Abstract

The cows are one of the ruminants that have the largest contribution as a producer of meat and for meeting the needs of animal protein. Some of the factors that can affect the lack of success of a cattle farm are the extensive rearing, limitation of cage’s fasilities, bad quality of sanitation, and lack of reproduction knowledge by farmers. The purpose of this service activity was to provide knowledge and skills of local farmers in terms of toward intensive farming, based on cowshed quality, sustainable of feed and roughage, and optimal reproduction of the cows. It hopes to increase the income of local farmers further. The activity was carried out by survey method as preparation step, counseling, training, coaching, and evaluation of program. The activity was attended by members of farmer group of Batu Kalang Indah and personnel of Nagari Ampang Pulai, Pesisir Selatan Regency. The community servant shared good practices system of cow’s farm including maintenance of cages, sanitation, feeding management and roughage’s fermentation, and reproduction skills. The participants showed enthusiasm learning during training and counseling about the livestock management. It is hoped that can provide proper information about cattle farming intensively that can be applied sustainably by targeted farmer’s group.

Abstract

The administration of hormone GnRH will affect the reproductive organs and can improve the reproductive performance of buffalo. The decrease of the buffalo population is caused by various factors including genetic and environmental factors. The development of buffaloes is relatively slow because the reproduction rate of buffaloes is lower than that of cattle, besides that it is also difficult to detect estrus, the pregnancy period is relatively long and longer calving rate. This study aims to determine the best dose of GnRH on the swamp buffalo producing of dadih in Indonesia. The materials used were 16 swamp buffalo. A completely randomized design was used in this study. Buffalo is synchronized with the injection of 5 ml of GnRH hormone n on the first and 11th day, on the 10th to 12th day all donor buffaloes are injected with GnRH. Four different doses of GnRH (200, 225, 250 and 275 ml/buffalo) and each treatment was replicated five times. The speed of estrus and estrus length were the measured variables. The results showed that the doses of GnRH significantly (P<0.05) increase of estrus speed and estrus length. Progesterone levels have increased significantly starting from before treatment until after treatment with GnRH. It is concluded that the best GnRH doses for estrus synchronization in the swamp buffalo producing of dadih was 2. 25 ml/buffalo with estrus speed 18.2 h and estrus length 18 h.

Abstract

Tujuan penelitian ini yaitu untuk meningkatkan keterampilan menulis karangan melalui metode field trip kelas IX A di SMPN 4 Tapung Hilir. Jenis penelitian yang ditetapkan dalam penelitianini adalah penelitian tindakan kelas (PTK). Ada empat tahapan dalam penelitian tindakan, yaitu (1) perencanaan, (2) pelaksanaan, (3) pengamatan, dan (4) refleksi, sedangkan penelitian ini terdiri dari dua siklus dengan masing-masing siklus terdiri dari dua kali pertemuan. Subjek penelitian dalam penelitian tindakan  kelas ini adalah siswa kelas IX A SMP N 4 Tapung Hilir dengan jumlah siswa yang diteliti sebanyak 31 siswa yang terdiri dari 13 siswa dan 18 siswi. Kesimpulan dalam penelitian ini adalah metode field trip dapat meningkatkan keterampilan menulis karangan atas siswa kelas IX A di SMPN 4 Tapung Hilir. Terjadi peningkatan keterampilan menulis karangan atas siswa dari siklus I dan siklus II. Hasil evaluasi sikus I dari 31 siswa yang berhasil mencapai KKM yaitu sebanyak 12 siswa (38,71%), dan hasil evaluasi nilai siklus II dari 31 siswa yang berhasil mencapai KKM yaitu sebanyak 26 siswa (83,87%).

