Abstract

The high demand for rice increases the chances of intensive rice cultivation (2 to 3 times a year). If these cultivation activities are not managed properly, it can reduce the quality of rice cultivation land due to ecosystem imbalances. Responding to this challenge, the Directorate of Food Crops Protection is present through the Healthy Plant Cultivation program as an alternative to rice cultivation techniques that are environmentally friendly and sustainable. A quantitative approach is used in this research. This study aims to analyze the sustainability of healthy rice farming in Wlingi District, Blitar Regency. The method used is Multidimensional Scaling (MDS) analysis. The value of the sustainability of healthy rice farming in Wlingi District as measured from the economic, ecological, social, technological, and institutional dimensions indicates a "quite sustainable" status, so to improve sustainability, it is necessary to increase the role of the attribute as a lever in the Leverage analysis. Abstract

Controlling is always needed in every public policies and programs, through both of monitoring and evaluation on the process, the conformability between its process and the purposes, outputs, and impacts. The aim of this research was to monitor and to evaluate the overcoming feminism on poverty program during 2018 in some cities in East Java Province, used descriptive-qualitative method. The data was collected by visiting and surveying the sites. Research locations were determined by proportional sampling method considering kinds of business process run by targeted households in each village. Two hundred and three households from twenty villages and seven cities were monitored and evaluated, which were represented about ten percent of the population. The research showed that the success rate of the program was 96.50%. as measured by the accuracy of the target (100%), the accuracy of receiving the amount of aid and its use (100%) as well as aspects of the benefit of the aid (89.49%). From the aid provided, 87.19% led to self-motivation in doing business, increasing in income, capital, and assets were 84.24%, 84.24%, 87.68% as well as environmental supporting was 88.67%. There was no similar activity from the regional government in term of equal distribution of the aid and program sustainability. The family supporting from the households ensure the success of this program.

Abstract

This study aims to describe and map the differences in ethnobotany of residential gardens with cases of stunting and without stunting. This research is both quantitative and qualitative. Data collection was carried out using semi-structured interviews and field observations at 20 points of the squared plot of vegetation observations which also became a source of information. The data obtained was then analyzed for vegetation using the Shannon-Wiener index method and the use value of species. The results showed that there were differences in preferences for the use of plants in local community settlements with cases of stunting and non-stunting. Utilization of plants with the highest UVs were in settlements with cases of stunting, namely Zingiber officinale, Pandanus amaryllifolius and Amorphophallus muelleri, while in the yards without stunting cases were Actinocarpus commansi Blanco, Magnolia acuminata (L.) , Moringa olefira Lam, and Artocarpus altilis (Parkison ex F.A.Zorn) Fosberg. Communities in settlements without stunting cases consume intensely sources of food from their garden plots, while people in settlements with stunting cases do not. In particular, Moringa plants are only found in residential yard gardens without stunting and are not found in settlements with stunting cases.

Abstract

Enzim L-asparaginase (E.C 3.5.1.1) adalah protein tetramer yang termasuk dalam kelompok keluarga amidohydrolases homolog. ). L-asparaginase secara selektif menghidrolisis L-aspargin (L-Asn) menjadi amonia (NH3) dan asam aspartat. Penelitian ini bertujuan untuk mendapatkan spesies bakteri penghasil enzim L-asparaginase yang diisolasi dari mangrove Avicennia marina yang berasal dari Pantai Bajul Mati, Malang, Jawa Timur, Indonesia. Penelitian ini dilakukan beberapa tahapan yaitu pengambilan dan preparasi sampel, skrining L-asparaginase, dan identifikasi menggunakan uji mycrobact system. Hasil penelitian menunjukkan bahwa isolat dengan aktivitas enzim L-asparaginase terbesar adalah isolat DAM6 yang berasal dari daun Avicennia marina. Isolat DAM6 adalah bakteri merupakan bakteri gram negatif, berbentuk basil, bentuk koloni oval, tepi koloni tidak rata dan elevansi cembung. Berdasarkan analisis microbact,  isolat DAM6 adalah bakteri spesies Pseudomonas aeruginosa.                                               AbstractThe L-asparaginase enzyme (E.C 3.5.1.1) is a tetramer protein belonging to the family group of amidohydrolases. L-asparaginase selectively hydrolyzes L-aspargin (L-Asn) to ammonia (NH3) and aspartic acid. This study aims to obtain species of L-asparaginase enzyme producing bacteria isolated from Avicennia marina mangroves from Bajul Mati Beach, Malang, East Java, Indonesia. This research carried out several stages such as took and prepared samples, screened L-asparaginase, and identified using the mycrobact system test. The results showed that the isolates with the largest L-asparaginase enzyme activity were DAM6 and it was originated from the leaves of Avicennia marina. DAM6 isolate is a gram-negative with an oval shape, uneven edges, and convex elevation. Based on microbact analysis, DAM6 isolates are Pseudomonas aeruginosa.

