Abstract

Indonesia is one of the world’s most biodiverse countries, including in medicinal plants that contain active compounds with potential as sources of modern medicine, particularly for treating bacterial infections. The use of herbal plants as antibacterial agents offers an alternative to overcome antibiotic resistance and the side effects of synthetic drugs. Bay leaf (Syzygium polyanthum) and nanochitosan are known for their antibacterial properties due to their bioactive compounds. However, studies on their combined effects are still limited. This study aimed to evaluate the antibacterial effectiveness of combinations of bay leaf extract (15%, 25%, and 35%) and nanochitosan (0.5% and 1%) against Escherichia coli and Bacillus subtilis. A Completely Randomized Design (CRD) was applied, using sterile distilled water as a negative control and 0.1% chloramphenicol as a positive control. The antibacterial activity test was performed using the Kirby-Bauer disk diffusion method. Results showed the positive control produced inhibition zones of 25.45 ± 1.27 mm for E. coli and 25.05 ± 0.39 mm for B. subtilis. The most effective combination was K3 (35% bay leaf extract and 0.5% nanochitosan), which produced inhibition zones of 12.65 ± 0.52 mm and 12.91 ± 0.55 mm, respectively. Statistical analysis using the Tukey HSD post hoc test confirmed significant differences among treatments. These findings suggest that the combination of bay leaf extract and nanochitosan holds promise as an alternative antibacterial agent

Abstract

The spike protein of SARS-CoV-2 is crucial for initiating infections by binding to host cells and mediating membrane fusion. In this study, HEK-293T cells were transfected with plasmids encoding three structural proteins of SARS-CoV-2, i.e., Spike (S), Membrane (M), and Envelope (E). This transfection enabled the formation of SARS-CoV-2 Virus-Like Particles (VLPs), which allows for safer studies of the virus and its proteins. To serve as a marker for expression, an enhanced green fluorescent protein (EGFP) was fused to the spike protein, resulting in a Spike-EGFP (S-EGFP) fusion protein. The characteristics of SARS-CoV-2 spike protein expression in the transfected HEK-293T cells were then investigated using polymerase chain reaction (PCR) and flow cytometry. The PCR analysis revealed non-specific DNA band smearing, which did not provide conclusive confirmation of spike protein expression. However, flow cytometry analysis demonstrated that approximately 30% of the transfected cells exhibited green fluorescence, indicating the expression of the Spike-EGFP fusion protein. These findings, obtained through flow cytometry, confirmed the successful spike protein expression in transfected HEK-293T cells.

Abstract

This study exakines the role of the COMT Val158Met genetic polymorphism in variations of children's cognition and learning character through a systematic literature review. This approach aims to understand how internal biological factors influence differences in children's learning styles and academic performance, as well as to highlight the importance of the interaction between genetic factors and the learning environment (gene x environment interaction). Through the search and analysis of relevant scientific articles from various international and national databases, this study found that genetic variations in COMT affect dopamine activity and neurotransmitter mechanisms, impacting executive function, emotion regulation, and responses to academic stress. The research findings conclude that understanding molecular genetics in the context of education can support the development of more personalized and adaptive learning approaches and support the implementation of neurodiversity-based educational strategies. This study also indicates the need for further research on the direct relationship between COMT polymorphism and learning characteristics in Indonesia, which is still limited at present.

Abstract

Kappaphycus alvarezii merupakan salah satu rumput laut strategis dalam industri marikultur Indonesia karena perannya sebagai sumber utama karagenan. Namun, tantangan seperti pencemaran, perubahan iklim, dan praktik budidaya yang intensif telah mengancam keberlanjutan dan keragaman genetik spesies ini. Kajian ini bertujuan untuk menganalisis secara sistematis pemanfaatan DNA barcoding sebagai pilar pengembangan bank genetik K. alvarezii di Indonesia. Metode yang digunakan adalah studi literatur sistematis berdasarkan protokol PRISMA, dengan sumber utama dari database Scopus dan Google Scholar. Hasil seleksi menghasilkan tujuh artikel relevan yang dianalisis menggunakan pendekatan tematik. Studi-studi tersebut menunjukkan bahwa DNA barcoding dengan gen penanda seperti rbcL, COI, cox1, dan cox2-3 spacer efektif mengidentifikasi K. alvarezii secara akurat (≥99% kesamaan sekuens dengan referensi GenBank), serta mengungkap keberadaan variasi genetik lokal dalam bentuk haplotipe. Beberapa studi juga menggabungkan pendekatan molekuler dengan analisis kimiawi (FTIR) untuk memperkuat keabsahan identifikasi dan mendukung sistem ketertelusuran (traceability). Temuan ini menegaskan bahwa DNA barcoding merupakan fondasi penting dalam strategi konservasi berbasis bukti dan pembangunan bank genetik nasional, guna menunjang pemuliaan bibit unggul dan pelestarian hayati laut secara berkelanjutan.

