Abstract

A selective separation techniques with Molecularly Imprinted Polymer (MIP) for High-Performance Liquid Chromatography (HPLC) has been developed for the assay of Dimethylamylamine (DMAA) doping compounds. Molecular imprinted polymer (MIP) is a technique to produce a polymer having the cavity due to the disposal of the templates, in which the cavity serves to recognize the molecules of the same size, structure, chemical and physical properties. The selectivity and affinity of the templates itself will increase, while the concentration value is increasing. MIP is made by DMAA as template, acrylamide as functional monomer, ethylene glycol dimethacrylate (EGDMA) as cross linking, azobisisobutyronitrile (AIBN) as the initiator and chloroform as a porogen solvent; using bulk method. The aim of research are conduct the MIP for the DMAA compound analysis, then the formed MIP is characterized by using Fourier Transform Infra Red (FTIR) and Scanning Electron Microscopy (SEM) to find out the polymer complexes formed and the morphological form of the MIP. The MIP formed then was analyzed by using High-Performance Liquid Chromatography (HPLC) to know the amount of the DMAA, the adsorption capacity, and the adsorption condition found in the MIP. The result of analysis on the content of DMAA in the MIP by using UV-Vis Spectrophotometer is 1.957 mg. Scanning Electron Microscopy (SEM) shows that the MIP has irregular and rough morphological structure; while the NIP has irregular morphology structures and smooth surfaces shape

Abstract

Air pollution induces the production of reactive oxygen species (ROS), which can cause tissue damage when excessive. Antioxidants help counteract this damage, and purple sweet potatoes, rich in anthocyanins, are a promising natural antioxidant source. This study aimed to determine the effect of solvent acidity variations on the antioxidant activity of purple sweet potato (PSP) tuber extract and identify the optimal pH condition. Using the DPPH (2,2-diphenyl-1-picrylhydrazyl) scavenging assay, ethanol solvents at different pH levels (non-acidified ethanol, pH 2.5, pH 2, and pH 1.5) were tested. The results showed that higher acidity enhanced antioxidant activity, with the strongest activity observed at pH 1.5, yielding an IC50 value of 9.74±0.23 ppm. Although less potent than Vitamin C (IC50 1.22±0.04 ppm), the extract demonstrated significant potential as a natural antioxidant source. Further studies on anthocyanin content are recommended to better understand its contribution to antioxidant activity.

Abstract

Personalized therapy is a treatment approach that considers genetic, environmental, and lifestyle variations of each individual to optimize the effectiveness and safety of therapy. This study aims to analyze the role of medicinal chemistry in the development of personalized therapy through the perspective of rational design of drug molecules, structural modification, and optimization of pharmacokinetic-pharmacodynamic properties. The method used is a systematic literature review with the PRISMA approach to scientific publications in 2013-2023 that focus on the integration of medicinal chemistry with pharmacogenomics and personalized therapy. The results of the study showed that medicinal chemistry plays an important role in four main areas: (1) drug design based on specific genetic targets with high selectivity (76% of publications); (2) development of prodrugs and drug delivery systems that are responsive to individual biomarkers (63% of publications); (3) optimization of pharmacokinetics based on genetic variations of drug metabolizing enzymes (58% of publications); and (4) development of molecular probes for companion diagnostics (52% of publications). Further analysis identified the development trends of personalized therapy including multi-target drug design based on patient molecular profiles and big data-based medicinal chemistry strategies. The main challenges identified include the complexity of molecular modification to achieve pharmacokinetic profiles that match specific genetic subtypes and the limitations of experimental models that represent individual heterogeneity. In conclusion, medicinal chemistry plays a fundamental role in realizing the concept of personalized therapy through the design and optimization of drug molecules that match individual patient characteristics. Further research is needed for the development of medicinal chemistry methodologies integrated with omics technology and artificial intelligence to accelerate the translation of the concept of personalized therapy into real clinical applications in Indonesia.
