Abstract

Mangrove litter production is important in the displacement of organic matter from mangrove vegetation to water such as in the coast of West Muna Regency. However, one factor influencing it is the frequency of rainfall. This research, therefore, aims to determine the influence of the frequency of rainfall in mangrove ecosystem by obtaining data through the fractional wet weight. The results showed that the biomass fraction with the largest amount were Rhizophora mucronata, Rhizophora apiculata, and Sonneratia alba with of 31.59; 31.07; and 30.96%, respectively, while the lowest was Rhizophora stylosa, Bruguiera cylindrica, and Bruguiera gymnorrhiza with of 30.07; 30.10; and 31.95%, respectively. The species with the largest leaf litter production was Bruguiera cylindrica, with of 1.72 g.m-2.day-1 which occurred in April and the lowest was Rhizophora apiculata, 0.24 g.m-2.day-1 in August. The species with the largest leaf litter production was Bruguiera cylindrica, 1.96 g.m-2.day-1 which occurred in May and the lowest was Sonneratia alba, 0.29 g.m-2.day-1 in August. In addition, the species with the largest fruit litter production was Rhizophora apiculata, 1.76 g.m-2.day-1 in April and the lowest was Sonneratia alba, 0.32 g.m-2.day-1 in September. The total litter production was 18,75 g.m-2.day-1 consisted of 6,69 g.m2.day-1 leaf litter, 6,54 g.m-2.day-1 branch litter, and 5,52 g.m-2.day-1 fruit litter. Abstract

Kaur Regency is a high-potential vannamei shrimp (Litopenaeus vannamei) aquaculture area in Bengkulu Province that frequently experiences high mortality rates due to vibriosis caused by pathogenic Vibrio spp. The high shrimp mortality rates resulting in decreased production or crop failure due to attacks or infections of vibriosis disease. Data on these Vibrio spp. in traditional Kaur ponds is required for illness prevention. To identify pathogenic Vibrio spp., this study included purposeful sampling and testing, as well as data analysis. Seven Gram-negative isolates (OALF 1, 2, 3, and 5 from shrimp organs; WALF 7, 8, and 10 from pond water) produced green/yellow colonies on TCBSA medium. Biochemical tests (Gram KOH 3%, catalase, oxidase, fermentation (O/F), TSIA, MIO, lysine decarboxylase, gelatin, Simon citrate, methyl red, urease, glucose, maltose, lactose, mannitol, and sorbitol) revealed that OALF isolates from vannamei shrimp organs have morphological and physiological similarities to the pathogenic species Vibrio parahaemolyticus, whereas WALF isolates from pond water are similar to the pathogenic species V. pluvialis. The study identified Vibrio spp. as the cause of vibriosis in L. vannamei in traditional Kaur ponds, emphasizing the need for improved water and feed management, as well as proper pond environmental practices, potentially including probiotics derived from healthy local shrimp digestive bacteria, to reduce losses in vaname aquaculture in Kaur Regency, Bengkulu. Abstract

Microbe study on degraded trash has been widely conducted, with the intention of improving the efficiency and effectiveness of waste processing. The purpose of this study was to select and identify hydrolytic Bacillus from litter composting. The samples were the litter composting outcomes with five different treatments, namely the inclusion of nutrient broth (NB), commercial formula, molasses, tryptic soy broth (TSB), and talcum, with each formula containing a microbial consortium. Bacteria were isolated using the serial dilution method. Each isolate was evaluated for its capacity to lyse red blood cells on blood agar media. Cellulolytic, amylolytic, and proteolytic bacteria were grown on nutritional agar media containing carboxy-methyl cellulose (CMC), starch, and skim milk. The antagonistic test involved testing isolates that hydrolyze all three types of substrates. Isolates that were not antagonistic to one another were identified based on phenotypes (colony and cell morphology using Gram staining) and genotypes (16S rRNA gene). Fourteen isolates of Bacillus sp. had cellulolytic, amylolytic, and proteolytic properties. Four isolates were chosen; two of them were comparable to Bacillus stercoris, while the other two, 5.5 and 5.9R, are similar to Calidifontibacillus erzurumensis and Bacillus amyloliquefaciens.