Abstract

Waste management in Indonesia is still dominated by open burning and landfilling, causing environmental and health problems. In Jetak Village, Getasan District, Semarang Regency, located in the rural highlands of Mount Merbabu, organic waste dominates household waste streams. Although a TPS3R facility has existed since 2022, its operation remains suboptimal due to low community participation and odor issues. This condition highlights the urgent need for innovative, community-based solutions. Black Soldier Fly (BSF, Hermetia illucens) farming offers an effective alternative, capable of reducing organic waste by 50–60% while producing valuable by-products. This community service program targeted village officials, farmer groups, women’s groups, youth, and TPS3R managers. Activities included awareness sessions on circular economy, training in BSF cultivation, field demonstrations using appropriate technology, digital marketing workshops, and operational assistance at TPS3R. Impact was assessed through participant questionnaires and scenario modeling of household-level BSF adoption based on waste generation data from SIPSN (2025) and BPS (2025). Twenty-five participants engaged actively throughout the program. Evaluation showed 100% agreement that BSF is effective for organic waste management, with 89% willing to adopt it at home. Products such as fresh and dried larvae and frass were successfully introduced, with estimated market value up to IDR 52,000/kg. Scenario analysis indicated potential reductions of 89–268 tons of organic waste annually and mitigation of 2.6–7.8 tons CO₂e, depending on adoption rates. The program proved that BSF technology is feasible, socially acceptable with proper education, and economically promising. It strengthened local waste management capacity, supported income generation, and contributed to circular economy transition while aligning with SDG 12 and Indonesia’s low-carbon development goals.

Abstract

Tepung tempe dapat dijadikan sebagai substitusi makanan atau suplemen antara lain minuman sari tempe. Namun, minuman sari tempe saat ini belum banyak diproduksi. Padahal minuman sari tempe memiliki efek fungsional untuk menjaga kesehatan tubuh. Perlu adanya pengujian terhadap kualitas minuman sari tempe meliputi nilai aktivitas antioksidan, kadar protein, dan tingkat kesukaan terhadap produk minuman sari tempe berbahan dasar tepung tempe original dan tepung tempe kelor dengan konsentrasi yang berbeda yaitu 1%, 2%, dan 3%. Tujuan penelitian ini adalah mengetahui nilai aktivitas antioksidan dan kadar protein minuman sari tempe berbahan dasar tepung tempe original dan tepung tempe kelor dengan konsentrasi 1%, 2%, dan 3%, dan mengetahui minuman sari tempe yang paling disukai konsumen. Penelitian ini merupakan penelitian eksperimen Rancangan Acak Lengkap (RAL) dengan enam perlakuan berupa minuman sari tempe berbahan dasar tepung tempe original konsentrasi 1%, minuman sari tempe berbahan dasar tepung tempe original konsentrasi 2%, minuman sari tempe berbahan dasar tepung tempe original konsentrasi 3%, minuman sari tempe berbahan dasar tepung tempe kelor konsentrasi 1%, minuman sari tempe berbahan dasar tepung tempe kelor konsentrasi 2%, dan minuman sari tempe berbahan dasar tepung tempe kelor konsentrasi 3%. Masing-masing perlakuan dilakukan dua kali ulangan. Pengujian aktivitas antioksidan menggunakan metode DPPH, pengujian kadar protein menggunakan metode Kjeldhal, dan pengujian tingkat kesukaan menggunakan metode hedonik. Data kualitatif dianalisis secara deskriptif, sedangkan data kuantitatif dianalisis menggunakan uji Anova dua arah dan uji lanjut Duncan. Hasil penelitian menunjukkan minuman sari tempe yang mempunyai nilai aktivitas antioksidan sangat kuat adalah minuman sari tempe berbahan dasar tepung tempe kelor dengan konsentrasi 1% yang ditunjukkan dengan nilai (IC₅₀ < 50), minuman sari tempe dengan kadar protein tertinggi ada pada minuman sari tempe berbahan dasar tepung tempe kelor dengan konsentrasi 3%, dan minuman sari tempe yang paling disukai oleh konsumen adalah minuman sari tempe berbahan dasar tepung tempe original dengan konsentrasi 1%.

Abstract

Rambutan (Nephelium lappaceum L.) is one of the many tropical plants found in Indonesia. The leaves of the rambutan plant are known to be used as medicine, so they have the potential to be a source of endophytic bacteria that can produce beneficial compound. This study aims to isolate, identify of endophytic bacteria from rambutan leaves, and test their activity as an antibacterial with two different secondary metabolite extraction methods, namely maceration with 70% methanol and without maceration. This study used experimental research with a Completely Ramdomized Design (CRD). Isolation of endophytic bacteria from rambutan leaves produced two isolates (D1 and D2). The results of identification through macroscopic, microscopic, and biochemical test showed that two pure isolates had similarities to genus Bacillus sp. Antibacterial activity testing was carried out using well diffusion method. Based on the results of antibacterial test showed that extraction method with 70% methanol produced an extract that was more effective as an antibacterial with average diameter of clear zone formed for Escherichia coli bacteria were 13.55 mm (Isolate D1) and 12.13 mm (Isolate D2) and that of Bacillus subtilis bacteria were 16.27 mm (Isolate D1) and 19.69 mm (Isolate D2). This finding contributes to the fact that the type of secondary metabolite extraction method of rambutan leaf endophytic bacteria influences the sensivity as an antibacterial.

