Abstract

Pesisir bulls from West Sumatra, Indonesia, have the potential to be valuable meat producers due to their adaptability and unique characteristics, but reproductive challenges necessitate the use of artificial insemination (AI) technology. The success of AI relies on the quality of frozen semen, which is influenced by thawing conditions, making the evaluation of sperm kinematics through Computer-Assisted Sperm Analysis (CASA) crucial for a comprehensive assessment of sperm function. This research evaluated the kinematics of frozen semen spermatozoa from Pesisir bulls. Frozen semen samples (n = 20) were thawed at 37°C for 10, 20, and 30 s and at 25°C for 30, 45, and 60 s. Kinematic parameters were observed using a Computer-Assisted Sperm Analyzer (CASA), including total motility (M), progressive motility (PM), the velocity of the average path (VAP) (?m/sec), the velocity of curvilinear (VCL) (?m/sec), the velocity of straight line (VSL) (?m/sec), straightness (STR) (%), linearity (LIN) (%), wobble (WOB) (%), amplitude of lateral head displacement (ALH) (?m), and beat/cross-frequency (BCF) (Hz). Thawed frozen semen at a temperature of 37°C for 20 s significantly (p<0.05) increased M, PM, and ALH. Furthermore, thawed frozen semen at 37°C for all durations and at 25°C for 60 s showed better quality for VAP, VCL, and VSL (p<0.05). Meanwhile, thawed frozen semen at 25°C increased STR, LIN, and WOB. However, thawing temperature and duration had no significant effect on BCF. Correlation analysis conducted on semen thawed at a temperature of 37°C for 20 s found that VAP was correlated with VCL, VSL, and ALH, while semen thawed at a temperature of 25°C for 60 s found that VAP was correlated with VCL, VSL, STR, LIN, and ALH. Most kinematic parameters were significantly better in the thawing treatment at 37°C compared to 25°C, suggesting careful consideration

Abstract

This study aimed to evaluate the effect of different vitamin C concentrations (0, 0.1, 0.2, and 0.3 g/100 mL) in egg yolk citrate extender and storage time (0, 24, 48, 72, and 96 hours) on the quality of Bangkok rooster semen stored at 5°C. Using a split-plot-in-time design with four replications, semen quality was assessed for motility, viability, abnormality, intact plasma membrane (IPM), and longevity. Results showed a significant interaction (P<0.01) between vitamin C concentration and storage time on motility, and significant effects (P<0.05) on viability, abnormality, and IPM. The 0.2 g vitamin C dose preserved >40% motility and >50% viability for 96 hours and extended sperm longevity up to 11.25±0.95 days. These findings suggest that 0.2 g/100 mL vitamin C is the optimal concentration for preserving rooster semen quality during short-term cold storage.

Abstract

This study aimed to find the optimum conditions for extracting the carotenoid content in orange sweet potato in an applied application. The extracted carotenoids will be used as raw materials to produce rose-scented aromatic compounds (b-ionone). The extraction process was carried out on dry matter using maceration technique and using hexane as solvent. Other solvents such as methanol, ethanol, isopropanol and choloroform were also used to extract carotenoids from orange sweet potato. The use of wet samples and dry samples was also carried out in this study. The ratio of the solvent used with the material was 1:3. Based on the yield and spectrophotometric characterization, it was known that the extraction process with hexane solvent can extract carotenoid compounds from dry orange sweet potato powder if the extraction process is carried out 7 times