Abstract

Siomay, a popular Indonesian snack traditionally made with high-fat chicken or beef fillings, can be made healthier by using mackerel fish instead. This modification, combined with the addition of potato starch, aims to improve the nutritional profile of siomay while maintaining its appeal to consumers, as mackerel is both tasty and nutritious.  This study aims to improve the nutritional profile and consumer acceptance of siomay filling through the integration of mackerel meat and potato starch. Data were analyzed using a Completely Randomized Factorial Design with two factors (mackerel meat and potato starch) mixed in different proportions in the siomay filling dough. Parameters tested included nutritional composition (protein, fat, carbohydrate, fiber, and mineral), physicochemical properties (texture, color, and aroma), and consumer acceptance test using a hedonic scale. The results showed that the most preferred treatment was K2D2 (14g mackerel meat and 20g potato starch), which was 5.6±1.5. The nutritional content of the siomay filling preferred by the panelists increased (water 29.07 g, protein 12.89 g, fat 3.65 g, ash 3.41 g, sugar 1.35 mg, vitamin B1 0.26 mg). However, the carbohydrate content decreased to 50.98 g, compared to the content of siomay filling without the addition of yellow potato starch (56, 74 g). This study shows that mackerel flour and potato starch can potentially be used to improve the nutritional profile and consumer acceptance of siomay filling.

