Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak hipofisa sapi terhadap pertambahan bobot badan dan umur pubertas mencit betina. Materi penelitian adalah mencit betina strain swiss webster yang berumur 21 hari, sebanyak 48 ekor dengan kisaran berat badan (BB) 11-13 gram (±12 gram). Rancangan percobaan adalah rancangan acak lengkap dengan 4 perlakuan dan 12 ulangan yaitu penyuntikan ekstrak hipofisis dengan dosis 0 mg/g BB (P0, kontrol); 0,05 mg/g BB (P1); 0,10 mg/g BB (P2); 0,15 mg/g BB (P3). Penyuntikan dilakukan dua kali seminggu sejak umur lepas sapih hingga pubertas, secara intraperitoneal. Data pertambahan bobot badan dan umur pubertas dianalis dengan ANOVA dan dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan bahwa perlakuan ekstrak hipofisa berpengaruh nyata (P<0,05) terhadap PBB dan umur pubertas mencit betina. Dari hasil penelitian dapat disimpulkan bahwa penyuntikan esktrak hipofisa sapi dapat meningkatkan pertambahan bobot badan dan mempercepat umur pubertas mencit betina, dengan dosis ekstrak hipofisa terbaik adalah 0,15 mg/g bobot badan.

Abstract

Determination of estrous cycle especially the length of estrous is an important role in improvingthe fertility and reproductive performance of farm animals. This study was aimed to observe thelength of estrous cycle and estrous period based on cytologic and estrogen (E2) and progesterone (P4)hormone profiles. Samples of vaginal smears were collected daily and stained with Giemsa. Bloodsamples were collected every two days using jugular vein implant catheter. The hormone profileswere analized by radio immuno assay. All data were analyzed descriptively. Results of this studyshowed that there were four lowest points percentage of parabasal epithelial cells with the averageof 14.25%, while the superficial epithelial cells showed four highest points (85.75%). The intervalsbetween lowest point of parabasal epithelial cell and the highest point of superficial cells were at 20,18, and 16 with the average of 18 days. This phenomenon showed a cyclical processed with almostthe same length of time of the actual an estrous cycle pattern in timor deer. The range of E2 concentration was 7.06 to 18.14 pg/ml and P4 concentration was 2.58 to 7.48 ng/ml. The intervaltime between the peak of E2 was 17 days which represented the estrous cycle of timor deer. It isconcluded that analysis of estrous cycle in timor deer can be detected by vaginal cytological andhormone analysis profiles.

Abstract

The objective of this experiment was to examine the fertility of timor deer (Rusa timorensis) frozensemen by artificial insemination (AI) with intracervical technique after estrus synchronization withControlled Internal Drug Release, for Goat (CIDR-G®). Six adult, healthy, and cycling hinds aged 3 to 4years were used in this experiment. The percentage of estrus hind, onset and duration of estrus wereobserved. Pregnancy diagnosis was conducted using ultrasound scanner (USG) at day 120 afterinsemination. The response of estrus was 82.22 %, the onset of estrus was 25.33 hours after CIDR®withdrawal with the length of estrus was 28 hours. The pregnancy rate at first AI trial under sedation was16.7% (1/6), at second and third without sedation was 60% (3/5) and 100% (2/2) respectively. The durationof pregnancy was 248 to 285 days; with the average calving rate was 50%. The result concludes thatinsemination in hind without sedation was better than with sedation.

Abstract

Penelitian ini bertujuan untuk menguji efek pemberian ekstrak pituitari (EP) terhadap produktivitas induk sapi bali bunting. Sebanyak 12 ekor sapi bali bunting 2,5 bulan dibagi ke dalam empat perlakuan yaitu: tanpa injeksi EP (P0, kontrol), injeksi EP satu kali pada umur kebuntingan 2,5 bulan (P1), injeksi EP dua kali pada umur kebuntingan 2,5 dan 5,0 bulan (P2), dan injeksi EP tiga kali pada umur kebuntingan 2,5, 5,0 dan 7,5 bulan (P3). Dosis EP yang diberikan adalah 20 mg/kg bobot badan. Parameter yang diamati adalah jumlah dan morfologi corpus luteum (CL), bobot lahir anak, dan produksi susu induk. Data yang diperoleh dianalisis dengan uji sidik ragam dan dilanjutkan dengan Uji jarak berganda Duncan. Hasil penelitian menunjukkan bahwa injeksi EP tidak memengaruhi (P>0,05) jumlah dan morfologi CL. Bobot lahir anak (19,87 kg) dan produksi susu induk harian (1.707,18 mL) tertinggi (P<0,05) dihasilkan oleh sapi bali yang diinjeksi EP tiga kali. Penelitian ini menyimpulkan bahwa penyuntikan EP tiga kali efektif meningkatkan bobot lahir anak dan produksi susu induk sapi bali.