Abstract

ABSTRAK. Studi ini bertujuan untuk mengidentifikasi keragaman gen FSH bagian ekson 2 menggunakan enzim restriksi TasI pada sapi Pesisir. Sampel darah yang digunakan yaitu 78 sampel darah dengan dua macam yaitu 13 jantan serta 65 betina yang berumur 1-5 tahun dari sapi pesisir yang didapat dari BPTUHPT Padang Mengatas. Protocol genomic DNA Purification system Kit dari Promega digunakan untuk mengisolasi DNA sampel darah. Amplifikasi gen FSH dilakukan dengan menggunakan teknik PCR atau Polymerase Chain Reaction. Enzim TasI digunakan untuk meretriksi produk amplifikasi. Hasil penelitian gen FSH bagian exon 2 pada sapi pesisir yaitu diperoleh terdapat dua macam genotip. Dua macam genotip tersebut yaitu heterozigot (+/-) serta homozigot tidak terpotong (-/-), untuk jantan berturut turut yaitu 0,38 dan 0,62 beserta frekuensi alel (+) dan alel (-) yaitu 0,19 dan 0,81, sedangkan untuk betina yaitu berturut-turut 0,43 dan 0,57 beserta frekuensi alel (+) dan alel (-) yaitu 0,21 dan 0,79. Sebaran genotip gen FSH|TasI exon 2 pada populasi Sapi Pesisir sifatnya polimorfik (beragam) dengan frekuensi alel yaitu kurang dari 0,99.Identification of Exon 2 FSH Gene Diversity Using TasI Restriction Enzymes in Coastal CattleABSTRACT. This study aims to identify the diversity of the exon 2 FSH gene using the restriction enzyme TasI in Coastal cattle. The blood samples used were 78 blood samples of two types, namely 13 males and 65 females aged 1-5 years, from coastal cattle obtained from BPTUHPT Padang Atas. The protocol genomic DNA Purification System Kit from Promega was used to isolate DNA from blood samples. The amplification of the FSH gene was carried out using the PCR or Polymerase Chain Reaction technique. The TasI enzyme was used to restrict the amplification product. The study results on the exon two portion of the FSH gene in coastal cattle showed two types of genotypes. The two types of genotypes are heterozygous (+/-) and untruncated homozygous (-/-); for males, 0.38 and 0.62, respectively, and the frequencies of allele (+) and allele (-) are 0.19 and 0. .81, while for females, it was 0.43 and 0.57, respectively, and the frequencies (+) and allele (-) were 0.21 and 0.79. The FSH|TasI exon two gene genotype distribution in the Coastal Cattle population is polymorphic (various) with an allele frequency of less than 0.99.Keywords: FSH Gene, Pesisir Cattle, TasI Enzyme

Abstract

The purpose of this study was to identify genetic diversity in the early exon 10 of the FSHR gene or follicle stimulating hormone receptor in Pesisir cattle. The blood samples used were 70 blood samples from female Pesisir cattle aged 2-5 years which were obtained from BPTU-HPT Padang Mengatas, West Sumatera. The isolated DNA was then amplified using a pair of primers L and R with PCR technique which produced a fragment with a length of 847 bp. The services of 1st Base Singapore are used for sequencing the amplified product. The results of the research on the identification of the early exon 10 FSHR gene in Pesisir cattle were found that there were 5 SNPs at positions --53T>C, +17A>G, +650C>T, +706A>C and +707 ins>A in some introns 9 to exon 10 first part. The results showed that the genotypic frequency of the Pesisir cattle population was in Hardy-Weinberg imbalance and polymorphisms were found in the early exon 10 FSHR gene diversity.