Abstract

Ikan Tuna Madhidihang (Thunnus albacares) merupakan salah satu jenis ikan ekonomis penting di dunia dan komoditas ekspor perikanan adal Indonesia. Pemanfaatan sumber daya perikanan umumnya milik bersama (common property) artinya upaya pemanfaatan dapat dilakukan secara bersama dalam waktu yang bersamaan oleh pelaku usaha perikanan (open acces). Semakin meningkatnya upaya pemanfaatan yang dilakukan dan banyaknya hasil tangkapan ikan tuna dibawah ukuran pertama kali matang gonad mengakibatkan terganggunya proses rekruitmen ikan-ikan muda di alam. Untuk mendukung pengambilan keputusan yang tepat dalam upaya pengelolaan perikanan yang berkelanjutan, maka ketersediaan data dan informasi yang terukur mutlak diperlukan untuk mengetahui status sumber daya perikanan tuna dewasa ini. Tujuan dari penelitian ini adalah untuk mengetahui (1) distribusi panjang ikan dan (2) hubungan panjang berat ikan tuna madhidihang di wilayah perairan selatan Jawa Timur (Sendang Biru).  Pengambilan data dilakukan di Tempat Pelelangan Ikan (TPI) Pelabuhan Pondokdadap Sendangbiru Malang, pada Bulan Agustus – Oktober 2022.  Sampel ikan tuna madhidihang yang didaratkan kemudian diukur panjang dan beratnya, dianalisis dengan menggunakan regresi linier untuk melihat hubungan antara panjang dan berat ikan.  Ada 347 ekor ikan madhidihang yang berhasil diukur panjang cagak dan beratnya pada bulan Agustus hingga Oktober 2022. Berdasarkan data yang telah diolah, rata-rata panjang cagak sekitar 109 cm, panjang cagak dominan antara 136 – 145 cm dengan panjang maksimal 195 cm dan minimal 86 cm.  Berdasarkan data Fish Base ukuran ikan pertama kali matang gonad atau Length at first maturity (Lm) tuna sirip kuning berada pada ukuran 103,3 cm. Hasil penelitian menunjukkan bahwa 55%  hasil tangkapan tuna sirip kuning nelayan Sendangbiru sudah layak tangkap, dan 45% belum layak tangkap.  Hasil analisis hubungan panjang dan berat ikan tuna madhidihang di lokasi penelitian memperlihatkan pertumbuhan bersifat alometrik negatif dimana pertumbuhan panjang lebih dominan daripada pertumbuhan berat ikan.   Yellowfin Tuna (Thunnus albacares) is one of the economically important fish species in the world and an Indonesian fishery export commodity. Utilization of fishery resources is generally shared property (common property) meaning that efforts to exploit can be carried out jointly at the same time by fishery business actors (open access). Increasing utilization efforts are being made and the large number of tuna catches below the size at first maturity has disrupted the recruitment process for young fish in nature. To support the right decision making in efforts to manage sustainable fisheries, the availability of measurable data and information are absolutely necessary to determine the current status of tuna fishery resources. The aims of this study were to determine (1) the distribution of fish length and (2) the relationship between the length and weight of yellowfin tuna in the southern waters of East Java (Sendang Biru). Data collection was carried out at the Fish Auction Place (TPI) Pondokdadap Port Sendangbiru Malang, from August to October 2022. The yellowfin tuna samples that were landed were then measured for length and weight, then analyzed using linear regression, so that a relationship between the length and weight of the fish could be seen. The number of yellowfin tuna or yellowfin tuna whose fork length and weight were successfully measured from August to October 2022 was 347 individual fish. Based on the data that had been processed, the dominant fork length of yellowfin tuna was between 136 – 145 cm with a maximum length of 195 cm and a minimum of 86 cm, and an average fork length of around 109 cm. Based on Fish Base data, the length at first maturity (Lm) of yellowfin tuna was 103.3 cm. This showed that the catch of yellow fin tuna by Sendangbiru fishermen had a percentage of 55% that was suitable for catching, while those that were not yet worth catching had a percentage of 45%. Fish length and weight data can be used to see the morphometric condition of a fish species. From the results of the analysis of the relationship between length and weight of yellowfin tuna at the study site, it was known that growth was negative allometric where length growth was more dominant than fish weight growth.

Abstract

Population growth accompanied by activities in all fields affects the quantity and quality of waste produced. Community patterns and culture in managing waste currently still follow the old paradigm of collect-transport-dispose. This results in a buildup of waste which can shorten the life of the final landfill site (TPA) and impact the environment. Therefore, it is necessary to know the public's perception and participation in waste management to reduce the volume of waste. This research aims to determine and analyze the relationship between public perception and participation in household waste management. This research was conducted in the work area of the Waste Management Technical Implementation Unit, Tumpang District, Malang Regency. This research uses a quantitative approach supported by qualitative descriptive data collected through surveys and questionnaires. Based on research, it shows that the public's perception of household waste management is cumulatively positive and no negative perceptions were found. This means that there is no rejection from the community towards waste management efforts, but this positive perception is not supported by community participation. Abstract