Abstract

Dimethyl Sulfoxide (DMSO) is one type of cryoprotectant which has a low molecular weight so that it is easier to enter cells when cryopreservation. The purpose of this study was to explore the optimal concentration of DMSO in modified coconut water (mCW) extender that were able to maintain frozen sperm quality of bali bulls. Semen was collected from two four-year old bali bulls by artificial vagina. Good quality semen diluted with mCW (young coconut water + 20% egg yolk + 7.5 % moringa leaf extract) and supplemented by 3, 5, or 7% DMSO. Semen was filled into 0.25 ml ministraw, and was incubated in a refrigerator at 5°C for four hours, frozen on the surface of liquid nitrogen for 10 minutes and then dipped into liquid nitrogen. The quality of post thawing sperm was measured 24 hours later by placing the ministraw of frozen semen into water at 37oC for 30 seconds. Data were analyzed by analysis of variance and continued with Duncan test. Postthawing observations showed that bali bulls sperm cryopreserved at 3% DMSO yielded higher motility and viability (p<0.05) i.e. 36 and 44.15%, than DMSO 5% i.e. 18 and 23.65%, and DMSO 7% i.e. 7 and 12.62%. The recovery rate of sperm cryopreserved at 3% DMSO was also higher (p<0.05) than DMSO 5 and 7%, successively 45.65, 23.06, and 8.86%. The results of this study concluded that the optimal concentration of DMSO in mCW diluent to maintain frozen sperm quality of bali bulls was 3%.

Abstract

The aim of the research was to study trehalosa supplementation in Beltsville Thawing Solution (BTS) and MIII extender on the quality of frozen boar semen. Semen was collected from nine boars age 5-6 years using glove hand method. The semen were then evaluated macro and microscopically. Semen having >70% sperm motility and >200x106/mL sperm concentration were diluted in two tubes containing BTS and two tubes containing MIII, then kept at room temperature (20-22oC) for 2 hours (holding time). Following this the semen were centrifugated at 2000 RPM for 10 minutes and the supernatant was discarded, the pellet was re-diluted with BTS, BTS-Trehalose 100 mM (BTS-T), MIII, and MIII-Trehalose 100 mM (MIII-T) with 4% glycerol, packed into 5 mL macrotube, equilibrate at 5oC for 2 hours. After equilibration macrotube were frozen horizontally 4 cm above the liquid nitrogen vapor for 20 minutes and then submerged in liquid nitrogen (-196oC) for futher evaluation. After 24 hours the semen than thawed at 52oC for 45 seconds. The results demonstrated that sperm motility in BTS-T (30.00±4.47%) was higher (P<0.05) then MIII-T (26.67±8.16%), BTS (24.17±9.17%), or MIII (21.67±7.53%). This research conclude that suplementation of 100 mM trehalosein BTS extender improved motility of boar sperm after freezing and thawing. ABSTRAK Penelitian ini bertujuan untuk mempelajari penambahan trehalosa dalam pengencer Beltsville Thawing Solution (BTS) dan MIII terhadap kualitas semen beku babi. Semen dikoleksi dari sembilan ekor babi berumur 5-6 tahun menggunakan teknik glove hand method. Semen dievaluasi secara makro dan mikroskopis. Semen yang memiliki spermatozoa motil di atas 70% dengan konsentrasi di atas 200x106/mL dibagi ke dalam empat tabung, dua tabung pertama diencerkan dengan BTS dan dua tabung lainnya dengan MIII. Semen yang telah diencerkan disimpan pada suhu ruangan (20-22oC) selama dua jam (holding time). Semen kemudian disentrifugasi 2000 rpm selama 10 menit. Supernatan dibuang dan pellet diencerkan kembali. Semen yang disentrifugasi dengan BTS, diencerkan dengan BTS gliserol (BTSg) dan BTS gliserol Trehalosa 100 mM (BTSgT) dan yang disentrifugasi dengan MIII diencerkan dengan MIIIg dan MIIIgT. Gliserol yang digunakan adalah 4%. Semen yang telah diencerkan dikemas dalam macrotube (5 mL), diekuilibrasi pada 5oC selama dua jam. Setelah equilibrasi macrotube dibekukan 4 cm di atas permukaan nitrogen cair selama 20 menit dan disimpan dalam container nitrogen cair (-196oC) untuk pengujian lebih lanjut. Setelah penyimpanan 24 jam, terhadap semen beku dilakukan thawing pada suhu 52oC selama 45 detik. Hasil penelitian menunjukkan motilitas spermatozoa dalam BTSgT (30,00±4,47%) paling tinggi (p<0,05) daripada MIIIgT (26,67±8,16%), BTSg (24,17±9,17%), atau MIIIg (21,67±7,53%). Simpulan dari penelitian ini adalah penambahan 100 mM trehalosa dalam pengencer BTSg dapat mempertahankan motilitas spermatozoa babi lebih baik daripada pengencer lainnya.

