Abstract

This study aims to know the effect of pH water immersion on the germinated cowpea flour characteristics and to know which pH water that can produce a germinated cowpea flour with the best characteristics. This research used a Complete Randomized Design with a different pH water for immersion as the treatment, which is pH 3, pH 5, pH 7,pH 9 and pH 11. The experimentation was repeated 3 times resulting 15 experimental units. The data obtained were analyzed using an analysis of variance and if the treatment had a significant effect, it was followed by a Duncan Multiple Range Test (DMRT). The results showed that the effect of pH water for immersion had a significant effect on total rendement, water content, protein content, carbohydrate content, water absorption capacity, ash content, fat content, crude fiber content and oil absorption capacity. The best characteristic of the flour in this study was the cowpea flour that has been soaked pH 3 with 55.07% rendement, 1.80% water content, 3.66% ash content, 25.40% protein content, 1.09% fat content, 68.03% carbohydrates content, 10.68% fiber content, 2.11 ml/g water absorption and 1.66 ml/g oil absorption.

Abstract

This research was conducted to determine the effect of Milk Clotting Enzyme concentration from Lactobacillus rhamnosus SKG34 on the quality of soft cheese and to determine the concentration of Milk Clotting Enzyme that produced the best quality of soft cheese. This research used a Completely Randomized Design (CRD) with single factor of Milk Clotting Enzyme concentration (16%, 18%, 20%, 22% and 24%). Each treatment was repeated 3 times resulting 15 experimental units. The data were analysed by Analysis of Variance (ANOVA) Test and if the treatment had a significant effect on the variable (P<0,05), then followed by Duncan Multiple Range Test (DMRT). The results showed that Milk Clotting Enzyme concentration had a significant effect on water content, protein content, cheese hardness, and yield. The concentration of 24% Milk Clotting Enzyme produced the best quality of soft cheese with water content of 58.01%, yield 9.34%, protein content 2.68%, cheese hardness 2.68 N, taste was slightly liked, flavour was liked, texture was liked and overall acceptance was liked.

Abstract

Prebiotik adalah komponen makanan yang tidak dapat dicerna di dalam tubuh dan mendorong pertumbuhan atau aktivitas mikroba usus yang bermanfaat seperti bakteri asam laktat (BAL). Namun, kemampuan Lactobacillus yang diisolasi untuk memanfaatkan prebiotik masih sedikit diketahui. Penelitian ini bertujuan untuk mengetahui pengaruh berbagai jenis prebiotik sebagai sumber karbon terhadap metabolisme isolat Lactobacillus indigenous dari teh kombucha, dadih (susu fermentasi tradisional) dan bambu yang digunakan untuk wadah dadih. Kemampuan isolat Lactobacillus dalam memetabolisme berbagai prebiotik sebagai sumber karbon dilakukan dengan menumbuhkan BAL dalam media MRSB dimana kandungan glukosa diganti dengan prebiotik yaitu fruktooligosakarida (FOS), galaktooligosakarida (GOS) dan inulin, kemudian penurunan kadar glukosa diukur setelah inkubasi 24 jam. Pengukuran total glukosa dilakukan dengan metode fenol sulfat. Hasil penelitian menunjukkan keseluruhan isolat Lactobacillus mengalami peningkatan jumlah sel selama inkubasi 24 jam yaitu dari 0,52 menjadi 1,26 log. Penurunan kadar glukosa berkisar antara 3,80 ± 0,44% sampai 91,33 ± 4,83%. Penurunan kadar glukosa tertinggi terjadi pada isolat L. plantarum1 RB210 yaitu 91,33 ± 4,83% dan tidak berbeda nyata (P>0,05) dengan L. paracasei RK41 dan L. paracasei RL2 yang ditumbuhkan pada media inulin. L. plantarum1 MK2 menunjukkan penurunan pH tertinggi yaitu 1,91 ± 0,08 dan tidak berbeda nyata (P>0,05) dengan L. rhamnosus MY2 pada media yang mengandung GOS. Hasil penelitian ini menunjukkan bahwa isolat Lactobacillus Indigenus secara definitif dapat menggunakan prebiotik sebagai sumber karbon untuk metabolismenya. Hasil penelitian ini memberikan dasar pengaruh penggunaan probiotik dan prebiotik untuk aplikasi sinbiotik sehingga dapat memodulasi mikrobiota usus.

