Abstract

Biofilm is one of parameters for determining pathogenic pathways of a bacterium. Bacterial virulence activities can be attenuated by antibiotics with inhibiting biofilm. The aim of this study was to examine the characteristics and activities of antibiotics against the biofilm of Stapylococcus aureus isolate which taken from Aceh cattle prepuce. The study was exploratory and experimental, using a Completely Randomized Design (RAL) factorial pattern using three types of antibiotics, three concentrations and replications. Antibiotics were tetracycline, oxytetracycline and phosphomycin with concentrations of 10%, 20% and 30% (mg/mL), respectively. Therefore, the aim of this study was to examine the characteristics and activities of antibiotics to the biofilm of Stapylococcus aureus from Aceh cattle prepuce isolate. U-bottom polystyrene 96 wells microplate and Nutrient Broth (NB) medium was used to biofilm bacterial while biofilm was stained on crystals violet 1%. Optical Density (OD) of biofilm were λ 595 nm. Data were analyzed by using Analysis of Variance (ANOVA). The results showed that the activity of Staphylococcus aureus was moderately positive, i.e 2,667cell/μL. It showed that antibiotics and concetrations have significant effect (P<0.05) to inhibit biofilm staphylococcus aureus. In additioon, tetracycline 30% (mg / mL) has the best ability to inhibit biofilms from 52,18% to 1,262 cell/μL. Based on that fact, it can be concluded that the virulence of Staphylococcus aureus was moderate positive to be false positive.

Abstract

Center for Agrotechnology Innovation, Gadjah Mada University (PIAT), one of the place for development and breeding ground of spotted deer in Yogyakarta, Indonesia. The study of blood profiles was intended to allow veterinarian to understand the normal profile of the spotted deer. Fifteen healthy spotted deers belonging to PIAT were used as experimental animals. The deers were drawn its blood through the jugular vein without the use of anaesthesia. Blood was then accommodated in an EDTA tube, centrifuged at 2500 RPM and analyzed using a Mindray BC-2800 haematology analyzer machine. Based on the results of the study of the blood it were known that  Haemoglobine (Hb) was: 11.5 ± 1.703 g/dl, Red blood cells (RBC) was: 9.3 ± 3.580 106/µl, Packed cell volume (PCV) was: 30.8 ± 6.035 %, Mean corpuscular Volume (MCV) was: 36.8 ± 11.102 fl, Mean corpuscular haemoglobine (MCH) was: 15.0 ± 7.313 pg, Mean corpuscular haemoglobine concentration (MCHC) was: 40.0 ± 14.657 g/dl, White blood cells (WBC) was: 6.4 ± 3.096 103/µl , Neutrophils was: 43.4 ± 21.646, % Basophils was: 0.2 ± 0.168 %, eosinophils was: 0.4 ± 0.447 %, lymphocytes was: 53.4 ± 21.546 % and monocytes was: 2.6 ± 2.394 %. From all of the datas it indicated that blood profiles of PIAT’s spotted deers are different from similar spotted deer blood profiles in India.

Abstract

Liver fibrosis is the excessive accumulation of extracellular matrix. The prevalence of liver fibrosis in small animal was reported by 10% of all systemic diseases occured in the United States. One of the toxic materials that cause liver fibrosis is carbon tetrachloride (CCl4). This study aims to determine the preventive effect of ethanol extract of leaves of cashew (Anacardium occidentale) against the production of Interleukin-6 (IL-6) and increase the levels of superoxide dismutase (SOD). Leaves cashew contain of flavonoids which have anti-inflammatory and antioxidant effects. This research is an experimental study, post-test control only design using a completely randomized design. Mice (Mus musculus) were divided into 5 groups, namely the negative control group, positive control group, preventive group dose of 500 mg/kg BW, 1000 mg/kg BW, and 1500 mg/kg BW. Interleukin-6 production is calculated using flowcytometry, while SOD was measured using spectophotometer. Data were statistically analyzed within one way ANOVA (α=0.05) with advanced test Post Hoc Tukey. Conclusions from this research that the preventive use of ethanol extract of leaves of cashew to prevent an increase of IL-6 production and increasing the levels of SOD in mice models of liver fibrosis. Production of IL-6 in group of 1500 mg/kg BW is 2.58%. SOD levels in group of 1500 mg/kg BW is 4.23%.

