Abstract

ABSTRACT This study aims to determine the effect of a multilevel dose of ethanol extract of seaweed (Eucheuma cottonii J. Agardh) on the decreasing blood glucose levels and kidney tissue regeneration of hypercholesterolemia-diabetic male white rats. 24 rats were divided into 6 groups, namely groups 1-3 (normal control, negative control, and positive control/Glibenclamide, respectively) and groups 3-6 (seaweed extract doses of 100, 200, and 400 mg/kg BW per oral, respectively). Rats were induced by high fat diet and Streptozotocin. Histopathological picture of the kidney was observed by HE staining using 400 magnifications. Data scoring of the level of kidney damage were analyzed by One Way ANOVA at a 95% confidence level and a Least Significant Difference (LSD) test was conducted. Seaweed ethanol extract at a dose of 100 mg/kg BW was effective in reducing blood glucose levels with an average value of 122.25 mg/dL and effective in regenerating the kidney tissue of male white rats with an average value of damage of 1.9. Therefore, seaweed extract has potency to be developed and further investigated as an antidiabetic agent.

Abstract

Ginjal merupakan organ yang mempunyai kemampuan menyaring darah dengan membuang limbah alami seperti urea. Limbah alami yang diperluas bisa menyebabkan kerusakan ginjal. Penelitian ini bertujuan untuk menentukan kandungan metabolits pilihan alkaloids, flavonoids, saponins, dan tanins dalam konsentrat etanol daun pepaya, dan untuk menentukan dampak dan porsi konsentrat yang layak pada melawan sel silinder ginjal pada tikus yang digerakkan oleh streptozotocin. Hewan pengerat diisolasi ke dalam 6 kelompok yang terdiri dari control tipikal, control negatif, control positif, perlakuan lepas tandan dengan dosis 100, 200, 300mg/kg BB. Penelitian selama 28 hari. Studi ini mencatat perkembangan yang terjadi pada sel tubullus ginjal tikus putih jantan yang dipicu oleh streptozotocin sebagai tingkat kerusakans dengan nilai yangs ditentukan sebagai persentase (%) pada setiap bagian tubullus yang rusak atau rusak. Hasil penelitian menunjukkans bahwa daun pepaya secara terpisah mengandung metabolits opsional alkaloids, flavonoids, saponins dan tanins. Pemberian konsentrat etanol daun pepaya dalam porsi 300mg/kg BB secara preventif bisa mempengaruhi sel silinder ginjals pada tikus yang diinduksis STZ, dengan nilais bahaya tipikal 0,4 dari batas 4.

Abstract

Background: Earthworm extract (Lumbricus rubellus) has antimicrobial activity because it produces high levels of the lumbricine compound but is easily denatured in the stomach so that its solubility and biological activity change. Therefore its Application can be developed in the form of gastrorentetif mucoadhesive preparation. Objectives: The purpose of this study was to optimize and evaluate the antibacterial activity of gastroretentive mucoadhesive preparation matrix of earthworm extract (Lumbricus rubellus). Material and Methods: The matrix utilized a preparatory technique of applying wet granulation mucoadhesive polymers such as chitosan, HPMC, and sodium CMC with concentrations determined based on simple lattice design resulting in 7 granule formulas. The granule formulas obtained were then evaluated including flow time and dwell angle tests, particle size distribution tests, compressibility tests, dissolution tests, and antibacterial activity tests. Results: The results showed that the flow rate on the F3 granule formulation met the requirements of flow rate >10 g/s, a consequence of measuring the angle of repose in the F1-F5 formula had good flowability which was below 30o, in contrast to F6 and F7 which had an angle of repose of 30-40o favorable to flowability. The F1-F7 formula was mostly preserved in mesh number 20 sieve with a size of 426-850 ?m, according to the results of particle size distribution utilizing the multistage sieve method, the compressibility index of the F1-F7 granule formula was 10% and the measurement results at the 30 minute showed that the average dissolution of the F1-F3 granules was 97.77%, 97.77%, and 101.22% respectively while the F4-F7 granule formulas did not show slow drug release because at the 30th minute the drug released ranged and the antibacterial activity test of Formula 1-Formula 7 granules is respectively 6 mm, 5.5 mm, 8 mm, 3.5 mm, 4.7 mm, 2.1 mm and 3.3 mm. Conclusions: The results of the evaluation of the characteristics of earthworm extract mucoadhesive gastroretentive granule preparation demonstrated that the preparation met the requirements for granule preparation and exhibited antibacterial properties. Among the seven formulas tested, formula 3 had the highest efficacy of antibacterial activity.

