Abstract

A candidate marker that influences the flavor and odor of Indonesian sheep that is the Potassium Channel Tetramerization Domain Containing 2 (KCTD2) gene. This study aims to examine the polymorphism and association between the KCTD2 gene and lamb flavor and odor in Indonesian local sheeps. This study used DNA taken from the longissimus dorsi (LD) muscle of 100 rams including 75 Javanese Thin-Tail sheep (JTTS), 10 Javanese Fat-Tail Sheep (JFTS), and 15 Jonggol Sheep (JS), with ranged in weight from 20 to 35 kg and were 10 to 12 months old. The PCR-RFLP technique and GLM test analysis were used to identify polymorphisms and association of KCTD2 gene. The restriction enzyme BafI digests the PCR-RFLP product of the KCTD2 gene. The results showed that the KCTD2 gene was polymorphic (CC and CT). KCTD2 gene analysis showed a significant (P<0.05) association with 3-Methylphenol (MP). This research provide information regarding the role of the KCTD2 gene in lamb flavor and odor, especially in 3-Methylphenol (MP) compounds, and explain KCTD2 as a functional gene in the selection of premium sheep with low flavor and odor in lamb meat.

Abstract

The objective of this study was to evaluate the effect of exercise levels on the hematological and metabolical status, and carcass quality of Garut sheep. In total 24 of Garut sheep used in this study. The levels of exercise contain three treatments namely non exercise, semi exercise  and exercise. Blood collection through the jugular vein of sheep samples was carried out at the end of the study, using a 5 mL EDTA tube to measure the haematological condition of the animal sample. Likewise, collecting muscle samples to determine the physical quality of meat.The data was analyzed by analysis of variance and continued by Duncan test for a post hoc test. The results showed that the models of exercise were not significantly (P0.05) affected to hematological level and also carccas quality including juiceness, tenderness, drip loss and water holding capacity. Organoleptic sensory  using semiexercise was more favourable meat  compare to exercise and non exercise. While for the colour of meat, non exercise treatment was significantly (P0.05) affected to the colour of meat compare to exercise and semi exercise. Furthermore the semi exercise  was the best treatment to produce fresh taste meat with good sensory characteristic.

Abstract

The apolipoprotein A-V (APOA5) gene is a candidate gene that plays an important role in carcass characteristics and lamb quality. This study aimed to association and polymorphism of the APOA5 gene with carcass characteristics and lamb quality in sheep. The breed of sheep were used 100 sheep consisting of Thin Tailed Sheep (TTS) (n=55), Compass Agrinac Sheep (CAS) (n=10), Garut Sheep (GS) (n=18), Garut Composite Sheep (GCS) (n=8), and Barbados cross sheep (BCS) (n=9). Identification of APOA5 gene polymorphism was performed using PCR-RFLP method and association study was performed using GLM analysis. The results showed that the APOA5 (g.26929941 C>T) gene polymorphism was polymorphic in TTS, CAS, GS, and GCS, while BCS was monomorphic. The APOA5 gene has three genotypes (CC, CT, and TT). Association showed that the APOA5 was a significantly (P<0.05) associated with carcass percentage and carcass length. The polymorphism of the APOA5 (g.26929941 C>T) was also significant (P<0.05) associated with lamb quality of the pH dan tenderness. The TT genotype showed greater carcass length and pH than the CC and CT genotypes. The CT genotype showed a greater percentage of carcass and tenderness than the CC and TT genotypes. The APOA5 gene helps improve sheep carcass characteristics and lamb quality.

Abstract

On the freezing semen chicken cryoprotectants required to overcome the damage of spermatozoa due to cold shock. This study aims to get the best concentrations of cryoprotectants glycerol concentration of 5%, 7% and 9% in freezing Sentul chicken semen. The semen used in this study came from three chickens Sentul and be repeated nine times. Semen was collected by  messase methods for three times a week. Semen was evaluated macroscopic and microscopic. Furthermore spermatozoa diluted with egg yolk and the addition of three concentrations of cryoprotectants glycerol (5%, 7% and 9%). Semen diluted 0:25 ml is packed into straw. Then equilibrated at a temperature of 5°C for two hours. After equilibration to evaluate the motility and viability of spermatozoa. Furthermore, frozen in liquid nitrogen vapor for 10 minutes. Frozen semen is then stored in liquid nitrogen containers with temperature -196°C. After 24 hours, semen is thawed at 37°C for 30 seconds. The results showed that the percentage of sperm motility and viability of frozen semen cock Sentul using glycerol cryoprotectants 5% better P (0.05) compared with the use of glycerol 7% and 9%. The use of glycerol 5% at this stage of equilibration and storage can reduce the damage of spermatozoa in the semen of chicken Sentul. Neither glycerol 5% could increase recovery rate after thawing

