Abstract

Semakin tinggi kadar trigliserida meningkatkan kekakuan arteri sehingga resiko kardiovaskular semakin meningkat. Kata kunci : trigliserida, kekakuan arteri, denyut jantung, kardiovaskular   ABSTRACT Epidemiological studies report that triglycerides are an independent risk factor for cardiovascular disease even though the target level of low density lipoprotein (LDL) has been achieved with statin drugs. It is suspected that there is a relationship between triglyceride levels and arterial elasticity. Arterial stiffness has been recognized as being closely related to cardiovascular disease. This study aims to determine the effect of hypertriglyceridemia on arterial stiffness in animal models of Wistar rats induced by a high-fat diet and 25% fructose in drinking water. A total of 10 rats were randomly divided into 2 groups consisting of a normal control group receiving normal feed and a positive control group receiving a high-fat diet and 25% fructose in drinking water for 28 days. Measurements of pulse wave velocity (PWV), heart rate, and serum triglyceride levels were carried out on day 28. The positive control group experienced an increase in serum triglyceride levels accompanied by an increase in PWV and heart rate that was statistically significantly different (p <0.05) compared to the group normal. The results concluded that there was a positive relationship between triglyceride levels and arterial stiffness. Higher triglyceride levels increase arterial stiffness. it increases the risk of cardiovascular disease. Keywords : triglyceride, arterial stiffness, heart rate, cardiovascular

Abstract

Cardiovascular disease is the leading cause of death worldwide. One of the major risk factors for cardiovascular disease is hyperlipidemia. This study aimed to determine the potential of Zingiberaceae (10 species) as inhibitor of HMG-CoA reductase enzyme activity and lipid peroxidation. This study was conducted by 2 methods including assay of HMG CoA reductase inhibition and lipid peroxidation test. The study was performed by in vitro method, using 20% rat liver homogenate. The inhibition of HMG CoA reductase enzyme was done by reacting liver homogenate, HMG CoA substrate, which added the Zingiberaceae extract compared with simvastatin as standard drug. The absorbance of the mixture was measured by a Microlab 300 spectrophotometer at a 340 nm wavelength. Lipid peroxidation assay was induced by the FeSO4.7H2O solution. The absorbance value measured using a spectrophotometer at a 532 nm wavelength. Lipid peroxidation inhibition was characterized by absorbance of the test extract, compared with the control group. The obtained data was calculated as percent of inhibition and was used to calculate IC50 extract test. The results showed that the 10 ethanolic extracts of Zingiberaceae rhizomes have activity as enzyme inhibitor HMG-CoA reductase with IC50 value range 65.8±4.1 – 203.3±15.2 ppm, and inhibition of lipid peroxidation with IC50 value range 13.5±5.0 – 219.6±4.3 ppm. This study can be concluded that the Zingiberaceae rhizomes have potential role as antihyperlipidemic agents through inhibition of HMG CoA reductase enzyme activity and preventing lipid peroxidation.

Abstract

Abstract

A strong link exists between arterial stiffening and cardiovascular conditions, particularly hypertension. Hypertension, a significant contributor to mortality rates, particularly affects elderly populations. The processed form of Allium sativum, known as black garlic, is renowned for its medicinal properties, including its ability to lower blood pressure. This study examined the blood pressure-lowering effects of black garlic extract, employing male Wistar rats aged two to three months. The experiment involved six distinct rat groups, each comprising of four animals. The two groups served as controls, whereas the remaining four underwent different treatments. Captopril was administered to one group at a dose of 2.5 mg/kg, whilst the remaining three groups were given black garlic extract as BG50, BG100, and BG200. All groups, with the exception of the control normal, underwent a 28-day induction protocol that included 25% fructose in drinking water, cholesterol (200 mg/kg), and propylthiouracil (12.5 mg/kg). The test substances were administered starting on day 14 concurrently with the induction phase. The evaluated parameters included blood pressure, arterial stiffness, nitric oxide (NO) levels, and renal histopathology. Black garlic extract significantly lowered blood pressure, enhanced arterial elasticity, and increased NO levels (p < 0.05) compared with the controls. Renal histopathological analysis revealed protective effects, suggesting prevention of kidney damage. These findings suggest that black garlic may have cardiorenal protective effects and could be useful for managing hypertension and arterial stiffness.

Abstract

Hypertension is a global health problem requiring effective and safe management. Conventional therapies often face challenges, such as drug resistance, side effects, and patient non-compliance, increasing the need for alternative therapies. This article aims to review the effectiveness of plant extract combinations as antihypertensive therapies. This study used a literature review approach to studies published in the last 10 years, focusing on the mechanism of action, effectiveness, and synergistic potential of plant combinations. The results showed that combinations such as Anredera cordifolia (ten.) v. Steenis and Sonchus arvensis L. provided a decrease in systolic blood pressure (SBP) by 27.95 mmHg and diastolic blood pressure (DBP) by 33.89 mmHg, approaching the effectiveness of standard drugs such as valsartan. The main mechanisms include the inhibition of angiotensin-converting enzyme (ACE), vasodilation through the release of nitric oxide (NO), and diuretic effects. The combination of plant extracts shows potential as a safe and effective alternative; however, further clinical trials are needed to strengthen these findings. Keywords: Combination of Plant Extracts, Antihypertensive, Alternative Therapy.
