Abstract

This study aims to determine the sensitivity level of gentamicin, chloramphenicol and penicillin antibiotics to inhibit the bacterial growth of P. aeruginosa taken from preputium isolate of Aceh cattle. This Stock of Pseudomonas aeruginosa isolate from Microbiology Laboratory of FKH USK, re-identified its purity through: indole test, methyl red test, sugar test and as well as on Nutrient Broth (NB) media; incubated at 37ºC for 24 hours; observed the turbidity level of 0.5 Mc Farland solution. Then, the antibiotic sensitivity test was carried out using the method of Mueller Hinton Agar (MHA). The observations were made by measure the inhibition zone using the calipers in millimeters (mm) against antibiotics. The results of the observation of the inhibition zone on the gentamicin antibiotic was averaged of 25.5 mm, 23.7 mm chloramphenicol, and 12.1 mm penicillin. This study concluded that the gentamicin, chloramphenicol, and penicillin antibiotic were still effective against P. aeruginosa bacteria with the highest level of sensitivity seen in gentamicin antibiotic.

Abstract

The purpose of this study is to determine the effect of the red dragon fruit peels (Hylocereus polyrhizus) extract on the growth of Salmonella pullorum. This research was conducted at the Laboratory of Microbiology, Veterinary Faculty, University of Syiah Kuala, Banda Aceh. Red dragon fruit peels that used in this study is obtained from  ripe fruit and Salmonella pullorum cultures is  obtained from  Microbiology Laboratory of Veterinary Faculty Unsyiah. Salmonella pullorum culture added evenly on the surface of Mueller Hinton Agar, the number of the bacteria have been standarized using Mc Farland'1 method. Discs soaked in the red dragon fruit peels extracts at different concentrations (20 mg/ml, 40 mg/mL, and 60 mg/ml). Ampicillin is used as positive controls and DMSO (dimethyl sulfoxide) was placed on the surface of Mueller Hinton Agar. The parameters in this study is the inhibition zone diameter. Data were analyzed descriptively. The results showed that the red dragon fruit peel extract is able to inhibit the growth of Salmonella  pullorum and the average of  inhibition diameter zone at a concentration 60 mg / ml is 9.6 mm, 40 mg / ml is 9.4 mm and 20 mg / ml is 9.3 mm

Abstract

This research was aimed to study the presence of ketone bodies in urine of sumatran orangutan as a medical record of orangutan at new reintroduction centre of pine nature reserve Jantho, Aceh Besar. Urine was collected from 8 individual sumatran orangutan (Kiskis, Amin, Mawasudin, Marconi, Ruben, Ayu, Yusniar, and Coty) which placed in individual cage and semiwild orangutan. Sampling was carried out in the morning after the sample going out of nest or before given the first feed for individual inside the cage. Sample was collected 4 times with three repetitions in 4 weeks from June to July 2013. The examination of urine was conducted subsequently after urine collection by dipped the striptest in 5 mL of urine for 60’second. Data were analyzed using descriptive qualitative method. The semiquantitative results of ketone body was obtained by striptest with 5 scale of colors change: negative (0 mg/dL), trace (5 mg/dL), small (15 mg/dL), moderate (40 mg/dL), and large (60 mg/dL-180 mg/dL). The results showed that three individuals placed in the cage (Kiskis, Amin, and Mawasudin) were negative for ketone bodies. In contrast for 5 semiwild orangutans namely Marconi was negative for ketone bodies, Ruben was positively detected for ketone bodies found in the third week (M3) with levels of 5 mg/dL, Ayu was positively detected for ketone bodies in the first week (M1) with levels of 15 mg/dL and in the second week (M2) with a concentration of 5 mg/dL, Yusniar was positively detected for ketone bodies with levels of 5 mg/dL in the first week (M1) and fourth week (M4), and Coty was positively detected for ketone bodies with levels of 5 mg/dL in the second week (M2) and fourth week (M4).Key words: orangutan, ketone bodies, reintoduction, urinalysis

