Abstract

Efforts to cultivate snakehead fish (Chanidae) still experience many problems, especially the availability of high-quality fry. To improve the quality of fry, improving the quality of broodstock is important. Genetic factors play an important role in the selection and breeding of snakehead fish parents. Therefore, it is necessary to explore sources of superior parents with good genetic characteristics. To achieve this goal requires studies from various fields such as ecology, biology, and genetics. Genetic studies can be started by examining the identity and relationships of snakehead fish from Merauke, as a first step in finding a source of germplasm that has high diversity. The aim of this research is to analyze the cytochrome oxidase subunit 1 (COI) gene fragment of snakehead fish from Merauke, Papua. DNA isolation was carried out using the Promega kit with standard protocols, amplification of the COI gene with primers F2 and R2 with the Go Taq Green amplification kit. A total of 653 base pairs of the COI gene fragment were obtained from the sequencing process, with a nucleotide composition of T (29.9%), C (28.8%), A (24.0%), G (17.3%). BLAST results on the NCBI database show that this sequence has 100% similarity to the Channa striata sequence from Papua with sequence ID OQ405388.1. This shows that the identification results of the snakehead fish from Merauke are Channa striata. Phylogenetic tree analysis also showed consistent results, namely the formation of a clade that was the same as the Channa striata fish group derived from GenBank data.

Abstract

Ikan nilem merupakan ikan air tawar asli Indonesia yang persebaranya hanya di daerah Jawa Barat. Induk betina ikan nilem memiliki jumlah telur yang bervariasi sehingga perlu dilakukan seleksi. Seleksi yang dapat dilakukan pada induk betina ikan nilem yaitu dengan cara melihat karakter morfometrik. Sampai saat ini penelitian tentang ikan nilem khususnya yang berkaitan dengan reproduksi juga masih sangat jarang dilakukan. Tujuan dilaksanakanya penelitian ini adalah untuk menganalisis hubungan dan mengetahui karakter morfometrik yang paling berhubungan terhadap fekunditas. Metode yang digunakan dalam penelitian iniyaitu deskripstif komparatif dengan pendekatan kuantitatif. Parameter yang diamati meliputi panjang standar, tinggi badan, lingkar tubuh, dan bobot tubuh. Kelompok sampel ikan uji diukur berdasarkan panjang standar yang telah ditentukan, yaitu 06,1-09 cm, 09,1-12 cm, 12,1-15 cm, 15,1- 18 cm, 18,1-21 cm. Hasil penelitian menunjukkan bahwa morfometrik berpengaruh terhadap fekunditas. Karakter morfometrik terhadap fekunditas diperoleh hasil tertinggi pada lingkar tubuh dengan fekunditas sebesar 99%terhadap fekunditas yang dihasilkan pada induk betina ikan nilem. Faktor yang dapat mempengaruhi jumlah fekunditas diantaranya yaitu umur ikan, lingkungan, makanan, dan diameter ikan. Diameter ikan sangat berpengaruh terhadap jumlah fekunditas yang di hasilkan, semakin besar diameter telur dalam suatu gonad maka akan semakin sedikit jumlah telur yang dihasilkan.

