Abstract

Rhizobacteria isolated from plant roots have the ability to produce siderophore compounds. These compounds play a role in inhibiting the growth of pathogens by binding to iron (Fe3+) which is needed by pathogens in their development. This research was aimed to find out the siderophore-producing bacteria isolated from local Kamba rice plants and their potential as biocontrol agents. Among the 28 isolates that were successfully isolated from the Kamba local rice rhizosphere, 10 isolates had the potential to produce siderophores with different morphological characters. The test was carried out to see the bacterial isolates capabilities to produce siderophores from two types of siderophores namely catechol and salicylate types. For the catechol type, the highest concentration of siderophore was found in the KBA8 bacterial isolate with 10.990 mg L-1, while the lowest was in the KBA1 bacterial isolate with only 5.876 mg L-1. The salicylate type siderophore with the highest concentration produced 9.493 mg L-1 was from the RKGU15 isolate and the lowest was found in KBU14 isolate which produced only 2.994 mg L-1. The isolates included in the Gram-positive group were 4 isolates while the Gram- negative group were 6 isolates and 90% isolates were able to produce the enzyme catalase. The results of this study indicate that all bacterial isolates can produce siderophores so that they have the potential as biocontrol agents to support environmentally friendly and sustainable agriculture.

Abstract

Morphological characterization of Phytophthora isolates commonly was done on the basis of the colony and hyphal swelling characteristics, form, and shape of chlamydospore, sporangia as well as oogonia and antheridia. Molecular characterization was conducted using specific primer pairs Pal1 s and Pal2a for Phytophthora palmivora and universal primer ITS4 and ITS5 followed by sequencing of isolates. The virulence of Phytophthora palmivora isolates was tested on a cocoa pod of the Sulawesi2 clone. A total of 21 Phytophthora isolates were collected from Palopo, Gowa, and Pinrang Districts. Based on colony morphology and microscopic features i.e. chlamydospore, sporangium, and its papilla form, all isolates indicated Phytophthora palmivora. Molecular identification using a specific primer showed that all ten representative group isolates had the same band in size of 650 bp, whereas using universal ITS4 and ITS5 primers, all isolates generated one band in the size of 900bp. The sequencing result showed, that all isolates were placed in the group P. palmivora. The virulence of P. palmivora isolates on the Sulawesi2 clone revealed a varying degree of virulence, from less virulent to highly virulent. All isolates from Pinrang were classified as highly virulent because induced necrotic in more than 50% of the tested cocoa pod.

Abstract

Burkholderia glumae and Xanthomonas oryzae pv. oryzae are the primary causes of bacterial disease in rice, capable of reducing production and posing a threat to food availability. This research aims to assess the efficacy of a consortium of five bacteria contained in Mikrobat as bioactivators. The goal is to enhance rice yields, mitigate the severity, and suppress grain rot disease and bacterial leaf blight in rice. The research employed a split-plot design encompassing 12 treatment combinations replicated three times. The experimental setup involved two factors: the main plot consisted of rice varieties-Ciherang, Inpari 4, and Trisakti-while the subplots represented types of bioactivators-Mikrobat, EM4, Trichoderma asperellum, and a control. Results demonstrated Mikrobat’s positive impact compared to the control, with an average of 17.37 productive tillers and higher yields than other bioactivator treatments. Notably, Mikrobat exhibited 50.04% spikelets per panicle and a 1000-grain weight of 19.77 g. The severity of disease with Mikrobat treatment was notably lower than with EM4 and T. asperellum, featuring 4.20% for grain rot disease and 4.41% for bacterial leaf blight. This was significantly different from the control, which showed severity rates of 9.94% for grain rot disease and 7.75% for bacterial leaf blight. Among the three tested varieties, Ciherang demonstrated the best response, displaying higher resistance to both tested diseases. The varietal treatments did not significantly differ from each other. Although Mikrobat’s effects were not significantly distinct from EM4 and T. asperellum, its usage still reduced the severity and suppression of grain rot and leaf blight diseases

