Abstract

Brucella abortus Strain Specific-Polymerase Chain Reaction (BaSS-PCR) is a single multiplex PCRtechnique which able to identify and differentiate between Brucella abortus field strains (biovar 1, 2, and4), B. abortus vaccine strains, Brucella species, and non-Brucella species. In this study, BaSS-PCR wasapplied to identify local isolates of B. abortus in order to investigate the B. abortus strains that infectedcattle in Indonesia. Fifty local strains of B.abortus isolated from infected cattle in Java (Jakarta andBandung), South Sulawesi (Maros), East Nusa Tenggara (Kupang and Belu) were used in this study. TheDNA bands were observed by agarose gel in the presence of ethidium bromide. Identification was performedbased on the size and number of DNA products amplified by PCR from each isolates. The results showedthat the 50 isolates were of B. abortus field strains. This finding showed that the cause of bovine brucellosisin Indonesia is B. abortus field strains.

Abstract

Administration of pregnant mare serum gonadotropin (PMSG) hormone prior to mating increasesquality of lamb born on one and two litter sizes, but in three litter size the motality is higher. Administrationof traditional herbal medicine “jamu” consisting of ginger,green chiretta, cinnamon, Zingiberzerumbet, andpepper during gestation increases performance of lambs. This research was conducted to explore theeffectivity of PMSG injection prior to mating and jamu administration during gestation to increase lambsperformance. Eighteen priangan fat-tail ewes with weight around 20-25 kg were injected with PGF2á at adose of 10 mg/ewe twice with 11 days interval to synchronize estrous cycle. Injection of PMSG at a dose of200 IU/ewe was conducted at the second PGF2á injection. The experimental ewes were mated naturally anddivided into a randomized design with a 2 x 3 factorial arrangement with three replications. The firstfactor was dose of PMSG with two levels i.e., 0 and 200 IU/ewe. The second factor was dose of jamu withthree levels i.e., 0, 15, and 30 mL/ewe. Jamu was administered orally every week during gestation. Injectionof PMSG and administration of jamu decreased prenatal mortality, increased ratio of lamb per ewe, increased total lamb born with average birth weight and total birth weight were higher in jamu at a doseof 15 and 30 mL/ewe by 30.02 and 31.76%, repectively. During the first month postnatal, lambs born toewes injected by PMSG and administered jamu had higher number of lambs survive, average weight, totalweight around 3 times as compared to control. It was concluded that injection of PMSG prior to mating andadministration of jamu during pregnancy increase lamb birth weight and improve the quality of lamb.

Abstract

The aim of this study was to identify the environmental factors of chickens based on the different altitudebreeding against the microclimatic and performance production in Takalar district, South Sulawesi. Atotal of 180 Lohmann strain chickens consisted of 90 male chickens and 90 female chickens. A randomizedblock design with two factorial patterns was used in this study. The first factor was based on altitude (low<300 mabove sea level, medium 300-600 m, and high >700 m ), the second factorwas based on gender(male and female). Data were analyzed using analysis of variance (ANOVA) and then the significant effectwas followed with Tukey test. The results showed that the altitudehad a significant effect against thephysiological status (rectal temperature), production performance (feed intake), water intake, final bodyweight, and FCR. The male and female chickens had a significant effect against feed intake, water intake,breeding final body weight, and carcass weight. There was interaction between altitude and sex on weightgain performances. The altitudes may influence the effects of physiological status and productionperformance. The production performance of male seems to behigher than the female.

Abstract

Coccidiosis is one of the important diseases in poultry industry. In Indonesia the morbidity of thedisease is between 80 to 90%. A rapid and prompt diagnosis would be one of the essential steps ineradication and control of the disease. The objective of this study is to determine the protein profile ofsporozoite of Eimeria tenella isolated in Yogyakarta using sodium dodecyl sulphate polyacrilamide gelelectrophoresis (SDS-PAGE) protein analysis. Protein analysis was performed in 12% polyacrilamide geland further electrophoresis at 100 volts and over-staining with Coomasie brilliant blue. The resultsshowed that the sporozoite of E. tenella isolated in Yogyakarta contained five proteins with molecularweights of 15, 26, 32, 80, and 91 kDa, respectively.

