Abstract

Traditional farmers in Pasuruan City face challenges in increasing pond productivity due to declining land quality and reliance on natural feed. Milkfish growth is often hampered by lack of feed and increased disease due to environmental changes. Although manufactured feed and probiotics are available, they are too expensive for traditional farmers. Pasuruan City has great potential for the development of milkfish farming, and the Banyu Urip Fish Farmers Group (POKDAKAN) is an experienced community in this field. The use of probiotics made from spices, such as ginger, turmeric, and curcuma, is proposed as an eco-friendly and economical solution as these ingredients are abundant and effective in improving pond water quality and fish disease resistance. The results of probiotic application showed significant improvement, with 85% of farmers reporting up to 30% increase in production, and 90% recording a decrease in fish disease cases. Pond water quality also improved, particularly in ammonia and dissolved oxygen levels. The use of this natural probiotic not only positively impacts the pond environment, but also supports the sustainability of milkfish farming productivity in Pasuruan City.

Abstract

Edwardsielliosis is a disease that commonly infects freshwater fish including catfish (Clarias sp). Edwardsielliosis is caused by Edwardsiella tarda infection and causes mass mortality in catfish farming. Overcoming diseases in freshwater fish can use drugs or antibiotics that are classified as safe and their use is in accordance with government regulations. This study aims to determine the appropriate dosage of Oxytetracycline in catfish infected by E. tarda bacteria through histological analysis. The study design used Completely Randomized Design with 3 treatments, 1 control and 3 replications in each treatment. Catfish used were ± 25 cm in size and the dose of oxytetracycline used were 15, 30, and 45 ppm. The results showed tissue damage edema, vacuolization, necrosis, and haemorhage in fish infected with Edwardsiella tarda bacteria with the lowest damage at 45 ppm of oxytetracycline treatment. The results indicate that oxytetracycline has an effect on muscle histology and recovery of fish health.

Abstract

Sargassum cristaefolium dried powder can be used as food and fed additives as well as  a beverage powder, contained some minerals and terpenoid active ingredients. This study aimed to determine the anti-bacterial activity of Sargassum cristaefolium dried powder on the inhibition of Escherischia coli and Salmonella thyposa bacteria in vitro. The research showed that the dried powder of Sargassum cristaefolium had better inhibition on Salmonella thyposa (2.3 mm) compared to Escherischia coli (1.2 mm) although it was classified as weak bactericidal. Sargassum cristaefolium dried powder showed positive testing for alkaloids, flavonoids, terpenoids and tannins and does not contain saponins, had low toxicity (LC50 322 ppm). The LCMS test showed a peak at 2.16 retention time with a molecular weight of 339.39 m/z. The structure of the Sargassum cristaefolium antibacterial active ingredient should be clarified using C-NMR and H-NMR.

Abstract

Tuberculosis (TB) remains a significant public health issue in Indonesia, including in Malang City. Despite various strategies, the disease poses significant challenges due to social, economic, and environmental factors. This study aims to evaluate the role of Community Health Center (Puskesmas) policies in TB management and prevention efforts in Malang City while identifying factors that influence program success. A descriptive quantitative method was employed, utilizing Structural Equation Modeling (SEM), SWOT analysis, and the Analytic Hierarchy Process (AHP). Primary data were collected through questionnaires and interviews with TB patients, health workers, and the community, while secondary data were obtained from local health office reports. The findings revealed that implementing Puskesmas policies has generally been effective, particularly regarding mandatory reporting, active case finding, and free TB treatment. However, several barriers persist, including inconsistent treatment adherence, stigma surrounding TB, inadequate living conditions, and limited healthcare access in certain areas. SEM analysis identified key factors affecting TB control success: patient knowledge, environmental conditions, treatment experience, and the role of health workers. SWOT analysis highlighted strengths such as strong community engagement and policy frameworks, alongside weaknesses like insufficient environmental interventions and healthcare resource limitations. Optimization strategies proposed include enhancing public education on TB prevention, improving the home environment to support recovery, strengthening health worker capacity, integrating digital technologies, and fostering multi-sector collaboration. The study emphasizes the need to address biomedical and socio-environmental determinants for effective TB control. These findings provide strategic recommendations for enhancing Puskesmas policy implementation toward Indonesia’s 2030 TB elimination goals. Abstract

