Abstract

UMKM atau usaha mikro, kecil, dan menengah merupakan bagian yang sangat penting dan strategis dalam peningkatan perekonomian nasional, terutama berperan dalam mendorong pertumbuhan ekonomi negara, apalagi saat ini pemerintah sedang berupaya memulihkan keadaan ekonomi pasca Covid-19. UMKM yang tangguh diawali dengan pemahaman pengelolaan keuangan yang baik. Kecamatan Bandar merupakan salah satu kecamatan di Kabupaten Batang yang memiliki banyak potensi sumberdaya yang dapat dikembangkan. UMKM di kecamatan Bandar kabupaten Batang Jawa Tengah yang terdiri dari berbagai jenis usaha yang semakin berkembang. Permasalahan yang dialami UMKM tersebut adalah masalah dan kendala dalam hal pengelolaan keuangan, hal ini disebabkan dengan adanya keterbatasan pemahaman pengelolaan keuangan baik secara sederhana maupun digital. Berlatar belakang dari permasalah ini, kegiatan digitalisasi pengelolaan keuangan UMKM Kabupaten Batang sebagai mitra dalam program ini. Program ini menggunakan metode ceramah dan diskusi. Kegiatan ini diawali dengan memberikan pre tes dan postes. Dilihat dari hasil penyebaran kuesioner peserta yang berprofesi sebagai wirausaha, dan memiliki berbagai jenis usaha. Dilihat dari sebelum sosialisasi digitalisasi pengelolaan keuangan terhadap peserta pelaku UMKM memiliki tingkat pemahaman sebesar 4-18% dan sesudah sosialisasi kegiatan mencapai tingkat pemahaman sebesar 100% meskipun masih tahap awal penggunaan digital. Dengah dilakukannya kegiatan sosialisasi ini, dapat dilihat bahwa tingkat pemahaman dan pengetahuan pengelolaan keuangan secara digital bagi pelaku UMKM unbu kecamatan Bandar mengalami peningkatan sehingga lebih meningkatkan kualitas dan produktivitas UMKM tersebut sehingga dapat meningkatkan perekonomian masyarakat pedesaan.

Abstract

Lead is a heavy metal contamination that is released into sea waters and cannot be decomposed so it accumulation and magnification along the food chain. Efforts to explore bacteria that have tolerance and have the potential to become lead reduction agents are a strategic step in remediating lead contamination. One mechanism for lead bioremediation by bioaccumulation is to utilize metallothionein protein which can potentially be obtained from isolates of green mussel associated bacteria. Bacterial isolates associated with green mussels were obtained by isolating the bacteria and testing the tolerance of the bacterial isolates using Luria Bertani media which was added with Pb(NO3)2 at a concentration of 100 ppm and its multiples to determine their tolerance to lead. The bacterial isolates obtained were characterized based on colony and cell morphology. The results of the lead resistance test carried out showed that 11 bacterial isolates were obtained and 3 bacterial isolates had tolerance to lead with a lead concentration of 400 ppm. Bacterial isolates that are tolerant to lead show changes in cell morphology to reduce the impact of exposure to lead which has a lethal effect, because the resulting decrease in cell surface area can reduce lead absorption.

Abstract

Tanjung Emas Port is the entry and exit point for trade commodities, both regional and international filled with many ships. This condition makes biofouling a very massive process in that place by various types of marine bacteria. The initial formation of a biofilm is relevant to bacterial diversity, colonization and adhesion. The objective of the study was assessing bacterial diversity in relation to with biofouling within Tanjung Mas Port Semarang, by using 16S rRNA eDNA metabarcoding Next-Generation Sequencing (NGS). Seawater samples from aquatic sites of Tanjung Mas harbor was used for DNA extraction and amplification of the 16S rRNA V-3-V4 hypervariable region, followed by sequencing and library construction of eDNA Metabarcoding. Sequence processing and analysis was performed in QIIME 2 and RStudio using DADA2 for advanced sequencing processing and Phyloseq. The results of this research showed that bacteria is the predominant taxon constituting 100% of the community. The taxon consists of Proteobacteria (49.38%), Bacteroidota (8.67%), and Firmicutes (8.88%). Alphaproteobacteria (20.92%) and Gammaproteobacteria (12.39%) dominate at the Class level, emphasizing their versatility and ecological influence. At the Order and Family levels reveals the prevalence of Rhodobacterales (10.04%), Chitinophagales (2.53%), Rhizobiales (3.61%), Rhodobacteraceae (5.67%), Saprospiraceae (3.63%), and Rhizobiaceae (2.15%). It was found that the unculturable taxa dominance in Tanjung Mas Port was 44.66%. These taxonomic entities contribute significantly to the taxonomic and functional diversity of the microbial community, influencing nutrient cycling, organic matter degradation, ecosystem stability and biofilm formation.

