Abstract

Species identification is one of the most basic and important factors in fisheries management. Oreochromis sp., such as tilapia (Oreochromis mosambicus) and tilapia (Oreochromis niloticus) are introduced species of fish originating from outside Merauke which are now the dominant fish and are commonly found in inland waters of Merauke. The purpose of this study was to identify the economically important fish species Oreochromis sp. which landed at Merauke Traditional Market with DNA Barcoding approach. DNA amplification with the COI gene resulted in a DNA sequence length of 655 bp with the identification result being Tilapia (Oreochromis niloticus). Based on the phylogenetic tree data and genetic distance, the DNA sequences of the samples formed the same clade as the Oreochromis niloticus sequences from GeneBank with 0% genetic distance and it was estimated that these species came from the same parents. Identification with DNA Barcoding makes it easy to identify species quickly and accurately.

Abstract

The biological nature of tilapia which causes uncontrolled mating and has an impact on reducing the quality of the fish has become one of the problems in tilapia aquaculture production. 5 times faster than female tilapia. The production of male monosexual tilapia can be obtained in several ways, including the application of low doses of hormones. The purpose of this study was to determine the effectiveness of using low doses of 17α Methyl Testosterone (MT) in immersing tilapia larvae and to obtain superior quality male tilapia results. This study used tilapia larvae that were 1 week old and were stocked in a tank measuring 30x60cm with a dose of MT 1 mg / l, 3 mg / l and 5 mg / l with 3 replications and 1 control. stocking density of 250 birds per cage. Soaking is carried out for 8 hours. The results of immersion of tilapia larvae using MT at a dose of 3 mg / l until the 60th day showed the highest results with the proportion of 72% compared to the untreated dose, 1 mg / l and 5 mg / l. It can be ignored that the use of low doses of 17α Methyl testosterone can affect sex changes in tilapia larvae. The results showed that the dose of 5 mg / l was the best dose with the proportion of male genitalia at 97%.

Abstract

The Cenderawasih Bay is a marine habitat for whale sharks (R. typus) which appear almost all year round. The appearance of this whale shark is triggered by various factors, including the food. Anchovy is one of the attractions for the emergence of whale sharks, so it is necessary to conduct genetic, biological and ecological studies. Anchovy has a small in size, making it difficult to identify morphologically. The purpose of this study was to genetically identify anchovy samples obtained from Cenderawasih Bay and compare the sequences with the GenBank database. The COI gene fragments were amplified by PCR method, using primer jg-LCO and jg-HCO. Sequencing is carried out from two directions, forward and reverse with the sanger termination dideoxy method. The resulting DNA sequence has a length of 669 base pairs encoding 223 amino acids. The results of homological comparisons with the NCBI and BOLD System databases show that this sample has similarities to the COI sequence of Spratelloides gracilis with a similarity number up to 99%. The results of the phylogenetic tree analysis showed that the anchovy samples from Cenderawasih Bay were in one clade with S. gracilis from Japan and separated from the clade of S. gracilis from the Red Sea, with a distance between clades is 0.104. This result is in line with the identification by homological comparison in the NCBI and BOLD System.

Abstract

Pemanfaatan perikanan secara lestari sangat diperlukan agar sumberdaya perikanan yang ada saat ini bisa terus dimanfaatkan dan juga dalam rangka pemulihan stok perikanan yang telah rusak. Ikan nila (Oreochromis niloticus) dan ikan mujair (Oreochromis mossambicus) secara luas telah dikenal sebagai salah satu spesies ikan air tawar yang penting dalam perikanan budidaya. Penelitian ini dilakukan dengan tujuan identifikasi spesies ikan Oreochromis spp. dari perairan daratan Merauke dengan menggunakan marka gen COI. Amplifikasi DNA dengan gen COI menghasilkan panjang sekuen DNA 656 bp dengan hasil identifikasi adalah O. niloticus dan O. mussambicus dengan tingkat kemiripan 100 % pada masing-masing spesies. Berdasarkan analisis filogenetik dan jarak genetik, sekuen DNA yang dianalisis membentuk clade sesuai dengn hasil indentfikasi dengan jarak genetik 0,000 - 0,002 pada ikan nila dan 0,000 pada ikan mujair. Analisis keragaman genetik ditemukan hanya 1 haplotype dari kedua jenis ikan tersebut. DNA barcode dan filogenetik dapat digunakan dalam mengidentifikasi spesies dalam pengelolaan dan konservasi.