Abstract

This research aims to know the influence of the equilibration to the quality of the swamp buffalo (Bubalus bubalis) cement after thawing including: motility, and percentage of live spermatozoa. This research used the fresh cement from a 4 years old water buffalo was collected using an artificial vagina,and evaluation cement for macroscopic and microskopic to see if cement was worthy or not worthy to frozen. Good quality cement was diluted by adding Andromed, then semen were incorporated into the straw using the machine filling sealing followed by equilibrated with 3 treatment groups. Group I (n = 3 straw) with equilibration time of 3 hours, the Group II (n = 3 straw) with a time of equilibration was 5 hours, and group III (n = 3 straw) with equilibration a time of 7 hours. After equilibrated, straw frozen and stored in liquid N2 (BIBD Tuah Sakato procedures). The results show that the length of time the real equilibration effect (P 0.05) on the motility and percentage life of swamp buffalo spermatozoa. Equilibration 5 hours of time differs markedly with equilibration time 3 and 7 hours, motility, and percentage of live spermatozoa (44.15±1.70; 46.50±1.36 vs 14,55±1,58; 1.42±37.38 vs. 15.37±1.58; and 37.50±1.25). In conclusion, equilibration time is showed on the cement of swmp buffalo after 5 hours thawing.Key words: equilibration, swamp buffalo, thawing

Abstract

Penelitian ini bertujuan mengetahui keberhasilan inokulasi Trypanosoma evansi (T. evansi) pada mencit Mus musculus strain Balb-C yang berasal dari darah sapi lokal di Rumah Potong Hewan kota Banda Aceh. Dari 205 sampel darah sapi lokal, 15 sampel positif mengandung T.evansi dengan metode hematokrit dan semua darah disuntikkan masing-masing pada  mencit secara intraperitoneal untuk memperbanyak jumlah parasit. Semua mencit diperiksa secara regular 2 kali seminggu dengan mengambil darah dari ujung ekor. Hasil penelitian menunjukkan bahwa dari 15 mencit yang diinokulasi, satu ekor mati pada minggu ke-3, satu ekor mati pada minggu ke-5, satu ekor mati minggu ke-8, dua ekor mati minggu ke-9, dan tiga ekor mati pada minggu ke-14, sementara 7 ekor masih hidup sampai minggu ke-20. Dari ke-8 ekor yang mati semuanya tidak ditemukan berkembangnya T. evansi pada mencit

Abstract

Halal slaughter is a way to kill animals by the process of draining blood properly, perfectly, and according to the sharia. One of the critical points of halal in the slaughtering process is the timeof death of the broilers after slaughter. The objective of this study was to determine the difference between the time of death of hanged broilers and unhanged broilers, in order to find out the right time before the scalding. The samples used were 10 male broilers (Gallus domesticus) five weeks old with body weight 1.6-1.7 kg/head. The variable observed was the time of death of hangedand unhanged broilers after slaughter looking at the signs of  death,  such  as  heart  rate  stopped,  pupil or  eyelid  reflexes,  and  stopped breathing. This study used a completely randomized design (RAL) with two treatments, such as slaughter of chickens in a hanging position (P1) and chickens without hanging (P2). The test data were analyzed using the independent T-test SPSS 25. The results obtained that the average time of death of hanged broilers after slaughter was 82,80 seconds, while the average time of death of unhanged broilers was 162.60 seconds. The test results of the two treatments with a significant level of 0.05 showed a significant difference. The conclusion of this study was time of death of hanged broilers is faster than that of unhanged broilers after slaughter

Abstract

This research aim to study the effect of curry leavesaddition in extracts form with different concentration and storage time to the total microbes on fresh beef. This study use beef sample obtained from Lambaro Market Aceh Besar. The sample was divided into 36 sections, each wieght 25 grams and curry leaf extracts add with 0%, 25% and 50%concentrations, and were stored at 4-10oC. Microbiological examination of beef using pour plate method. Observations were made every 3 days in day 0, 3, 6 and 9. Measured parameters is total bacteria (CFU / gr). Data processing is using analysis of variance and the significantly different results tested with Duncan Test. Observation total of microbes on day 0 the concentration of 0% (without the addition of curry leaf extract), 25% and 50% was 2.7 x 102 CFU / gr, 2.2 x102 CFU / gr and 6.8 x101. It still can be consumed because it is still below the safety zone1x104 CFU / gr accordance with SNI 01-6366-2000 (SNI 2008). Meanwhile the observations at 0% concentration after 3 days already exceeded the standard of SNI 4.2 x106 cfu / gr. Meat with 50% curry leaf extract addition is still safe to eat up to day 9. Interaction effect between the concentration and the curry leaf extract addition and storage timeto the total microbe is not significantly different (P 0.01). Curry leaf extract addition and storage time factors is very significant (P 0.01) to the total microbes. These results indicated that curry leaf effective  into a preservative against fresh meat.