Abstract

Vannamei shrimp is one of Indonesia's leading aquaculture sector commodities due to its high survival rate, short cultivation time, and disease resistance. Vannamei shrimp cultivation in Indonesia adopts an intensive system favoured for high productivity. In intensive cultivation systems, shrimp ponds typically utilize concrete or high-density polyethylene (HDPE) ponds. Each type has its own positive and negative side that affects the growth and production of Vannamei shrimp. Apart from the type of pond, water quality also played an important role in the growth of Vannamei shrimp. Water quality is one of the important factors in intensive shrimp cultivation. Pond type is categorical data and has a nominal measurement scale, while water quality is numerical data with a ratio measurement scale; ordinary regression analysis is unsuitable for analyzing the effect of pond type and water quality on the growth of Vannamei shrimp. Therefore, a dummy regression model accommodates these measurement scale differences. This research aims to determine the effect of different types of ponds and water quality dynamics on the growth of Vannamei shrimp and to select the best type of pond for intensive cultivation of Vannamei shrimp. Additionally, it aims to identify the water quality parameters that most influence the growth of Vannamei shrimp so that it can serve as a reference for Vannamei shrimp farmers to achieve high productivity efficiently. The study was conducted from November 2022 to January 2023 in Banyuwangi Regency, East Java Province, Indonesia. There are three concrete ponds and 10 HDPE ponds. The research method employed in this study is a survey method utilizing simple random sampling. The parameters in this research are the quality of cultivation water, including pH, temperature and dissolved oxygen (DO). The water samples taken are then measured for water quality in the laboratory. The results of this study indicate that the average values for water quality are still within the optimal range. The average ABW in HDPE and concrete ponds ranges from 2.30 to 9.39 grams per shrimp and 2.22-9.73 grams per shrimp.  Based on the study's results, it can be concluded that different pond types and water quality dynamics, specifically pH and temperature, affect shrimp growth based on ABW. However, when considering SGR values, the type of pond does not have a significant impact. The water quality parameters that affect SGR are pH and temperature.

Abstract

Axolotl (Ambystoma mexicanum) merupakan hewan unik yang populasinya sudah terancam punah. Axolotl merupakan yang unik, salah satu keunikannya adalah dapat meregenerasi hampir seluruh bagian tubuhnya. Di Indonesia sendiri masih sedikit sekali yang membudidayakan hewan satu ini. Di Indonesia kegiatan budidaya axolotl banyak menemui hambatan terutama karena faktor lingkungan yang sangat berbeda dengan habitat aslinya. Salah satu kendala utama untuk membudidayakan axolotl adalah suhu. Untuk membuat lingkungan yang sesuai untuk axolotl tumbuh dan berkembang diperlukan bantuan alat. Salah satu alat yang digunakan adalah ultra recirculate chiller system (URCS). Penelitian ini bertujuan untuk mengetahui pengaruh manipulasi suhu yang berbeda terhadap pertumbuhan axolotl. Penelitian ini menggunakan metode eksperimen dengan penggunaan tiga perlakuan suhu yang berbeda (18°C, 22°C dan 26°C). Parameter utama yang diuji adalah tingkat kelulushidupan (SR), laju pertumbuhan spesifik (SGR) dan rasio konversi pakan (FCR). Hasil penelitian menunjukkan suhu berpengaruh terhadap tingkat kelulushidupan, laju pertumbuhan spesifik dan rasio konversi pakan. Perlakuan terbaik didapat pada perlakuan suhu 18°C dengan SR 100%, SGR 0,49%.

Abstract

Ikan kerapu Epinephelus areolatus adalah salah satu spesies ikan laut dalam genus Epinephelus yang penting secara ekologi, ekonomi, biologi, dan pangan. Genus ini tergolong tinggi tingkat keanekaragamannya. Meskipun demikian, informasi keragaman genetik spesies E. areolatus di bagian selatan bentang laut kepala burung Papua (Bird’s Head Seascape, BHS) masih terbatas. Tujuan penelitian ini adalah menentukan keragaman genetik ikan kerapu E. areolatus baik secara individu, populasi, dan antar populasi di selatan BHS. Untuk mencapai tujuan ini, peneliti telah mengumpulkan sampel ikan dari Fakfak dan Kaimana pada Agustus 2024. Analisis laboratorium dilakukan pada September 2025 menggunakan penanda genetik Cytochrome c oxidase sub unit 1 (COI) DNA mitokondria. Isolasi genom, amplifikasi gen COI, elektroforesis dan sekuensing fragmen gen COI adalah tahapan dalam analisis laboratorium ini. Hasil sekuensing pada 13 individu memperoleh panjang fragmen 665 panjang basa (pb). Hasil blast dan boldsystem terhadap ke-13 sekuens individu memastikan 12 sampel diantaranya adalah E. areolatus. Total teridientifikasi 35 sisi nukleotida yang berbeda antar individu spesies E. areolatus. Total 6 haplotipe yang teridentifikasi pada penelitian ini dengan keragaman haplotipe (Hd) populasi E. areolatus di Kaimana 0,85714 dan keragaman nukleotida (Pi) 0,0193, sedangkan keragaman haplotipe di perairan Fakfak sebesar 0.8333 dan keragaman nukleotida 0,0272. Jarak genetik rata-rata antar individu adalah 0.000 – 0.053. Ikan kerapu Fakfak dan Kaimana memiliki hubungan yang sangat dekat yang ditunjukkan oleh pohon filogenetik dan jaringan haplotipe. Populasi ikan BHS ini juga mengalami pencampuran genetik dengan spesies yang sama asal perairan Pasifik seperti China, Philipina, Canada, dan lainnya.

