Abstract

The use of red ginger (Zingiber officinale var. rubrum) as the primary ingredient of kombucha is a diversification to increase the antioxidant activity of kombucha. The presence of gingerol dan shogaol in red ginger acts as a potential antioxidant, while the use of palm sugar serves as an additional source of antioxidants and alternative source of sucrose from regular granulated sugar. The fermentation time and the amount of sugar used are two variables that affect the kombucha’s fermentation process. This study aims to examine the effect of various fermentation times and palm sugar concentrations on the antioxidant activity, pH, alcohol content, and organoleptic properties of red ginger kombucha. The methods used were antioxidant test with DPPH, pH test, alcohol content, and organoleptic test with 20 panelist. The results indicate that variations in fermentation time and concentration of palm sugar have a significanr effect (α = 0.05) on the antioxidant activity with IC50 values ranging from 27.89 ppm to 46.77 ppm, categorized as very strong antioxidant activity. The characteristics of red ginger kombucha includes pH 3.6 to 4.6, alcohol content 0.2% to 0.3%, and organoleptic properties varying from moderately liked to liked. Red ginger kombucha fermented for 12 days with 20% palm sugar showed an IC50 value of 40.28 ppm, which categorized into very strong antioxidant activity and is most preferred by the panelists with organoleptic scores of color attribute at 4, aroma at 3.65, and taste at 4.

Abstract

Natural carotenoid synthesis has never exceeded synthetic products on a commercial scale. Lack of understanding of the microbiological and ecophysiological aspects of carotenoid-producing isolates leads to misidentification of species. One local isolate of green algae is used as a natural food source of carotenoids in the food industry, namely Chlorella pyrenoidosa. Carotenoid accumulation of the nonMVA pathway in green algae is determined by the enzyme D-1-deoxyxylulose 5-phosphate synthase, which is encoded by the D-1-deoxyxylulose 5-phosphate synthase (DXS) gene. The main purpose of this study was to detect the DXS gene as a carotenoid biosynthetic key enzyme encoder in C. pyrenoidosa whether or not to follow the non-MVA pathway for carotenoid biosynthesis or not. Morphological and ecophysiological characterization methods are carried out based on periodic observations and DXS gene detection using the guide Kuzuyama (2000). The results of the analysis of the similarity of the C. pyrenoidosa DXS gene in sustainable areas show that it can detect partial DXS genes from Chlamydomonas reinhardtii. The absence of growth inhibition in C. pyrenoidosa with lovastatin shows a non-MVA pathway that is the pathway used in carotenoid biosynthesis.

Abstract

Filariasis limfatik merupakan suatu penyakit yang disebabkan oleh nematoda dari jenis Wuchereria bancrofti dan Brugia malayi dewasa yang menghuni jaringan limfatik. Filariasis limfatik ini disebarkan oleh empat genus nyamuk, yaitu, Anopheles, Culex, Aedes dan Mansonia. Respon imun terhadap parasit filaria terdiri dari Thelper2 (Th2) dan melibatkan produksi cytokin –IL-4, IL-5, IL-9, IL-10, dan IL-13, isotype antibody—IgG1, IgG4 and IgE, dan meningkatnya populasi eosinophils dan makrofag yang aktif. Polimorfisme pada daerah promotor gen IL-10 diidentifikasi berhubungan dengan tinggi rendahnya produksi IL-10 pada sel darah perifer pada stimulasi in vitro dengan lipopolysacharide (LPS). Sampel diambil dari penderita filariasis, kemudian diekstraksi dan diamplifikasi pada posisi -1082, -819, -592. Hasil amplifikasi kemudian di restriksi dan di sekuensing. Hasil penelitian dari sampel penderita filariasis yang diteliti menunjukan memiliki haplotip ACA pada analisis RFLP. Analisis hasil sekuensing menggunakan dua aplikasi, yaitu DNA baser dan Bioedit. Analisis menggunakan DNA baser dan Bioedit serta dibandingkan dengan tag SNP didapatkan satu SNP. Perbandingan antara sekuens DNA yang didapatkan dengan tag SNP mandapatkan haplotip CC.