Abstract

ABSTRAK Jeruk bali merah (Citrus maximaMerr) merupakan tanaman lokal yang tersebar di wilayah Indonesia. Keunggulan tanaman tersebut adalah nutrisi yang tinggi yang terdiri dari vitamin C996,773 mg/L, riboflavin, natrium (Na), kalsium (Ca), serta senyawa metabolit lainnya yang merupakan sumber antioksidan yang baik. Dalam masa pandemi ini, kebutuhan vitamikhusunya dalam menghadapi Pandemi Covid19.Pengembangan potensivarietas unggul dari jeruk bali perlu dilakukan salah satunya dengan analisis genetik. Tahap awal analisis genetik suatu tanaman adalah isolasi DNA tanaman tersebut. Penelitian ini bertujuan untuk mengisolasi DNA pada daun jeruk bali dengan metode Doyle & Doyle yang dimodifikasi dengan penambahan CTAB untuk mengatasi kadungan polisakarida dan senyawa polifenol yang tinggi dalam tanaman jeruk bali. Sampel DNA jeruk bali yang telah diisolasi di analisis menggunakan NanoDrop dengan absorbansi (A260/A280). Hasil pada penelitian ini, sampel DNA yang dianalisis menggunakan NanoDrop memiliki konsentrasi 220,8ng/µlserta kemurnian sebesar 1,93.Kata kunci : Jeruk bali (Citrus maxima), isolasi DNA, CTAB

Abstract

SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2) is a virus responsible for the infectious disease of COVID-19 (Coronavirus Disease 2019), whose development is still being monitored. One way to deal with the virus’s development is by searching for natural-based medicines that prevent and treat SARS-CoV-2 infection. The abundant biodiversity can be used as a source of treatment. Among many natural resources, seaweed is one of the natural resources rich in bioactive components. Sulfated polysaccharide is one of the potential bioactive compounds in seaweed because it has antiviral effects and the potential to treat SARS-CoV-2. This research aims at increasing the potential of Karimunjawa green seaweed sulfate polysaccharides for antiviral capabilities in SARS-CoV-2 through molecular docking. This research began with identifying the morphology of Karimunjawa seaweed. Identify the components of sulfated polysaccharide compounds based on literature studies according to the species that have been identified. Analysis of the antiviral ability of SARS-CoV-2 based on its binding ability to the SARS-CoV-2 target protein through a molecular docking computational program and testing drug compounds using the Lipinski rule. Based on the research results, it is known that the seaweed obtained from Karimunjawa based on morphology, belongs to the genus Kappaphycus. Molecular docking with a ligand and spike glycoprotein (6LZG) resulted in the hexadecanoic acid compound having a binding free energy of -5.3 kcal.mol-1, which was the compound with the lowest yield compared to other test compounds. The prediction of the physicochemical properties of all test compounds fulfills Lipinski's five rules and has the potential to be used as medicinal compounds.

Abstract

In the lagoon area of the Mangrove Information Center (PIM), Pekalongan, annual flooding leads to inundation in some land areas, particularly pond areas, lagoons, and residential areas. The area of submergence due to tidal floods in 2020 reached 783.99 hectares. The construction of embankments to the south of the lagoon as protection against flooding has transformed the northern area into a lagoon. Our research, crucial for understanding and managing these environmental changes, aims to determine and analyze the content and distribution of nitrate in the Lagoon Area of Mangrove Information Center Waters, Pekalongan. This was achieved through a thorough process of in situ observations based on purposive sampling at six sampling sites on June 22, 2023. The nitrate distribution data was then interpolated with a 2D current hydrodynamic model, ensuring the most accurate representation of the data. The value of nitrate content in the Mangrove Information Center lagoon ranges between 0.0065-0.1072 mg/L. This distribution pattern, influenced by deposits of mangrove litter, the depth of the lagoon, and water circulation, reflects the accumulation of physical and chemical influences in the waters.