Abstract

Genetic diversity is one level of biodiversity that is very importan for the resilience of biota in the face of environmental pressures such as cilmater change. However, information on the genetic diversity of red snapper in Kaimana water is still very limited. This study determines the genetic diversity of red snapper (Lutjanus erythropterus) in Kaimana waters, West Papua, to support conservation and sustainable management. The methods used include PCR, DNA sequencing, and phylogenetic analysis using MEGA11, with samples taken from the Kaimana fish market. The results of this study show differences in the percentage of adenine and guanine in sample B81.01, with an average codon composition of 218 codons, where UCU(G) Serine has the highest percentage of 13.8% and RSCU 2.38%, and 20 types of amino acids were detected. Lutjanus erythropterus has 3 haplotypes, with 2 universal haplotypes and 1 specific. In Kaimana waters, the haplotype diversity (hd) value is 0.50000 and the nucleotide diversity (π) is 0.00091, which is influenced by the large population size and high migration ability.

Abstract

The potential of freshwater fisheries in local species such as hampal fish (Hampala macrolepidota) presents several research gaps, particularly in specific localities. Research on H. macrolepidota in East Java may serve as a foundation for future management and development of local species, especially given that H. macrolepidota aquaculture is rarely carried out. Morphological studies based on meristics and morphometrics can be used for broodstock selection in H. macrolepidota domestication efforts. In this study, 21 Hampala specimens were assessed based on morphometrics and meristics, and analysed using a significance test, PCA and dendrogram. The analysis results by PCA and dendrogram showed morphometric differences in the Probolinggo and Banyuwangi populations compared with the four Brantas River populations. In contrast, based on meristics, the entire population does not show apparent differences based on the population. The clear differences demonstrated in the Probolinggo and Banyuwangi populations morphometrically indicate local adaptation due to differences in water conditions.

Abstract

The community service program in Boyolali Regency aims to increase the productivity of the fisheries sector through assistance with laserpuncture technology, the application of microdiet and natural feed, and the production of synbiotics. The methods used include counseling and direct practice involving more than 40 local fish farmers. The results of the program show that the application of laserpuncture and nutritional engineering through microdiet and synbiotics tecknology are increase fish growth. Participation and positive responses from the community show high enthusiasm for new technology which has a direct impact on productivity and the local economy. It is hoped that this program can be implemented independently in sustainable fisheries businesses.

Abstract

Penaeid Shrimp (Crustacea: Decapoda) is an important fishery commodity in human life both in terms of economy, ecology, and nutrition. Penaeid shrimp are widely distributed and abundant on the coast of Merauke and become one of the leading commodities in the fisheries sector. The identification problem in the Penaeid shrimp group is a problem that still often occurs today. Identification problems are often not carried out because they are considered unimportant even though they can be important information for conservation and sustainable management. This study was conducted to identify the Penaeid Shrimp from Payum Beach, Merauke Regency, Papua by using a DNA barcoding approach with the Cytochrome Oxidase I Gene. The results of the analysis showed that the species of specimens was Mierspenaeopsis sculptilis (Heller, 1862) with a similarity level of 97.38%. Genetic distance and level of DNA similarity are very influential in phylogenetic reconstruction. Identification with DNA barcoding has succeeded in identifying species quickly and accurately and can be used to confirm previously identified species based on morphological characters.