Abstract

Kontaminasi bakteri pada semen sapi merupakan salah satu faktor yang memengaruhi kualitas dan keberhasilan program inseminasi buatan. Penelitian ini bertujuan untuk mengidentifikasi jenis bakteri yang terdapat pada semen beku sapi simental dan efektivitas variasi antibiotik dengan perlakuan pengencer berbeda. Penelitian dilaksanakan melalui tiga tahap, yaitu koleksi semen segar dari delapan ekor sapi simental di Unit Pelaksana Teknis Daerah (UPTD) Balai Pengembangan Teknologi dan Sumber Daya (BPTSD) Tuah Sakato, Payakumbuh, Sumatra Barat. Pengenceran semen dilakukan dengan tiga perlakuan (P1= tanpa antibiotik; P2= antibiotik Penicillin-Streptomycin; P3= kombinasi Gentamisin, Tilosin, Lincomisin, Spektinomisin/GTLS), serta isolasi dan identifikasi bakteri pada semen cair dan semen beku di Laboratorium Bakteriologi, Balai Veteriner Bukittinggi, Sumatra Barat. Metode isolasi menggunakan media selektif dan identifikasi dilakukan melalui pewarnaan Gram, uji motilitas, katalase, oksidase, dan uji biokimia. Hasil penelitian menunjukkan bahwa dari semen segar diisolasi 12 jenis bakteri, sedangkan dari semen beku diidentifikasi bakteri Alcaligenes spp., Chromobacterium spp., Corinebacterium spp., Klebsiella spp., Micrococcus spp.,  Alcaligenes spp., Haemophilus spp., Mycoplasma spp., Proteus spp., dan Serratia spp. Semen tanpa antibiotik (P1) memiliki tingkat kontaminasi bakteri tertinggi dengan lima jenis bakteri terdeteksi, sedangkan pada perlakuan P2 teridentifikasi tiga jenis bakteri dan P3 lima jenis bakteri namun dengan populasi lebih terkendali. Bakteri oportunistik seperti Klebsiella spp. dan Proteus spp. masih ditemukan meskipun menggunakan GTLS. Simpulannya, pada semen beku yang diencerkan dengan antibiotik masih ditemukan bakteri. Penambahan kombinasi antibiotik GTLS pada pengencer lebih efektif menekan pertumbuhan bakteri patogen dibandingkan Penicillin-Streptomycin, serta berpotensi meningkatkan mutu semen beku dan keberhasilan inseminasi buatan.

Abstract

Animal pregnancy testing is one of the techniques/tools used to detect animal pregnancy. This livestock pregnancy test kit can identify early pregnancy in beef cattle, which is useful for farmers to take appropriate actions in reproductive management and cattle care. The objectives of this activity are: 1. to improve farmers' knowledge of early pregnancy detection and the cattle reproduction process to increase the birth rate of calves, 2. to enhance farmers' skills in early pregnancy detection in cattle to improve efficiency in farm operations, and 3. to disseminate early cattle pregnancy technology using pregnancy test kits to farmer groups within the campus community in Padang City. This activity was conducted in a cattle group within the campus community located in Pauh Subdistrict, Padang City. The methods of this activity included counseling, training/demonstration, and evaluation. Participants in this activity were the Taruko Sakato Farmer Group, lecturers, and students from the Faculty of Animal Husbandry, Andalas University. The results of this activity were that participants understood the principles of the test kit, collected necessary urine samples, and improved their skills and understanding in independently and accurately detecting animal pregnancy. The application of this cattle pregnancy test kit technology is expected to assist farmers in enhancing productivity and efficiency in cattle reproductive and production management, thereby positively impacting farmers and local food security. In conclusion, this activity provided knowledge, understanding, and practical skills to farmer groups/partners in adopting innovative technology for improving their livestock farming outcomes.

The application of sperm sexing technology in cattle breeding is often limited by reduced sperm quality due to processing and storage conditions. This study evaluated the effect of extender storage duration on post-thaw sperm quality and the stability of X- and Y-bearing spermatozoa in Bali cattle (Bos javanicus) using the Bovine Serum Albumin (BSA) column method. Fresh semen with acceptable initial quality was subjected to sperm sexing. The extender was stored for 0, 1, and 2 days before use. Post-thaw sperm quality was assessed based on motility, viability, plasma membrane integrity, acrosome integrity, and abnormalities. Molecular verification was performed using duplex Polymerase Chain Reaction (PCR), targeting the glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and sex-determining region Y (SRY) genes. Data were analyzed using analysis of variance and regression models. Extender storage duration significantly affected plasma membrane integrity, viability, and motility (p < 0.05). Both sperm fractions showed decreased membrane integrity with increasing storage duration. Viability declined more in X-bearing spermatozoa, whereas motility decreased more in Y-bearing spermatozoa. In contrast, acrosome integrity and sperm morphology remained stable (p > 0.05). Regression analysis indicated negative trends, although moderate R² values suggest that storage duration contributed partially to sperm quality variation. Duplex PCR confirmed successful sperm separation through detection of the SRY gene exclusively in the Y-bearing fraction. In conclusion, extender storage for up to two days induces moderate changes in sperm quality without critical deterioration. These findings support the practical use of short-term extender storage in sperm sexing. Further studies integrating fertility trials and quantitative assessment of sexing accuracy are required to validate the reproductive performance of the processed semen under field conditions.