Abstract

Fish is part of the diet which is a source of animal protein which is very beneficial for health. To meet the availability of fish-based food, it can be processed into various processed products, one of which is shredded fish. Shredded fish still has a drawback, such as the low fiber content. To enrich the fiber content of shredded fish, fortification with banana blossoms can be an alternative. The process of making shredded fish does not yet have a standard because many methods and spices are added, so there are variations in the types and amounts of spices used. This causes the quality of shredded fish to vary. Therefore, this study aimed to evaluate the chemical properties of the effect of different concentrations on banana blossoms fortification in the processing of shredded tuna. The research was conducted in June-August 2022 at the Fish Product Technology Laboratory and the Chemical Engineering Laboratory, Universitas Brawijaya. The study design was a non-factorial Completely Randomized Design (CRD). The results were analyzed using ANOVA (α 5%) and Tukey's post hoc test. The results showed that banana blossoms fortification had no significant effect on the peroxide value and water content of shredded tuna. Banana blossoms fortification has a significant effect on the hedonic value of the appearance, taste, and texture of shredded tuna. However, it has no significant effect on the hedonic aroma value and overall acceptance. The value of the proximate composition of shredded tuna with the best treatment level was 70%, i.e. protein content of 14.51%, fat content of 36.19%, ash content of 6.82%, carbohydrate content of 29.15%, and total dietary fiber of 20.80%. The fat and protein content of shredded tuna does not meet the Indonesian National Standard (SNI) for shredded products.

Abstract

Ekowisata pantai merupakan salah satu sektor ekonomi yang penting di Kawasan Malang Selatan, salah satunya adalah pantai kondang merak. Tujuan kegiatan pengabdian masyarakat strategis ini adalah untuk meningkatkan amenitas berupa pasar tradisional yang menjadi kebutuhan masyarakat kondang merak, meningkatkan tata Kelola wisata kuliner di Kawasan wisata Pantai Kondang Merak. Kegiatan ini dilaksanakan dengan melibatkan 6 mahasiswa dalam kuliah kerja nyata tematik. Pengambilan data dilakukan dengan pengamatan langsung, survey dan wawancara. Pasar yang dibangun memiliki ukuran 8x5 m, dengan tiang terbuat dari kayu dan atap digunakan sebagai tempat meletakkan solar panel untuk sumber energi terbarukan. Pendataan aspek sosial ekonomi di Pantai Kondang merak menunjukkan adanya beberapa hoestay, warung kuliner dan beberapa potensi wisata yang dapat dikunjungi masyarakat. Pengunjung pantai Kondang Merak lebih banyak ditemukan pada weekend dan hari libur daripada pada hari kerja.

Abstract

Tuna is one of the fish source of nutrition for humans because it contains high-quality protein and omega-3 fatty acids, which are beneficial for health. Tuna can be processed into various products, such as tuna-shredded. But it still has a drawback, i.e., the lower-fiber content. To enrich the fiber of tuna-shredded, fortification with banana blossoms can be developed as functional food such as preventing gout arthritis. The aims of this study were to develop a diversified product of tuna-shredded fortified banana blossoms and to determine the antioxidant activity in vitro and anti-arthritis gout through inhibition of the xanthine oxidase (XO) enzyme in silico. The method used was a simple, completely randomized design. The formulation of tuna-shredded used fortification and active compounds analyzed by LC-HRMS. The antioxidant activity was analyzed by the DPPH. Inhibition of the XO enzyme was analyzed by molecular docking in silico. The results showed that tuna-shredded extract contained 32 compounds, which had total phenolic was 0.00134 mg GAE/g, total flavonoid was 0.0006670 mg QE/g, and IC50 was 4.38 ppm. Ferulic acid had the potential to inhibit the XO enzyme with binding affinity was -9.70 kcal/mol through hydrogen bonds and hydrophobic interactions.

