Abstract

Kedabu leaves (Sonneratia ovata Backer) are known contain secondary metabolites, one of which is phenolic, flavonoid and antioxidant activity. Phenolic, flavonoids and antioxidant are known to inhibit the tyrosinase enzyme. The tyrosinase enzyme is involved in the process of melanogenesis which produces melanin. Melanin can cause hyperpigmentation. To prevent hyperpigmentation, tyrosinase inhibitors are needed. This study aimed to test the tyrosinase enzyme inhibitor in ethanol extract, n-butanol fraction, ethyl acetate fraction, n-hexane fraction of kedabu leaf (Sonneratia ovatta Backer) using L-tyrosine as a substrate. The method used to get ethanol extract using ethanol maceration, while some fractions are obtained using the fractionation method. The test was carried out using L-tyrosine and kojic acid as a positive control with absorption measurements using microplate reader at a wavelength of 492 nm. The results showed that the best tyrosinase inhibitory activity was produced by the ethyl acetate fraction with an IC50 value of 18,82 g/mL, followed by the n-butanol fraction with an IC50 value of 24,42 g/mL, and the n-hexane fraction with an IC50 value. 28,77 g/mL, ethanol extract with an IC50 value of 99,09 with a strong category for all samples compared with kojic acid as a positive control with an IC50 value of 30,09 g/mL.

Abstract

The sweet orange (Citrus sinensis (L.) Osbeck) is a fruit that is loved by all walks of life because of its sweet taste. The flesh part of the fruit is consumed, while the fruit peel is discarded. However, the fruit peel also contains secondary metabolites, which have various biological activities. The peel of this sweet orange fruit contains secondary metabolites of alkaloids, flavonoids, phenolics, steroids, and terpenoids. This study aims to determine the antifungal activity of the n-hexane fraction against Trichophyton mentagrophytes and Candida albicans using the disc diffusion method and the Minimum Inhibitory Concentration (MIC). The results of measuring the diameter of the inhibition zone formed on the fungus Trichophyton mentagrophytes at a concentration of 50; 25; 12.5; 6.25; 3.125 and 1.5625% w/v, respectively 12.29±0.08; 11.14±0.12; 9.54±0.17; 7.01±0.12; 6.19±0.03; 6.06 ± 0.03 mm, with a MIC of 1.5625 %. Results of measuring the diameter of the inhibition zone formed against Candida albicans fungi respectively 9.97 ± 0.02; 9.05±0.18; 8.32±0.38; 7.09±0.07; 6.00±0.00; 6.00±0.00 mm, with a MIC of 6.25%. The results of data analysis using one-way ANOVA statistics showed that there was a significant difference (p<0.05) between variations in the concentration of the test fraction with positive control and negative control. Based on the research that has been carried out, it can be concluded that the n-hexane fraction of sweet orange peel has activity in inhibiting the growth of the test fungus, with the best antibacterial activity at a concentration of 50% w/v.

Abstract

Food is one of the most important needs for humans because it relates to daily nutritional needs. The solution to this problem is to consume food with good nutritional content supported by appropriate food processing innovations. Milk can be drunk directly or processed into various forms of food and drinks with milk-based ingredients, such as yogurt. This study aims to make yogurt products from soy milk using red dragon fruit juice as a substitute for sugar and to test their antibacterial activity in inhibiting Salmonella typhi and Shigella dysenteriae bacteria. This study used a completely randomized design (CRD) with 3 treatments, namely the addition of red dragon fruit juice with concentrations of 5%, 10%, and 15%, 3 replications, and the diffusion method for testing antibacterial activity. The results showed that the best yoghurt formula was obtained from F3 (yoghurt with the addition of 15% concentration of red dragon fruit juice). The panelist hedonic test gives values ranging from slightly like to like for color, aroma, taste, and texture; for organoleptic F3, these are: color (dark pink), taste (sour), texture (liquid-thick), and aroma (typical); pH 3.49; TAT 1.39%; Lactic Acid Bacteria (LAB) 2.09 x 1010 CFU/mL; and also had an average diameter of inhibition of 18.96 mm against Salmonella thypi (P<0.05) and 20.10 mm against Shigella dysenteriae (p<0.05). Soy milk yogurt with the addition of varying concentrations of red dragon fruit juice has antibacterial activity.

Abstract

Abstract

Shallots (Allium cepa L.) are widely known by the public as a kitchen spice and traditional medicine. Shallots have antibacterial activity. Compounds that have antibacterial properties can be obtained by isolating endophytic microbes. Endophytic microbes are microbes that live in plant tissue, are able to form colonies in plant tissue without having detrimental effects on the host, and can produce secondary metabolites like the host. Phytochemical screening of shallot tubers showed that they contained alkaloids, flavonoids, and phenolics. This research aims to isolate and identify endophytic fungi from shallot tubers and determine the antibacterial activity of these endophytic fungal isolates. Endophytic fungi were isolated using the direct planting method, and then pure isolates were identified macroscopically and microscopically. Antibacterial activity testing was carried out using the agar diffusion method against the bacteria Staphylococcus aureus, Staphylococcus epidermidis, Bacillus cereus, Escherichia coli, Salmonella typhi, and Shigella dysenteriae. There were 4 isolates of endophytic fungi produced during the isolation process, namely FEAC-U1 (Aspergillus), FEAC-U2 (Penicillium), FEAC-U3 (Trichophyton), and FEAC-U4 (Rhizopus). FEAC-U1 isolate showed the highest activity against Staphylococcus epidermidis, Bacillus cereus, and Shigella dysenteriae. FEAC-U2 isolate showed the highest activity against Salmonella typhi. FEAC-U3 isolate showed the highest activity against Staphylococcus aureus and Escherichia coli. The results of antibacterial activity testing on four isolates of shallot tuber endophytic fungi were able to inhibit the growth of the test bacteria.