Abstract

The objectives of this research were to determine the optimum incubation time of the LAB isolated from kombucha to produce the best bile salt hydrolase activity (BSH) and to better understand its in vitro cholesterol-lowering ability. The experimental design used in this research was Completely Randomized Factorial Design, with 4 isolates of LAB which were L. pentosus MB23; L. pentosus MS21; L. paracasei ssp. paracasei RK41; L. plantarum 1 RB210 and 3 levels of incubation time which were 12, 24, and 36 h were given as the treatments. The 12 combinations were obtained and were done thrice, resulting in 36 experimental units. The data obtained were analyzed using ANOVA and if it showed a significant result, the Duncan Test will be carried out. The results showed that L. plantarum 1 RB210 isolate exhibited its best anti hypercholesterolemic potential at 36 h of incubation with the total LAB of 8.47 log cfu/ml, BSH activity with precipitation diameter of 7.61 mm, and cholesterol-lowering ability of 62.33%.

Abstract

The objectives of this research were to determine the antibacterial activity of Lactobacillus plantarum 1 RN9 on several metabolites with different incubation times against E. coli ATCC 25922 and to determine its optimum incubation times in producing the highest antibacterial activity against Escherichia coli ATCC 25922. This research used L. plantarum 1 RN9 which was isolated from dadih (Dadih is a typical Minang fermented milk made from buffalo milk, keep 2 nights in a bamboo tube covered with banana leaves), incubation times (12, 24, 36, and 48 hours), with three types of supernatant namely cell-free supernatant (CFS), neutral cell-free supernatant, and heated neutral cell-free supernatant. The supernatants were tested for its antimicrobial activity against E. coli ATCC 25922. There were two methods used in this research, namely agar-based disk diffusion and contact method. The highest antimicrobial activity of L. plantarum 1 RN9 against E. coli ATCC 25922 with cell-free culture supernatant was shown by isolate RN9 at incubation time of 24 hours with a decrease of 1.76 log cycle and 6.50 mm inhibition diameter.

Abstract

The purpose of this research was to know the effect of mulberry leaves powder addition on marshmallow characteristic and to determine the right amount of mulberry leaves powder addition that produced marshmallow with the best characteristics. This research used a completely randomized design with treatment consisting of 5 levels of concentration ratio of mulberry leaves powder (0%, 2%. 4%, 6%, 8%). Each treatment repeated 3 times in order to obtain 15 experimental units. The data obtained were analysed by analysis of variance and if there was an effect, then it will be followed by the Duncan Multiple Range Test (DMRT). The result showed that the addition of mulberry leaves powder had significant effect (p>0.05) on ash content, antioxidant activity, vitamin C, color intensity, color sensory, scent, texture, taste, and overall acceptance. The result showed that 4% concentration of mulberry leaves powder addition in marshmallow had significant effect on ash content, antioxidant activity, vitamin C, Color intensity, color sensory, scent, texture, taste, and overall acceptance. The 4% addition of mulberry leaves powder produced the best characteristics with 27.67% water content; 0.55% ash content; 15.82% antioxidant activity; 0.79 mg/g vitamin C; 46.30 L* score; -5.93 a* score; 20.77 b* score; color liked; scent liked; texture liked; taste liked; all acceptance liked.

Abstract

The study aimed to know the effect of the ratio maltodextrin and gum arabic on the antioxidant activity and color of the copigmentation tamarillo powder and to proper ratio of maltodextrin and gum arabic to get the copigmentation tamarillo powder with the best antioxidant activity and color by foam mat drying method. The study used a completely randomized design (CRD) with ratio of maltodextrin and gum arabic treatment consisting of five levels, there were 5:0, 4:1, 3:2, 2:3, 1:4. All treatments were repeated three times in order to obtain 15 experimental units. The data were analyzed statistically using the variance test and if the treatment had a significant effect to observed variables, it was continued with Duncan's Multiple Range Test (DMRT). The result showed that the ratio of maltodextrin and gum arabic had a significant effect (P<0,05) on water content, ash content, solubility, total anthocyanin, total phenol, antioxidant activity, and color intensity. The ratio of maltodexstrin and arabic gum (1:4) was the best treatment to produce copigmentation tamarillo powder with the total anthocyanin 41.97 mg/100g, total phenol 125.57 mg GAE/100g, antioxidant activity 78.05%, value L* 47.10, a* 44.03, b* 23.97, chroma 50.14, °Hue 28.56 (red color), water content 3.92% and solubility 83.90%