Abstract

Diacylglycerol acyltransferase 1 (DGAT1) is a potential candidate gene for improving Indonesian lamb’s quality and nutrient value.  The study aimed to identify the diversity of the DGAT1 gene with SNP g.8539 CT and its relation to Indonesian lamb's flavor and odor, cholesterol, and mineral.  Total of 254 ten to twelve months old sheep consisted of 20 Javanese fat-tail sheep (JFTS), 107 Javanese thin-tail sheep (JTTS), 10 Garut composite sheep (GCS), 10 Compass agrinak sheep (CAS), 10 Barbados cross sheep (BCS), 20 Garut sheep (GS), 27 Jonggol sheep (JS), and 50 Jambi local sheep (JLS).  One hundred sheep were used to analyze the association of the DGAT1 gene with flavor and odor content, cholesterol, and mineral.  The diversity of DGAT1|AluI was analyzed with the PCR-RFLP method.  The association of the DGAT1 gene with the nutritional value of meat was analyzed using the GLM (General Linear Model) method.  The results showed that the DGAT gene was polymorphic in JTTS, GS, and JFTS and monomorphic in CAS, BCS, JLS, and GCS.  CC and CT genotypes were found in JTTS, GS, and JFTS.  SNP g.8539 CT of DGAT1 gene had a significant association (P0.05) with flavor and odor, 4-Ethyloctanoic Acid (EOA).  However, the DGAT1 gene had no significant association (P0.05) with cholesterol and mineral.  The DGAT1 gene might be marker-assisted selection for improving lamb flavor and odor in Indonesian sheep.

Abstract

The Complement Factor H Related 5 (CFHR5) gene is speculated to have an important role in regulating fatty acid composition in sheep. The aim of this study was to investigate the polymorphism of the CFHR5 gene and its association with fatty acid composition and cholesterol of sheep in Indonesia. A total of 172 rams from 83 priangan sheep (PS), 20 sapudi sheep (SS), 19 Garut sheep (GS), 20 jonggol sheep (JS), 10 Garut composite sheep (GCS), 10 compass agrinac sheep (CAS), and 10 Barbados cross sheep (BCS) were used for this study. Identification of CFHR5 gene polymorphism were performed by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) using AciI restriction enzyme. The results showed that the CFHR5 gene in all sheep populations were polymorphic producing three genotypes, e.g. CC, CT and TT, respectively. The polymorphism had a significant effect (p<0.05) on fatty acid composition (miristoleic [C14:1], ginkgolic [C17:1], tridecanoic [C13:0], and heptadecanoic [C17:0] acids) and cholesterol. The breed and polymorphism had a significant effect (p<0.05) on fatty acid composition (JS with tridecanoic acid [C13:0] and BCS with miristoleic acid [C14:1]). The CC genotype is the preferred genotype and as it exhibits reduced levels of saturated fatty acids and cholesterol. The BCS sheep is the preferred genotype and as it exhibits high PUFA/SFA ratio. The CFHR5 gene (SNP c.1011C>T) has the potential to be used as a genetic marker for the selection of low saturated fatty acid composition and cholesterol in sheep

Abstract

The purpose of this study was to determine the effect of the Payroll Control System and Performance Benefits on Employee Performance at Bappeltibangda Cianjur Regency. The research method used is descriptive research, which analyzes research results without drawing broader conclusions. This research involves independent variables, namely Payroll Control System (X1) and Performance Allowance (X2), as well as the dependent variable, namely Employee Performance (Y). Data collection techniques were carried out through field studies with questionnaires distributed to 84 respondents, and literature searches. Based on the results of the study, it can be seen that there is a significant relationship between the Payroll Control System, Performance Benefits, and Employee Performance at Bappeltibangda Cianjur Regency. The results of statistical calculations using SPSS show the value of F = 51.503, and the value of Sig. = 0,000. With the calculated F value above the F table (3.11) and the Sig value. <0.05, H0 is rejected, so Ha is accepted. This means that the Payroll Control System (X1) and Performance Allowance (X2) variables together have a significant effect on the Employee Performance variable (Y).