Abstract

The aim of study was to find out the best concentration and exposure time of calcimycin andionomycin in order to produce parthenogenetic embryos. Female Swiss Webster mice were fisrtlyprimed with Pregnant Mare’s Serum Gonadotropin (PMSG) and Human Chorionic Gonadotropin (hCG)with an interval of 48 hours. Sixteen hours after injection of hCG oocyte was collected by Dulbecco’sPhosphate Buffer Saline (dPBS) as a flushing medium. To separate the eggs from cumulus cells wereused hyaluronidase enzyme. The good quality oocytes were incubated in activation medium that isionomycin or calcimycin with a concentration of 3, 6, or 9 ìM and exposure time 1, 4, or 7 minutes. Toyield diploid embryos were used 5 ?g/ml cytochlasin B for four hours at 37°C, 5% CO2. Activatedoocytes characterized by the formation of pronuclei washed three times in Potassium SimplexOptimization Medium (KSOM) and subsequently cultured in the same medium until blastocyst stage.The results showed that oocytes activated at calcimycin, the best results was presented at concentration6 ?M and exposure time four minutes, i.e. activation rate reached 96%, cleavage rate 82% and blastocystrate 28%. On the other hand, oocytes activated in ionomycin, the best results was presented atconcentration 3 ?M and exposure time four minutes, i.e. activation rate reached 82%, cleavage rate64% and blastocyst rate of 4%. It was concluded that the best concentration and exposure timecalcimycin on mice oocytes were 6 ?M for four minutes, whereas ionomycin were 3 ?M for four minutes.

Abstract

The purpose of this research was to examine the effect of lactose in Tris extender on theviability of Etawah crossbreed goat spermatozoa which is preserved in the plasma using Prianganram seminal plasma at 3–5oC. Semen was collected using artificial vagina once a week. Freshsemen was divided into four tubes, and centrifuged at 3,000 RPM for 30 min. Supernatant wasremoved and replaced which an equal volume with Priangan ram seminal plasma. Semen in firsttube was diluted with Tris extender-20% egg yolk without lactose (control). Semen in second,third, and fourth tubes were diluted with Tris extender-20% egg yolk and 0.3% (0.3 g per 100 mlextender) (L0.3), 0.6% (L0.6), and 1.2% (L1.2) lactose, respectively. Diluted-semen were stored inrefrigerator at 3–5oC. Quality of diluted-semen including percentages of motile spermatozoa (MS),live spermatozoa (LS), and intact plasma membrane (IPM) were evaluated daily during storage at3–5oC for four days. The data were analyzed by usi analysis of variance (ANOVA). Means werecompared significant difference test at 0.05 significant level. Results of this study showed thatmean volume, colour, consistency, pH, mass activity, spermatozoa concentration, MS, LS,spermatozoa abnormal, and IPM of Etawah crossbreed goat fresh semen were 0.68 ml, cream,thick, 7, ++/+++, 4,148.57 million cell/ml, 70%, 83.89%, 7.12%, and 84%, respectively. Addition oflactose in Tris extender could maintain the quality of semen during preservation. At day-5 ofstorage, the percentages of MS, LS, and IPM for treatment L0.6 (40, 55.6, and 53.8%) weresignificantly (P<0.05) higher than treatment L1.2 (36, 50.2, and 51.8%), treatment L0.3 (34, 51.2,and 48.4%), and control (29, 41.4, and 41.8%). In conclusion, addition of 0.6% lactose in Trisextender is the best dose in maintaining the quality of Etawah crossbreed goat spermatozoa, andcould be maintaining percentage of motile spermatozoa 40% for four days after preserved at 3–5oC.

