Abstract

The aim of this study was to evaluate the effect of different levels of PGF2α (Estron ™ bioveta) induction on estrus performance and artificial insemination of kacang does. Sixteen parous kacang does (3-4 years) were used in the study. The aniamls were housed in group following a block randomized design of 4 treatments with 4 blocks. The 4 treatments offered were: injection with physiological NaCl (P0); 0.25 mL PGF2α (P1); 0.50 mL PGF2α (P2); and 0.75 mL PGF2α (P3). Variable measured were: estrus intensity, estrus percentage, estrus duration, service per conception, and conception rate. All data collected were descriptively analyzed. Kacang does injected with PGF2α 0.5-0.75 mL resulted in higher estrus (100%) for P2 and P3, respectively than those injected with 0, 25 mL (P1). The higher estrus intensity was recorded for does had treatment P1 and P2 (score 3) than those treated with P0 and P1 (score 1-2). Goats injected P2 and P3 had longer estrus duration (38.75 - 45.50 hours) than P1 and P0 (32.67- 37.33 hours). Furthermore, the higher conception rate were recorded for P1 (66.67% S/C 1.33), followed by P0 (33.33% S/C 1.67), P2 (0.75 S/C 1.75), and P3 (25% S/C 1.75), respectively. Therefore, it can be concluded that does injected with 0.25 mL PGF2α (Estron ™, Bioveta) had highest conception rate. ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh induksi PGF2α dengan berbagai dosis yang berbeda terhadap tampilan estrus dan tingkat keberhasilan inseminasi buatan (IB) kambing kacang. Penelitian ini menggunakan kambing kacang betina sebanyak 16 ekor dengan umur 3-4 tahun. Ternak dikandangkan secara koloni mengikuti pola rancangan acak kelompok (RAK) dengan 4 perlakuan dan 4 ulangan. Perlakuan yang dicobakan adalah injeksi menggunakan NaCl fisiologis (P0), 0,25 mL PGF2α (P1), 0,50 mL PGF2α (P2), dan 0,75 mL PGF2α (P3). Parameter yang diukur adalah intensitas estrus, persentase estrus, lama estrus, service per conception, dan persentase kebuntingan. Hasil penelitian menunjukkan bahwa injeksi PGF2α sebanyak 0,5 mL (P2) dan 0,75 mL (P3) pada kedua perlakuan menghasilkan persentase estrus 100%, lebih tinggi daripada yang dinjeksi 0,25 mL (P1) hanya 75%. Tampilan intensitas estrus lebih tinggi pada perlakuan P1 dan P2 dengan skor 3, sedangkan P0 dan P1 hanya menunjukkan intensitas estrus dengan skor 1-2. Demikian juga dengan lama estrus P2 dan P3 mencapai 38,75 – 45,50 jam pada perlakuan, lebih lama daripada P1 dan P0 dengan lama estrus 32,67 – 37,33 jam. Angka kebuntingan tertinggi ditunjukkan pada perlakuan P1 66,67% dan 1,33 untuk nilai services per conception diikuti dengan P0 yaitu 33,33% dan 1,67; dan angka kebuntingan terendah terdapat pada perlakuan P2 dan P3 dan 0,75 yaitu 25% dan 1,75. Dari hasil penelitian ini dapat disimpulkan bahwa injeksi PGF2α ( Estron™, Bioveta ) sebanyak 0,25 mL menghasilkan angka kebuntingan tertinggi