Abstract

Integrated Marketing Communication (IMC) Omnichannel has become a key strategy for retail companies in Indonesia. This research aims to analyze the impact of IMC Omnichannel implementation on the satisfaction of retail products and services in Indonesia using an open access approach. The rapid development of technology and high internet penetration in Indonesia has prompted retail companies to shift to digital technology-based marketing. The objective of this study is to understand how the integration of various communication channels in IMC Omnichannel affects consumers' perceptions and levels of satisfaction with retail products and services in Indonesia. The research method used is descriptive qualitative, with primary data sources derived from relevant journals, books, and articles. Analysis was conducted by collecting and evaluating relevant information about IMC Omnichannel implementation and its impact on consumer satisfaction in the retail context in Indonesia. The research findings indicate that IMC Omnichannel implementation has a significant impact on the satisfaction of retail products and services in Indonesia. The integration of various communication channels allows companies to provide a more comprehensive and consistent shopping experience for consumers. Additionally, service quality, responsiveness to consumer needs, and personalized shopping experiences also play a crucial role in influencing consumer satisfaction in the context of IMC Omnichannel.

Abstract

Scabies does not endanger human life, but it is very disturbing to feel comfortable and can reduce activity and productivity, this skin disease is often found in highly populated environments such as in Islamic boarding schools. Health education is needed to provide information about scabies to students, because the lack of knowledge and behavior can be one of the factors causing scabies. This study was to determine the effect of health education about scabies on the behavior of students in Al-Ulfah Rancah boarding school. This research design is Pre-Experimental one group pretest-posttest design. The number of samples in this study were 43 respondents. Sampling using purposive sampling technique. Data were collected by questionnaire. It is known that before being given health education about scabies to 43 respondents, there were 14 respondents (32.55%) who fell into the bad category and 29 respondents (67.45%). This study shows that after being given health education about scabies, 2 respondents (4.65%) were in the bad category and 41 respondents (95.35%) were in the good category with an average increase of 11.79. It can be concluded that there is a significant influence between health education about scabies on the behavior of students. Thus it is expected to make a sustainable program, especially regarding the behavior of preventing scabies by paying attention to the environment and personal hygiene.

Abstract

Kandungan asam lemak dipengaruhi oleh sejumlah gen dan di antaranya ialah selenoprotein W1 (SEPW1). Eksperimen ini bermaksud untuk mengetahui polimorfisme gen SEPW1 serta hubungannya terhadap kandungan asam lemak di domba. Sejumlah 102 ekor domba yang tersusun dari 20 domba ekor gemuk (DEG), 30 domba ekor tipis (DET), 9 domba komposit garut (DKG), 10 domba compass agrinak (DCA), 10 domba barbados cross (DBC), dan 23 domba jonggol (DJ) digunakan dalam penelitian ini. Analisis polimorfisme gen SEPW1 dicari dengan memanfaatkan Polymerase Chain Reaction-Restriction Fragment Lenght Polymorphism (PCR-RFLP), enzim BanI, serta produk amplifikasi berukuran 478 bp. Analisis Chi-square memberikan hasil bahwa populasi berada dalam keseimbangan Hardy-Weinberg, kecuali sampel domba jonggol. Polimorfisme gen SEPW1 teramati berada di semua sampel rumpun domba. Single nucleotide polymorphism (SNP) g.53617063 G>A gen SEPW1 berasosiasi (P<0,05) dengan asam laurat (C12:0), asam palmitat (C16:0), asam arakidat (C20:0), asam henekosilat (C21:0), asam miristoleinat (C14:1), asam oleat (C18:1n9c), asam linoelat (C18:2n6c) dan asam lemak total. Genotipe GG memiliki kandungan asam lemak tak jenuh yang paling tinggi dibandingkan genotipe lainnya. Gen SEPW1 memiliki kapasitas untuk dikembangkan sebagai penanda asam lemak di domba.