Abstract

ABSTRAK. Penelitian ini bertujuan untuk menentukan kualitas oosit sapi Pesisir yang akan diproduksi secara in vitro dan untuk menentukan dosis penambahan Insulin pada media maturasi in vitro efektif untuk meningkatkan kematangan oosit sapi Pesisir. Sampel yang digunakan yaitu ovarium sapi Pesisir yang diambil di RPH kemudian dilakukan koleksi oosit dengan metode slicing di Laboratorium Bioteknologi. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) yang terdiri dari 4 perlakuan dan 4 ulangan. Perlakuan yang digunakan adalah penambahan insulin pada media maturasi in vitro yang terdiri dari tanpa insulin (P1), insulin 10 µg/ml (P2), insulin 15 µg/ml (P3) dan insulin 20 µg/ml (P4). Oosit sapi Pesisir dimaturasi dalam media maturasi selama 24 jam di dalam inkubator CO2 dengan suhu 38,5 ᵒC. Sampel yang sudah dimaturasi diwarnai dengan 2% aceto-orcein selama 5 menit kemudian zat pewarna dihilangkan dengan campuran (asam asetat, gliserin dan aquadest) selanjutnya pengamatan inti diamati dengan menggunakan mikroskop. Hasil penelitian menunjukkan bahwa oosit yang paling banyak ditemukan adalah oosit grade B (75,59%), A(13,53%), C (8,48%), dan yang paling sedikit adalah grade D (2,40%). Penambahan Insulin dengan dosis 10 µg/ml dan 15 µg/ml pada media maturasi in vitro dapat meningkatkan pematangan oosit sapi pesisir yang mencapai metaphase II (MII) daripada tanpa insulin, sedangkan penambahan dosis insulin 20 µg/ml menurunkan pematangan oosit sapi pesisir yang mencapai MII. Berdasarkan hasil penelitian tersebut disimpulkan bahwa oosit yang dimaturasi dengan penambahan insulin 10 µg/ml dan 15 µg/ml sangat berpengaruh (P<0,01) terhadap pematangan oosit mencapai MII dan hasil terbaik adalah penambahan insulin 15 µg/ml sebesar 84,58%. Kata kunci: Insulin, Oosit Sapi Pesisir, Pematangan Inti.Effect of Insulin Addition on In Vitro Maturation Media on Pesisir Cattle Oocyte MaturationABSTRACT. This study aims to determine the quality of Pesisir cattle oocytes to be produced in vitro and to determine that adding different insulin to in vitro maturation media effectively increases the maturity of pesisir cattle oocytes. The sample was bovine ovaries of Pesisir cattle taken at the RPH and then bovine oocytes collected at the Biotechnology Laboratory. This study used a randomized block design (RBD) which consisted of 4 treatments and 4 replications. The treatment used was adding of insulin to in vitro maturation media consisting of no insulin (P1), insulin 10 µg/ml (P2), insulin 15 µg/ml (P3) and insulin 20 µg/ml (P4). Pesisir bovine oocytes were matured in maturation medium for 24 hours in a CO2 incubator with a temperature of 38.5 ᵒC. Samples that had been matured were stained with 2% aceto-orcein for 5 minutes and then the dye was removed with a mixture (acetic acid, glycerin and distilled water) then the observations of the nucleus were observed using a microscope. The results showed that the most common oocytes were grade B oocytes (75.59%), A (13.53%), C (8.48%), and the fewest were grade D (2.40%). The addition of insulin at a dose of 10 µg/ml and 15 µg/ml in in vitro maturation media could increase the oocyte maturation of coastal bovines that reached metaphase II (MII) than without insulin, while the addition of 20 µg/ml insulin decreased the oocyte maturation of coastal bovines that reached MII. Based on the results of this study it was concluded that oocytes which were matured with the addition of insulin 10 µg/ml and 15 µg/ml had a very significant effect (P<0.01) on oocyte maturation reaching MII and the best treatmentin this study was an insulin dose of 15 µg/ml of 84.58%.

Abstract

ABSTRAK. Tujuan penelitian ini untuk menilai kualitas semen beku sapi jenis Simmental, Limousin dan Friesian Holstein yang meliputi persentase motilitas, viabilitas dan abnormalitas post thawing. Semen yang pakai pada penelitian ini berasal dari Balai Inseminasi Buatan Lembang sebanyak 90 mini straw. Metode yang dipakai dalam penelitian adalah metode eksperimen dengan menggunakan Rancangan Acak Kelompok tiga perlakuan suhu dan waktu thawing dan 15 kali ulangan sebagai kelompok. Hasil penelitian menunjukkan bahwa pengaruh suhu dan waktu thawing berpengaruh nyata terhadap motilitas dari ketiga semen beku yaitu 49,33±5,62% pada Simmental. Selanjutnya persentase viabilitas juga berpengaruh nyata pada P3 yaitu 71,06±5,89%. Sedangkan pada persentase abnormalitas tidak berpengaruh disetiap perlakuan. Kata kunci: Semen beku, sapi, thawing, suhuSimmental Bull, Limousine, and Friesian Holstein Frozen Cement Quality with Different Thawing MethodsABSTRACT. This study aims to determine the quality of frozen semen of Simmental, Limousin and Friesian Holstein cattle which includes the percentage of motility, viability and post thawing abnormalities. The frozen semen used in this study came from the Lembang Artificial Insemination Center with 90 mini straws. The method used in this study was experimental using a randomized block design with three treatments of temperature and thawing time and 15 replications as a group. The results showed that the effect of temperature and thawing time had a significant effect on the motility of the three frozen cements, namely 49.33±5.62%.Furthermore, the percentage of viability also had a significant effect on P3, namely 71.06±5.89%. While the percentage of abnormalities did not affect each treatment.