This study aimed to evaluate the ability of Heterotrigona itama worker bees to return to the nest from various test distances. This research is an observational study using the translocation method. There were 550 individuals of H. itama worker bees from 5 colonies reared for over two years. Each worker bee sample was identified with a mark in the form of water-based paint before being released at a testing distance every 100 meters until a distance where no more worker bees returned. The camera with a macro lens is installed at the entrance to the beehive to record worker bees returning to the hive. The video recording process was carried out over 90 minutes. The parameters observed included environmental temperature and humidity, the number of worker bees returning, and the time required for worker bees to return from each test distance. The data obtained were analyzed using Chi-square, Kruskal-Wallis, and Mann-Whitney U-test. The research showed that the maximum distance that H. itama worker bees can travel to return to the nest was 1000 meters. However, the most effective return distance was more than 500 meters because although some worker bees can return from greater distances, the number of returns significantly decreases after passing 500 meters. Therefore, the ideal distance for placing a colony of H. itama is less than 500 meters from the source of bee food plants. The minimum distance to move a colony from the initial location to a new location is around 1100 meters.

Abstract

Abstract: The demand for beef in Indonesia continues to rise, while beef production remains below this demand. This study aims to evaluate the estrus response, the onset of estrus, and the intensity of estrus in Simmental and Limousin cattle experiencing repeat breeding after the administration of the PGF2? hormone. A total of 24 cattle, consisting of 11 Simmental and 13 Limousin that met the research criteria, were injected with PGF2? hormone. The results showed that both cattle breeds exhibited a 100% estrus response and a high onset of estrus, particularly within the 73-96 hour interval after injection. Statistical analysis indicated no significant differences in the onset and intensity of estrus between the two breeds after hormone administration. These findings suggest that the application of estrus synchronization with PGF2? is effective in enhancing reproductive efficiency. Further research is recommended to explore other factors that may influence reproductive outcomes, such as nutrition and feed management.  Keyword: PGF2?, Simmental, Limousin, Estrus Synchronization. Abstrak: Kebutuhan akan daging sapi di Indonesia terus meningkat, sementara produksi daging sapi masih berada di bawah permintaan. Penelitian ini bertujuan untuk mengevaluasi respon berahi, kecepatan munculnya berahi, dan intensitas berahi pada sapi Simmental dan Limousin yang mengalami repeat breeding setelah pemberian hormon PGF2?. Sebanyak 24 ekor sapi yang terdiri dari 11 ekor Simmental dan 13 ekor Limousin yang memenuhi kriteria penelitian disuntik dengan hormon PGF2?. Hasil penelitian menunjukkan bahwa kedua ras sapi memiliki respon berahi 100% serta kecepatan muncul berahi yang tinggi, terutama pada interval 73-96 jam setelah penyuntikan. Analisis statistik menunjukkan tidak ada perbedaan signifikan dalam kecepatan dan intensitas berahi antara kedua ras setelah pemberian hormon. Temuan ini menunjukkan bahwa penerapan sinkronisasi berahi dengan PGF2? efektif untuk meningkatkan efisiensi reproduksi. Penelitian lebih lanjut dianjurkan untuk mengeksplorasi faktor lain yang dapat mempengaruhi hasil reproduksi, seperti nutrisi dan manajemen pakan. Kata kunci: PGF2?, Simmental, Limousin, Sinkronisasi Berahi.