Abstract

Akuaponik memiliki konsep menyatukan akuakultur (budidaya ikan) dengan hidroponik (budidaya tanaman dengan media selain tanah). Sistem akuaponik dapat memperbaiki kualitas air kolam, sehingga dapat meningkatkan kelangsungan hidup dan pertumbuhan ikan yang pada akhirnya dapat meningkatkan produktivitas budidaya ikan. Penelitian ini bertujuan membandingan dua metode akuaponik dalam meningkatkan kualitas air kolam ikan nila secara daily basis. Dua metode tersebut adalah Deep Flow Technique (DFT) dan Dutch Bucket System (DBS). Metode penelitian yang digunakan adalah eksperimental deskriptif dengan percobaan di lapangan, dengan perlakuan pertama (A) DBS, dan perlakuan kedua (B) DFT, dan diulang sebanyak 3 kali. Pengamatan dilakukan selama 30 hari, pada pukul 06:00; 12:00; dan 18:00, yang terdiri dari kualitas air berupa Dissolved Oxygen, pH, suhu, Total Dissolved Solids, kadar amonia, serta kadar nitrat. Kesimpulan dari penelitian ini adalah terbuktinya kolam akuaponik dengan menggunakan sistem DBS memiliki kualitas air lebih baik dalam pengukuran harian, dibanding dengan menggunakan sistem DFT, hal ini disebabkan oleh media tanam yang digunakan oleh sistem DBS menggunakan hidroton yang dapat membantu menjadi filter mekanis. Sistem DBS dalam hal ini memiliki dua kelebihan dalam satu waktu, yaitu sebagai filter biologis (tanaman) dan filter mekanis (hidroton). Sedangkan sistem DFT hanya memiliki kelebihan filter biologis yang membantu menyerap ammonia dan menyediakan nitrat bagi ikan.   Aquaponics has the concept of uniting aquaculture (fish farming) with hydroponics (plant cultivation using media other than soil). The aquaponic system can improve the quality of pond water, thereby increasing the survival and growth of fish which in turn can increase the productivity of fish farming. This study aims to compare two aquaponic methods in improving the water quality of tilapia ponds on a daily basis. The two methods are the Deep Flow Technique (DFT) and the Dutch Bucket System (DBS). The research method used was descriptive experimental with field experiments, with the first treatment (A) DBS, and the second treatment (B) DFT, and repeated 3 times. Observations were made for 30 days, at 06:00; 12:00; and 18:00, which consists of water quality in the form of Dissolved Oxygen, pH, temperature, Total Dissolved Solids, ammonia levels, and nitrate levels. The conclusion of this study is that it is proven that aquaponic ponds using the DBS system have better water quality in daily measurements, compared to using the DFT system, this is due to the planting medium used by the DBS system using hydroton which can help become a mechanical filter. The DBS system in this case has two advantages at one time, namely as a biological filter (plant) and a mechanical filter (hydroton). Meanwhile, the DFT system only has the advantage of a biological filter that helps absorb ammonia and provides nitrate for fish.

Abstract

Mantis shrimp is one of the marine organisms that has a very abundant diversity of species. The similarity of morphology of this group make it difficult to identify morphologically. Therefore, identification was carried out using a DNA barcoding approach. The purpose of this study was to identify the mantis shrimp species from Manokwari waters using molecular techniques. The CO1 gene fragments were amplified by using LCO and HCO primers. The DNA sequences obtained were analyzed for homology, phylogeny, and genetic distance using MEGA X software. A total of 537 base pairs of CO1 gene fragments of mantis shrimp from Manokwari were obtained. The results of the homology analysis on GenBank, show the highest similarity of 91.18% with sequences from the genus Gonodactylus. Analysis of phylogenetic tree and genetic distance showed the same results and supported each other with homology analysis on GenBank data.

Abstract

The high market demand for nilem fish must be balanced with good hatcheries. One of the success factors of hatcheries is the hatchability of fish eggs. Egg hatchability is the percentage of eggs that hatch after a certain time. The hatchability of fish eggs is certainly influenced by several factors, one of which is pH which affects the chorionase enzyme in eggs. The purpose of this study was to determine the effect of different pH in the media on the hatching of nilem fish eggs. The research method used is an experimental method with a complete randomized design (RAL). The mainmain parameters used in this study are embryo development, hatching rate (HR), larval abnormality, and larval survivale (SR) and the supporting parameters used in this study are temperature and DO. The results showed that pH treatment had a significantly different effect on the hatching of nilem fish eggs. The best result of HR in nilem fish was 95% at pH (9) and the lowest result of HR in nilem fish was 15% pH (11). The hatchability relationship of eggs given different treatments formed a quadratic equation. The results showed that pH treatment affects the SR value. The best SR result of nilemfish larvae was 93% at pH (9) and the lowest SR result was 0% at pH (11). The survivale rate relationship of eggs given different treatments formed a quadratic equation.