Staphylococcus aureus becomes a normal flora in humans, especially on the skin and in the nose. However, if it becomes excessive or if there are pathogenic variants, it can cause various health problems. The purpose of the study is to develop a molecular-based detection method for Staphylococcus aureus using the norA primer gene. The norA gene in Staphylococcus aureus is known to play a role in pathogenesis with its antibiotic resistance ability. This type of research is analytical observational with a cross-sectional method. The methods in the study include the isolation and identification of Staphylococcus aureus from healthcare facility equipment. Isolation and identification include bacterial isolation using Blood Agar Plate (BAP) media; isolate purification, Gram staining; biochemical tests using Mannitol Salt Agar (MSA) media, glucose tests, Voges Proskauer (VP) tests, catalase tests, and coagulase tests. Furthermore, S. aureus isolates were tested using a molecular-based method, namely Polymerase Chain Reaction (PCR). This method includes DNA isolation stages, qualitative testing with agarose gel electrophoresis, semi-quantitative testing with image J software, amplification with Real-Time Polymerase Chain Reaction (RT-PCR) using norA gene primers. The Mann-Whitney test results gave a value of p = 0.334 (p> 0.05) indicating the suitability between the culture method and the PCR method with the developed protocol in detecting Staphylococcus aureus. The developed method includes the use of base sequences in the norA gene primer, optimization of annealing and extension temperatures, and the concentration of DNA templates used.

Abstract

Laju urbanisasi, pertumbuhan penduduk, peningkatan pendapatan, dan perilaku tidak ramah lingkungan mengakibatkan timbulan sampah semakin meningkat. Maka dari itu, diperlukan optimalisasi dalam upaya penanganan sampah pada kawasan urban untuk meminimalisasi dampak yang ditimbulkan. Diantaranya dengan melakukan pengolahan sampah secara 3R (reduce, reuse, recycle) di unit TPS-3R dengan melibatkan peran aktif masyarakat melalui kegiatan pemilahan dan pengolahan sampah dari sumber. Hal ini didasarkan pada Jakstrada Kabupaten Sidoarjo yaitu target pengurangan hingga 30% dan penanganan 70% sampah pada tahun 2025 sebelum masuk ke TPA Jabon. Tujuan dalam penelitian ini untuk mengevaluasi potensi reduksi sampah pada TPS-3R dan rumah tangga di Desa Janti serta potensi daur ulangnya. Penelitian ini menggunakan analisis mass balance dengan pendekatan observasi dan penyebaran kuesioner ke 190 responden rumah tangga secara propotional random sampling. Hasil dari penelitian ini memperlihatkan bahwa skenario 1 menunjukan dengan adanya unit TPS-3R dapat mereduksi sampah Desa janti ke TPA Jabon sebesar 2035.1 kg/hari (743.4 ton/tahun) atau dengan recovery factor 55.1%. Sedangkan hasil pada skenario 2, unit TPS-3R dengan melibatkan partisipasi masyarakat memilah dari sumbernya dapat mereduksi sampah sebesar 2624.7 kg/hari (958.7 ton/tahun) dengan recovery factor 78.0%. Hal ini menunjukan bahwa skenario 2 dapat mengoptimalisasi reduksi sampah dari Desa Janti ke TPA Jabon, Sidoarjo secara signifikan. Dengan menggunakan skenario 2 dapat menjadi landasan dalam menentukan kebijakan di tingkat pengelolaan sampah kota dan pengembangan penanganan sampah di TPS-3R lainnya di Kabupaten Sidoarjo.

Abstract

Degenerative diseases such as stroke and cancer arise due to oxidative stress by a large number of reactive oxygen species (ROS) in the body. Free radical scavenging (antioxidant) compounds are needed to neutralize the ROS. The natural antioxidant potential of mangrove species Sonneratia caseolaris has been widely explored. However, its extraction is usually carried out with polar solvents such as methanol. Besides being more toxic than ethanol, methanol can cause blindness and death when inhaled in large quantities. Additionally, Indonesia, as the country with the largest Muslim population, requires the use of halal products for food and cosmetics. This study aims to analyze the scavenging activities of S. caseolaris with methanol and distilled water as the polar solvents for extraction. Antioxidant activity was tested with the free radical assay method of DPPH (2,2-diphenyl-1-picrylhydrazyl radicals). The test results were calculated as the IC50 value, a concentration capable of reducing free radicals by 50%. The antioxidant activity test showed that the IC50 values of methanol and distilled water were 18.9 ppm and 36 ppm, respectively. Therefore, both are considered very strong antioxidants. It indicates that distilled water is an acceptable substitute for methanol in the extraction process. Keywords: antioxidant, extraction, mangrove, solvent, Sonneratia.