Abstract

The purpose of this study was to determine the efect of extra virgin olive oil on the quality of duroc pig spermatozoa during preservation. Semen was collected twice a week using the golve hand method of two 3 and 4 years old male duroc boar wiht healthy body and reproductive organs. Good quality semen was diluted with citrate-egg yolk (C-EY) supplemented with extra virgin olive oil (EVOO) at various consentration: 0. 6. 8. 10 and 12 %. Diluted semen is stored at 18-20°C and sperm quality was evaluated every 8 hours. The resulsts showed that the duroc spermatozoa in CEY diluents supplemented with EVOO 12% (P4) showed higher quality (P <0,05) compared to others treatment. with motility 41.50±2.24%. viability 47.53±1.59%. abnormalities 5.27±0.19% and survival 65.6±0.44 hours. The conclusion of this study is that 12% extra virgin olive oil in citric-yolk diluents is more effective in maintaining the quality of liquid sperm duroc pigs. ABSTRAK Tujuan dari penelitian ini adalah untuk menguji pengaruh penambahan minyak zaitun extra virgin terhadap kualitas spermatozoa babi duroc selama preservasi. Semen ditampung dua kali seminggu menggunakan metode glove hand dari dua ekor babi duroc jantan berumur 3 dan 4 tahun dengan kondisi tubuh dan organ reproduksi yang sehat. Semen yang berkualitas baik diencerkan dengan sitrat-kuning telur (S-KT) yang disuplementasi dengan minyak zaitun extra virgin (MZEV) pada konsentrasi: 0,6,8,10 dan 12%. Semen yang telah diencerkan disimpan pada suhu 18-20 ᶱ C dan kualitas spermatozoa dievaluasi setiap 8 jam. Hasil penelitian menunjukkan bahwa spermatozoa yang dipreservasi dalam SKT yang disuplementasi oleh (SKT-MZ12) mempunyai kualitas yang lebih tinggi (P<0,05) dibandingkan dengan keempat perlakuan lainnya. Kualitas spermatozoa yang dipreservasi dalam pengencer SKT+MZEV 12% dan disimpan selama 64 jam menghasilkan motilitas 41,50±2,24%, viabilitas 47,53±1,59%, abnormalitas 5,27±0,19% dan daya tahan hidup 65,6±0,44 jam. Kesimpulan dari penelitian ini adalah penambahan minyak zaitun extra virgin 12% dalam pengencer sitrat- kuning telur lebih efektif dalam mempertahankan kualitas sperma cair babi duroc.

Abstract

The purpose of this study was to determine the effect of giving pituitary extract (EH) and probiotics ABG-O on piglets crossing to increase weaning weight, linear size of body and blood metabolites. The research material was a crossbred of landrace and duroc aged 2 weeks, as many as 24 birds. The experimental design was a completely randomized design with three treatments and eight replications. The three treatments are P0 (control), P1 (EH injection 1 mL), and P2 (injection EH 1 mL + probiotic ABG-O 2 mL). EH injections are carried out every 5 days and administration of probiotics is given orally using spoit every day until the piglets are weaned. The parameters observed were weight gain, linear size of body and blood metabolites. This data was analyzed by ANOVA and continued with Duncan test. The results of this study indicate that the average daily weight gain of crossbred piglets in treatments P0, P1, and P2 are 0.11; 0.13; 016 kg, body length (0.40; 0.45; 0.44 cm), chest circumference (0.29; 0.27; 0,30 cm), and shoulder height (0.25; 0.24; 0.25 cm). Blood glucose levels (117.31; 112.85; 117.25) and total plasma protein (4.60; 4.90; 4.80). The results of the statistical analysis showed that the EH and ABG probiotic treatments affected the UN (P<0.05) but did not affect (P> 0.05) the size of the linear body and blood metabolites. It can be concluded that the administration of pituitary extracts and ABG probiotics can increase the weaning weight but does not affect the linear size of the body blood metabolites and total protein plasma of crossing piglets.