Abstract

One of the constraints in rice production is grain rot disease caused by Burkholderia glumae, which can be carried by seeds grain. An observation to determine the presence of B. glumae in different grain yield classes of seeds was conducted by taking samples derived from several seed producers in South Sulawesi. This research was carried out by first taking seed samples at the South Sulawesi Agricultural Technology Study Center (BPTP), Tungro Research Workshop, South Sulawesi Main Seed Center (Balai Benih Induk), PT. Sang Hyang Seri, PT. Pertani and PT. Harmoni were then tested at the Agricultural Quarantine Laboratory of Makassar. Based on the observations, it was concluded that all samples in the sowing seed class tested positive for B. glumae, supported by an average percentage of disease incidence of 25.13%, namely foundation seeds from the Balai Benih Induk, as well as foundation seeds and stock seeds from AIAT.

ABSTRACT Bananas are an important commodity in South Sulawesi, but their productivity still faces various challenges, including suboptimal cultivation techniques and pest and disease attacks. The 2024 Unhas Community Service Program-Partnership Program (PPMU-PK) aims to improve the quality and productivity of banana seedlings through environmentally friendly seedling technology innovations, particularly by utilizing tribacompost and tissue culture technology. The activities were carried out in Mattiro Bulu Subdistrict, Pinrang Regency, involving local farmers in practical training and technology implementation. This activity showed a significant increase in producing healthy and uniform banana seedlings using tissue culture technology. Using tribacompost improved soil fertility and structure and reduced dependency on chemical fertilizers. In addition, introducing antagonistic microbes such as Trichoderma spp. has effectively controlled fusarium wilt disease. Through this approach, it is expected that farmer groups will be able to produce high-quality and environmentally friendly banana seedlings independently. This activity is also expected to make a significant contribution to increasing banana production in the region, as well as having a positive impact on community welfare through increased income and sustainable agricultural enterprises. Abstract

Fundamentalists are one of the streams that do not want to accept change in the sense that they are against reform. So, they are careful to emphasize that the announcement of the prophethood of Muhammad is not something new, but only continues the series of prophets and apostles who preceded him. Meanwhile, radicalism is a group that is often viewed by the West as a terrorist which aims to undermine political authority by means of jihad. That is, these radical religious movements make jihad as a method to achieve their goals, namely the order of the Islamic system (al-nizām al-Islāmi). In addition, radicals are considered narrow-minded, ultra zeolous, or want to achieve goals by using violent means. Because it is understandable why most Muslim scholars view radicalism as an unfavorable term and cause misunderstanding. This view is also found in Western orientalists and scholars who understand Islam.

Abstract

Bacterial leaf blight (BLB), caused by Xanthomonas oryzae pv. oryzae (Xoo), is a major disease of rice. This study aimed to determine the Xoo pathotypes present in rice-growing areas of South Sulawesi and to evaluate the resistance of selected rice varieties. The research was conducted at the Agricultural Biotechnology Research and Development Center and the Plant Pests and Diseases Laboratory, Hasanuddin University, Makassar. Xoo isolates were cultured on Wakimoto medium and inoculated using the clipping method. Five differential varieties (Kinmaze, Wase Aikoku, Tetep, Java 14, and Kogyoku) were used to classify the pathotypes, while five commercial rice varieties (Inpari 6, Ciliwung, Ciherang, IR 64, and Pioneer) were tested for resistance. Six Xoo isolates were classified into pathotypes VI, VII, VIII, and XI, with pathotype VIII being the most dominant (33.3%). Inpari 6 and Ciliwung showed resistance to all tested pathotypes. Ciherang was resistant to most pathotypes but moderately susceptible to VIII. IR 64 and Pioneer displayed varying degrees of resistance and moderate resistance. These findings highlight the presence of diverse Xoo pathotypes and identify rice varieties with broad-spectrum resistance, providing valuable information for the development of BLB management strategies suited to specific regional conditions.