Abstract

This study aimed to determine the inputs given, the process of implementing the UPPO program, and the results of the UPPO program in Solok Regency. Respondents in the study were Farmer Groups who received assistance from the Organic Fertilizer Processing Unit (UPPO) program in 2017, namely the Kelompok Tani Hulu Gawan, Kandih Maju Bersama, dan Kelompok Tani Sawah Rambahan. The method that we used in this study is a survey method. The observed variables are inputs  and outputs of implementation of the UPPO program. The results of this study indicate that the inputs for the UPPO program assistance carried out by the research farmer groups are match accordance with the technical instructions for processing organic fertilizers. The livestock breeds used were Simmenal Cattle, Ongole Crossbreed, Limousin and Coastal Cattle. The feed given to livestock is field grass and beef, livestock rearing is carried out intensively, disease prevention that has been carried out is cage sanitation and treatment of intestinal worms by giving Albendazole bolus drug and in kelompok tani Hulu Gawan, injured cattle are given Gusanex Antiseptic spray. For the cattle marketing is generally through intermediary trading. The production of organic fertilizer for the Kelompok Tani  Hulu Gawan  is 2.1 tons/month with 9 livestock, Kandih Maju Bersama 1.4 tons/month with 5 livestock and Sawah Rambahan 1.7 tons/month with 7 livestock. Birth rate of cattle in the Kelompok Tani Hulu Gawan in  the number of children born was 2, Kandih Maju Bersama and Sawah Rambahan was 1 cattle. None of the cattle of the Kandih Maju Bersama and Sawah Rambahan farmer groups died, while the Kelompok Tani Hulu Gawan had 1 death. Abstract

This study aims to examine the composition of the pH, cooking loss and water holding capacity of Bali cattle, at different ages and muscle locations. The locations of the muscles observed were the Longissimus dorsi (LD), Pectoralis superficialis (PS), Semitendinosus (ST) and Triceps Brachii (TB) muscles. Cattle aged 1-1.5 years and 3.5-4 years with moderate body condition, 250 g of each sample was taken for every 6 Bali cattle. This study used a 4x2x3 factorial design with a total of 24 experimental units, in a randomized block design (RBD), factor A was muscle type (A1 (LD), A2 (PS), A3 (ST), and A4 (TB)). Factor B is age (B1 aged 1-1.5 years and B2 aged 3.5-4 years). The treatment showed a significant effect, so further tests were carried out using the Duncan Multiple Range Test (DMRT) and how to analyze the data using SPSS. The variables measured were pH, cooking losses and water holding capacity. The research data was processed using analysis of variation (ANOVA). The results of this study concluded that there was no interaction between the type of muscle and the age of the cattle in the meat quality test on pH, cooking loss and water holding capacity of Bali beef. Muscle type has no effect on pH, cooking loss and water holding capacity of Bali beef. Age greatly influences the pH of Bali beef and affects the water holding capacity of Bali beef, but has no effect on cooking loss of Bali beef. Abstract

Madu dan kuning telur merupakan salah satu feed suplement yang mengandung antioksidan sangat tinggi. Penelitian ini bertujuan untuk mengetahui Kualitas Spermatozoa ayam jantan Sentul yang diberi madu dan kuning telur sebelum Inseminasi Buatan terhadap kualitas spermatozoa, fertilitas, dan daya tetas telur.Penelitian ini dilakukan menggunakan metode Rancangan Acak Lengkap Pola Faktorial dengan 3 kali ulangan.Faktor A pemberian madu pada ayam jantan Sentul dengan 4 level yaitu 0 ml (kontrol),3 ml madu,5 ml madu, dan 7 ml madu.Faktor B pemberian kuning telur pada ayam jantan Sentul dengan 4 level yaitu 0 ml kuning telur (kontrol), 3 ml kuning telur, 5 ml kuning telur, dan 7 ml kuning telur.Hasil penelitian menunjukkan analisis statistik terdapat interaksi sangat nyata (P< 0,01) antara kombinasi perlakuan yaitu pemberian madu dan kuning telur terhadap volume semen (0,85ml), konsentrasi spermatozoa (3,92 x 109), motilitas spermatozoa (90,00%), abnormalitas spermatozoa (5,00%), fertilitas telur (91,67%), mortalitas embrio (7%), dan daya tetas telur (93%). Dari hasil penelitian dapat disimpulkan bahwa pemberian 5 ml madu dan 5 ml kuning telur yang terbaik.