Abstract

The increasing beef cattle population in Indonesia is not significantly with high demand of meat every year. The insignificant increasing in population is caused by fertility decreasing, infectious disease and the reduction amount of forage land. The factor which causes cattle fertility decreasing can’t be separated from the ovarian and follicular diameter which have an effect on high various reproductive appearance. Parameters that used for knowing reproductive appearance of cattle are Service per Conception (S/C) and Calving Interval (CI). The aims of this study was to describe ovary sizes and follicular diameter in cattle, to describe ovary and follicular diameter toward S/C and CI variation. This research uses female cattle, 8 Simmental-PO (SimPO) and 9 Peranakan Ongole breeds (PO), multiparous, are not pregnant, 4-8 years old, Body Condition Score (BCS) 2.5-3.5, peak phase estrus, has no reproductive problems, clear recordings and live in the area of farmer in Jatinom District, Klaten Regency, Central Java Province. Cattle are examined for ovarian size and follicle size using ultrasonography. S/C and CI data were obtained from records of artificial insemination cards (AI). The data obtained were analyzed by t test. The results of this study showed there were no differences in the diameter of ovaries in the size of SimPO and PO cattle (P> 0.05), there were no differences in the diameter of SimPO and PO cattle follicles (> 0.05), there were differences in ovarian and follicular diameter sizes with respect to S/C and CI in cattle with good and bad reproductive performance (P <0.05). The conclusion of the study showed that there was no difference in ovarian and follicular diameter size in SimPO and PO cattle (P> 0.05) but there was an influence on S/C and CI (P <0.05).

Abstract

Maninjau lake is the largest center for the freshwater floating-net-cage fisheries system in West Sumatra. Every year, during the up-welling season, a huge number of fish suffered to death in the lake including nile tilapia (Oreochromis niloticus), a common species cultivated in the floating-net cages. This current study aimed to determine the factors contributing to the death of fish by measuring the concentration of toxic chemical substances in the blood plasm of nile tilapia as well as in the subtrate collected from the bottom of the lake. The samples were collected in the sites with higher number of floating-net cages as well as higher number of mass death of fish (Tanjung Sani, Pakan Raba’a dan Maninjau) during the up-welling season. The result revealed that the levels of phosphate, nitrate, nitrit, and amonium were higher in the blood plasm of the fish (87, 216, 13.9, 0.6, dan 5.5 mg/l). Moreover, the levels of those chemical substances were also higher in the substrate of the lake (82.150, 0.546, 33.166, 23.954 dan 0.0776 mg/l). The higher concentration of the toxic chemical substances might contribute to the mass death of fish living in the floating-net cages during the up-welling season.

Abstract

Molecular sexing for bird with polymerase chain reaction-based method have been developed, but the samples used as a sources of DNA are blood and feathers which are considerably invasive. The aim of this study was to study the efficiency of buccal swabs as a resource of DNA for sexing canary bird and pigeon. This study used 10 canaries (Serinus canaria) consisting of 6 adult canaries (3 males and 3 females) and 4 young nestling canaries (14 - to 18-day old) and 6 adults (3 males and 3 females) pigeons (Columba livia) and 7 young nestling pigeons (14- to 25-day-old). All birds were taken their buccal swab samples, then DNA were extracted, mixed with PCR-mix to be amplified for sexing genes with CHD1F/CHD1R primer pairs. The amplification results showed that all of adult male birds produced single band (± 500 bp), whereas all of adult female birds produced double bands (± 500 bp and ± 300 bp). The PCR method for nestling canaries showed 2 males and 2 females. whereas nestling pigeons 6 males and 1 female. Based on this study it can be concluded that buccal swabs are efficient as a source of DNA for birds sexing especially young nestling birds.

Abstract

Resident in Kokap, Kulon Progo, Yogyakarta Province has another income sector, that is dairy goat (Etawah Crossbreed) farming and they have been joining into farm group, which is called Kelompok Ternak. There are many bacteria that causing dairy goat intra mamary infection, thus the milk production will be contaminated. Two hundred and four (204) milk samples from 103 lactation goat in four farm groups in Kokap, Kulon Progo, Yogyakarta is used in this study. Out of these, 21 samples (10,29%) were detected strong positive (++ and +++) for subclinical mastitis by using California Mastitis Test (CMT). Among this positive result, the next step was bacteria detection. This tests were held in Preclinic Laboratory Animal Health Study Program, Vocational School, UGM. The samples were inoculated in blood agar, then colony was identify by Gram staining. The next steph is biochemical tests using McConkey Agar (MCA) and eosin methylene blue (EMB) as a selective media for Gram negative bacteria, manitol salt agar (MSA) as selective media for Staphylococcus, sugar fermented broth (glucose, lactose, mannitol, maltose, and sacharose), Kigler iron agar (KIA), lysine iron agar (LIA), motility indole ornithin (MIO), and Simmon’s citrate. The result was Staphylococcus aureus (80%), Staphylococcus epidermidis (20%), Escherichia coli (10%), and Klebsiella pneumoniae (10%).