Abstract

The purpose of this study was to find out whether the refill water (AMIU) was contaminated with coliform or not. The methods used were presumptive, confirmed, and completed test. Sample of this study was refill water (AMIU) from five refill water shops located in Ilie village, sub-district of Ulee Kareng, Banda Aceh. The sample was taken 3 times with interval period of one week. Sample was culture in lactose broth and incubated at 37° C for 48 hours. If it did not form gas in Durham tube, presumptive test result was considered negative and the samples were not proceed to the other test. The data was presented using descriptive analysis. It could be concluded that AMIU from 5 shops in Ilie Ulee Kareng, Banda Aceh is negatively contaminated with coliform bacteria.

Abstract

Penelitian ini bertujuan mengetahui potensi getah batang pisang kepok dalam proses penyembuhan luka operasi. Penelitian ini menggunakan 10 ekor anjing lokal (Canis familiaris) sehat yang berumur rata-rata 6 bulan dengan bobot badan 6-7 kg. Sebelum perlakuan dilaksanakan, semua anjing diadaptasikan selama tujuh hari di dalam kandang individu, setiap anjing dilakukan operasi laparotomi. Anjing dibagi dalam dua kelompok secara acak. Kelompok I diinjeksi penisilin-G dan dioleskan salep oksitetrasiklin pagi dan sore sebagai kontrol, kelompok II dioleskan getah batang pisang kepok pagi dan sore hari sebagai perlakuan. Pengamatan dilakukan setiap hari pada pagi hari sampai luka operasi sembuh. Data yang diperoleh dianalisis secara deskriptif berdasarkan pengamatan makroskopis terhadap luka kelompok I dan dibandingkan dengan kelompok II berdasarkan adanya kemerahan, kebengkakan, dan pertautan tepi luka. Pada kelompok II terjadi penyembuhan luka operasi yang sama dengan kelompok I. Dapat disimpulkan bahwa, penggunaan getah batang pisang kepok dapat menyembuhkan luka, sama halnya dengan proses kesembuhan luka yang menggunakan injeksi penisilin-G dan dioleskan salep oksitetrasiklin.

Abstract

The aim of this research was to find out the effect of Aspergillus niger-fermented soybean residue in ration on the histomorphometry of the small intestine villi of local chickens (Gallus domesticus).This study used a completely randomized design. Nine female chicken aged 15-16 months were divided into three groups. Each group consisted of three chikens. All chicken were given 324-2 ration and water ad libitum. Group P1 (control group) was given 100 g ration each day, group P2 was given a combination of 85 g ration and 10 g unfermented soybean residue everyday whereas group P3 was given 85 g ration and 10 g Aspergillus niger-fermented soybean residue everyday. Treatment was performed for 30 days. On day 31, chickens were slaughtered and subsequently three parts of the small intestine that consists of duodenum, jejunum, and ileum were collected and fixed in 10% neutral buffered formaline (NBF) solution for histological preparation. To measure the surface area of three regions of intestine, all histological sections were stained with Masson’s trichrome Goldner's modification. Based on the statistical analysis, there was no difference in height of villi among three parts of small intestine (P0.05), but there were statistical different (P0,05) in ileum basal and duodenum apical width of three groups after supplementation with fermented soybean residue and non-fermented. Based on the results of this experiment, it could be conluded that supplementation non-fermentation and the fermentation of soybean residue with A. niger has no impact on the height of vili ileum, but it increase the width of vili ileum and apical vili duedenum intestin of local chickens. ____________________________________________________________________________________________________________________ Key words: chicken, soybean residue, fermentation, small intestine