Abstract

Research was conducted at Bukit Jimbaran, Badung Bali from September to October 2007 to investigatethe lenght and circular body dimension of Bali cattle. Ten young male bali cattle 0.5-1.5 year old weremeasured for three times at 2 weeks interval. Collected data was analysed with regretion-corelation byallometric growth lines Y = aXb. F test and coeficient-correlation was then used to chek the validity andacurate models. Result showed that the lenght head, neck, body anterior and posterior had medium potentialgrowth. Circular neck anterior grew early with a high potential. The circular neck posterior growth potentialwas medium, where as circular thorax and abdomen was law poitential growth.

Abstract

An experiment has  conducted to investigate the effect level of fermented banana peels by Rhyzopusoligosporus   in  ration to growth of broiler. The experiment was designed use Completely RandomizedDesign with four treatments: as a control ration (0%  fermented banana peel); 5% fermented banana peel;10%  fermented banana peel; 15% fermented banana peel, with four replicates. There were six ayampedagings a weeks with average   weight  112.30 ± 2.74 g in each replicate. The result there was a 15%fermented banana peels in broiler ration was not significant (P>0,05)  to feed consumption, but itsignificantly (P<0,01) effected gain, and feed conversion . Fermented banana peels by R. oligosporus couldbe used as much as 10% in broiler ration.

Abstract

The aim of this study was to evaluate the capability of two human commercial enzyme-linkedimmunosorbent assay (ELISA) kits(DRG International Inc.,Germany and GBC Taiwan) for measuringestradiol (E2) and progesterone (P4) in plasma of kacang goat. Three healthy and non pregnant femalekacang goats aged 2-3 years with regular estrous cycles were used in this study. Blood samples werecollected from the jugular vein using a 21G venoject every two days and it was intensified every day duringthe period before heat. Collected plasma were stored at -20ºC until the analysis. Capability validationwas conducted by measuring accuracy (parallelism test), sensitivity and precision.Parallelism test usingDRG commercial kit showed that sample curvewas parallel with standard curve of E2 and P4. In contrastit was not parallel with standard curve of GBC commercial kit. Sensitivity was measured from the lowesthormones concentration of E2 and P4 at 90% binding that were 25 pg/ml and 0.14ng/mL in DRG kit, whilein GBC kit were 5 pg/mL of E2 and 0.2 ng/mL of P4, respectively. Coefisien of variation of intra- andinterassay for both ELISA commercial kits were less than10%. It can be concluded that DRG commercialELISA kit E2 and P4 can be used to analyse female kacang goat blood plasma, while GBC commercialELISA kit E2 and P4 are not recommended.

Abstract

Catharanthus roseus is one plant recognized as medical potential which can decrease cholesterol. Thepresent study was carried out to evaluate the effect of aqueous leaf extracts of C. roseus on plasma lipidprofil levels in rats cholesterol rich diet and atherogenic index. This study was carried out on 15 SpraqueDawley male rats randomly distributed into five groups (n=3). Rats hypercholesterolemic wereadministrated cholesterol rich diet containing 1% (w/w). Normal control group with normal diet whitoutextracts (K1), positive control hypercholesterolemic group (K2) with cholesterol rich diet whitout extracts,and others three groups (K3-K5) were feed high cholesterol and 1 mL of aqueous leaf extracts of C. roseuswith a dose of 20% (w/v), 40% (w/v) and 80% (w/v) respectively, administered twice daily 1 mL orally.Treatment was given for 28 days. After treatment, the plasma lipids profile such cholesterol total, highdensity lipoprotein (HDL), low density lipoprotein (LDL), triglyceride (TGA) level were measured. Theresults showed that during the period of treatment with cholesterol rich diet have produced under condition hypercholesterolemic of rats with average cholesterol levels of 124.33 ± 4.04 mg/dL (K2 group). Treatmentwith extract dose of 80% showed the best results among the other doses in reducing levels of total cholesterol,triglycerides, LDL and raise HDL levels. Extract treatment dose of 80% up to four weeks resulted incholesterol total (79.33±3.51 mg/dL), TGA (72.33±6.65 mg/dL), HDL (55.00±3.60 mg/dL), and LDL(9.87±5.34mg/dL). In addition, value ratio of cholesterol:HDL was 1.4: 1 and atherogenic index value was0.44. These results suggest that extracts of leaf C. roseus optimum doses 80% (w/v) has hipolipidemicactivity in hypercholesterolemic rats and it has low atherogenic index value.