Abstract

Indonesia has quite a serious nutritional problem which is characterized by many cases of malnutrition. Malnutrition is an impact of nutritional status. 2016 National Health Service data recorded that the prevalence of stunting reached 33.6%, this is an important health problem because the stunting problem is above the 20% threshold. Meanwhile, stunting in children under five is caused by multifactors, such as nutritional consumption during pregnancy, maternal knowledge about nutrition, limited access to services, inadequate access to sanitation and clean water. This counseling aims to help increase public knowledge, about preventing stunting in toddlers and is a form of the Tri Dharma of Higher Education. Counseling is provided using an interactive lecture method through distributing leaflets, health checks, providing nutritious food, providing TOGA books and social service. The effectiveness of the extension was tested by administering a questionnaire on community satisfaction with the activities carried out. The results of this service activity provide the Sejahtera village community with knowledge about what stunting is, the causes of stunting, the dangers of stunting in the long term and how to overcome the impact of stunting so that stunting can be minimized in children.

Abstract

Coconut (Cocos nucifera) is an Indonesian commodity that has high economic value tall. Virgin Coconut Oil (VCO) is one of the processed coconut products whose selling value is very high, because The composition of VCO consists of medium-chain fatty acids that can maintain a healthy body and prevent various diseases. The process of making VCO used in This research is an enzymatic method using pineapple weevil as a bromelain enzyme. This study aims to determine the effect of enzymatic administration of VCO and an enzymatic dose of VCO which is effective in reducing urea and creatinine levels in hypercholesterolemic-diabetic male white rats (Rattus norvegicus). This study was an experimental laboratory with a modified pretest and posttest randomized controlled group design using 30 test animals which were divided into 6 treatment groups. Each group consisted of 5 test animals, namely normal control, negative control, and positive control, with doses of 0.2, 0.4, and 0.8 mL/kg BW. The data obtained were analyzed using a One Way Anova and non- parametric statistical test by Kruskal Wallis. test and followed by a further Mann Whitney test to determine differences between treatments. The results showed that enzymatic VCO at a dose of 0.8 mL/kg BW was an effective dose in reducing urea and creatinine levels with an average decrease of 17.40 mg/dL and 0.36 mg/dL. The novelty in this study showed that the enzymatic VCO had an effect on reducing urea and creatinine levels in diabetic hypercholesterolemic male white rats.

Abstract

Purple leaves are one of the plants that are widely used as medicine. Purple leaves have 3 types of plant varieties, namely purple leaves, green leaves, and white mottled leaves.  Purple leaves have pharmacological activities that can be used as medicine, including antioxidants (improving the deterioration of cells in the body), anti-inflammatory (reducing or suppressing inflammation), antidiabetic (lowering blood glucose levels), analgesics (relieving pain), immunomodulators (increasing the work of the immune system in the body), antihemorrhoids (treating symptoms and complaints of hemorrhoids), and antibacterial (as an antiseptic). The purpose of this study was to determine the antioxidant activity of purple leaf ethanol extract (Graphtophyllum pictum (L.) Griff) in vitro using the DPPH method and to determine its effectiveness as an analgesic and anti-inflammatory against Wistar rats.The results showed that antioxidant activity showed an IC50 value of 72.31 ppm (strong antioxidant). Meanwhile, in the analgetic test, the highest percentage of analgesic effectiveness was found at a dose of 100 mg/kg BW (129.64%) and showed anti-inflammatory activity at a dose of 100 mg/kgBW with a percentage decrease in edem volume of 28.73% at the sixth hour.