Staphylococcus aureus becomes a normal flora in humans, especially on the skin and in the nose. However, if it becomes excessive or if there are pathogenic variants, it can cause various health problems. The purpose of the study is to develop a molecular-based detection method for Staphylococcus aureus using the norA primer gene. The norA gene in Staphylococcus aureus is known to play a role in pathogenesis with its antibiotic resistance ability. This type of research is analytical observational with a cross-sectional method. The methods in the study include the isolation and identification of Staphylococcus aureus from healthcare facility equipment. Isolation and identification include bacterial isolation using Blood Agar Plate (BAP) media; isolate purification, Gram staining; biochemical tests using Mannitol Salt Agar (MSA) media, glucose tests, Voges Proskauer (VP) tests, catalase tests, and coagulase tests. Furthermore, S. aureus isolates were tested using a molecular-based method, namely Polymerase Chain Reaction (PCR). This method includes DNA isolation stages, qualitative testing with agarose gel electrophoresis, semi-quantitative testing with image J software, amplification with Real-Time Polymerase Chain Reaction (RT-PCR) using norA gene primers. The Mann-Whitney test results gave a value of p = 0.334 (p> 0.05) indicating the suitability between the culture method and the PCR method with the developed protocol in detecting Staphylococcus aureus. The developed method includes the use of base sequences in the norA gene primer, optimization of annealing and extension temperatures, and the concentration of DNA templates used.

Abstract

Siomay, a popular Indonesian snack traditionally made with high-fat chicken or beef fillings, can be made healthier by using mackerel fish instead. This modification, combined with the addition of potato starch, aims to improve the nutritional profile of siomay while maintaining its appeal to consumers, as mackerel is both tasty and nutritious.  This study aims to improve the nutritional profile and consumer acceptance of siomay filling through the integration of mackerel meat and potato starch. Data were analyzed using a Completely Randomized Factorial Design with two factors (mackerel meat and potato starch) mixed in different proportions in the siomay filling dough. Parameters tested included nutritional composition (protein, fat, carbohydrate, fiber, and mineral), physicochemical properties (texture, color, and aroma), and consumer acceptance test using a hedonic scale. The results showed that the most preferred treatment was K2D2 (14g mackerel meat and 20g potato starch), which was 5.6±1.5. The nutritional content of the siomay filling preferred by the panelists increased (water 29.07 g, protein 12.89 g, fat 3.65 g, ash 3.41 g, sugar 1.35 mg, vitamin B1 0.26 mg). However, the carbohydrate content decreased to 50.98 g, compared to the content of siomay filling without the addition of yellow potato starch (56, 74 g). This study shows that mackerel flour and potato starch can potentially be used to improve the nutritional profile and consumer acceptance of siomay filling.

Abstract

Malang City faces several environmental challenges, including inadequate sanitation and water management, which have not been fully addressed across all aspects of life. This is not in line with the Sustainable Development Goals, which aim to ensure the availability and sustainable management of clean water and sanitation for all people. Malang City is also experiencing increasingly massive land use changes, from green open land to housing, which can impact clean water quality. This study aims to assess the quality of clean water from the non-Local Water Supply Utility in Merjosari Village, Lowokwaru Subdistrict, Malang City, which is undergoing land-use transformation. A quantitative observational approach was employed using the National Sanitation Foundation's Water Quality Index (NSF-WQI) method, which evaluates water quality based on physical, chemical, and biological parameters. The results showed that the water quality index in Merjosari Village, Lowokwaru District, Malang City, analyzed using the NSP-WQI method, has a value of 77.53 and falls within the good criteria. However, several parameter values exceed the clean water standard. The average value of the DO parameter, at 5.85 mg/,L is less than the quality standard of 6 mg/L. The average value of the BOD parameter, at 8.12 mg/,L exceeds the quality standard of 2 mg/L. The pH parameter at several sample points exceeds the quality standard of above 8.5, and the Nitrate Parameter at several sample points exceeds the quality standard of 20 mg/L. In conclusion, the land cover change in the Merjosari area, Lowokwaru District, Malang City, does not significantly affect the quality of clean water. However, the values of several water parameters exceed the clean water standard, which has the potential to impact public health negatively.