Abstract

Cabai merah (Capsicum annuum L.) merupakan komoditas pertanian penting di Indonesia yang sering terkena serangan OPT ( organisme pengganggu tanaman) misalnya Fusarium oxysporum. Tanaman cabai merah memiliki sifat ketahanan kimiawi aktif yaitu mengekspresikan peroksidase sebagai respons terhadap infeksi patogen. Penelitian ini bertujuan untuk mengetahui ekspresi gen penyandi peroksidase pada tanaman cabai merah sebagai respons terhadap jamur F. oxysporum. Penelitian ini diawali dengan inokulasi jamur pada tanaman cabai merah dilakukan dengan metode rendaman akar. Analisis ekspresi gen penyandi peroksidase menggunakan qRT-PCR dilakukan pada gen CaPer dengan gen 18s rRNA sebagai gen pembaku, pada interval 6, 48, dan 96 jam setelah inokulasi. Hasil penelitian gen CaPer terekspresi paling tinggi pada 6 jam setelah inokulasi.

Abstract

Tanaman atsiri merupakan unggulan ciri khas Kabupaten Batang dimana produk minyak atsirinya menempati peringkat ke 3 di Indonesia dan utamanya terdiri dari minyak cengkeh, nilam, dan serai wangi. Minyak atsiri Indonesia mempunyai pangsa pasar lebih dari 90% kebutuhan dunia atau sekitar 35-40% dari total nilai ekspor minyak atsiri. Kabupaten Batang memiliki sekitar 30 UMKM yang bekerja secara sinergis di bidang wirausaha produksi dan pengolahan minyak atsiri. Meskipun demikian, harga minyak atsiri sangat mudah naik turun bergantung pada pasar. Hal ini menyebabkan perlunya upaya diversifikasi berbagai produk berbasis atsiri yang dapat dikerjakan oleh UMKM (Usaha Menengah Kecil dan Mikro) dan Kelompok Wanita Tani (KWT)  dan diversifikasi produk berbasis atsiri yang mempunyai nilai ekonomi cukup tinggi. Pengabdian Masyarakat yang dilakukan bertujuan melatih pembuatan diversifikasi produk berbasis atsiri.  Kegiatan dilakukan melalui pelatihan diversifikasi produk atsiri berupa pembuatan bahan tambahan cat aromaterapi antimikroba. Hasil kegiatan memperlihatkan UMKM dan KWT mengerti dan mampu membuat produk yang disuluhkan dengan baik dan bernilai ekonomi. Selain itu mereka juga sangat tertarik untuk mengaplikasikan cat aromaterapi antimikroba di lingkungan mereka agar tercipta lingkungan yang lebih sehat.

Abstract

Patchouli (Pogostemon cablin Benth.) is an essential oil-producing plant from Batang Regency that has excellence in patchouli alcohol contents and highly survives in any condition. Molecular identification has been done in the ITS region because DNA sequences in the ITS rRNA region evolved faster than in other areas. This study aimed to get the molecular and morphological identity of patchouli (Pogostemon sp.) from BPP Kabupaten Batang. The study consisted of sample preparation, DNA isolation, amplification, electrophoresis, sequence data analysis, and phylogenetic analysis using MEGA X. All parts of plant were morphologically identified and compared with patchouli from Sidikalang, Aceh, Java, and China. Extraction DNA produced 301.7 ng ?L–1 concentration and 1.93 purity. Amplification of ITS fragment patchouli produced a 670 bp-sized single band. Phylogenetic analysis showed patchouli BPP related to Pogostemon cablin (KR608752.1) with 98% coverage identity. BPP patchouli showed 62,5% morphological similarity with Sidikalang patchouli compared to Java patchouli. In conclusion, BPP patchouli is a Sidakalang patchouli P. cablin that has undergone environmental adaptation.

Abstract

Cardamom and Sambiloto are phytopharmaceutical plants that produce phytochemical compounds that have the potential to be used to increase immunity against COVID-19. because they contain carotenoids, phenols, anthocyanins, saponins, alkaloids and steroids. This research aims to obtain the molecular characteristics of Cardamom and Sambiloto plants from the Gunungpati area, Semarang by ITS primer, testing phenolic phytochemicals, tannins, flavonoids, saponins and alkaloids followed by molecular docking tests with the 6WX4 protein. SARS-CoV-2. Molecular characterization results show that Cardamom and Sambiloto are similar to Wurfbainia compacta MF802556.1 (100%) and Andrographis paniculate LC646073.1 (84.47%). The results of the phytochemical test screening showed that both plants contain flavonoids. Molecular docking tests were carried out with the compounds Quercetin, Avicularin, Naringenin, 5-hydroxy-7,8,2',5' tetramethoxyflavone, and Retinoic Acid. Retinoic Acid as a test ligand has the greatest potential in inhibiting the 6WX4 protein in the SARS-CoV-2 virus with a binding affinity value of -7.28 and RMSD 0.00.