Abstract

This study aimed to evaluate study the influence of cattle hypophysis extract (HE) on the reproductive performance of post partum sows. Twenty landrace sows, 45 days after partum were divided into four groups, five sows each. All were injected with HE intramuscularly. Group I (P0) was injected with 3 ml of saline solution, Group II (P1) was injected with 3 ml contain of 10 mg/EH/Kg body weight (BW), Group III (P2) was injected with 3 ml contain of 20 mg/EH/Kg BW, Group IV (P3) was injected with 3 ml contain of 40 mg/EH/Kg BW. Every sows that showed the estrus symptom were inseminated artificially on the second days, twice each in the morning and evening. The intensity of estrus, length of estrus, number of service preconception (S/C), conception rate (CR), litter size, weight of piglet and milk production were recorded. The result showed that the score of estrus intensity in Group I, II, III and IV were 1.40; 2.40; 2.80 and 2.80 respectively with the length of estrus in each group were 4.40; 5.00; 5.00; 5.60 days respectively. The number of service in Group I, II, III and IV of each pregnancy were 4.40; 1.40; 1.00 and 1.20 with the CR were 0; 60; 100; and 80% respectively. The litter size produced of Group I, II, III and IV were 4.00; 6.00; 10.60; 8.40 respectively. The average of piglet body weight of Group I, II, III and IV were 1.20; 1.36; 1.56 and 1.35 kg respectively. The milk production of each sows of Group I, II, III and IV were 13; 41; 93; 88 g respectively. It can be concluded that cattle hypophysis extract improves the performance of post partum sows with the best level was 20 mg EH/kg of body weight. ABSTRAK Penelitian ini bertujuan mempelajari pengaruh pemberian ekstrak hipofisis terhadap performans reproduksi induk babi betina post partum. Penelitian ini menggunakan 20 ekor induk babi betina jenis landrace, dibagi kedalam empat kelompok perlakuan masing-masing terdiri atas lima ekor induk babi. Babi betina yang baru 45 hari menyapih anaknya diinjeksi dengan ekstrak hipofisis (EH) secara intramuskuler. Kelompok I (P0) diinjeksi dengan 3 mL NaCl fisiologis; Kelompok II (P1) diinjeksi dengan 3 mL NaCL mengandung 10 mg EH/kg bobot badan (BB); Kelompok III (P2) diinjeksi dengan 3 mL NaCl mengandung 20 mg EH/kg BB: Kelompok IV (P3) diinjeksi dengan 3 mL NaCl mengandung 20 mg EH/kg BB. Setiap induk babi yang menunjukkan gejala estrus diinseminasi pada hari kedua dengan interval dua kali, pagi dan sore. Peubah yang diamati adalah intensitas estrus, lama estrus, jumlah perkawinan per kebuntingan (S/C), angka konsepsi (conception rate/CR), litter size, bobot lahir anak, dan produksi susu. Hasil penelitian menunjukkan skor intensitas estrus kelompok I, II, III, dan IV masing-masing 1,40; 2,40; 2,80; 2,80 dengan lama estrus masing-masing kelompok secara berturut-turut 4,40; 5,00; 5,00; 5,60 hari. Nilai S/C masing-masing adalah 4,40; 1,40; 1,00; 1,20 dengan CR masing-masing 0; 60; 100; dan 80%. Total jumlah anak yang dihasilkan kelompok I, II, III, dan IV masing-masing adalah 4,00; 6,00; 10,60; dan 8,40 ekor. Rataan bobot lahir anak per ekor pada kelompok I; II; III; dan IV masing-masing adalah 1,20; 1,36; 1,56; dan 1,35 kg. Produksi susu per induk pada kelompok I, II, III, dan IV masingmasing adalah 13; 41; 93, dan 88 g. Simpulan dari penelitian ini adalah ekstrak hipofisis dapat meningkatkan performans induk babi post partum. Dosis terbaik adalah 20 mg EH/kg BB.

Abstract

Penelitian ini bertujuan untuk menguji efektivitas ekstrak hipofisa sapi (EHS) terhadap produktivitas ayam petelurtipehisex brown periode afkir. Penelitian ini menggunakan 60 ekor ayam petelur tipe hisex brown yang berumur 88 minggu dengan berat badan 2,01 ± 8,30 kg. Ayam-ayam tersebut diacak dengan rancangan acak lengkap ke dalam lima grup dan diinjeksi EHS dengan konsentrasi:  0; 7,5; 15; 22,5 atau 30 mg/kg bobot badan. Injeksi dilakukan secara intramuskuler pada otot dada dengan frekuensi satu kali per minggu. Masing-masing perlakuan diulang empat kali, dengan tiga ekor ayam pada setiap ulangan. Data penelitian dianalisis dengan sidik ragam dan dilanjutkan denganuji Duncan. Hasilpenelitian menunjukkan bahwa injeksi EHS tidak mampu meningkatkan produksi telur, konversi pakan, indeks telur, indeks putih telur, indeks kuning telur, haugh unit, dan tebal kerabang (p>0,05), menurunkan bobot telur dan konsumsi pakan (p<0,05), dan meningkatkan indeks warna kuning telur (p<0,05). Disimpulkan bahwa injeksi EHS tidak efektif untuk meningkatkan produktivitas ayam petelur tipe hisex brown periode afkir.Kata kunci :ekstrak hipofisa sapi, hisex brown, produktivitas, period eafkir