Abstract

Veterinary Authority (Otovet) is a government institution, formed in order to deal with decision making of anything related to animal health, by involving veterinarians and mobilizing all related professional lines. The purpose of this study was to describe the Regional Organizations’ (OPD) assessment of the implementation of Veterinary Authority (Otovet) in rabies management policies. Research material included 36 Regional organizations (OPD), 9 Technical services, supervising animal health (keswan) and veterinary public health (kesmavet), 9 Health services, 9 Regional development planning services, 9 Regional staffing services, Banten Province. The results comprised eight questions: 1). Are the organization bureaucratic? 2). Positions, roles, and authority of veterinarians? 3). Is the veterinary authority regarding rabies control already in line with the implementation? 4). Strategic policy 5). Cooperations OPD, 6). Regional Policy 7). Veterinarian openings, and  8). Enforcement and improvement of otovet institutions. The answers were: Technical services of animal health and veterinary public health: 0%, 33,33%, 77,78%, 100%, 33,33%, 44,44%, 77,78%, 100% answered yes; 100%, 66,67%, 22,22%, 0%, 66,67%, 55,56%, 22,22%, 0% answered no. Abstract

Avian pathogenic Escherichia coli (APEC) merupakan penyebab kolibasilosis pada unggas, salah satu penyakit pernapasan yang menyebabkan permasalahan yang serius di industri perunggasan. APEC dapat mengakibatkan tingginya jumlah kematian dan angka afkir, penurunan produksi, dan tingginya biaya pengobatan. Manifestasi kolibasilosis yang biasa terjadi adalah airsacculitis, perihepatitis, dan perikarditis. Serotipe APEC yang banyak diidentifikasi di lapangan adalah O1K1, O2K1, dan O78K80. Embryo lethality assay (ELA) adalah salah satu metode yang dapat digunakan untuk mengetahui virulensi serotipe APEC. Tujuan dari penelitian ini adalah untuk mengetahui karakteristik virulensi isolat APEC berbagai serotipe. Lima isolat APEC serotipe O1K1, O2K1, O78K80, O157H7, dan unknown serotype digunakan untuk pengujian virulensi dengan menginokulasikan bakteri ke dalam cairan alantois telur ayam berembrio spesific pathogenic free (TAB SPF) umur 12 hari. Masingmasing serotipe dibutuhkan telur sebanyak 10_butir, dengan dosis bakteri 100-500 CFU/ 0.1 ml. Candling dilakukan setiap hari, sampai umur embrio 18 hari, untuk menentukan jumlah kematian dan perubahan lesi patologi. Setelah diamati selama 6 hari, persentase TAB yang mati setelah diinokulasi dengan APEC serotipe O1K1, O2K1, unknown serotype adalah 100% (10/10), serotipe O78K80 90% (9/10), dan serotipe O157H7 70% (70%). Seluruh ayam yang mati mengalami lesi patologi adanya hemoragi daerah kranial dan ektremitas. Seluruh isolat E. coli dalam penelitian ini memiliki virulensi tinggi.

Abstract

In the tropical area such as in Indonesia, ruminant productivity is relatively low due to, among others, the low quality of nutrition that leads to low-efficiency metabolism at the level of ruminal fermentation, post rumen digestibility, and intermediary metabolism. This study was conducted with the objective to analize effect of methionine hydroxyl analog (MHA) supplementation on ruminal fermentation profiles of Indigenous sheep specifically in the increase of ruminant productivity. In vitro utility test was conducted using rumen fluid of the indigenous sheep and sample of ration having a proportion of grass and concentrate of 30%:70%, on dry matter basis. The treatments were three levels of MHA supplementation; T0: 0%, T1: 3%, and T2: 6% of dry matter (DM) concentrate. Variables measured were dry matter digestibility (DMD), organic matter digestibility (OMD), production of VFA, NH3, as well as total protein, and molar proportion of partial VFA of rumen fluid. Data were analyzed using analysis of variance (ANOVA) in a completely randomized design (CRD). The 6% MHA supplementation increased OMD with the highest production of total protein from 28.57 mg/g (T0) to 40.49 mg/g (T2) (P<0.05). Meanwhile, the lowest ratio of acetate : propionate was from 2.74 (T0) to 2.33 (T2) (P<0.05). It can be concluded that supplementation of MHA up to 6% in the concentrate increases the performance of Indigenous sheep ruminal fermentation and feed utility.