Abstract

Global consumption trends show a tendency to choose natural food preservatives, especially those derived from plants, especially to avoid chemical and synthetic preservatives that are harmful to health. Nanotechnology is an important development and an efficient choice to extend the shelf life of food. This study aims to create and carry out seafood preservation using herbs edible coating from garlic nanoparticles, pandan leaves and lemongrass to determine its use as a preservative for salted mackerel and tigawaja salted fish. As a comparison, a mixture of the three ingredients. The study used four treatments where each was repeated 3 times. A hedonic test was also carried out in the form of observations of dryness, color, texture and odor of salted fish. The research method consisted of making garlic nanoparticle extract, pandan leaf extract, lemongrass extract, and a mixture of the three ingredients followed by microscopic observation. Antibacterial activity was carried out using the TPC test. The hedonic test was carried out to see the potential of consumers for the product. Furthermore, a PASS analysis was carried out to see the potential of antimicrobial compounds in the three ingredients. The results showed that the edible coating layer was made into nanoparticle form. Edible coating from pandan is most suitable for use on salted mackerel. Edible coating from garlic is most suitable for use on tigawaja salted fish. The TPC test results showed that garlic, pandan and lemongrass were able to prevent microbial growth effectively for 48-96hours and have the potential to be used as edible coating.

Abstract

Ketersediaan sumber daya alam yang melimpah di KHDTK Wanadipa Undip, khususnya tanaman penghasil atsiri belum banyak dimanfaatkan. Kelompok Wanita Tani (KWT) mengalami kesulitan dalam memanfaatkan tanaman atsiri untuk memnghasilkan produk khas hutan yang berkualitas dan bernilai jual tinggi. Kegiatan bertujuan memberikan aplikasi TTG melalui pelatihan pembuatan produk  sabun pembersih lantai berbantuan atsiri hutan dan Eco-enzyme yang mempunyai nilai ekonomi beserta teknik pemasarannya melalui e-commerce. Pengabdian dilakukan dengan metode observasi, dan pelatihan langsung kepada anggota KWT. Hasil yang didapatkan yaitu KWT dapat menghasilkan sabun pembersih lantai atsiri beraroma atsiri hutan dan berbantuan Eco-enzyme secara mandiri yang bernilai jual dan peningkatan keterampilan dalam pemasaran melalui platform e-commerce. Kegiatan yang dilakukan telah terbukti meningkatkan kesejahteraan ekonomi masyarakat dan KWT di sekitar KHDTK Wanadipa Undip secara berkelanjutan.

Abstract

Heavy metal contamination, particularly by copper (Cu) and lead (Pb), in aquatic ecosystems poses a significant threat to ecological balance, aquatic organisms, microbial diversity and human health. The Kali Baru Estuary in Semarang, Indonesia, has been reported as a heavily polluted area with Cu and Pb concentrations far exceeding environmental safety thresholds, highlighting the urgent need for sustainable remediation strategies. This study aimed to isolate and characterise bacterial strains resistant to Cu and Pb from sediment samples collected from the Kali Baru Estuary in Semarang as potential bioremediation agents. Bacterial isolation was performed using a serial dilution technique up to 10?², in triplicate, spread onto Marine Agar medium, and incubated at 37°C for 24 h. The obtained isolates were purified and characterised based on their macroscopic and microscopic morphological features. Resistance profiling of the isolates was assessed by exposing them to Cu and Pb at concentrations of 100, 200, and 300 mg/L. A total of 32 bacterial isolates were successfully obtained. Among these, two isolates demonstrated resistance to Cu, and 14 isolates exhibited resistance to Pb at the highest concentration tested (300 mg/L). These metal-resistant bacterial isolates exhibit substantial promise for bioremediation applications aimed at remediating Cu and Pb-contaminated aquatic environments.