Abstract

The natural ingredients of rosella flowers have the potential as natural anti-cancer and chemopreventive agents, contain secondary metabolite compounds with cancer therapeutic mechanisms with inhibiting and killing cancer cells and their development through inhibition of apoptosis (cell death program) and in vitro inhibited cell flow. The study aims to identification of secondary metabolite compounds and also determine the cytotoxic activity of ethanol extract from the rosemary plant against HepG2 cancer cells. This study used by identification of secondary metabolite compounds of ethanol extract from the rosemary plant (Hibiscus sabdariffa L.) and the cytotoxicity test used with MTT assay on HepG2 liver cancer cells with a test concentration series of 12,5; 25; 50; 100; 200; 250; 500 and 1000 µg/ml. : The results showed the secondary metabolite compounds of rosella flowers have flavonoids, alkaloids, saponins and tannins and data of 50% cell growth inhibition activity, which was showed that rosella flower had IC50 values 629,919 μg/mL. The ethanol extract of rosella flowers have activity on HepG2 cells growth inhibition had considered moderately active anticancer activity

Abstract

Medicinal plants are considered a rich source of bioactive metabolites and have the potential for the discovery and development of new drugs, one of which is the purple leaf plant which is known to treat various diseases. The aim of this research was to determine the effect of administering purple leaf ethanol extract over a certain period of time at doses of 500 mg/kgBB, 2.000 mg/kgBB and 5.000 mg/kgBB on the histopathology of the Wistar rat pancreas. In this study, an experimental method was used with a randomized block design, using 20 male Wistar rats which were divided into 4 research groups, 1 control group which was given Na-CMC. and 3 treatment groups were given purple leaf ethanol extract at a dose of 500 mg/kgBB (P1), dose of 2.000 mg/kgBB (P2) and  dose of 5.000 mg/kgBB (P3). The test preparation was administered orally, for 28 days. The results of subchronic toxicity testing of purple leaf ethanol extract on Wistar rats carried out for 28 days, up to a dose of 5.000 mg/kg BB, showed that at a dose of 2.000 mg/kg BB – 5.000 mg/kg BB it was toxic to the histopathology of the Wistar rats pancreas, which was characterized by the presence of cell degeneration, increased organ index and the presence of symptoms of toxicity

Abstract

This study aims to determine the toxic effects of purple leaf ethanol extract on lymph organs at doses of 500; 2.000; and 5.000 mg/kgBB administered for 28 days and and to determine the antioxidant activity of purple leaf Graptophyllum pictum (L.) This study is an experimental study with a random group design, using 20 test animals divided into 4 groups consisting of two treatment groups, namely in the normal control group given Na-CMC 0.5% and the experimental group given PLEE (k1: dose 500 mg/kgBB, k2: dose 2.000 mg/kgBB and k3: dose 5.000 mg/kgBB hematoxylin-eosin staining (HE) using a computer-connected Motic BA210 microscope with 40X magnification in 5 fields of view and antioxidant test using DPPH method with UV-Vis Spectrophotometer at 516 nm wavelength with concentration 20, 40, 60, 80, and 100 ppm. The results showed that leaf ethanol extract was toxic at a dose of 2.000 mg/kgBB – 5.000 mg/kgBB against the diameter of lymphatic organ pulp, organ index and organ weight, there were symptoms of toxicity and purple leaf ethanol extract had strong antioxidant activity with 66.26 μg/mL.

Abstract

Traditional herbal medicine has become increasingly popular as an alternative to conventional medical treatments. Although the plant offers numerous health benefits, caution should be exercised regarding its long-term use. Long-term use of traditional herbal remedies can have detrimental effects on the kidneys. This study was designed to assess the histological changes in the kidneys of male rats following 28 days of purple sage leaf extract administration and to identify potential harmful effects. The study utilized 20 male Wistar rats and utilized a randomized group design for the experimental methodology. Rats were categorized into four distinct groups, consisting of one control group and three treatment groups. The control group received solely sodium chloride, whereas the treatment group 1 (P1) received 500 mg/kg BB of purple sage ethanol extract. Treatment group 2 received up to 2000 mg/kg BB of purple sage ethanol extract, while treatment group 3 received up to 5000 mg/kg BB. The oral examination was conducted for a duration of 28 days. The treatment groups showed an increase in weight and organ index of the right kidney. The findings suggest a potential for toxicity resulting from the administration of purple sage leaf extract over a duration of 28 days.