Abstract

The purpose of this study was to determine the effect of different levels of leaf aging on the characteristics of soursop leaf herbal tea and to determine the level of soursop leaf aging which can produce soursop herbal tea with the best characteristics. This study used a completely randomized design (CRD) with 3 levels of leaf aging treatment, namely young leaves, half old leaves and old leaves. Parameters observed in this study included water content, ash content, crude fiber content, extract content in water, total phenol, total flavonoid, tannin content, antioxidant activity, and sensory assessment which included color, aroma, taste, and overall acceptance ( hedonic test) soursop herbal tea and color and taste (scoring test) soursop herbal tea. The results showed that the difference in the level of leaf aging had a very significant effect on water content, ash content, crude fiber content, extract content in water, total phenol, total flavonoid, tannin content, antioxidant activity, and sensory characteristics including color and aroma. Soursop herbal tea prepared from young leaves had the best characteristics with criteria of water content 7.69%, ash content 5.86%, crude fiber content 17.62%, extract in water 121.25%, total phenol 2.76%, total flavonoids 2.66%, tannin content 3.26%, antioxidant activity 55.87%, the hedonic sensory characteristics of color, aroma, taste, and overall acceptability were neutral, and the scoring sensory characteristics of color was reddish yellow and taste was a little bitter.

Abstract

This study aims to determine the effect of the ratio of coconut pulps on the characteristics of Kintamani Siamese Orange jam, as well as to determine the optimal ratio between coconut pulps and kintamani siamese orange to produce jam with desirable characteristics amd preference. This study was an experimental study using a Randomized Block Design consisting of six treatments, namely the ratio of coconut pulps and kintamani siamese orange 0: 100, 5: 95, 10: 90, 15: 85, 20: 80, and 25: 75. Each treatment was repeated 3 times, resulting in total of 18 experimental units. The obtained data were analyzed using Analysis Of Variance (ANOVA) and is there were significant effects, the Duncan Multiple Range Test (DMRT) was conducted. The results of the study showed that the ratio of coconut pulps and kintamani siamese orange in jam had a significant effect (P<0.05) on water content, viscosity, syneresis, vitamin C content, crude fiber content, total dissolved solids, and hedonic tests on color, aroma, taste and overall acceptance. The ratio of coconut pulp and Kintamani siam orange 5 : 95 has the best jam characteristics with a water content 17,24 %, total dissolved solids 51,66 %, vitamin C content 51,47 mg/g, syneresis 0%, viscosity 4.66 Pa.S, crude fiber content 1,86%, with orange color, aroma, taste, texture, and overall acceptance preferred by the panelists

Abstract

Lime is a fruit that contains high vitamin C. Holding time could decreased the vitamin C, and to prevent that, the addition of honey is needed. The aim of this study is to determine the effect of holding time and honey concentration on vitamin C content, and to determine the holding time and honey concentration that can produce the lime water beverage with the best characteristics. This study used a completely randomized block design with 2 factors, which are holding time with 3 levels of treatment 0, 30, and 60 minutes, and honey concentration with 3 levels of treatment 0, 5 and 10%. All treatments were combined and repeated 3 times to obtain 27 experimental units. The data was analysed by analysis of variance and the treatment that had a significant effect on the parameters was continued with DMRT. The results showed that the interaction of both factors had a significant effect on vitamin C, antioxidant activity, reducing sugar and total acid. Holding time caused decreased in vitamin C, antioxidant activity, total acid, reducing sugar, hedonic and scoring of taste. Whereas, honey concentration caused increased in all parameters except total acid that experience a degradation. Lime water beverage with the best characteristic is the lime water beverage with 30 minutes of holding time and 10% of honey concentration with the characteristics of vitamin C 1112.95 mg/100g, antioxidant activity 86.57%, reducing sugar 4.93%, total acid 1.97%, sweet taste, yellow brownish colour, taste was liked and colour was liked.