Abstract

The purpose of this study was to examine the effect of adding various doses of virgin coconut oil to the quality of bali bull spermatozoa during preservation. Semen is accommodated twice a week using an artificial vaginal method from two 3-4 year old bali bull with normal body condition and reproductive organs. Semen with motility> 70%, concentration> 1000 × 106, and abnormality <15% was diluted with Tris-egg yolk (T-KT) diluent added with virgin coconut oil (VCO) at a concentration of: 0% (T0), 2 % (T1), 4% (T2), and 6% (T3). The diluted semen was stored at 3-5ᶱC. Evaluated every 24 hours for motility, viability, abnormalities and vitality of spermatozoa up to a motility of at least 40%. The results showed that semen diluted with T-EY with 6% VCO (T3) had a higher quality (P <0.05) compared to the other three treatments, with motility reaching (43.10 ± 2 , 83%), viability (48.68 ± 1.59%), abnormalities (5.92 ± 0.90%) and survival (6.00 ± 0.00 days). The conclusion of this study is the addition of 6% virgin coconut oil in Tris-egg yolk diluent is more effective in maintaining the quality of bali cattle liquid semen.

Abstract

This study aims to determine the effect of adding various doses of carrot juice into citrate-egg yolk diluents (C-EY) on bligon buck semen quality. Semen was collected twice a week using an artificial vaginal method of three buck with normal body condition and reproductive organs. Semen was 70% of motility, konsentration ≥1000x106mL and abnormality ≤ 15 diluted with C-EY added with carrot juice at a concentrations: 0% (CJ-0), 10% (CJ-10), 12.5% (CJ-12,5), 15% (CJ-15), 17.5% (CJ-17,5), and 20% (CJ-20), and stored in a refrigerator at 3-5o C. The quality of spermatozoa was evaluated every 24 hours. The Variables measured were spermatozoa motility, viability, and abnormalities. The results showed that spermatozoa preserved by C-EY supplemented by 17.5% carrot juice (CJ-17.5) had a higher quality (P<0.05) compared to the other four treatments, with motility (49.04 ± 1.0%), viability (55.70 ± 1.22%), dan abnormalities (4.57 ± 0.88 %) on the 4th day of storage. This study concludes that 17.5% carrot juice into citrate eggyolk extender improves the bligon buck semen

Abstract

The purpose of this study was to examine the effect of adding various concentrations of dragon fruit peel extract (DFPE) in Tris-egg yolk diluent (TEY) on the quality of bligon goat spermatozoa. Semen was collected twice a week using the artificial vaginal method from three year old bligon male goats with normal body conditions and reproductive organs. After the macroscopic and microscopic evaluation, good quality semen with a sperm motility of > 70%, a concentration of > 800 x 106 mL-1, and abnormality of < 15 %, was diluted with TEY + DFPE at a concentration of 0% (P0), 2% (P1), 4% (P2), 6% (P3), 8% (P4), dan 10% (P5). The semen which still had a motility ≥ 70% was then stored at a temperature of 3-5oC. Evaluation of the quality of spermatozoa was carried out every 24 hours until the motility decrease to 40%. The results showed that from day 1 to day 5 of preservation, spermatozoa preserved in TEY + 6% DFPE diluent (P3) had higher motility and viability (P<0,05) than other treatments. The abnormalities of spermatozoa until the 5 day of preservation were below 6%, but showed a significant difference (P<0,05) between treatments that received DFPE suplemention with control (P0). It was concluded that the addition of DFPE in TEY up to 6% could reduce the decrease in sperm quality of bligon goats during in vitro preservation.

Abstract

The purpose of this study was to determine the effect of adding onion extract (OE) to Tris-egg yolk (T-EY) diluent on the quality of bali bulls spermatozoa. The semen of three bali bulls aged 3-4 years were collected twice a week using the artificial vaginal method. Semen with good quality, motility 79%, viability 84.63%, abnormality 2.93% diluted with T-EY diluent with the addition of OE: T0: 0%; T1 1%; T2: 2%; T3: 3%; T4: 4%; and T5: 5%. The diluted semen was preserved in a refrigerator at 3-5 °C and evaluated for the quality of the spermatozoa every 24 hours. The results showed that the addition of OE in T-EY diluent had a positive effect on the quality of bali bulls spermatozoa. The addition 3% of OE (T3) in T-EY diluent resulted in higher spermatozoa quality (P <0.05) than the other treatments, namely motility 42.01%, viability 48.74%, and abnormalities 4.13% on the sixth day of preservation. It was concluded that the addition of onion extract in Tris - egg yolk diluents could improve the quality of bali bulls spermatozoa, with the best level of onion extract being 3%.