Abstract

Cardamom and Sambiloto are phytopharmaceutical plants that produce phytochemical compounds that have the potential to be used to increase immunity against COVID-19. because they contain carotenoids, phenols, anthocyanins, saponins, alkaloids and steroids. This research aims to obtain the molecular characteristics of Cardamom and Sambiloto plants from the Gunungpati area, Semarang by ITS primer, testing phenolic phytochemicals, tannins, flavonoids, saponins and alkaloids followed by molecular docking tests with the 6WX4 protein. SARS-CoV-2. Molecular characterization results show that Cardamom and Sambiloto are similar to Wurfbainia compacta MF802556.1 (100%) and Andrographis paniculate LC646073.1 (84.47%). The results of the phytochemical test screening showed that both plants contain flavonoids. Molecular docking tests were carried out with the compounds Quercetin, Avicularin, Naringenin, 5-hydroxy-7,8,2',5' tetramethoxyflavone, and Retinoic Acid. Retinoic Acid as a test ligand has the greatest potential in inhibiting the 6WX4 protein in the SARS-CoV-2 virus with a binding affinity value of -7.28 and RMSD 0.00.

Abstract

Cadmium (Cd) heavy metal pollution not only affects aquatic ecosystems but also has toxic effects on human health. Bioremediation using microalgae Nannochloropsis sp. is considered more economical and sustainable for overcoming heavy metal pollution. This study aims to molecular characterization of Nannochloropsis sp. with tufA gene markers, determine the effect of different Cd concentrations on the growth and morphology of Nannochloropsis sp. and the ability of Nannochloropsis sp. to absorb Cd concentrations. The methods used include DNA isolation, quantitative and qualitative DNA analysis, amplification of tufA gene, sequencing result analysis, phylogenetic tree analysis, and bioremediation test. The results of molecular characterization showed that Nannochloropsis sp. has similarities with N. oceanica strain BR2 plastid (CP044614.1) with 60% bootstrap value. Nannochloropsis sp. grew at different Cd concentration (0, 2, 4, and 6 ppm) but optimal at 0 ppm Cd concentration. Morphology of Nannochloropsis sp. showed morphological differences in the cell structure. The cell was broken and the color turned increasingly clear. Different concentrations of Cd metal on Nannochloropsis sp. growth significantly affected the decline in Nannochloropsis sp. growth. Nannochloropsis sp. has the potential to absorb heavy metal Cd with an efficiency that increases as the concentration reaching 62.6% at 6 ppm. It can be concluded that molecular characterization using the tufA marker was effective in showing that Nannochloropsis sp. had closed relation with N. oceanica strain BR2 plastids. Cadmium (Cd) exposure significantly reduced growth and caused cell damage, while Nannochloropsis sp. effectively adsorbed Cd and lowering Cd levels by 3.76 ppm.

Abstract

Heavy metal lead (Pb) pollution in aquatic environments poses a serious threat to aquatic ecosystems and human health. The microalga Thalassiosira  sp. has potential as a bioremediation agent for heavy metals; however, further studies are needed to maximize its capability. This study aims to molecularly characterize Thalassiosira  sp. using the tufA gene, evaluate the effects of Pb on growth and cell morphology, and assess its Pb uptake efficiency. Methods included DNA isolation, phylogenetic analysis, growth measurement using a hemocytometer, Pb uptake testing via ICP-OES, and cell morphology analysis. Results showed that the tufA gene could molecularly characterize Thalassiosira  sp., with a phylogenetic similarity of 99.53% to Conticriba weissflogii (MH571875.1). Pb significantly inhibited growth at concentrations of 0.03 ppm and 0.05 ppm and caused cell aggregation and pigment degradation at higher concentrations (2 ppm). This microalga exhibited the highest Pb uptake efficiency at lower concentrations (0.03–0.05 ppm), reaching 100%, while at 2 ppm, the efficiency decreased to 40%. This study highlights the potential of Thalassiosira  sp. in remediating Pb, providing a foundation for the development of microalgae-based biotechnological applications.
