubicin 7.5 mg/kgBW intraperitoneally twice (day 1 and 6); (3) doxorubicin + chloroform extract of A. flava leaves 250 mg/kgBW/day orally for 11 days; (4) Chloroform extract of A. flava leaves 250 mg/kgBW/day orallly for 11 days. At the 12th day, the rats were sacrificed; the heart organ was taken to make histopathological preparations and analyzed using HE staining. Vacuolization and necrosis are the parameters used in evaluating this effect. The phytochemical screening was also done to determine the compounds in chloroform extract of A. flava leaves. Based on the HE staining, chloroform extract of A. flava leaves decreased the cardiotoxicity caused by doxorubicin. The phytochemical screening showed that chloroform extract of A. flava leaves contains flavonoid, tannin, alkaloid, and triterpenoid. The cardioprotective effect of chloroform extract of A. flava leaves was suggested to be contributed by the flavonoid, tannin, and triterpenoid.

Abstract

Antioxidants are compounds that inhibit free radicals. Many plants have antioxidant activity because they contain antioxidant compounds such as flavonoids. This study aimed to evaluate the antioxidant activity, phytochemical contents, total flavonoid content and to establish the relationship of total flavonoid content and antioxidant activity of extract and fractions of walnut (Canarium indicum L.) leaves. Antioxidant activity of ethanolic extract, n-hexane fraction, ethyl acetate fraction, and ethanolic-water fraction of walnut leaves was examined by using the DPPH method and determination of total flavonoid content was determined spectrophotometrically. The results showed that antioxidant activity of ethanolic extract, n-hexane fraction, ethyl acetate fraction, and ethanolic-water fraction based were 25.294 ± 0.055; 175.245 ± 0.4999; 20.135 ± 0.009; and 28.806 ± 0.0424 μg/ml, respectively. The phytochemical content of ethanol extract of walnut leaves are saponins, flavonoids, tannins, and polyphenols. The total flavonoid content of ethanolic extract, n-hexane fraction, ethyl acetate fraction and ethanolic-water fraction were 2.624 ± 0.012; 0.499 ± 0.023; 3.846 ± 0.006; and 1.596 ± 0.006 gram quercetin equivalent/gram extract, respectively. Correlation between antioxidant activity and total flavonoid content revealed that 63.2 % of antioxidant activity was influenced by the presence of flavonoid compounds.

Abstract

A previous study showed that Tithonia diversifolia ethanolic extract has antiproliferative properties against HeLa cervical cancer cells. It has also shown anti-inflammatory properties in vivo, one of the approaches to cancer chemoprevention. This research aimed to determine the relation of in vitro anti-inflammatory and antioxidant activity of T. diversifolia leaves ethanolic extract and fractions with total phenolic and flavonoid content. The anti-inflammatory was evaluated in vitro using red blood cell (RBC) membrane stabilization and inhibition of protein denaturation method, while antioxidant activity was done using the DPPH method. Phytochemical screening was done using the TLC method. Total phenolic and flavonoid content were also counted using the colorimetry method. ANOVA followed by LSD post hoc, multiple regression analysis, and Pearson correlation were used to analyze the data. Compared to its fractions, the highest anti-inflammatory and antioxidant activity was obtained from T. diversifolia leaves ethanolic extract. The T. diversifolia leaves ethanolic extract contained terpenoids, alkaloids, polyphenols, and flavonoids. Overall, the antioxidant property contributed more to RBC membrane stabilization than protein denaturation inhibition. The phenolic and flavonoid content contributed most to antioxidant and red blood cell membrane stabilization rather than protein denaturation inhibition.

Abstract

The exploration of secondary metabolites for various human needs has now extended to aquatic environments. The fluctuating and dynamic nature of aquatic environments can stimulate marine bacteria to adapt by producing secondary metabolites. The diversity of marine bacteria has become increasingly promising as a target for obtaining various potential secondary metabolites. This study aims to isolate potential marine bacteria that produce secondary metabolites from the Papuma Beach area in Jember. Marine bacterial characterization can be carried out through morphological and molecular approaches. In this study, bacterial isolates were collected from marine algae at aquatic sites. Bacteria were isolated from the algae using the spread plate and streak plate methods on nutrient agar dissolved in artificial seawater as the medium until single colonies were obtained. Morphological characterization of the single colonies was conducted by observing colony shape and color, as well as performing Gram staining. Molecular characterization of selected single colonies was performed by amplifying the 16S rDNA as a bacterial molecular marker. The results of this study showed that marine bacteria that grew well and remained stable on artificial media were successfully isolated from green and red algae samples, labeled GaN 2 and RaN 2, respectively. The GaN 2 bacterial isolate was identified as Fictibacillus barbaricus strain SQ5-4, while the RaN 2 bacterial isolate was identified as Bacillus altitudinis strain 41KF2bT.26. According to previous studies, Fictibacillus barbaricus is known to have potential as a bioremediation agent, while Bacillus altitudinis has potential applications as an antitumor, antimicrobial, and antioxidant-producing agent.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh sistem informasi akuntansi terhadap kualitas laporan keuangan pada CV Pajajaran Ciamis. Latar belakang dari penelitian ini adalah pentingnya penyajian laporan keuangan yang akurat dan relevan sebagai dasar pengambilan keputusan ekonomi, serta kebutuhan perusahaan terhadap sistem informasi akuntansi yang efisien dalam mendukung pengelolaan data keuangan. Penelitian ini menggunakan metode kuantitatif dengan pendekatan deskriptif analisis. Teknik pengumpulan data dilakukan melalui penyebaran kuesioner kepada karyawan yang terlibat dalam penyusunan laporan keuangan. Hasil penelitian menunjukkan bahwa sistem informasi akuntansi yang terdiri dari komponen perangkat keras, perangkat lunak, brainware, prosedur, dan jaringan komunikasi secara signifikan berpengaruh terhadap kualitas laporan keuangan yang diukur melalui indikator relevansi, keandalan, keterpahaman, dan kemampuan dibandingkan. Dengan demikian, dapat disimpulkan bahwa penerapan sistem informasi akuntansi yang baik akan meningkatkan kualitas laporan keuangan dalam perusahaan.