Abstract

This study aims to determine the effect of equilibration time on the quality of landrace spermatozoa both pre- and post-freezing and to find the best equilibration time. The materials used is fresh semen collected from a 2,5 years old landrace boar. Collecting was carried out twice per week using the glove hand method. Semen of good quality (sperm motility ≥70%, sperm concentration ≥200 x 106 cells/ml, and percentage of sperm abnormalities ≤15%) was diluted with durasperm that had been modified with lontar fruit juice, while the cryoprotectants used glycerol and sucrose. Holding time was carried out for 2 hour at 27-28°C, centrifuged at 2000 rpm for 15 minutes, and the sperm sediment was diluted with the same diluent. After that, the liquid semen was packed in a 0.5 mL straw and equilibrated at 3-5°C for 1 (P1), 2 (P2), or 3 hours (P3). The freezing of semen was carried out on the surface of liquid nitrogen at a distance of 5 cm for 10 minutes, and then stored in a liquid nitrogen container. Thawing is done by placing the frozen semen straws in warm water (37°C) for 30 seconds. The research data were analyzed with analysis of variance and continued with the Duncan test. The results showed that the equilibration time at 2 hours (P2) gave the best results (P<0,05) with the mean percentage of motility, viability, abnormality, intact plasma membrane and recovery rate respectively were 30,62%, 58,21%, 4,68%, 58,94% and 43,74%. It was concluded that the equilibration time did not affect the quality of pre-freezing spermatozoa, but it did affect the quality of post-freezing spermatozoa with the best equilibration time to maintain the quality of frozen semen of landrace boar was 2 hours.

Abstract

A study was carried out in one of Indonesia native cattle, Sumba Ongole (SO) Cattle for determination of statuses of various biochemical parameters, aspartate aminotransferase (AST or SGOT) and alanine aminotransferase (ALT or SGPT). Apparently blood from randomly selected cattle (n= 36) were collected aseptically. Biochemical analysis of AST and ALT was performed on UPT. Laboratorium Kesehatan, Kupang, NTT. The results showed that the variations of parameters were not significant (p>0.05) between male and female SO cattle. The AST/SGOT (142.56±20.55 U/l) was slightly higher in female compared with the male (137.00±16.24 U/l), while level of ALT/SGPT were 46.44±10.99 U/l in male and 46.41 ± 10.23 U/l in female. Although, the result was not significant (p>0.05), level of AST and ALT in SO cattle was higher compared to standard which is 60-125 U/l for AST and 6.9-35 U/l for ALT. Results showed possibility of seasonal effect on AST/ SGOT and ALT/SGPT level in male and female SO cattle.

Abstract

This study aims to determine the effect of adding watermelon juice (WJ) into citrate-egg yolk (C-EY) diluents on the spermatozoa quality of bali catlle. Semen was collected twice a week using an artificial vaginal method of three bali bulls with normal body condition and reproductive organs. Semen that has motility > 70%, concentration >1000 x 106 and abnormalities ≤ 20 % was used in this study. Semen then divided into five treatments: C-EY 100% (T0), C-EY-25% WJ (T1), C-EY-50% WJ (T2), C-EY-75% WJ (T3), WJ 100% (T4) and then stored in a refrigerator at temperature 3-5oC. The quality of spermatozoa was evaluated every 24 hours for sperm motility, viability, and abnormalities. The results showed that spermatozoa diluted with S-KT supplemented with 75% WJ (T3) had a higher quality (P<0,05) compared to the other treatments, with motility 40,53 %, viability 47,35%, and abnormalities 4,21% on the 6th day of preservation. It was concluded that the addition of WJ to C-EY diluent was able to maintain the quality of bali bull spermatozoa, with the best SBS concentration is 75%.