Abstract

Liberica coffee is one of the leading commodities in several regions so it has quite high economic potential. Coffee beans can be stored for quite a long time, more than one year, even more than 4 years before going through the roasting process. However, during long storage periods coffee beans can be attacked by microorganisms such as mold (ochratoxigenic and non-ochratoxigenic). This research aims to determine the effect of storage time on the quality of Liberica coffee beans, both microbiologically and chemically. The benefit of this research is to provide information on the quality of Liberica coffee beans. This research is a laboratory experimental research. The long storage time for coffee beans was 36 and 39 weeks. The results of the research showed that the yeast mold rate (AKK) on Liberica coffee beans stored for 39 weeks was greater than those stored for 36 weeks. The increase in AKK during storage is thought to be due to the presence of insects, temperature and humidity factors. A total of three mold isolates were found at week 36 and there were six mold isolates at week 39. The percentage of fungal isolate attacks at both weeks 36 and 39 was dominated by fungi of the Mucorales order (Mucor sp. and Absidia sp.). In the 36th week the attack of Mucor sp. amounted to 36.67% while in the 39th week the Mucor sp attack. 20%. and Absidia sp. 23.33%. Ochratoxin A (OTA) levels from week 36 to week 39 did not increase but instead decreased. This occurs due to OTA degradation by ochratoxin-degrading molds which are thought to be Aspergillus sp. during storage.

Abstract

This study aims to determine the impact of utilization Computer Assisted Instruction (CAI) based learning media in improving digital literacy skills and student learning outcomes. In this study, data collection used observation sheets to improve students' digital literacy skills through observations of experimental and control groupes. It is known that the score of students' digital literacy skills is 81% in the experimental group, while the control group is 67%, meaning that the criteria for students' digital literacy skills in the experimental group are classified as better.  In collecting data to determine the effect of media on student learning outcomes, the Pretest Posttest control group design method is used with a pretest, treatment, and posttest pattern. In practice, 2 groupes are needed as experimental groupes with treatment and control groupes without treatment, and observations are made of students' digital literacy skills during learning activities. From the results of the analysis of student pretests and posttests, it is known that there is an increase in scores in the experimental group as indicated by the average N-Gain value of 57.54%, thus the use of CAI-based learning media is interpreted in the category quite effective in improving student learning outcomes. Then based on the results of the one-way anova analysis for the relationship between the improvement of learning outcomes with digital literacy skills in experimental and control groupes, it is known that the significance value is 0.00 <0.05, as well as the results of the post-hoc test with the Turkey method which shows the significance value between digital literacy and learning outcomes is less than 0.05, which means that the use of CAI-based learning media has an influence on digital literacy as well as student learning outcomes. So it can be concluded that the use of CAI-based media has an impact in improving digital literacy skills and student learning outcomes.

Abstract

Stunting adalah suatu kondisi dimana anak mengalami gangguan pertumbuhan, sehingga tinggi badan anak tidak sesuai dengan usianya, sebagai akibat dari masalah gizi kronis yaitu kekurangan asupan gizi dalam waktu yang lama. Stunting berdampak negatif terhadap kecerdasan anak dan meningkatkan risiko penyakit tidak menular pada anak, seperti diabetes mellitus, kanker, dan penyakit jantung koroner pada saat dewasa nantinya. Banyak faktor yang mempengaruhi kejadian stunting pada anak di perkotaan maupun di pedesaan. Penelitian ini bertujuan untuk menganalisis pengaruh pola asuh, pelayanan kesehatan, dan kesehatan lingkungan terhadap stunting di Kelurahan Bandarharjo Semarang. Desain penelitian adalah cross sectional dengan teknik pengumpulan data menggunakan kuesioner. Penelitian dilakukan di Kelurahan Bandarharjo, Kota Semarang pada bulan Juli-Agustus tahun 2023 dengan sampel balita sebanyak 95 dengan usia 11-59 bulan. Metode pengambilan sampel dalam penelitian ini adalah pengampilan sampel acak bertingkat. Analisis data menggunakan software SPSS 16.0 dengan uji univariat, uji bivariat, dan uji multivariat menggunakan uji regresi logistik. Hasil penelitian menunjukkan bahwa pola asuh berpengaruh sebesar 14,8%, pelayanan kesehatan 2,2%, dan kesehatan lingkungan 8,4% terhadap stunting. Disimpulkan bahwa pola asuh, pelayanan kesehatan, dan kesehatan lingkungan berpengaruh terhadap kejadian stunting di Kelurahan Bandarharjo Semarang.
