Abstract

The aim of this research was to determine the histopathological finding of rat (Rattus novergicus) spleen infected with Trypanosoma evansi and treated with willow bark (Salix tetrasperma Roxb) extract. This research used 25 male rats divided into four treatment groups and fed ad libitum. Group 0 (K0) was control group, group I (K1) was infected with 103/0.3 ml. T. evansi, group II (K2), III (K3), and IV (K4) were infected with103/0.3 ml T. evansi and treated with willow bark extract 30, 45, and 60 mg/kg bw, respectively. All groups were treated for 3 consecutive days orally using a stomach tube. On day four, rats were euthanized using chloroform and necropsied, then spleen were collected for histophatological examination. The results showed that all spleen on group K1, K2, K3, and K4 showed hyperplasia of white pulp, hemosiderin, and necrosis with different severity levels compared to normal spleen structure. In conclusion, the administration of willow bark extract at the dose of 30 mg/kg bw has a potency to prevent spleen from T. evansi infection. However, the higher the dose of willow bark extract the higher the level of spleen damage.Key words: spleen, T. evansi, willow bark extract

Abstract

This study aimed to find out the parasitemia of DDY white mice infected with T. evansi of Pidie and Pemalang isolates obtained from Bbalitvet Bogor. A total of 12 mice were divided into 3 treatment groups. Group 1 (K1) without any treatment, group 2 (K2) was inoculated with 104 T. evansi Pidie isolate, and group 3 (K3) was inoculated with 104 T. evansi Pemalang isolate. Parasitemia examination was carried out every two days and the level of parasitemia was observed as well. Parasitemia examination was conducted until all mice died. Parasitemia of mice infected with Pidie isolate was characterized by rapid rise of parasitemia in blood (107-108/mL of blood) in a short time (2-4 days) since first parasitemia was detected and followed by death at day 4. The parasitemia of mice infected with Pemalang isolate increased in the blood (108-109/mL of blood) on day 4 and maintained for a few more days and then fluctuated for a few more days before the animal was dead. In conclusion, there was the difference in parasitemia level between Pidie and Pemalang isolates. Key words: Trypanosoma evansi, parasitemia, Pidie isolate, Pemalang isolate

Abstract

Abstract

Microbial contamination of fish has the potential to cause product quality degradation and cause fishborne disease. Microbial contamination can occur at any stage of fish processing. This study aimed to determine the total plate count and coliform at each stage process of milkfish (Chanos chanos) boneless. Milkfishes were obtained from three processing stage, namely raw material (fresh fish) stage, the fish bone removal stage and the baking fish stage. The parameters observed were the number of colonies growing as total plate count and coliform as the most probable number (MPN). The results showed that the highest ALT contamination was found at the bone removal stage with an average of 1.7x104 CFU/g but this figure did not exceed the standard limit and the lowest was at the firing stage with an average of 1.2x103 CFU/g. The highest coliform was found in the fresh fish stage, namely 129 MPN/g and the lowest was 3 MPN/g. From this research, it can be concluded that the total microbial plate number does not exceed the SNI 7288: 2009 standard and the coliform exceed the SNI 7388: 2009 standard.