Abstract

Flood disasters cause significant damage and losses in various cities around the world. One of the contributing factors is poor drainage system management. This literature review study aims to explore the effectiveness of drainage systems in addressing flood problems in major cities across the globe. The study utilizes a systematic literature review design with the PRISMA method. Data is sourced from the Scopus database. The document analysis process involves several stages: identification, screening, eligibility, and inclusion, ultimately resulting in 25 articles that align with the objectives of this study. The results indicate that there are three aspects to optimizing drainage systems as an effective flood management strategy in major cities: 1) Smart Technology-Based Drainage System Management, 2) Policy and Regulation-Based Drainage Management, 3) Community and Social Adaptation-Based Drainage Management, and 4) Geomorphology and Natural Process-Based Drainage Management. This literature review is expected to provide practical recommendations for improving drainage systems to be more adaptive and sustainable in addressing increasingly complex flood challenges in the future, particularly in Indonesia.

Abstract

Abstrak: Salah satu penyebab menurunnya produktivitas tambak di Kelurahan Blandongan Kota Pasuruan yaitu penurunan kualitas lingkungan tambak karena tingginya ketergantungan terhadap pakan dan probiotik pabrikan yang mahal serta kurangnya inovasi budidaya ramah lingkungan. Kegiatan ini bertujuan untuk meningkatkan pengetahuan petambak tentang penggunaan probiotik alami berbahan rempah sebagai alternatif yang lebih murah dan ramah lingkungan. Kegiatan disampaikan dengan metode dialog interaktif dan praktik, diikuti oleh 3 dosen dan 3 mahasiswa Universitas Brawijaya. Sebagai mitra yaitu Kelompok Pembudidaya Ikan Banyu Urip sebanyak 27 peserta dan Dinas Perikanan Kota Pasuruan sebanyak 10 peserta. Evaluasi kegiatan menggunakan kuesioner dengan 6 pertanyaan untuk mengukur tingkat pemahaman peserta sebelum dan sesudah kegiatan. Hasil evaluasi menunjukkan bahwa lebih dari 80% peserta belum mengetahui kandungan dan proses pembuatan probiotik alami. Setelah sosialisasi, sebanyak 95% responden menyatakan telah memahami fungsi probiotik, kandungan bahan aktif, serta tahapan pembuatannya. Dengan pendekatan edukatif berbasis dialog interaktif dapat meningkatkan kapasitas petambak dalam mengadopsi teknologi tepat guna dan berkontribusi pada keberlanjutan produktivitas tambak tradisional.Abstract: One of the causes of declining pond productivity in Blandongan Village, Pasuruan City is the decline in the quality of the pond environment due to the high dependence on expensive feed and probiotics and the lack of environmentally friendly cultivation innovations. This activity aims to increase farmers' knowledge about the use of natural probiotics made from spices as a cheaper and environmentally friendly alternative. The activity was delivered using interactive dialog and practical methods, attended by 3 lecturers and 3 students of Universitas Brawijaya. The partners were the Banyu Urip Fish Farmer Group with 27 participants and the Pasuruan City Fisheries Service with 10 participants. The evaluation of the activity used a questionnaire with 6 questions to measure the level of understanding of the participants before and after the activity. The evaluation results showed that more than 80% of the participants did not know the content and process of making natural probiotics. After the socialization, 95% of respondents stated that they understood the function of probiotics, the content of active ingredients, and the stages of making them. This proves that an educative approach based on interactive dialog can increase the capacity of farmers to adopt green technology and contribute to the sustainability of traditional pond ecosystems.

Abstract

Traditional ponds in Pasuruan City are dominated by milkfish (Chanos chanos). Milkfish take food from the top layer of the seabed in the form of phytoplankton and zooplankton. This study aims to determine the abundance, diversity and dominance of plankton and water quality in traditional milkfish ponds in Pasuruan City. Sampling locations were selected based on the distance of the pond from the sea. Station 1 (near the sea), station 2 (1–3 km from the sea), and station 3 (furthest from the sea). The results showed the availability of natural food for milkfish in the form of phytoplankton and zooplankton. Phytoplankton was found more than zooplankton. The most common phytoplankton division was Bacillariophyta and the least was Euglenozoa with diversity index ranging from 1.64–1.82 and dominance index ranging from 0.24–0.31. The most common zooplankton phylum is Arthropoda and the least is Molluscs with diversity index ranging from 0–1.83 and dominance index ranging from 0.21–1.00. The measurement results of pond water quality parameters such as pH (8.51–9.02), temperature (30.1–32.5°C), brightness (25.5–29.6 cm), DO (3.7–5.7 ppm) and salinity (32.5–33.7 ppt) are still in accordance with the optimal standards of milkfish farming.
